Molecular Characterization of Five Medicinally Important Species of Typhonium (Araceae) through Random Amplified Polymorphic DNA (RAPD)

The interrelationship of five medicinally important species of Typhonium (Araceae) including T. venosum, which was previously placed under the genus Sauromatum, was inferred by analysis of random amplified polymorphic DNA (RAPD). DNA from pooled leaf samples was isolated and RAPD analysis was performed using 20 decamer oligonucleotide primers. Out of a total of 245 bands amplified, 12 were found to be monomorphic while 233 bands were polymorphic including 86 species-specific bands. The genetic similarities were analyzed from the dendrogram constructed by the pooled RAPD data using a similarity index. The dendrogram showed two distinct clades, one containing T. roxburghii, T. trilobatum and T. venosum and the other containing the remainder two species, i.e. T. diversifolium and T. flagelliforme. Both the clusters shared a common node approx. at 23.7% level of similarity. The maximum similarity of 31.2% was observed between T. venosum and T. trilobatum. In view of its close genetic similarity with other members of Typhonium, transfer of Sauromatum venosum to the genus Typhonium and merger of the two genera was supported.

Genetic variation and relationship among 28 mango germplasm were analyzed using Random Amplified Polymorphic DNA (RAPD). Out of 20 primers screened, four were selected, which gave 50 clear and bright fragments, out of which 48 fragments were considered polymorphic. The proportion of polymorphic loci and gene diversity values across all loci were 96% and 0.29, respectively. The UPGMA dendrogram based on genetic distance segregated the 28 mango germplasm into two main clusters. Sukul alone formed one cluster and the rest germplasm were grouped together into another cluster. Mallika and Amrapali cultivar pair was very close to each other with the highest intervarietal similarity index (87.30%) and lowest genetic distance (0.08). On the other hand, Sukul and Meghnath pair was more distant to each other with the lowest intervarietal similarity index (14.29%) and highest genetic distance (0.87). Classical taxonomic studies of the bamboos are based on floral morphology and growth habit, which can cause problems in identification due to erratic flowering. Identification and genetic relationships in 18 species of bamboo were investigated using SDS-PAGE and RAPD analysis. Protein profiles of 18 bamboo species revealed clear polymorphism which can be used to identify species. Dendrogram based on SDS-PAGE data separated 18 species into two major clusters with Bambusa nutans & B. polymorpha, Gigantochloa andamanica & Dendrocalamus longispathus, G. andamanica & D. gigangteus showed maximum similarity of 71.4% in each case while genotype B. tulda and B. nutans (10.4%) showed lowest similarity. On the other hand, RAPD analysis proved to be a better tool with 14.67 as an average number of bands per polymorphic primers. Dendrogram based on RAPD separated 18 species into 2 major clusters with D. membranaceous and B.multiplex showed maximum similarity of 83% while genotype D. asper and B. balcooa showed least similarity (10.6%).

This study examined the degree of variations in DNA fingerprints associated with slight altitudinal change of Schinus molle grown in Abha region, Saudi Arabia. Seven populations from Schinus molle plants located at 2193.0, 2246.0, 2197.7, 2441.0, 2372.0, 2250.6 and 2175.0 meters had been investigated. The degree of genetic variability was evaluated using random amplified polymorphic DNA (RAPD), mixed RAPD and intersimple sequence repeat markers (ISSR). The genetic similarity coefficients from RAPD analysis revealed the maximum similarity value (89.9%) was between population at 2250.6 m and population at 2175.0 m. The genetic similarity coefficients from mixed RAPD primers displayed the highest similarity value (87.6%) between population at 2246.0 m and population at 2197.7 m. Similarity coefficients from ISSR analysis revealed the highest similarity value (86.2%) among populations at 2193.0 m, 2246.0 m, 2441.0 m and at 2250.6 m. Super tree analysis (RAPD + mixed RAPD + ISSR) showed the highest similarity value (85.5%) between population at 2441.0 m and population at 2250.6 m. In conclusion, marker systems including RAPD, mixed RAPD and ISSR, alone or combined can be effectively used in determining the genetic relationship among Schinus molle plants even at very close populations.Bangladesh J. Random amplified polymorphic DNA (RAPD) markers were employed for characterization, assessment of genetic variation and inferring relationships among six variants of Canna indica L. A total of 198 RAPD bands ranging from 200 bp to 3 kbp were generated by all the six variants. Among them, most of the bands were found to be polymorphic, four band were unique of which two bands (OPA022000 and OPA043000) were observed in the variant 2 (small red) and the other two (OPA013000 and OPA053000) were noticed in the variant 4 (orange), and the remaining bands were found to be monomorphic. The pair-wise genetic distance was determined among the six variants that ranged from 0.1446 to 0.6554. A dendogram was constructed based on the RAPD profiling to infer the relationship among the six variants of C. indica that resulted in two major clusters: the first one contained two variants, viz. variant 1 (local red) and variant 2 (small red), while the second cluster composed of the remaining four variants. The genetic relationships among the five groups of hexaploid wheat: common, spelta, macha, vavilovii, and semi-wild wheat (SWW) are not clear. Random amplified polymorphic DNA (RAPD) analysis was used to assess phylogenetic relationships among these five morphological groups of hexaploid wheat. RAPD data were analyzed using the NTSYS-PC computer program to generate Jaccard genetic similarity coefficients. A dendrogram based on RAPD analysis grouped 15 accessions into five distinct clusters. These results are in agreement with those based on morphological classification, suggesting that common wheat is most closely related to SWW, followed by spelta, vavilovii, and macha.Key words: RAPD, macha, spelta, vavilovii, semi-wild wheat, phylogenetic relationships.

RAPD analysis of 19 Ponkan mandarin accessions was performed using 25 random primers. Of 112 amplification products selected, only 32 were polymorphic across five accessions. The absence of genetic variability among the other 14 accessions suggested that they were either clonal propagations with different local names, or that they had undetectable genetic variability, such as point mutations which cannot be detected by RAPD.

Vegetable grafting is a practice employed worldwide since it helps prevent biotic and abiotic disorders, and watermelon is one of the most important species grafted. The objective of this study was to set critical limits for the characterization of quality categories for grafted watermelon seedlings. Specifically, watermelon (scion) seedlings were grafted onto squash (rootstock) seedlings, moved into a healing chamber for 7 days, and then transferred into a greenhouse for seven more days. At 7 and 14 days after grafting, experienced personnel assessed grafted seedling quality by categorizing them. The categories derived were Optimum and Acceptable for both time intervals, plus Not acceptable at 14 days after grafting. Optimum seedlings showed greater leaf area, and shoot and root fresh and dry weights at both time intervals. Moreover, they had greater stem diameter, root-to-shoot ratio, shoot dry weight-to-length ratio and Dickson’s quality index compared to the other category at 14 days after grafting. Therefore, Optimum seedlings would likely develop into marketable plants of high quality, with better establishment in the field. Not acceptable seedlings showed considerably inferior development, while Acceptable seedlings were between the other categories, but were still marketable.

Abstract The Rhipicephalus (Boophilus) microplus complex currently consists of five taxa, namely R. australis, R. annulatus, R. (B.) microplus clade A sensu, R. microplus clade B sensu, and R. (B.) microplus clade C sensu. Mitochondrial DNA-based methods help taxonomists when they are facing the morpho-taxonomic problem of distinguishing members of the R. (B.) microplus complex. The purpose of this study was to perform molecular characterization of ticks in all five regions of Brazil and infer their phylogenetic relationships. Molecular analysis characterized 10 haplotypes of the COX-1 gene. Molecular network analysis revealed that haplotype H-2 was the most dispersed of the studied populations (n = 11). Haplotype H-3 (n = 2) had the greatest genetic differentiation when compared to other Brazilian populations. A Bayesian phylogenetic tree of the COX-1 gene obtained strong support. In addition, it was observed that the population of R. (B.) microplus haplotype H-3 exhibited diverging branches among the other Brazilian populations in the study. The study concludes that the different regions of Brazil have R. (B.) microplus tick populations with distinct haplotypes.
