Abstract The present research was planned to assess the occurrence of intestinal parasites in small ruminants of Upper Dir of Khyber Pakhtunkhwa Province of Pakistan. For this purpose, the faecal material was collected randomly with gloved fingers directly from the rectum region of sheep and goats and the faecal materials were then put in hygienic plastic bottles with 10% formalin. The overall 315 (n=184 sheep and n= 131 goats) faecal samples were collected out of 315 samples, 281 were found positive for different parasites. Patterns-wise prevalence of GI parasites of the study area was found. Overall Single parasitism 89.20% (281/315) with 94.0% (173/184) in sheep and 82.43% (108/131) in goats. Double parasitic infection in small ruminant recorded in which Fasciola+ Haemonchus. contortus in sheep were found their prevalence was 25.54% (47/184). While in goats, the double parasitic infection in which Haemonchus contortus+Trichuris spp were found and their prevalence were 23.43% (30/131). The species found in the sample of sheep were includes, i.e., Strongyloides papillosus (41.30%), Heamonchus controtus (21.73%), Trichuris ovis (17.39%), and Fasciola hepatica (13.58%), the corresponding value for goat were Strongyloides spp 33.33% (36/108), Haemonchus spp 28.70%, (27/108), Trichuris spp 25.20% (27/184) and Fasciola spp 10.68% (14/184). The sheep of the study area are more infected as compared to goats. This study suggested that gastrointestinal parasites are major health problems of small ruminants in the study area. Therefore, a comprehensive study on species of gastrointestinal parasites circulating in the area, control options, cost-effective strategies and awareness about gastrointestinal parasites among the farmers in the study area should be instituted.

Fasciola spp. liver fluke have significant impacts in veterinary and human medicine. The absence of a vaccine and increasing anthelmintic resistance threaten sustainable control and underscore the need for novel flukicides. Functional genomic approaches underpinned by in vitro culture of juvenile Fasciola hepatica facilitate control target validation in the most pathogenic life stage. Comparative transcriptomics of in vitro and in vivo maintained 21 day old F. hepatica finds that 86% of genes are expressed at similar levels across maintenance treatments suggesting commonality in core biological functioning within these juveniles. Phenotypic comparisons revealed higher cell proliferation and growth rates in the in vivo juveniles compared to their in vitro counterparts. These phenotypic differences were consistent with the upregulation of neoblast-like stem cell and cell-cycle associated genes in in vivo maintained worms. The more rapid growth/development of in vivo juveniles was further evidenced by a switch in cathepsin protease expression profiles, dominated by cathepsin B in in vitro juveniles and then by cathepsin L in in vivo juveniles. Coincident with more rapid growth/development was the marked downregulation of both classical and peptidergic neuronal signalling components in in vivo maintained juveniles, supporting a role for the nervous system in regulating liver fluke growth and development. Differences in the miRNA complements of in vivo and in vitro juveniles identified 31 differentially expressed miRNAs, notably fhe-let-7a-5p , fhe-mir-124-3p and, miRNAs predicted to target Wnt-signalling, supporting a key role for miRNAs in driving the growth/developmental differences in the in vitro and in vivo maintained juvenile liver fluke. Widespread differences in the expression of neuronal genes in juvenile fluke grown in vitro and in vivo expose significant interplay between neuronal signalling and the rate of growth/development, encouraging consideration of neuronal targets in efforts to dysregulate growth/development for parasite control.

The complement system is a first-line innate host immune defence against invading pathogens. It is activated via three pathways, termed Classical, Lectin and Alternative, which are mediated by antibodies, carbohydrate arrays or microbial liposaccharides, respectively. The three complement pathways converge in the formation of C3-convertase followed by the assembly of a lethal pore-like structure, the membrane attack complex (MAC), on the pathogen surface. We found that the infectious stage of the helminth parasite Fasciola hepatica, the newly excysted juvenile (NEJ), is resistant to the damaging effects of complement. Despite being coated with mannosylated proteins, the main initiator of the Lectin pathway, the mannose binding lectin (MBL), does not bind to the surface of live NEJ. In addition, we found that recombinantly expressed serine protease inhibitors secreted by NEJ (rFhSrp1 and rFhSrp2) selectively prevent activation of the complement via the Lectin pathway. Our experiments demonstrate that rFhSrp1 and rFhSrp2 inhibit native and recombinant MBL-associated serine proteases (MASPs), impairing the primary step that mediates C3b and C4b deposition on the NEJ surface. Indeed, immunofluorescence studies show that MBL, C3b, C4b or MAC are not deposited on the surface of NEJ incubated in normal human serum. Taken together, our findings uncover new means by which a helminth parasite prevents the activation of the Lectin complement pathway to become refractory to killing via this host response, in spite of presenting an assortment of glycans on their surface.

ABSTRACT: Fascioliasis is a zoonosis of global distribution caused by the parasitic trematode Fasciola spp. Infection in humans can occur in areas endemic for animal fascioliasis, suggesting the need for studies on this parasitosis and its determinants. This exploratory study aimed to analyze the spatial distribution of bovine fascioliasis in the state of Santa Catarina, Brazil, based on cases notified between 2015 and 2017 in a state abattoir, located in the municipality of Orleans, and to associate disease occurrence with altitude, temperature, and rainfall. The research was conducted at Centro Universitário Barriga Verde (UNIBAVE)/Orleans, SC and Fundação Oswaldo Cruz/Rio de Janeiro, RJ. During the study period, the abattoir received animals from 58 municipalities in that state. Of the animals slaughtered in that period, 10,81% were infected with Fasciola hepatica. Presence of bovine fascioliasis infection was identified in the South and Southeast regions of the state, with higher prevalence rates in the latter. There was an association between low altitudes and higher occurrence of cases in the municipalities assessed. In addition, prevalence of bovine fascioliasis was higher in municipalities with high temperatures and low rainfall. Therefore, these results should be considered for planning disease control measures in the South and Southeast regions of the state of Santa Catarina.

Abstract Pseudosuccinea columella is considered invasive and has become an important intermediate host of both Fasciola species in many regions of the world. This systematic review assessed the geographical distribution of P. columella, and its implications in the transmission of Fasciola hepatica and Fasciola gigantica, globally. A literature search was conducted on Google Scholar, JSTOR and PubMed databases using Boolean operators in combination with predetermined search terms for thematic analysis. Results show that P. columella has been documented in 22 countries from Europe (3), Africa (8), Oceania (2), North America (3) and South America (6). Furthermore, this snail species has shown to adapt to and inhabit a vast array of freshwater bodies including thermal lakes and ditches with acidic soils. Studies showed that P. columella transmits F. hepatica, with natural and experimental infections documented in sub-Saharan Africa, Europe, South America and North America. Experimental infection studies in Cuba showed the presence of P. columella populations resistant to F. hepatica infection. Furthermore, some populations of this invasive snail collected from F. hepatica endemic locations in Brazil, Venezuela, Australia, South Africa, Colombia and Argentina were found without Fasciola infection. As a result, the role played by this snail in the transmission of Fasciola spp. in these endemic areas is still uncertain. Therefore, further studies to detect natural infections are needed in regions/countries where the snail is deemed invasive to better understand the veterinary and public health importance of this snail species in Fasciola-endemic areas and determine the global dispersion of resistant populations of P. columella.

Fascioliasis is an infection of the trematodes of Fasciola hepatica and Fasciola gigantica which causes disease worldwide. This trematode infection commonly attacks ruminants, especially in cattle, buffalo, sheep, goats, pigs and humans, as well. This disease can cause significant economic losses in the livestock industry, mainly through death, liver damage, reduced production of meat, milk and wool, and expenses for deworming. Parasitological examination for identification of fasciola infection was carried out by visually observing fecal samples for adult worms and then examined by simple flotation (using saturated NaCl solution) and formalin-ethyl acetate sedimentation techniques. Here, we were systematically reviewed existing knowledge about sensitive and efficient fasciola observation methods for the identification of fasciola species. Four databases of articles that met the inclusion requirements were compiled for the literature review. This article was published between 2015-2020 and reports on the identification of fasciola species using one or a combination of sedimentation and flotation methods. As a result, the combined method of sedimentation and flotation has the best sensitivity and egg recovery rate (Ef) values for fasciola identification.   Abstrak Fascioliasis merupakan infeksi trematoda spesies Fasciola hepatica dan Fasciola gigantica yang menimbulkan penyakit di seluruh dunia. Infeksi trematoda ini umum menyerang ruminansia, terutama pada sapi, kerbau, domba, kambing, babi dan bisa terjadi pada manusia. Penyakit ini dapat menimbulkan kerugian ekonomi yang cukup besar dalam industri ternak, terutama melalui kematian, kerusakan hati, berkurangnya produksi daging, susu, dan wol, dan pengeluaran biaya untuk obat cacing. Pemeriksaan parasitologi untuk identifikasi infeksi fasciola dilakukan dengan mengamati sampel feses secara visual untuk cacing dewasa dan kemudian diperiksa dengan cara flotasi sederhana (menggunakan larutan NaCl jenuh) dan teknik sedimentasi formalin-etil asetat. Disini, kami meninjau secara sistematis pengetahuan yang ada tentang metode pengamatan fasciola yang sensitif dan efisien dalam identifikasi spesies fascioal. Empat data base artikel yang telah memenuhi syarat inklusi disusun untuk tinjauan literature review. Artikel ini terbit antara tahun 2015-2020 dan melaporkan identifikasi spesies fasciola menggunakan salah satu atau gabungan metode sedimentasi dan flotasi. Hasilnya, metode gabungan antara sedimentasi dan flotasi memiliki nilai sensitivitas dan nilai tingkat pemulihan telur (egg recovery rate, Ef) paling baik untuk identifikasi fasciola.

Background and Aim: Gastrointestinal parasites identified in the wild can negatively affect host fitness, lower performance, and growth. On the other side, sympatric mammals that share habitat and resources may also cross-transmit parasites, which are often zoonotic and can contribute to morbidity and mortality. This study aimed to characterize the diversity of gastrointestinal parasites circulating in mammalian hosts in Moukalaba-Doudou National Park. Materials and Methods: We screened a total of 25 fecal samples collected from nine wild mammalian species, namely, western gorilla (Gorilla gorilla gorilla), chimpanzee (Pan troglodytes), putty-nosed monkey (Cercopithecus nictitans), African forest elephant (Loxodonta cyclotis), African buffalo (Syncerus caffer), blue duiker (Philantomba monticola), bay duiker (Cephalophus dorsalis), and red river hog (Potamochoerus porcus) as well as people working as trackers (Homo sapiens) using direct microscopic observations following a sedimentation technique to concentrate the fecal material. Results: Of the total 25 fecal samples screened, 15 (60%) were positive for parasitic gastrointestinal infection. Based on the morphology of parasite eggs and cysts, we identified a rich diversity of nematodes, protozoans, trematodes, and cestodes, including unidentified strongyles (73%), Oesophagostomum spp. (53%), Ancylostoma spp. (27%), Trichuris spp. (13%), Ascaris spp. (13%), Mammomonogamus spp. (13%), Strongyloides spp. (47%), Balantidium coli (20%), Entamoeba coli (20%), Endolimax nana (6%), Fasciola hepatica (6%), Paramphistomum spp. (13%), and Taenia spp. (6%). Conclusion: All parasites were found at least once in one of the hosts, and most were potentially zoonotic and responsible for several diseases of public health concern. Because of the small sample size, our findings should not be considered conclusive. Nevertheless, they highlight the diversity of gastrointestinal parasites in this area.

La Distomatosis hepática también conocida como Fascioliasis es una enfermedad parasitaria de los animales herbívoros, de manera especial de la especie bovina y zoonótica. Por lo tanto, se planteó recopilar información de literatura sobre Distomatosis hepática o fascioliasis  en bovinos, zoonosis  y factores de riesgo e interés sobre la salud pública, de igual manera analizar el número de casos mediante un estudio retrospectivo sobre la prevalencia de hígados decomisados por la presencia del parásito Fasciola hepática en el Camal Municipal de Ambato entre los años 2019, 2020, 2021 (primer semestre). La metodología que se utilizó fue exploratoria y descriptiva a través de revisión bibliográfica de artículos científicos publicados en diferentes bases de datos publicados por varios autores que permite obtener información de la Distomatosis o fascioliasis hepática. Se tomó toda la población de reses que llegaron a ser faenadas; en el año 2019 de 21,276, en el 2020 de 12.396 este disminuyó por restricciones de la pandemia COVID-19 y para el 2021(primer semestre) de 6.570 bovinos. Los datos fueron ubicados en tablas de contingencia, se analizó la  prevalencia   de   Distomatosis   hepática o fascioliasis, para el año 2019 de 7.96%, para 2020 de 7.21% y para el 2021 de 6.49%; con un promedio de los tres años del 7.22%, de esta manera se demuestra la existencia Distomatosis en el Camal Municipal de Ambato-Ecuador; se recomienda un plan sobre el manejo, prevención y control, tratamiento y erradicación de dicho parásito y dar a conocer a la población sobre los factores de riesgo.
