Ganoderma lucidum (Fr.) Krast, commonly known as “Lingzhi” in Chinese, is a medicinal mushroom that is rich in biologically active substances. Polysaccharides and triterpenoids are the two major components responsible for the bioactivity of this fungus. In the present study, the ultrasonic-assisted co-extraction (UACE) of polysaccharides and triterpenoids from G. lucidum was optimized using response surface methodology with a desirability function, with the equal importance for the two components. Following single factor experiments, the optimal conditions were determine as ultrasonic power of 210 W, extraction temperature of 80C, ratio of liquid to solid of 50 mL/g, and 100 min extraction time, using aqueous ethanol (50%, v/v) as the extracting solvent. Under the optimal conditions, the extraction yields of polysaccharides and triterpenoids reached 0.63% and 0.38%, respectively. On the basis of the scavenging capacity of 2,2-diphenyl-1-picrylhydrazyl and evaluation of reducing power, the antioxidant capacities of the polysaccharides obtained by optimal UACE process were higher than those of polysaccharides extracted using traditional hot water extraction, whereas the triterpenoid-rich extracts showed antioxidant activities similar to those obtained using the ethanol maceration method. The present study is the first report on the simultaneous extraction of polysaccharides and triterpenoids from G. lucidum. The developed UACE process could be useful in preparation of a polysaccharide- and triterpenoid-rich ingredient that holds great promise for application in the Ganoderma industry.

The medicinal values of polysaccharides (PS) in Lingzhi have been shown to lie in many anti-cancer effects and good benefits for human health. Lingzhi, which is rich in healthy PS, has been used more and more commonly in Vietnam in recent years. In the present work, ultrasonic-assisted enzymatic extraction (UAEE) was used for extraction of PS from Lingzhi. The experiments were conducted according to a Box-Behnken design (BBD), with four independent variables: solvent temperature, ultrasonic power, pH, and ultrasonic irradiation time. The results showed that the best adequate extraction conditions were extraction time of 144 min, extraction temperature of 55°C, ultrasonic power of 240W, pH 7.9, and ultrasonic irradiation time of 144 min. Under these conditions, the predicted optimal yield was 3.716%. Whereas by following the optimized conditions, the experimental yield of PS was 3.65% ± 0.14, which was in good agreement with that of the prediction. Compared to the hot water extraction (HWE) method, ultrasonic-assisted extraction (UAE) method and enzyme-assisted extraction (EAE) method, the yield of PS obtained by UAEE was favorable. The PS yield obtained by HWE, and EAE were 1.96% and 3.10%, respectively. These results demonstrated that UAEE was an appropriate and effective extraction of polysaccharides from Lingzhi.

Ganoderma lucidum (Curtis) P. Karst., known as “Lingzhi” in China or “Reishi” in Japan, is a well-known medicinal mushroom and traditional Chinese medicine, which has been used for the prevention and treatment of bronchitis, allergies, hepatitis, immunological disorders and cancer. G. lucidum is rarely collected from nature and mostly cultivated on wood logs and sawdust in plastic bags or bottles to meet the demands of international markets. Diverse groups of chemical compounds with pharmacological activities, isolated from the mycelia and fruiting bodies of G. lucidum are triterpenoids, polysaccharides (β-D-glucans), proteins, amino acids, nucleosides, alkaloids, steroids, lactones, lectins, fatty acids, and enzymes. The biologically active compounds as primarily triterpenoids and polysaccharides of G. lucidum have been reported to possess hepatoprotective, antihypertensive, hypocholesterolemic, antihistaminic effects and antioxidant, antitumor, immunomodulatory, and antiangiogenic activities. Several formulations have been developed, patented and used as nutraceuticals, nutriceuticals and pharmaceuticals from G. lucidum’s water or ethanol extracts and rarely purified active compounds. As the result of clinical trials, various products have commercially become available as syrup, injection, tablet, tincture or bolus of powdered medicine and an ingredient or additive in dark chocolate bars and organic fermented medicinal mushroom drink mixes such as green teas, coffees, and hot cacaos. This review has intended to give and discuss recent knowledge on phytochemical and pharmacological compositions, therapeutic and side effects, clinical trials, and commercial products of G. lucidum.

An efficient ultrasonic-assisted extraction of inulin from Jerusalem artichoke was investigated by Response Surface Methodology (RSM). The results showed that the yield of ultrasonic-assisted extraction was 62.07 ± 0.39% over the microwave (40.85 ± 0.28%) and hot water extraction (27.42 ± 0.42%). The Box-Behnken Design (BBD) was used to optimize the effects of three parameters (temperature-X1, ultrasonic power-X2 and time-X3) on inulin yield. Analysis of variance showed that the contributions of X1, X3, X12, X13, X22 were significant. The optimal yield of inulin was 82.93 ± 1.03% at 82 °, 120 W and 18 min.

Calophyllum inophyllum (C. inophyllum) is a plant known as “Penaga Laut” in Malaysia. Its seeds contain valuable oil that serve as a potential alternative sources for vegetable oil. In this study, C. inophyllum seeds oil was extracted using an ultrasonic-assisted extraction (UAE) technique. The optimization of extraction parameters namely different types of solvent, extraction time, ultrasonic power, extraction temperature and liquid to solid (L/S) ratio were performed using one-factor-at-a-time (OFAT) method. The optimum extraction conditions obtained were n-hexane as a solvent, extraction time 20 min, ultrasonic power 210 W, extraction temperature 40°C and L/S ratio 20 ml/g, with an oil yield 55.44 ± 0.53 %.

Oleuropein was extracted fromFrantoioolive leaves using reduced-pressure boiling extraction coupled with ultrasound-assist (URPE). Four important factors, extraction temperature, ultrasonic power, ethanol concentration, and the ratio of solid to liquid, were selected to carry out the response surface methodology (RSM) for seeking optimal conditions on high oleuropein extraction yield at different levels. Box-Behnken design was employed to investigate the effects of the four factors on it. The results showed that the ratio of solid to liquid was the most significant factor of all on oleuropein yield. The optimal operation conditions were obtained as follows: ethanol concentration 75% (v/v), extraction temperature 53°C, ultrasonic power 600 W, and the ratio of solid to liquid 1 : 31. Under these optimal conditions, oleuropein extraction yield was 7.08%, which was close to the predicted value 7.121%. The scanning electron microscope (SEM) images of olive leaves after extraction were provided as well. It was seen that, compared with the untreated leaves, URPE could effectively break cells within the olive leaves.

Red Ganoderma lucidum (G. lucidum) is a popular medicinal herb commonly used in Vietnamese traditional remedies due to its potential value for health. In this study, polysaccharides were extracted from G. lucidum using ultrasound-assisted enzymatic extraction method. The response surface methodology and Box–Behnken design were employed to investigate the effects of pH, extraction temperature, extraction time, and ultrasonic power on the content of polysaccharides. Based on ultraviolet-visible spectroscopy analysis, the highest content of polysaccharides in the extract was 32.08 mg/g under optimum experimental parameters including enzyme concentration of 3%, pH of 5.5, extraction temperature of 45°C, extraction time of 30 min, and ultrasonic power of 480 W. The Fourier-transform infrared spectroscopy was also used to identify the functional groups in the extracts. The molecular weights of polysaccharides were determined by gel permeation chromatography. The obtained extract was then evaluated for anticancer activities by using (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, showing the anticancer activities with the half-maximal inhibitory concentration value of more than 512 μg/mL. This result suggested that UAEE could be considered as an appropriate and effective extraction method for bioactive crude polysaccharides from G. lucidum.

Commercial crude tea saponin as raw material, ultrasonic assisted extraction technology of tea saponin was investigated. Five influence factors of ethanol concentration, extraction temperature, extraction time, solid-liquid ratio, ultrasonic power on the extraction of tea saponin were studied in detail. Orthogonal experiment through the single factor experiment design, the experimental results show that the optimum operating conditions for the ethanol concentration of 80%, solid-liquid ratio 1: 40, extraction temperature 40°C, ultrasonic power of 300W, ultrasonic extraction time 45 minutes. Under the optimum conditions obtained tea saponin extract with ethanol- acetone precipitation method is further purified, tea saponin content increased from 52.1% to 93.6%. The result indicated that the purity of the tea saponin could be effectively improved through the ultrasonic assisted- precipitation combined method.

Ultrasonic-assisted extraction (UAE) method was used to polysaccharides from Radix Astragali (RAP), and the orthogonal test was applied to optimize extraction conditions. Furthermore, the antioxidant activities of RAP were evaluated by DPPH and superoxide anion radical scavenging activities assay. The results indicated that the optimal conditions for RAP extraction as followings: solid/liquid ratios of 1:20, extraction time of 10 min, ultrasonic power of 65 W and extraction temperature of 60°C. Meanwhile, RAP had relatively higher scavenging activities on DPPH and superoxide anion radicals.
