AbstractElectrogenerated chemiluminescence (ECL) involves the generation of species at electrode surfaces that then undergo electron-transfer reactions to form excited states that emit light. The ECL method is becoming an increasingly promising method in the life sciences, environmental analysis, and clinical analysis, owing to its good selectivity, high sensitivity, wide dynamic concentration response range, and potential and spatial controllability. Here, we give a mini-review on the advances of the ECL peptide-based bioassay using peptide as the molecular recognition element. First, a general history of ECL development is presented. Then, the general strategies of the ECL peptide-based bioassay are reviewed. Particular attention is paid to the related progress in the last 5 years. Finally, we conclude with the future challenges and prospects in the development of the ECL peptide-based bioassay.

All management standards have requirements for different aspects of improvements on the personal level, family level, company level, in business and life. What is about national level and country level? Is it possible for today’s generations to learn history of nations and of civilizations? If it is — ok, let’s apply it on actual time and people to have less problems and difficulties — especially if is actual in field of risk management. Majority of people are occupied by today’s problems. They don’t consider past and future challenges. People from each country strive for better quality, better and cleaner environment, higher safety etc. historically and today. But could we remember: How did Genghis Khan conquer many regions and how was he defeated? How did Mayas and Aztecs die out? How were Native Americans in North America drastically reduced in numbers? How did the Roman Imperium vanish? How was the Ottoman Imperium established and how it vanished? How many people were killed in the wars in XX century, etc? In all these catastrophic changes risks were not considered in an adequate way. Requirements of risk management — Principles and guidelines — ISO 31000:2009 are very consultative. They could be used on country level, national level, regional level, continental and intercontinental level.

AbstractCirculating cell-free DNA from blood plasma of cancer patients can be used to non-invasively interrogate somatic tumor alterations. Here we develop MSK-ACCESS (Memorial Sloan Kettering - Analysis of Circulating cfDNA to Examine Somatic Status), an NGS assay for detection of very low frequency somatic alterations in 129 genes. Analytical validation demonstrated 92% sensitivity in de-novo mutation calling down to 0.5% allele frequency and 99% for a priori mutation profiling. To evaluate the performance of MSK-ACCESS, we report results from 681 prospective blood samples that underwent clinical analysis to guide patient management. Somatic alterations are detected in 73% of the samples, 56% of which have clinically actionable alterations. The utilization of matched normal sequencing allows retention of somatic alterations while removing over 10,000 germline and clonal hematopoiesis variants. Our experience illustrates the importance of analyzing matched normal samples when interpreting cfDNA results and highlights the importance of cfDNA as a genomic profiling source for cancer patients.

Since its inception in 2004, the Swiss Industrial Biocatalysis Consortium (SIBC) has brought together scientists from the Pharma, Fine Chemicals, Agrochemicals and Flavor and Fragrance Industries with the goal of promoting biocatalysis inside and outside of Switzerland as well as providing mutual benefits in the form of pre-competitive knowledge sharing. One of the 'founding fathers' of the SIBC was of course Oreste Ghisalba, whom we are honoring here in this special edition. The history of the SIBC as well as current activities and future challenges will be presented.

The present paper provides a brief history of Electronic Data Processing Systems and their supporting user documentation. The problems inherent to paper documentation are highlighted. Techniques employed to improve on-line reject messages in addition to the development of an online user documentation system are discussed. Future challenges facing the human factors specialist involved in development of on-line documentation are outlined.

Introduction: Although the role of inflammation in DVT has been investigated in different studies, there is no definite answer as to whether increased systemic inflammation is the cause or the consequence of DVT. Aim: To follow inflammatory parameters in a cohort of patients with idiopathic DVT. Methods: Out of 49 patients with an acute idiopathic DVT, which were investigated four months after an acute episode (DEVTA 1), 43 patients were included in the follow-up study investigating inflammatory markers and hemostatic markers of endothelial damage five years after an acute DVT (DEVTA 2). A control group consisted of 43 sex and age matched healthy subjects (CONTROLS). Results: The levels of inflammatory markers were significantly higher in DEVTA 2 in comparison to CONTROLS: tumor necrosis factor alpha 2.0 pg/mL (1.1-2.3) vs 1.3 pg/mL (0.8-1.9), p < .001, high sensitivity C-reactive protein 3.2 mg/L (1.5-5.2) vs 1.7 mg/L (0.9-3.0), p = .008, interleukin-6 (IL-6) 2.7 pg/mL (2.0-3.5) vs 2.1 pg/mL (1.5-2.6), p = .025, IL-8 5.0 pg/mL (3.6-7.3) vs 2.4 pg/mL (1.8-2.8), p < .001. IL-10 was significantly decreased (0.9 pg/mL (0.7-1.8) vs 1.8 (1.5-2.2), p < .001. Most of the proinflammatory markers remained elevated in the DEVTA 2 in comparison to DEVTA 1. Markers of endothelial damage were higher in DEVTA 2 in comparison to CONTROLS and higher than in DEVTA 1. Conclusion: Patients with idiopathic DVT have long-term increased inflammatory markers and markers of endothelial damage. These findings favor the hypothesis that inflammation is a cause and not merely a consequence of acute DVT.

: The design of new Drugs, Molecules, Nano-, and Microstructures for targeted applications along the pass of the time within the history of Chemistry and Clinical Analysis showed to be of high impact on Life Sciences. In this context, and mainly for the actual Corona Virus Pandemic status, it should be highlighted the importance of a fast response. Thus, the developments of differents approaches looking for improved and higher sensitive solutions in the Biodetection field accompanied with new treatments of SARS CoV-2 are required. In this way, in this short communication based on the latest Research publications it was afforded and discussed the following topics: i) the importance of non-covalent interactions from antibody-antigen recognition events, ii) the genomic factor, and iii) the use of non-covalent interactions in different Optical detection systems and Lab-On particles. Moreover, it was afforded the theme related with the importance of new treatments in progress, such development of new vaccines; and current state of the pandemic situation. So, it was discussed and stimulated the improvement of innovative Multifunctional Nanophotonic approaches intended for SARS CoV-2 detection accompanied with potential targeted treatments.
