Gotu kola (Centella asiatica L.) is a medicinal crop, needed by local industry of pharmacy. Gotu kola in Indonesia is not enough to supply in market requisites. Tissue culture method was used to multiply gotu kola with a combination of medium from liquid organic fertilizer and pearl grass extract. The purposes of the research were to determine the effect of liquid organic fertilizer and pearl grass extract on the shoot growth of gotu kola in vitro, and to obtain the optimal concentration of liquid organic fertilizer and pearl grass extract combination on the shoot formation of gotu kola in vitro. The research was conducted in November 2010 to June 2011 in Plant Physiology and Biotechnology Laboratory, Faculty of Agriculture, Sebelas Maret University, Surakarta. The experimental design used was a Completely Randomized Design (CRD) with two treatment factors and three replications. The first factor was liquid organic fertilizer concentrations, i.e. 0 mL/L, 2 mL/L, 4 mL/L and 8 mL/L. The second factor was pearl grass extract, i.e. 0 mL/L, 3 mL/L, 6 mL/L and 12 mL/L. Variables observed were the percentage of shoot formation, the time of shoot formation, the number of shoot, the length of shoot, the percentage of leaf formation, the time of leaf formation and the number of leaf. The result of the research showed that not all of treatments provided shoot formation. The combination of liquid organic fertilizer concentrations 0 mL/L and pearl grass extract 12 mL/L showed the best results, in which the average of length of shoot was 2.16 cm, the number of shoot was 8 and the number of leaf was 8.

ABSTRAK<br />Pegagan (Centella asiatica L.) adalah tanaman obat yang<br />mengandung zat asiaticotik sebagai obat alzaimer dan penghalus kulit.<br />Tanaman ini telah diperbanyak sejak tahun 2000. Penelitian dilakukan di<br />Laboratorium Kultur Jaringan dan Rumah Kaca Kelompok Peneliti<br />Plasma Nutfah dan Pemuliaan, Balai Penelitian Tanaman Obat dan<br />Aromatik, Bogor dari bulan Januari 2000 sampai dengan Juni 2005.<br />Penelitian ini bertujuan untuk melihat daya multiplikasi tunas setiap<br />periode subkultur dimulai dari tahun kedua sampai periode lima tahun.<br />Media yang digunakan adalah MS + BA 0,1 mg/l. Pengamatan dilakukan<br />pada jumlah tunas, jumlah daun dan visual tunas pada umur 2, 3, 4 dan 5<br />tahun pada dua periode subkultur setiap tahunnya. Penelitian disusun<br />dengan rancangan acak lengkap, masing-masing terdiri atas 10 botol yang<br />merupakan ulangan dan setiap botol terdiri atas 1 eksplan. Untuk re-media<br />terhadap tanaman yang terlihat berubah digunakan media MS + BA (0;<br />0,1; 0,2; 0,3) mg/l. Selanjutnya untuk perakaran dilakukan pada media MS<br />+ IAA (0,1 dan 0,2); MS + NAA (0,1 dan 0,2) mg/l serta MS + IBA (0,1<br />dan 0,2) mg/l. Plantlet utuh yang terbentuk selanjutnya diaklimatisasi pada<br />media tanah + pupuk kandang dan tanah + sekam dengan perbandingan 1 :<br />1. Hasil penelitian menunjukkan bahwa daya multiplikasi tunas optimum<br />terjadi pada tahun ketiga, dan memasuki tahun keempat dan kelima<br />menurun yang diiringi dengan perubahan eksplan yang terlihat pada<br />tangkai daun yang terbentuk. Akar terpanjang dan terbanyak yang<br />terbentuk didapat pada media IAA 0,2 mg/l dengan penampilan yang<br />kurus dan rapuh. Keberhasilan aklimatisasi sangat rendah, tetapi plantlet<br />mampu beregenerasi dengan baik dan terlihat tumbuh normal. Dari hasil<br />perbanyakan terlihat bahwa jumlah anakan, jumlah daun, panjang stolon<br />dan jumlah bunga lebih tinggi dibandingkan yang tumbuh pada media<br />sekam, berturut-turut : 6,77; 7,30; 46,50 cm dan 8,31. Sementara pada<br />media sekam komponen yang dominan adalah panjang tangkai daun yakni<br />9,75 cm.<br />Kata kunci : Pegagan, Centella asiatica L., multiplikasi, tunas, aklima-<br />tisasi, penyimpanan, Jawa Barat<br />ABSRACT<br />Shoot multiplication and acclimatization of gotuloca<br />(Centella asiatica L.) five years after conservation by in<br />vitro culture<br />Gotuloca (Centella asiatica L.) is a medicinal crop containing<br />asiaticotic as alzaimer and skin revitalizer. This crop has been<br />multiplicated in vitro since 2000. This research was carried out in the<br />laboratory and glasshouse of Breeding and Germplasm Group in the<br />Indonesian Medicinal and Aromatic Crops Research Institute (IMACRI)<br />from January 2000 to June 2005. The objective of the research was to find<br />out the viability of shoots multiplication after two year to five year period,<br />with media MS + BA 0.1 mg/l. The parameters observed were the number<br />of shoots, the number of leaves at the age 2, 3, 4 and 5 years from two<br />subculture periods every year. The treatments were arranged in a<br />completely randomized design, each replication consisted of 10 bottles and<br />each bottle consisted of 1 explant. After subculture the ex-plant were re-<br />media in medium MS + BA (0; 0,1; 0,2; 0,3) mg/l. The rooting media<br />before glasshouse were : MS + IAA ( 0,1 and 0,2); MS + NAA ( 0,1 and<br />0,2) mg/l; and MS + IBA ( 0,1 and 0,2) mg/l. The plantlets formed were<br />acclimatized using soil + cattle manure and soil + rice husk with<br />comparison 1:1. Research result indicated that the optimum viability<br />multiplication was achieved in the third year, and it decreased after the<br />fourth and fifth years with change in explant forming the petiole. The<br />longest and plantlet roots were formed through media IAA 0.2 mg/l with<br />brittle and thin appearance, but the plantlets were able to regenerate better<br />and grow normal. The acclimatization was not very successful but the<br />plantlets could regenerate and grew normally. The multiplication showed<br />that the number of stumps, leaves, stolons and flowers were : 6,77; 7,30;<br />46,50 cm and 8,31 respectively. The Effect of Several Types of Cytokinin on Shoot Multiplication of Vanda douglas Orchid In VitroThe study was aimed to determine the response of Vanda douglas orchid on shoot-multiplication media to different cytokinin concentrations in vitro. A completely randomized design experiment was employed with one factor cytokinin, in which the cytokinins used were TDZ (thidiazuron), BAP (6-Benzylaminopurine) and kinetin at the concentrations of 0, 0.5, 1, and 1.5 mg/L. The results showed that kinetin 0.5 mg/L was the best concentration for shoot formation, occuring on average at 14.88 days after planting; while TDZ 0.5 mg/L was the best concentration for increasing the height of the plant, being on average 0.53 cm. TDZ at 0.5 mg/L concentration also had positive effect on shoot and leaf formation, which resulted in the highest number of shoots and leaves. The average number of shoots was 8.00 buds, and the average number of leaves was 12.25 sheets. Keywords: Vanda douglas, thidiazuron, BAP, kinetin, shoots multiplication ABSTRAKPenelitian ini bertujuan untuk mengetahui respon anggrek Vanda douglas terhadap media perbanyakan tunas pada beberapa konsentrasi jenis sitokinin secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan satu faktor, yaitu sitokinin. Sitokinin yang digunakan adalah TDZ (thidiazuron), BAP (6-Benzylaminopurine) dan kinetin, dengan konsentrasi 0, 0,5, 1, dan 1,5 mg/L. Hasil penelitian menunjukkan bahwa pemberian kinetin 0,5 mg/L menunjukkan hasil terbaik pada peubah waktu pembentukan tunas, dengan rata-rata 14,88 hari setelah tanam. Sedangkan konsentrasi TDZ 0,5 mg/L merupakan jenis sitokinin dan konsentrasi terbaik terhadap pertambahan tinggi tanaman, dengan rata-rata 0,53 cm. TDZ dengan konsentrasi 0,5 mg/L juga berpengaruh positif terhadap pertumbuhan tunas dan daun, dengan menghasilkan jumlah tunas tertinggi dan jumlah daun terbanyak. Kiwi fruit takes about 25 weeks from flower bloom until it reaches physiological maturity, so the time required to produce kiwi seeds from seeds in large quantities and uniform is very long. Tissue culture is one method that can be used to obtain a lot of kiwi seeds and uniforms with large quantities in a faster time. The purpose of this study was to examine various types of media compaction materials for the growth of kiwi shoots in vitro. This study was prepared based on Completely Randomized Design (RAL) with 5 treatments and repeated 4 times so that there were 20 experimental units, each experiment using 2 explants so that there are 40 eksplan. The treatments were: MA 1: Agar Swallow Globe 8 g / l, MA 2: Agar Swallow Globe 4 g / l + Agar Nutrijell 4 g / l, MA 3: Agar Swallow Globe 4 g / l + Agar Nutrijell 5 g / l, MA 4: Phytagel 2.2 g / l, MA 5: Agar Nutrijell 11 g / l. Observation variables are When shoots appear, Number of shoots, number of leaves, Number of Roots, number of root hair. The results showed Swallow Globe 4 g / l + Agar Nutrijell 4 g / l treatment gave the highest average number of shoots, the highest number of leaves and roots, this proved that the combination of Swallow Globe and Nutrijell agar gave a good condition for shoot growth kiwi plant.t.

Curcuma sp has been widely investigated for its anti-cancer properties. Conventionally, vegetative propagation needs a long time to produce a large number of planting materials, so that it is necessary to find an alternative approach through in vitro propagation.  The effect of BAP on the in vitro shoot formation of ‘temu putih’ and ‘temu putih’ has been investigated in this study. The experiment was a 4x2 factorial with 5 replications arranged in a completely randomized design.  The first factor was the concentration of BAP i.e. 0, 1.5, 3, and 4.5 ppm.  The second factor was the curcuma species consisting of ‘temu putih’ (Curcuma zedoaria Roch.) and ‘temu mangga’ (Curcuma mangga Val.).  Each experimental unit consisted of 2 in vitro bottles, each of which planted with 1 explant bud.  Analysis of variance was conducted on percentage of live explants, shoot height, number of roots, root length, wet weight, percentage of explants that sprouted, percentage of rooted explants and shoot color.  Mean comparison was performed by the Least Significant Difference (LSD).  The results showed that there was no interaction between BAP concentration and genotype on any variable observed.  The shoot growth of ‘temu putih’ was significantly higher than ‘temu mangga’ in vitro.  The best concentration of BAP for the growth of ‘temu putih’ and ‘temu mangga’ shoots was 1.5 ppm.

Biofarmasi 14: 47-55. Centella (Centella asiatica L) is one plant utilized as the traditional medicine because it has many benefits, one of which is to fulfill the brain requirement. The aims of this research was to know of media type, mahkota dewa and meniran extract, concentration of meniran, concentration of mahkota dewa, and concertation of liquid organic fertilizer what as well as shoot growth of Centella using tissue culture and filtering (2k design) that is expected it can be screened of the basic media types, organic material types (mahkota dewa and meniran extract), meniran concentration, mahkota dewa concentration and liquid organic fertilizer concentration. This research was taken place in Plant Physiology and Biotechnology Laboratory of Agriculture Faculty of Sebelas Maret University, from November 2010 to June 2011. This research used 2k design with RAL environment. Factor 1: Media type ( ½ MS and MS); Factor 2: Concentration of liquid organic fertilizer (2 mL/L and 20 mL/L); Factor 3: Concentration of meniran 5 mL/L and 25 mL/L); and Factor 4: Concentration of mahkota dewa (5 mL/L and 25 mL/L). Thus, there is 24 = 16 treatment combinations, each of which is repeated three times. The variables observed are shoot emerging duration, shoot number, shoot height, and leaves number. The data were analyzed descriptively. The result of research showed that basic media MS, that basic media MS, POC 20 mL/L, and meniran extract 5 mL/L provided the best explants growth of Centella through in vitro.

This research aims to obtain the combination of BAP and NAA concentration which give the best effect on explant growth of Vanda orchid in-vitro. The research was conducted at the biotechnology laboratory Faculty of Agriculture Riau Islamic University Pekanbaru from August to December 2013. The completely randomized design with two factorials (BAP and NAA) was used. The observed parameters included percentage of shoot growing, shoot age, shoot height, shoot number and abnormal embryo somatic. Data were analysed using statistics and BNJ Test at the 5% significance level. The result showed that,interactionally,BAP and NAA applications had a significant effect on height plant with the best concentration without BAP and 1.0 ppm NAA.  Individually, the application of BAP effected significantly on percentage of shoot growing, shoot growth, shoot height and shoot number with the best concentration without BAP and 0.1 BAP. Furthermore, the application of NAA influenced significantly on percentage of shoot growing, shoot age, shoot height and shoot number with the best concentration of 1.0 ppm NNA

Rauwolfia serpentine [L] Kurz ex Benth is a valuable plant yielding alkaloid of reserpine. The aim of this study was to find the most effective way of reserpine production. Growth regulators {naphthalene acetic acid [NAA] and Kinetin}were applied to promote and to regulate the growth of explant of R. serpentine cultured in vitro. The study was conducted at the laboratory of Bandung Institute of Technology [ITB]. Nine formulations of NAA and Kinetin were the treatments that were repeated twice in completely randomized design. Weight of callus was measured at one, two, three and four weeks after induction of explant. Reserpine analysis was performed by High Performance Liquid Chromatography [HPLC]. The result showed that the best callus induction was the treatment of 2.5 ppm NAA + 2.5 ppm Kinetin, and the highest content of reserpine was in root organ [0.0021 g / L], and in callus [0.0021 g / L] at the age of 4 weeks after induction. There was revealed that in vitro culture method was more productive in producing reserpine compound than the conventional plantation of R serpentine.  Production of reserpine by callus culture was more effective and may be the basic for recommended further effort.

<p>The demand for rice as an important food crop in Indonesia is still constrained by the attack of blast disease caused by <em>Pyricularia grisea</em>. Controlling using synthetic chemical pesticides has a negative impact on the environment so that the use of biological agents is an alternative option. This study aims to determine the effectiveness of several biological agents against the pathogen <em>P. grisea</em> that causes blast disease in rice plants in vitro. The study used a completely randomized design (CRD) with 6 levels of treatment and was repeated 3 times using the dooble culture method. The results of the study showed that the inhibition of <em>P. grisea</em> was the highest in the treatment of <em>P. grisea</em> × <em>Trichoderma</em> sp. and <em>P. grisea</em> × <em>Gliocladium</em> sp. namely 67.04% and 51.85% compared to other treatments. The <em>P. polymyxa</em> and <em>P. fluorescence</em> treatments showed low inhibition, namely 23.70% and 28.89%. Biological agents <em>Trichoderma</em> sp. and <em>Gliocladium</em> sp. able to inhibit the growth of the fungus <em>P. grisea</em>. Each biological agent has a different percentage of inhibition in that caused by the inhibitory mechanism of the biological agent.</p>

Mangosteen is considered as fruit of many benefits with great popularity, rendering it valuable as an export commodity. Unfortunately, it has a relatively slow growth  and long early fruiting time. Generative propagation of mangosteen by seed result in low-yielding productionof qualified seedswith recalcitrant nature which means it cannot be stored for a long time. Whereas the result of vegetative propagation using grafting, branch cuttings and budding has difficulty rooting out. Tissue culture method requires growth regulators to control organogenesis and morphogenesis so it can accelerate the growth of the mangosteen explants. This study was arrangedintwo- factor completely randomized design (CRD). The observed variables werethe strength of the medium (½ and 1 MS) and GA3 concentrations (0 ppm; 0.25 ppm; 0.5 ppm; 0.75 ppm and 1 ppm). The variatonson GA3 concentration (0 ppm; 0.25 ppm; 0.5 ppm; 0.75 ppm; 1 ppm) on  different strength MS medium (½ MS and 1 MS) did not significantly affect the growth of radicle and sooth in the  in vitro seeds germination of Mangosteen (Garcinia mangostana).

This study was aimed at investigating the effect of NAA and BAP on the development of basal slip slices in tissue culture of pineapple cv. Tangkit. The experiment was conducted at the Plant Biotechnology Laboratory Agricultural Faculty University of Jambi from July through to October 2016. Six level of NAA concentrations (0, 1, 2, 3 , 4 and 5 mgL-1) were tested in combination with six levels of BAP concentration (0, 1, 2, 3 , 4 and 5 mgL-1). A Completely Randomized Design with 5 replicates was employed in this trial. Data on the percentage of explants forming shoots, time required for shoot formation, number of shoots growing per explants, and callus proliferation were recorded. Results showed that without the involvement of plant growth regulators there had been no growth on cultured explants. However, shoot growth was found on explants cultured on medium supplemented with 1 – 3 mgL-1 NAA or 1 – 5 mgL-1 BAP. The application of NAA higher than 3 mgL-1 was found to suppress shoot growth even in combination with BAP. Shoot formation was found to be fast on medium supplemented with 1 mgL-1 NAA + 4 mgL-1 BAP, but took longer time on medium with or 1 mgL-1 NAA + 5 mgL-1 BAP as well as medium with 1 – 2 mgL-1 NAA only. The same phenomenon was also found on medium with 1 – 2 mgL-1 NAA only. This study also revealed that most of cultured explants regenerated only one shoot, but explants cultured on medium with 1 mgL-1 NAA along with 1 mgL-1 BAP produced 2.33 shoots on the average. The addition of 1 mgL-1 NAA without BAP and 1 mgL-1 BAP without NAA could increase average shoot growth by 1.78 and 1.44, respectively.
