Effect of long term water deprivation on body weight and water intake of breeding ewes under semi-arid conditions

SummaryThirty-nine adult ewes of identical body size and age were randomly taken from the Chokla breed. They were divided into four groups which were allowed water once in 24, 48, 72 and 96 h. Seven ewes from each of the first, third and fourth groups were mated for the first time in the spring and for a second time in winter, so that they lambed in the monsoon and summer season respectively. All the ewes were maintained on uncultivated pasture. Watering once in 72 and 96 h caused body weight loss up to 26%, compared with those watered daily. The ewes which failed to maintain pregnancy lost more than 30% of their body weights due to watering only once in 96 h and there were about 43 and 100% lambing in the first and second breeding cycle of the same ewes. The remaining groups displayed 100% lambing. The water-intake increased significantly in the third month of pregnancy in the group allowed water daily and water consumption was found to be about 13% of body weight, whereas the values for those watered once in 72 and 96 h were 9 and 8% respectively. The water-deprived animals were able to drink up to 32 % of their body weights within 2–3 min. It is concluded that breeding ewes could be watered once in 72 h without any loss of lambing during summer.

SUMMARYNon-breeding groups, I, II and III, each with six high-potassium (HK) and six lowpotassium(LK) Chokla ewes were given water once in 24, 48 and 72 h respectively. In the next summer, seven ewes (4 HK and 3 LK) from each group I, II and III were switched over to a watering schedule of once in 24, 72 and 96 h respectively and were naturally bred. All the animals were maintained on grazing alone.Water deprivation for 72 h caused 18·8 and 19·2% body weight loss in HK and LKewes respectively; an overall average maintenance of weight loss in LK ewes was significantly higher than in HK ewes from the same group. There were significant differences in water intake due to treatments only. The HK and LK ewes from groups I and III showed a similar trend.Pregnant ewes of HK and LK types given water once in 96 h lost 21·7 and 23·8% of their body weights respectively. Corresponding weight loss in aborted ewes were 23·8 and 33·3%. Two ewes, each from 3 LK and 4 HK animals aborted owing to water deprivation for 96 h. The water intake reached 30 and 36·5% of body weight in LK and HK pregnant ewes given water intermittently.The wool attributes of non-pregnant ewes did not indicate a significant influence of blood potassium types. Four out of five ewes of the LK phenotype died during 3 years, irrespective of watering schedule.

Abstract Background Cannabis use in pregnancy leads to fetal growth restriction (FGR), but the long-term effects on cardiac function in the offspring are unknown, despite the fact that fetal growth deficits are associated with an increased risk of developing postnatal cardiovascular disease. We hypothesize that maternal exposure to Δ9-tetrahydrocannabinol (Δ9-THC) during pregnancy will impair fetal development, leading to cardiac dysfunction in the offspring. Methods Pregnant Wistar rats were randomly selected and administered 3 mg/kg of Δ9-THC or saline as a vehicle daily via intraperitoneal injection from gestational days 6 to 22, followed by echocardiogram analysis of cardiac function on offspring at postnatal days 1 and 21. Heart tissue was harvested from the offspring at 3 weeks for molecular analysis of cardiac remodelling. Results Exposure to Δ9-THC during pregnancy led to FGR with a significant decrease in heart-to-body weight ratios at birth. By 3 weeks, pups exhibited catch-up growth associated with significantly greater left ventricle anterior wall thickness with a decrease in cardiac output. Moreover, these Δ9-THC-exposed offsprings exhibited increased expression of collagen I and III, decreased matrix metallopeptidase-2 expression, and increased inactivation of glycogen synthase kinase-3β, all associated with cardiac remodelling. Conclusions Collectively, these data suggest that Δ9-THC-exposed FGR offspring undergo postnatal catch-up growth concomitant with cardiac remodelling and impaired cardiac function early in life. Impact To date, the long-term effects of perinatal Δ9-THC (the main psychoactive component) exposure on the cardiac function in the offspring remain unknown. We demonstrated, for the first time, that exposure to Δ9-THC alone during rat pregnancy results in significantly smaller hearts relative to body weight. These Δ9-THC-exposed offsprings exhibited postnatal catch-up growth concomitant with cardiac remodelling and impaired cardiac function. Given the increased popularity of cannabis use in pregnancy along with rising Δ9-THC concentrations, this study, for the first time, identifies the risk of perinatal Δ9-THC exposure on early postnatal cardiovascular health.

Abstract Background Baloxavir acid (BXA), an active form of orally available prodrug baloxavir marboxil (BXM, formerly S-033188), is a novel small molecule inhibitor of cap-dependent endonuclease (CEN) of influenza A and B virus, and was recently launched for the treatment of acute and uncomplicated influenza with single dosing of BXM (the trade name XOFLUZA™) in Japan in March 2018. Here, we evaluated the prophylactic efficacy of BXA in mice lethally infected with influenza A virus. Methods T1/2 of BXA in human is more than 10 times longer than that in mice. Therefore, suspension of BXA was subcutaneously administered at 0.8 or 1.6 mg/kg in mice to maintain the plasma concentration of BXA as seen in humans, and then mice were intranasally inoculated with a lethal dose of A/PR/8/34 strain at 48, 72, or 96 hours after the administration of BXA. Survival time and body weight change were then monitored through a 28-day period after virus infection. Mice were euthanized and regarded as dead if their body weights were lower than 70% of the initial body weights according to humane endpoints. Results Single dosing of BXA (1.6 mg/kg) completely eliminated mortality in mice, when the mice were administrated the drug at 48, 72, or 96 hours before virus infection (Figure 1). BXA treatment also significantly prevented body weight loss, consistent with the prolonged survival. Conclusion Prophylactic dosing of BXA exhibited significant protective efficacy against mortality and body weight loss in mice following a lethal infection with influenza A virus. The significant prophylactic efficacy observed in our mouse model suggests the potential utility of BXM for the prophylaxis of influenza in human. Disclosures S. Our group showed that crude ethanol (CE) and butanol (BUT) extracts ofCapsicum baccatumpresented anti-inflammatory and antioxidant properties. Furthermore, the flavonoid and total phenolic contents were positively correlated with both of these properties observed forC. baccatumextracts. The present study demonstrated that 60 days of oral administration of CE and BUT (200 mg/kg) in mice did not cause significant differences in the following parameters evaluated: hematological profile, body weight and relative weight of visceral organs, systemic lipid profile, glucose homeostasis (GTT), kidney and hepatic biochemical markers, and spontaneous locomotion and anxiety-like behavior. Altogether, these results indicate for the first time that the long-term oral administration ofC. baccatumextracts does not affect specific aspects of CF1 mice physiology, suggesting their safety, building up the venue to test their efficacy in animal models underlying persistent activation of oxidative and inflammatory pathways.

SUMMARYAs part of a long-term breeding programme aimed at improving the performance of Omani goats and sheep, a 3-year performance trial was carried out between 1991 and 1993 to evaluate the reproductive and productive performance of three breeds of goat: Batinah (BT), Dhofari (DH) and Jabal Akhdar (JA), and one breed of local Omani sheep. In each year, flushing started on 1 September, mating started on 1 October and terminated on 15 November, and the kidding/lambing season started on average on 1 March. Kids and lambs were weaned at c. 3 months of age, then put on a 3-month performance test, fed on concentrates ad libitum and Rhodes grass hay. Some 3706 dam and 3011 progeny performance records were available over a 3-year period from 1991 to 1993. Ewes were lighter in body weight and lower in litter size weaned but higher in fertility than the does. Although the lambs had lighter birth weights, they were heavier at weaning and at 6 months of age. Among the goats, the DH does had lighter body weight but higher litter size weaned than both the BT and the JA goats. DH kids were somewhat lighter at birth, weaning and 6-month body weights than the other two breeds but had higher survival.The sheep had higher output per ewe exposed (3·28 kg, 33%) and per ewe lambed (3·27 kg, 26%) and were 35% more efficient in production than the goats. Although the DH goats had a lower output per doe exposed than the BT and JA goats (0·9 kg, 9 % and 0·76 kg, 7% respectively) and per doe kidded (1·11 kg, 9% and 1·29 kg, 10%), they had higher efficiency of production than both BT (0·05 units, 6%) and JA (0·11 units, 14%).In view of these results, attention should be paid to the sheep industry through the introduction of promotion schemes. The DH goats would be even more useful if involved in a crossbreeding scheme and the JA goats would benefit from selection for maternal care.

SummaryExternal balances of water, sodium and potassium were examined in carpet wool Chokla sheep given water once in 24, 48 and 72 h during the hot months. There was a significant decline (P < 0·01) in feed intake on the third day of dehydration. The water intake of this group approached 30% of body weight and there was also a significant (P < 0·05) decrease in urine volume after 2 days of water deprivation. There was a large balance for potassium in all treatments and most especially when the sheep were given water every 72 h. It is argued that this unusually high balance of potassium was probably a characteristic of sheep adapted to a hot climate.

AbstractThe question of effects of long-term melatonin (MEL) administration have not yet been explained sufficiently, especially its metabolic consequences in young persons and animals. The aim of the present study was to analyze the effects of MEL given during prolonged time (for 3 months) and chronically (for 6 months) at the dose of 4 µg/mL of tap water, on the selected metabolic and hormonal parameters in young female and male Wistar:Han (WH) rats. The weights of selected organs, tissues, body weight gains and food and water intake were registered. Six weeks aged rats were adapted to standard housing conditions and light regimen L:D=12:12 h, fed standard laboratory diet and drank tap water (controls) or MEL solution ad libitum; finally they were sacrificed after overnight fasting. Prolonged MEL administration decreased serum glucose concentration and increased triacylglycerol and malondialdehyde concentration/content in the liver in females. In males MEL increased concentrations of serum phospholipids, corticosterone and liver malondialdehyde. MEL treatment reduced the body weight in both sexes and weight of epididymal fat in males, without any alterations of food and water intake. Chronic MEL administration reduced serum glucose concentration and increased concentration/content of glycogen, triacylglycerol and cholesterol in the liver and glycogen concentration/content in heart muscle in males. In females, the significant rise of serum corticosterone concentration and liver malondialdehyde content was recorded. MEL significantly increased liver weight and decreased thymus weight in males. MEL administration increased temporarily water intake in males, body and epididymal fat weights were similar to that in controls. Body weight of MEL drinking females was reduced in the 1st half of experiment only; the food and water intake did not differ from control group. The response in WH rats on MEL was more prominent as in the Sprague-Dawley strain (our previous studies). Male rats were generally more affected, probably due to higher daily and total consumption of melatonin.

Some phenotypic and genetic trends on growth traits of lambs are observed. The research implicated lambs of two local breeds of sheep: Lipska (LP) and Svrljig (SV). All lambs were weighed for the first time at birth (BWB), and thereafter, regularly in two-week intervals. On the basis of the results during weighing the body weight (BW) were adjusted to the average age: 30 (BW30), 60 (BW60) and 90 days (BW90). Results of the effect of genotype on growth showed a significant effect on the growth traits. Sex of lambs had a significant effect at birth and very significant effect at 30 days, but had no significant effect on weight at 60 (BW60) and 90 (BW90) days. Birth type had significant effect on the body weight from birth to weaning and the highest body weights were recorded in singles. Phenotypic correlations were positive and very highly significant between the growth traits. Genetic correlations were significant, but not between all observed ages of lambs.

Cell kinetic characteristics of the epididymal adipose tissue of mice were examined during refeeding after prolonged food deprivation. Mature mice were given a glucose-electrolyte solution for 20-40 days, and subsequent to a body weight loss of 35%, they were given normal mouse food ad libitum. After refeeding, their body weight and adipose tissue weight returned to the levels before the deprivation. We examined the formation of new adipocytes in the epididymal adipose tissue with [3H]thymidine autoradiography. Flash labeling experiments revealed that cell proliferation was most active on the 6th day after refeeding and decreased thereafter until the 13th day when few labeled cells were seen. Cumulative labeling experiments showed that replicated poorly differentiated mesenchymal cells developed into adipocytes by storing fat droplets. New adipocyte formation was observed in adult mice during refeeding after long-term deprivation.

A study was undertaken to test whether the elimination of metabolic pathways strongly involved in growth and fatness, comprising thyroid hormones (TH) and growth hormone (GH), is responsible for a substantial part of the genetic change produced by selection. Lines used in this study have been selected for about 50 generations for high (PH) and low (PL) body weight at 10 weeks and for high (F) and low fat content (L) at 14 weeks, producing a 3-fold difference in body weights and a 5-fold difference in fat content. Thyroid ablation was achieved by repeated backcrossing into the four selection lines of a transgene comprising the HSV1-tk gene coupled to the promoter of the thyroglobulin gene. Hemizygous pregnant dams were treated with ganciclovir leading to thyroid-ablated dams and offspring and therefore to a lack of TH and subsequently of GH. In the absence of TH and GH, lines still differ in body weight over the period studied (10 d to about 100 d; e.g. at the end PH=32·1 g vs PL=10·2 g) and in fat content (F=16·2% vs L=3·8%) ; the corresponding values for the wild-type controls were PH=49·9 g vs PL=17·4 g and F=27·5% vs L=4·8%. The effect of the transgene depended on the genetic background for body weights at most ages and for relative gonadal fat pad weights, but less for fat content. The L line showed the lowest growth depression. The lit gene, which causes GH but not TH deficiency, was also transferred by repeated backcrosses into three of these lines (PH, PL, F). The combined deficiency of TH and GH had bigger effects on body weights at earlier ages than did GH deprivation. The data show that changes in the TH- and GH-systems are not the only cause of line differences in growth and fatness resulting from long-term selection, but both are involved to a significant extent. The interactions between the effects of the transgene and of the lit gene and the genetic background were, nevertheless, relatively small and therefore these results support a polygenic model of selection response.
