AbstractCaptive breeding is often a last resort management option in the conservation of endangered species which can in turn lead to increased risk of inbreeding depression and loss of genetic diversity. Thus, recording breeding events via studbook for the purpose of estimating relatedness, and facilitating mating pair selection to minimize inbreeding, is common practice. However, as founder relatedness is often unknown, loss of genetic variation and inbreeding cannot be entirely avoided. Molecular genotyping is slowly being adopted in captive breeding programs, however achieving sufficient resolution can be challenging in small, low diversity, populations. Here, we evaluate the success of the Vancouver Island marmot (Marmota vancouverensis; VIM; among the worlds most endangered mammals) captive breeding program in preventing inbreeding and maintaining genetic diversity. We explored the use of high-throughput amplicon sequencing of microsatellite regions to assay greater genetic variation in both captive and wild populations than traditional length-based fragment analysis. Contrary to other studies, this method did not considerably increase diversity estimates, suggesting: (1) that the technique does not universally improve resolution, and (2) VIM have exceedingly low diversity. Studbook estimates of pairwise relatedness and inbreeding in the current population were weakly, but positively, correlated to molecular estimates. Thus, current studbooks are moderately effective at predicting genetic similarity when founder relatedness is known. Finally, we found that captive and wild populations did not differ in allelic frequencies, and conservation efforts to maintain diversity have been successful with no significant decrease in diversity over the last three generations.

Sambar deer was up-listed from Least Concern to Vulnerable by the IUCN Red list in 2015. The local government has initiated Ex-situ conservation efforts to boost sambar deer numbers in captivity and reintroduce them into the wild. The reproductive success of sambar deer and their welfare management practices in captivity are important components for effective captive breeding programs. However, there has been a lack of study on sambar deer in recent years, especially about their behavior in captivity. This study aimed to identify environmental factors that may influence the behavior of the captive sambar. Three captive sites were selected and observed for an average of 40 days at each site (minimum 37 days to maximum 43 days, 6 hours/day). A Generalized Linear Model was used to determine the correlation between social behavior and extrinsic parameters. ‘Captive sites’ showed the strongest correlation in behavioral variability environmental settings, such as the size of the enclosure, could force the deer to spend more time in a herd, which increases the frequency in grooming, which was recorded to be highest in Zoo Negara compared to other captive sites. Time of day also significantly influenced certain behavior skewed towards morning slots. It could be due to an adaptive behavior to the feeding time in the captive sites being often in the morning, which caused the deer to rest towards the afternoon. A suggestion would be to create a more erratic feeding schedule to ensure that the deer adapt to behavior variations. An extensive study needs to be done on sambar deer to pinpoint the specifics and better understand these possible influential factors in their behavior.

AbstractFemales of many insect species are unreceptive to remating for a period following their first mating. This inhibitory effect may be mediated by either the female or her first mate, or both, and often reflects the complex interplay of reproductive strategies between the sexes. Natural variation in remating inhibition and how this phenotype responds to captive breeding are largely unexplored in insects, including many pest species. We investigated genetic variation in remating propensity in the Queensland fruit fly, Bactrocera tryoni, using strains differing in source locality and degree of domestication. We found up to threefold inherited variation between strains from different localities in the level of intra-strain remating inhibition. The level of inhibition also declined significantly during domestication, which implied the existence of genetic variation for this trait within the starting populations as well. Inter-strain mating and remating trials showed that the strain differences were mainly due to the genotypes of the female and, to a lesser extent, the second male, with little effect of the initial male genotype. Implications for our understanding of fruit fly reproductive biology and population genetics and the design of Sterile Insect Technique pest management programs are discussed.

Abstract Developing a captive breeding system for the widely hunted Caribbean species of opossum Didelphis marsupialis insularis can greatly aide in the management and conservation of this species in the neo-tropics. Although this species possesses ideal traits for captive breeding in this region (tolerance to high heat and humidity, high reproductive rate, and resistance to disease), challenges due to its aggressive behaviour and limited information on its breeding behaviour have prevented a system from being developed for this species. The present study describes a breeding system, and the reproductive behaviour of this species under captive conditions. Six (1 male; 5 females) adult opossums were maintained and managed for breeding over a ten (10) month period. Pouch litter sizes averaged 5±2.5 with a range of 2 to 8. Gestation length was found to be 13.25±0.96 days and 4 litters (n=23) were successfully weaned at 11-13 weeks. It was found that the male D. m. insularis exhibits behaviours of interest that can serve as indicators for receptivity of the female, and overall, that this species can be successfully reared and bred under captive conditions in the neo-tropics.

Abstract The Punjab urial (Ovis vignei punjabiensis) is a globally threatened wild sheep species. In Pakistan the species is legally protected and bred in captivity to release into natural habitats to reinforce wild populations. Vigilance and feeding are critical to survival, though the degree to which captive-bred animals exhibit these behaviours post-release has rarely been investigated. We monitored the daily behaviours of five adult urial after release and classifying behaviours from 0600 hrs to 1800 hrs, replicating each one-hour period twice and recording four observations of each animal per hour. At the herd level, vigilance and feeding behaviours were exhibited equally. Males and females invested equal time in feeding, males were more vigilant (p = 0.001) and more aggressive (p = 0.010), and females were more restful (p = 0.019) or engaged in grooming (p = 0.044) or locomotion (p = 0.002). Females spent more time feeding than being vigilant (p = 0.002). Males maintained high levels of vigilance throughout the day. Patterns of resting 1300 hrs to 1500 hrs and feeding in early morning and late afternoon were common for both sexes. Behaviours classified as ‘other’ were exhibited equally between sexes. Our results reveal positive indications of captive-bred urial balancing threat detection and energy acquisition post-release and exhibiting natural behaviours and activity patterns. We encourage assessment of survivorship to evaluate long-term effectiveness of captive breeding and release as a candidate restoration programme.

The Nguru spiny pygmy chameleon (Rhampholeon acuminatus) is endemic to the Nguru mountains in Tanzania. It is assessed as Critically Endangered and is collected from the wild for the pet trade. An ex-situ population of this species was recently established at The Wild Planet Trust, Paignton Zoo, in the hope of learning more about the husbandry and biology of this species. We report on the captive husbandry of adults and the successful breeding, hatching and rearing of juveniles. Females carried four eggs but laid them in pairs. When eggs were incubated at 19.2-22.8 °C, hatchlings emerged roughly 180 days after laying. The hatchlings had a total length of about 30 mm and weighed 0.2-0.3 g. To our knowledge, this is the first published account of captive breeding for this species. The husbandry methods described could be used to establish populations of this and other Rhampholeon species in captivity, which in turn would reduce the demand for wild caught (Rhampholeon), as well as inform future conservation breeding programmes for this species.

Troides helena and Pachliopta adamas utilize the same food plant species: Aristolochia acuminata. For the purpose of captive breeding and conservation, it is desirable to find out whether they can co-exist in captivity. Captive breeding research was conducted on the butterfly species within the period of October 2016 to September 2019. In total, 1,361 individuals were observed. Data on adult emergence of the species is presented to show population trends. Both species co-existed poorly at the facility when food plants were limited.  It took 45.9 days for T. helena helena and 32.6 days for Pc. adamas adamas from egg to imago stage. Habitat enrichment can encourage the species to come and establish the population.

This paper discusses paradoxes in our relationship to and treatment of birds in captive and conservation contexts. The paper identifies modern and new challenges that arise from declining bird numbers worldwide. Such challenges have partly changed zoos into providers of insurance populations specifically for species at risk of extinction. They have also accelerated fieldwork projects, but by using advanced technological tools and in increasing numbers, contradictorily, they may cause serious harm to the very birds studied for conservation purposes. In practice, very few avian species have any notable protection or guarantee of good treatment. The paper first deals with shortcomings of identifying problematic avian behavior in captive birds. It then brings together specific cases of field studies and captive breeding for conservation in which major welfare deficits are identified. Indeed, the paper argues that avian welfare is now an urgent task. This is not just because of declining bird numbers but because of investment in new technologies in field studies that may have introduced additional stressors and put at risk bird survival. While the paper documents a substantial number of peer-reviewed papers criticizing practices counter to modern welfare standards, they have by and large not led to changes in some practices. Some solutions are suggested that could be readily implemented and, to my knowledge, have never been considered under a welfare model before.
