Summary The aim of this study was to investigate the use of the anaesthetic 2-phenoxyethanol on the transmission factors of gyrodactylid and to ascertain how this may affect in the colonisation of new hosts using the Gyrodactylus gasterostei Gläser, 1974 - Gasterosteus aculeatus L. model which is a simple and successful system to examine aspects of transmission of parasites from live and dead fish. Laboratory experiments include determining the maturity (presence of male copulate organ) and reproductive (presence of daughter) status of transmitting worms, in order to consider the factors that influence parasite option to migrate to a new individual of the same host species. This study demonstrates that parasites with a Male Copulate Organ (MCO) present are more likely to abandon the host and attempt a host transfer. The use of the anaesthetic 2-phenoxyethanol does not affect transmission of gyrodactylids which leave the host to colonise a new host. Finally, the use of other anaesthetic although its relative importance with respect to transmission remains uncertain.

In order to study the relationships between mistletoes and their host species, comprehensive collections of both mistletoes and hosts are needed. The effect of sampling effort on the estimation of mistletoe host range was demonstrated in a comparison of an inventory of host mistletoe interactions published by Downey in 1998 and a 2019 inventory presented here, which is based on data from collections in the Australian Virtual Herbarium and information in the literature. New hosts were recorded for 93% of the 90 Australian mistletoes. There were 338 previously known hosts recorded to be parasitised by additional mistletoe species, and 317 new host species that were not previously known as mistletoe hosts (25 being alien species). These were from 78 new host genera and 13 new host families. The total number of host species was 1186 within 327 genera from 92 host families. A total of 63% of all Australian mistletoes parasitise species of either Eucalyptus or Acacia or both these genera. The large rise in host species recorded in less than two decades between inventories suggests that current knowledge of hosts is still incomplete, such that further new hosts will be discovered in future. Some mistletoe species show a strong preference to one host family or genus but due to insufficient collecting it is premature to conclude that any of the three species known from a single host are host specific.

This study evaluates through modeling the possible individual and combined effect of three populational parameters of pathogens (reproduction rate; rate of novelty emergence; and propagule size) on the colonization of new host species – putatively the most fundamental process leading to the emergence of new infectious diseases. The results are analyzed under the theoretical framework of the Stockholm Paradigm using IBM simulations to better understand the evolutionary dynamics of the pathogen population and the possible role of Ecological Fitting. The simulations suggest that all three parameters positively influence the success of colonization of new hosts by a novel parasite population but contrary to the prevailing belief, the rate of novelty emergence (e.g. mutations) is the least important factor. Maximization of all parameters result in a synergetic facilitation of the colonization and emulates the expected scenario for pathogenic microorganisms. The simulations also provide theoretical support for the retention of the capacity of fast-evolving lineages to retro-colonize their previous host species/lineage by ecological fitting. Capacity is, thus, much larger than we can anticipate. Hence, the results support the empirical observations that opportunity of encounter (i.e. the breakdown in mechanisms for ecological isolation) is an fundamental determinant to the emergence of new associations - in special of Emergent Infectious Diseases - and the dynamics of host exploration, as observed in SARS-CoV-2. Insights on the dynamics of Emergent Infectious Diseases derived from the simulations and from the Stockholm Paradigm are discussed.

Twelve species of parasitic helminths, seven trematodes, four nematodes and one acanthocephalan are reported from various hosts. Creptotrema lynchi, a parasite from Bufo marinus in Colombia, is described for the first time in fish and from Brazil, parasitizing two different species. A list of the host species, measurements and figures of most parasites are included with particular reference to the tegument of Bellumcorpus major recovered from a new host. The genus Zonocotyloides Padilha, 1978 is considered a synonym of Zonocotyle and the new combination: Zonocotyle haroltravassosi is proposed to the species Zonocotyloides haroltravassosi Padilha, 1978. The nematodes Cucullanus pinnai and Procamallanus (Spirocamallanus) inopinatus and the trematode Pararhipidocotyle jeffersoni are reported in new hosts. The description of the acanthocephalan Neoechinorhynchus curemais (new locality record) is supplemented. Other parasites recovered include the nematodes Travnema travnema (new locality record), Rondonia rondoni and the digenetic trematodes Cladocystis intestinalis, Pseudosellacotyla lutzi (new locality record), Teratotrema sp. and Zonocotyle bicaecata.

Stator limbatus is a phytophagous beetle native to warm regions of North and Central America, feeding on Fabaceae seeds and one of the most polyphagous species within the subfamily Bruchinae, here reported for the first time in Europe and on new hosts. Adult beetles emerged from Acacia spp. seeds collected in the islands of Corsica (France), and Sardinia (Italy). The wide presence in Sardinia and Corsica supports the hypothesis that this alien species was introduced several years ago. In both islands, S. limbatus emerged from Acacia mearnsii seeds, with infestation rates of up to 74.2 and 90.8% in 2019 and 2020, respectively. This seed beetle also emerged from two previously unreported host species, Acacia saligna and A. pycnantha, showing highest infestation rates of 4.0 and 95.1%, respectively. Both Acacia species are reported as new host associations with S. limbatus. Overall, seed infestation rates recorded in 2019 and 2020 indicate that S. limbatus is well established and that Mediterranean bioclimatic conditions are suitable for its population increase in size. This study lays the foundations for further research on known and potential host species and the spread and distribution of S. limbatus in Europe.

Female threespine sticklebacks (Gasterosteus aculeatus) spend only 9–15 days on the spawning grounds, an intertidal salt marsh at Isle Verte, Quebec, during a 2-month breeding season. Individuals average only one spawning. However, in the laboratory they lay clutches of several hundred eggs every 3–5 days for several months. We designed laboratory experiments to determine (i) whether daily temperature fluctuations similar to those encountered in the marsh affect reproduction (number of clutches, number of eggs per clutch, and size of eggs) and (ii) whether the amplitude of the fluctuations encountered by the fish affects reproduction. We compared the reproduction of females held in fluctuating temperatures with that of females kept at 20 °C. Fish kept under fluctuating conditions produced more eggs per clutch but had longer interspawning intervals than those at 20 °C. Total seasonal egg production and egg size did not differ between the two groups. Fish in fluctuating temperatures survived longer and were in better condition than those at 20 °C. We conclude that the amplitude of the fluctuations is less important than mean temperature in determining reproductive performance. Fluctuating temperatures on the spawning grounds are not responsible for the short residency there.

SUMMARYWhen a pathogen infects a number of different hosts, the process of determining the relative importance of each host species to the persistence of the pathogen is often complex. Removal of a host species is a potential but rarely possible way of discovering the importance of that species to the dynamics of the disease. This study presents the results of a 12-year programme aimed at controlling brucellosis in cattle, sheep and goats and the cascading impacts on brucellosis in a sympatric population of red deer (Cervus elaphus) in the Boumort National Game Reserve (BNGR; NE Spain). From February 1998 to December 2009, local veterinary agencies tested over 36 180 individual blood samples from cattle, 296 482 from sheep and goats and 1047 from red deer in the study area. All seropositive livestock were removed annually. From 2006 to 2009 brucellosis was not detected in cattle and in 2009 only one of 97 red deer tested was found to be positive. The surveillance and removal of positive domestic animals coincided with a significant decrease in the prevalence of brucellosis in red deer. Our results suggest that red deer may not be able to maintain brucellosis in this region independently of cattle, sheep or goats, and that continued efforts to control disease in livestock may lead to the eventual eradication of brucellosis in red deer in the area.

AbstractIn order to infect a new host species, the pathogen must evolve to enhance infection and transmission in the novel environment. Although we often think of evolution as a process of accumulation, it is also a process of loss. Here, we document an example of regressive evolution in the Irish potato famine pathogen (Phytophthora infestans) lineage, providing evidence that a key sequence motif in the effector PexRD54 has degenerated following a host jump. We began by looking at PexRD54 and PexRD54-like sequences from across Phytophthora species. We found that PexRD54 emerged in the common ancestor of Phytophthora clade 1b and 1c species, and further sequence analysis showed that a key functional motif, the C-terminal ATG8-interacting motif (AIM), was also acquired at this point in the lineage. A closer analysis showed that the P. mirabilis PexRD54 (PmPexRD54) AIM appeared unusual, the otherwise-conserved central residue mutated from a glutamate to a lysine. We aimed to determine whether this PmPexRD54 AIM polymorphism represented an adaptation to the Mirabilis jalapa host environment. We began by characterizing the M. jalapa ATG8 family, finding that they have a unique evolutionary history compared to previously characterized ATG8s. Then, using co-immunoprecipitation and isothermal titration calorimetry assays, we showed that both full-length PmPexRD54 and the PmPexRD54 AIM peptide bind very weakly to the M. jalapa ATG8s. Through a combination of binding assays and structural modelling, we showed that the identity of the residue at the position of the PmPexRD54 AIM polymorphism can underpin high-affinity binding to plant ATG8s. Finally, we conclude that the functionality of the PexRD54 AIM was lost in the P. mirabilis lineage, perhaps owing to as-yet-unknown pressure on this effector in the new host environment.Author SummaryPathogens evolve in concert with their hosts. When a pathogen begins to infect a new host species, known as a “host jump,” the pathogen must evolve to enhance infection and transmission. These evolutionary processes can involve both the gain and loss of genes, as well as dynamic changes in protein function. Here, we describe an example of a pathogen protein that lost a key functional domain following a host jump, a salient example of “regressive evolution.” Specifically, we show that an effector protein from the plant pathogen Phytopthora mirabilis, a host-specific lineage closely related to the Irish potato famine pathogen Phytopthora infestans, has a derived amino acid polymorphism that results in a loss of interaction with certain host machinery.

Data on host species and the distribution of the endoparasitic fungusMyrmicinosporidium durumincreased continuously in recent decades. Here, we add the antCardiocondyla elegansas new host species. Colonies of the monogynous species were found infested in the region of Languedoc-Roussillon (South France). Samples from the nest indicate high infection rates. All castes and sexes were infected by the spores. Variations of infection rates between sampling methods and species are discussed.

ABSTRACT The population of wild animals is regulated by many biotic and abiotic factors, and parasites are a biotic factor that affects the dynamic and density of host populations. From 2002 to 2014, 62 wild animals from the biomes Pantanal, Amazon, and "Cerrado" (or Savanna), which died in attendance in the veterinary hospital or have been road-killed, underwent necropsy for parasitological examination. Overall, 36 species of parasites were identified from 24 host species. Among the parasites, the most prevalent order was Oxyurida (29.1%), followed by Strongylida (20.9%), Spirurida (19.4%), Ascaridida (16.2%), Pentastomida (3.2%), Echinostomida (3.2%), Gygantorhynchia (3.2%), Rhabditida (1.6%), Plagiorchiida (1.6%), and Monilimorfida (1.6%), especially nematodes, which have more biotic potential and is more easily adapted to the environment than other classes. The occurrence of endoparasites was observed more frequently in endothermic than ectothermic animals, and herein is reported eleven new host occurrences for endoparasites in wild animals. The study has contributed to the knowledge on the biodiversity of parasites in wild animals from three biomes in central-western Brazil.
