Close examination of hormonal profiles and uterine morphology in the marsupial reproductive cycle highlights significant differences between pregnant and non-pregnant cycles. In the polyovular dasyurid marsupial Sminthopsis macroura, we identified changes associated with gestation by comparing ovarian and plasma progesterone concentrations, uterine weights, uterine epithelial mitoses, body weights and gestation lengths between pregnant and non-pregnant luteal phases. The plasma progesterone profile of S. macroura was biphasic, peaking during unilaminar blastocyst expansion and on the day of implantation. Periods of rapid embryonic development were associated with increasing plasma progesterone concentrations and animal body weight. For the first time in a polyovular marsupial, we identified 1) a correlation between ovarian progesterone concentration and conceptus number during the luteal phase just prior to implantation (total ovarian progesterone), indicating a conceptus influence on progesterone concentration; 2) a pulse of uterine epithelial mitotic activity at the time of implantation and 3) increased mitotic activity in pregnant animals during unilaminar blastocyst formation compared with non-pregnant animals. Gestation length was reduced by up to 15%, due to the loss of, or reduction in, the four-cell arrest and more rapid definitive blastocyst expansion. This is the first time a conceptus influence on gestation length has been identified in a dasyurid. This study provides further evidence for the modification of the luteal phase by pregnancy in S. macroura.

ABSTRAK.  Progesteron merupakan hormon yang berperan penting dalam proses pemeliharaan kebuntingan dan dihasilkan oleh corpus luteum. Penelitian ini bertujuan mengetahui hubungan antara kadar hormon progesteron pada fase awal luteal dengan kematian embrio pada sapi Aceh. Dalam penelitian ini digunakan empat ekor sapi betina dewasa berumur 3-5 tahun, bobot badan 150-250 kg, sehat secara klinis, dan memiliki reproduksi normal. Sapi disinkronisasi menggunakan 5 ml prostaglandin F2 alfa (PGF2α) dengan pola penyuntikan ganda berinterval 11 hari. Koleksi sampel darah untuk pengukuran konsentrasi progesteron dilakukan pada hari ke-5, 6, dan 7 pasca inseminasi. Pengukuran konsentrasi progesteron dilakukan menggunakan metode enzymelinked-immunoassay (ELISA), pemeriksaan kebuntingan dan kematian embrio menggunakan metode transrektal ultrasonografi pada hari ke-25 pasca inseminasi. Pemeriksaan diulang setiap 10 hari sampai hari ke-55 pasca inseminasi. Puncak sekresi progesteron pada sapi bunting dengan embrio yang bertahan hidup terdapat pada hari ke-7 (2,082 ng/ml), pada sapi Late Embryonic Mortality (LEM) di hari ke-5 (8,209 ng/ml) dan pada sapi tidak bunting di hari ke-7 (3,051±1,157 ng/ml). Sekresi progesteron sapi LEM pada hari ke-5 sampai dengan ke-7 cenderung menurun sedangkan pada sapi yang bertahan hidup cenderung meningkat.  (Correlation between progesterone levels in early luteal phase and embryonic death  in Aceh cattle) ABSTRACT. Progesterone is an important hormone that functions to maintain pregnancy and is produced by the corpus luteum. The aim of this study was to see a correlation between progesterone and the incidence of embryonic death in Aceh cattle. This study used four adult female cows, 3-5 years old, 150-250 kg body weight, clinically healthy, and have a normal reproduction. The synchronized with 5 ml prostaglandin F2 alfa hormone, and double injection pattern with 11-day intervals. The blood was collected for progesterone measurements on 5th, 6th, 7th day post artificial insemination. Measurement of progesterone concentration was carried out using an enzymelinked-immunoassay (ELISA), while pregnancy and embryo mortality was performed using the trans-rectal ultrasonography method on the 25th day after insemination. The examination was repeated every 10 days until day 55th after insemination. Progesterone secretion peaks in pregnant cows were on day 7th (2.082 ng/ml), in cattle Late Embryonic Mortality (LEM) on day 5th (8.209 ng/ml) and in cattle not pregnant on day 7th (3.051±1.157 ng/ml). The pattern of LEM progesterone secretion on days 5th to 7th tends to decrease while those that survive tend to increase.

SummaryThirty-nine adult ewes of identical body size and age were randomly taken from the Chokla breed. They were divided into four groups which were allowed water once in 24, 48, 72 and 96 h. Seven ewes from each of the first, third and fourth groups were mated for the first time in the spring and for a second time in winter, so that they lambed in the monsoon and summer season respectively. All the ewes were maintained on uncultivated pasture. Watering once in 72 and 96 h caused body weight loss up to 26%, compared with those watered daily. The ewes which failed to maintain pregnancy lost more than 30% of their body weights due to watering only once in 96 h and there were about 43 and 100% lambing in the first and second breeding cycle of the same ewes. The remaining groups displayed 100% lambing. The water-intake increased significantly in the third month of pregnancy in the group allowed water daily and water consumption was found to be about 13% of body weight, whereas the values for those watered once in 72 and 96 h were 9 and 8% respectively. The water-deprived animals were able to drink up to 32 % of their body weights within 2–3 min. It is concluded that breeding ewes could be watered once in 72 h without any loss of lambing during summer.

Maternal recognition of pregnancy in marsupials occurs in more subtle ways than it does in eutherians. For instance, unlike in eutherians, the plasma progesterone profiles of pregnant and non-pregnant animals are similar during the luteal phase. It is typically during the brief luteal phase that both gestation and parturition occur in marsupials. Yet histological and physiological changes have been documented between gravid and non-gravid uteri in certain monovular marsupials and between pregnant and non-pregnant animals in polyovular marsupials. Early pregnancy factor (EPF), a 10.8-kDa serum protein known to be homologous to chaperonin 10, is associated with maternal immunosuppression, embryonic development and pregnancy in eutherian mammals. It has been reported in two Australian marsupials: the dasyurid Sminthopsis macroura and the phalangerid Trichosurus vulpecula. This paper documents its occurrence in the New World didelphid Monodelphis domestica. EPF is detectable by rosette inhibition assay in the peripheral circulation of pregnant but not of non-pregnant or pseudopregnant animals. Our work focuses on the embryo–maternal signalling role of EPF during pregnancy. Because progesterone-driven changes are similar in pregnant and non-pregnant marsupials, these animals are an excellent laboratory model in which to investigate the role of EPF in orchestrating the physiological changes necessary to sustain pregnancy.

SUMMARY Progesterone concentrations in the peripheral plasma of cows were measured by a protein-binding radioassay method. The mean concentration was lowest at oestrus (0·44 ng/ml) and then increased to a maximum of 6·8 ng/ml about day 14 of the 21-day cycle. The concentration decreased rapidly during the last 4 days of the cycle, reaching low levels on the day before oestrus. There were no significant changes in progesterone concentration during oestrus. After ovariectomy the plasma progesterone concentration decreased to a very low level (< 0·4 ng/ml). After hysterectomy, progesterone concentrations remained high for longer than in a normal cycle. At puberty, plasma progesterone concentrations indicated cyclic ovarian activity before the first observed oestrus. Daily treatment of cows with oxytocin (0·4 u./kg body weight) from day 2 reduced the oestrous cycle length to 9 days, but did not significantly alter the slow increase in plasma progesterone concentration during the first 5 days of the cycle. Plasma progesterone concentrations decreased again after day 5 to low values. Plasma progesterone concentration during early pregnancy was similar to the luteal phase value (4–6 ng/ml), declined during mid-pregnancy and then increased to a maximum (7–8 ng/ml) at about 240 days gestation. The concentration declined 2–3 weeks before calving. During lactation progesterone concentrations were very low until the resumption of cyclic ovarian activity. The first post-partum cycle, whether accompanied by observed oestrus or not, was usually preceded by a small increase in plasma progesterone concentration 3–5 days before the start of the cycle. Undernutrition significantly increased plasma progesterone concentrations in mid- and late pregnancy. Undernutrition of non-pregnant cows increased progesterone concentrations during the luteal phase of the first cycle, but reduced it in later cycles.

The progesterone concentration in peripheral plasma has been measured sequentially in individual cows during the estrous cycle, gestation, at parturition, and post-partum. During the estrous cycle the concentration was lowest just before, during, and just after estrus when the level (0.1–0.4 ng/ml) was similar to that found in three ovariectomized cows. The concentration commenced to rise on the 4th–6th day (day of estrus = 1st day), reached a peak of 3–6 ng/ml on the 11th–13th day, and dropped rapidly over a 24–48-hr period to a basal value 24–72 hr before the next estrus. In early pregnancy, the plasma progesterone concentration was similar to the maximum levels found during the luteal phase of the estrous cycle. Between 90 and 150 days there was an indication of a decline to a relatively low plasma progesterone concentration followed by a variable rise. Prior to parturition, there was a gradual decline over a period of 35–70 days reaching a level of <2.0 ng/ml the day before parturition. Following parturition, the level remained at <0.5 ng/ml until the first sign of the resumption of cyclic activity. The time for this to occur was very variable (20–>60 days). Although not conclusive, the evidence favors the view that for estrous behavior to occur, estrus must be preceded by luteal activity. The discharge of mucus can occur without previous luteal activity.

Some phenotypic and genetic trends on growth traits of lambs are observed. The research implicated lambs of two local breeds of sheep: Lipska (LP) and Svrljig (SV). All lambs were weighed for the first time at birth (BWB), and thereafter, regularly in two-week intervals. On the basis of the results during weighing the body weight (BW) were adjusted to the average age: 30 (BW30), 60 (BW60) and 90 days (BW90). Results of the effect of genotype on growth showed a significant effect on the growth traits. Sex of lambs had a significant effect at birth and very significant effect at 30 days, but had no significant effect on weight at 60 (BW60) and 90 (BW90) days. Birth type had significant effect on the body weight from birth to weaning and the highest body weights were recorded in singles. Phenotypic correlations were positive and very highly significant between the growth traits. Genetic correlations were significant, but not between all observed ages of lambs.

ABSTRACT Three of 24 infertile women with a short luteal phase transiently showed chorionic gonadotrophin (HCG) like immunoreactivity in serum as measured by the HCG-beta subunit radioimmunoassay. The plasma progesterone concentration was elevated above the lower normal post-ovulatory level in 2 of these women who also had elevated basal body temperature at the time HCG was detected. Positive HCG reaction coincided with the pre-ovulatory surge of luteinizing hormone (LH) in one patient, but in 2 other cases the LH concentration was low suggesting absence of LH interference. Our results suggest that some infertile women may conceive during their cycles with a short luteal phase and maintain pregnancy until HCG becomes detectable.

SUMMARY The progesterone concentration in the peripheral plasma of ewes throughout pregnancy has been determined by a protein-binding method. Plasma progesterone concentrations during the first 50 days of pregnancy (2–3 ng./ml.) were not significantly higher than peak concentrations during the luteal phase in cycling non-pregnant ewes, but there was no decrease in the concentration 15–20 days after mating as occurs in non-pregnant ewes. Between 50 and 120 days after mating the plasma progesterone concentration increased steadily to values 2–5 times that found in early pregnancy. These high concentrations were maintained until lambing. A decrease in progesterone concentration during the week preceding lambing was usually, but not always, observed. Mean plasma progesterone concentrations during the last 50 days of pregnancy in ewes with twins were approximately twice those in ewes with a single foetus.

ABSTRACT Serial plasma progesterone determinations were performed by gas-liquid chromatography in healthy women during 24 hour periods during the luteal phase of the cycle and during pregnancy. In 10 women during the hyperthermic phase of the cycle and in 14 women during early pregnancy (week 8–18) no significant circadian variation in the peripheral plasma progesterone concentration was observed. During later pregnancy (week 20–41) in 15 women, a statistically significant rise in the plasma progesterone concentration, usually from 8.00 a.m. until between 4.00 and 8.00 p. m. followed by a decrease after 8.00 p. m. until between 4.00 and 8.00 a. m., was demonstrated. The data indicate a diurnal change in the peripheral plasma progesterone level during the last trimester of pregnancy.
