The aim of this study was to determine whether endogenous progesterone regulates synthesis and secretion of luteal oxytocin. In Expt 1, mature ewes (n = 5 per group) were assigned randomly to control or mifepristone (RU486) treatment groups. Ewes were injected s.c. twice a day with vehicle or 10 mg RU486 on days 5-7 of the oestrous cycle (oestrus = day 0). On day 8, after an i.v. injection with prostaglandin F(2alpha) (250 microg cloprostenol), venous blood samples were collected at frequent intervals to determine plasma oxytocin concentrations. Plasma oxytocin concentrations of RU486-treated ewes were not significantly different from those of control ewes. In Expt 2, ewes were injected s.c. each day with vehicle or 175 mg RU486 on days 2-5 of the oestrous cycle followed by administration of prostaglandin F(2alpha) on day 6. Four of five RU486-treated ewes showed 'split-oestrus' (oestrous behaviour for 36 h and then again at 84-108 h after the onset of initial oestrus). There was no significant difference in mean plasma oxytocin or progesterone concentrations between treatment groups. The mean masses of mature corpora lutea from control and RU486-treated ewes on day 6 of the oestrous cycle did not differ significantly (394.8 +/- 28.8 versus 319.5 +/- 48.3 mg). RU486-treated ewes contained mature corpora lutea, new corpora lutea (two of four ewes) and preovulatory follicles (>or= 10 mm, two of four ewes). The average interoestrous interval for RU486-treated ewes was 9 days more than that for control animals (26.2 +/- 2.9 versus 17 +/- 0.5 days; P < 0.025).

SUMMARY The response of the guinea-pig corpus luteum to the luteolytic influence of glass beads placed in the uterus, or to prostaglandin administration, was followed by assay of the progesterone content of blood samples collected daily. Following the introduction of glass beads into the uterus early in the cycle, the secretion of progesterone was curtailed. Treatment with prostaglandin F2α over days 4–6 or 6–8 of the cycle temporarily depressed progesterone release without shortening the life of the corpora lutea. When the drug was administered over days 8–10, 10–12 or 12–14 the depression in progesterone was not followed by any recovery. These observations indicate that the response of the corpora lutea to a luteolytic influence changes during the oestrous cycle.

This study was conducted to compare several methods for inducing pseudopregnancy in local rabbits. The study used 25 clinically healthy rabbits of local breeds, aged 1-1.5 years, with a bodyweight of 1.8-2.2 kg. The rabbits were divided into five treatment groups (n = 5): R1, R2, R3, R4, and R5. Each group was given a different treatment. Group R1 was injected intramuscularly with 100 IU PMSG and mated with males three days later, which was followed by an intravenous injection of 75 IU hCG. Pseudopregnancy in group R2 was induced through artificial copulation by inserting a 1 cm long cotton bud into vaginal lumen at 5 a.m. Pseudopregnancy in group R3 was induced using 50 IU hCG intramuscularly. Group R4 was injected intravenously with 5 g GnRH and then intramuscularly with 100 IU PMSG and three days later with 75 IU hCG. Blood samples were collected on days 2, 4, 6, and 8 for estrogen and progesterone measurement, whereas an ovarian histological observation of all groups was performed on day 8. The results show that the highest concentration of estrogen was obtained in group R2, which was significantly different from the other groups (P <0.05), but the difference in examination time was not statistically significant (P > 0.05). Furthermore, the concentration of progesterone in all groups fluctuated at various times of the day. Progesterone concentrations were higher in group R5, in which pseudopregnancy was induced with PMSG and hCG. Group R5 showed a significant difference (P <0.05) with R2, R3, and R4, but the difference with the positive control group (R1) was not significant (P > 0.05). The numbers of corpora lutea in groups R1, R2, R3, R4, and R5 were, respectively, 6.87 ± 1.58, 6.22 ± 2.71, 6.74 ± 1.94, 5.98 ± 2.04, and 8.8 ± 1.90, and the largest diameter was obtained in R5 (1.65 ± 0.37 mm). In conclusion, the best method of inducing pseudopregnancy in local rabbits is the administration of PMSG and hCG.

SUMMARYThe luteolytic effect of prostaglandin F2α (PGF2α) during the confirmed luteal phase of the oestrous cycle was evaluated in ten Boran and ten Boran × Friesian cross-bred heifers. Following injection with 25 mg Lutalyse, animals were bled every 6 h for 96 h and plasma progesterone (P4) determined by the ELISA technique. Borans had significantly (P < 0·05) smaller corpora lutea (12·01±0·72 ν. 17·03±2·10 mm) and responded faster to PGF2α injection (65·57±1·40 ν. 78·27±2·18 h) than the cross-bred heifers. However, there was no significant difference in either the initial P4 values (6.24±0·98 ν. 8·00±1·71 ng/ml) or the rate of its decline following PGF2α injection between the two breeds. Values declined sharply to basal levels (11% of the initial pretreatment values) within 48 h in both breeds. All ten cross-breds and eight Borans showed standing oestrus within a week of treatment. However, oestrus was better synchronized (P < 0·05) in Borans than cross-breds. It was concluded that PGF2α is effective for oestrus synchronization in both breeds, but oestrus occurred earlier and was more precise in Borans than in cross-breds.

SummaryTo characterize the extent of early activation of the hemostatic system following angioplasty, we obtained blood samples from the involved coronary artery of 11 stable angina patients during the procedure and measured sensitive markers of thrombin formation (fibrino-peptide A, prothrombin fragment 1.2, and soluble fibrin) and of platelet activation ((3-thromboglobulin). Levels of hemostatic markers in venous blood obtained from 14 young individuals with low pretest probability for coronary artery disease were not significantly different from levels in venous blood or intracoronary samples obtained prior to angioplasty. Also, there was no translesional (proximal and distal to the lesion) gradient in any of the hemostatic markers before or after angioplasty in samples obtained between 18 and 21 min from the onset of the first balloon inflation. Furthermore, no significant difference was noted between angioplasty and postangioplasty intracoronary concentrations. We conclude that intracoronary hemostatic activation does not occur in the majority of patients during and immediately following coronary angioplasty when high doses of heparin and aspirin are administered.

Background: The relationship between SLE and traditional risk factors for cardiovascular events was evaluated. Methods: The data regarding sixty patients with SLE and 30 healthy controls (age and sex matched) were gathered using SLEDAI forms. Venous blood (10mL) from all the participants was examined for hs-CRP, homocysteine, VCAM1, CBC, anti-DNA antibody, C3, C4, low-density lipoprotein (LDL), cholesterol, FBS and triglyceride. : The IMT of carotid arteries was determined bilaterally by ultrasound. Other measurements included insulin levels via Elisa (Linco/Millipore Corp) and the HOMA-IR index for insulin resistance. Results: The mean age (in years) in the test and control groups was 28.8±10.3 (18-52) and 33.8±9.13 (18-48), respectively. Results: The mean age (in years) in the test and control groups was 28.8±10.3 (18-52) and 33.8±9.13 (18-48), respectively. : The average IMT in the test group was directly related to serum levels of VCAM1 (p<0.001), homocysteine (p<0.001), cholesterol (p<0.009), LDL (p<0.001), TG (p<0.001), and FPG (p=0.004). The association between other risk factors, insulin resistance, carotid IMT and SLEDAI, was nonexistent. Mean insulin and insulin resistance levels in all the participants were 0.43±2.06 µU/mL and 0.09±0.44, respectively. There was no significant difference between the test and control groups regarding serum insulin and insulin resistance levels (p=0.42 and p=0.9, respectively). None of the risk factors, such as hsCRP, VCAM1, or homocysteine, were shown to be related to insulin resistance (p=0.6, p=0.6, p=0.09, respectively). Conclusion:: Our findings did not show an increase in the prevalence of atherosclerosis in patients with SLE. There was no association between IMT and insulin resistance. However, the former was associated with FPG, total cholesterol, LDL, TG, homocystein and VCAM1.

The purpose of this study is to compare whether or not there is a difference between venous and capillary blood samples in blood glucose measurements and investigate the effects of different aseptic methods used in skin cleaning before collecting blood samples on measurement results. This quasi-experimental study was conducted with 109 patients. The capillary first and second blood drop values taken from the patients after fasting and at 2 hours following 75 g oral glucose tolerance test (OGTT) and capillary and venous blood glucose values were compared. There was no significant difference between the median venous blood glucose value and the capillary second blood drop value taken after wiping the finger with alcohol. There was no significant difference between the first and second blood drop values of capillary blood glucose 2 hours after OGTT.

Background : Migraine is primary headache disorder characterized by recurring attacks of pain and associated symptoms. The management modality is still unsatisfactory due to poor understanding of its cause and pathogenesis. To assess the efficacy and safety of low dose Topiramate vs Propranolol in migraine prophylaxis. Materials and methods : A randomized clinical trial including 130 patients with frequent migraine headache >5 attacks per month was performed in the out patients Department of Medicine and Neurology, CMCH for a period of 12 weeks. The patients were randomly divided into two treatment groups – treated by Topiramate 50mg/day and Propranolol 80mg/day respectively. Topiramate started with 25mg/day for 7 days then increased up to 50mg/day in two divided dose. Propranolol started with 40mg/day for 7 days then increased up to 80mg/day in two divided dose. The patients were assessed at 0, 8 and 12 weeks of the study. Results: The Topiramate group showed a reduction in the mean (±SD) of frequency of migraine attack from 6.95(±2.88) to 1.75(±1.08) episode per month, headache days per month from 7.62(±4.14) to 1.83(±1.10), intensity of headache per attack from 8.98(±1.05) to 6.10(±2.50) based on VAS scale, duration of headache per episode from 11.56(±9.16) to 5.40(±2.97) per hour and MIDAS score from 16.19(±3.91) to 8.14(±3.93). In patient treated with Propranolol, the mean (±SD) of monthly frequency of migraine attack declined from 7.09(±2.87) to 1.92(±0.98) episode per month, headache days per month from 8.17(±4.52) to 1.86(±o.83), intensity of headache per attack from 8.47(±1.10) to 6.03(±2.05) based on VAS scale, duration of headache per episode from 11.16(±8.08) to 5.97(±3.45), MIDAS score from 15.48(±3.55) to7.77(±3.49). Pre- and post-treatment values were significantly different for individual groups but no significant difference observed between groups. Conclusion: This study demonstrated that both low dose Topiramate and propranolol could significantly reduce migraine frequency, intensity and duration. Background: This study was designed to investigate the effect of tranexamic acid and its side effects in preventing postpartum hemorrhage in patients referred to Ali ibn Abitaleb Hospital in Zahedan during 2020-2021. Methods: Patients were divided into two groups of 85 patients. Vital signs as well as drug side effects related to tranexamicity are monitored and recorded every15 minutes in the first hour and every 30 minutes in the second hour. Also, blood samples are taken from patients to measure hemoglobin and hematocrit within6 hours after delivery. The number and weight difference of droschitis used during 2 hours after delivery are also measured. In addition, PPH (Postpartum hemorrhage) cases in both groups were checked by repeated examination of the uterine fundus during the first 2 hours and after 6 weeks. Results: There was no statistically significant difference between the two groups in terms of the number of deliveries and further decrease in hemoglobin levels in the control group compared to the intervention group. Nausea, vomiting, and dizziness were observed in the intervention group and thrombosis was not observed in any group. According to the independent samples t-test, there was a statistically significant difference between the mean blood volume lost between the two groups. The mean hematocrit drop was statistically significant between the two groups. No blood transfusion was observed in any of the patients. Conclusion: Tranxamic acid injections can effectively reduce postpartum hemorrhage and reduce maternal hemoglobin and hematocrit following this complication.

Myoelectric recording techniques were used to study the motility of rabbit ileum during infusions of vasoactive intestinal peptide (VIP). VIP was infused intravenously at a rate of 300 pmol X kg-1 X h-1, and peripheral venous blood samples were obtained hourly for VIP assay. VIP was also infused intraluminally at a similar rate, and hourly portal vein blood samples were obtained for VIP assay. Alterations in motility were observed after both intravenous and intraluminal infusions of VIP. These alterations in motility consisted of the migrating action potential complex and repetitive bursts of action potentials. The VIP infusion rate used and the mean peripheral plasma VIP level of 267 +/- 29 pg/ml attained during intravenous VIP infusion were similar to those that induced intestinal secretion in other animal species. Portal venous VIP levels (93 +/- 21 pg/ml) were unchanged during the intraluminal infusion of VIP. These studies show that intravenous infusion of VIP causes alterations in motility of rabbit ileum. These alterations in motility with concomitant secretion of water and electrolytes may contribute to the diarrhea induced by VIP infusion. In addition, intraluminal infusion of VIP also induced alterations in myoelectric activity, which suggested that this peptide has a luminal effect as well as a hormonal effect.

Progesterone levels were estimated by radioimmunoassay in blood samples obtained by venipuncture on the day of estrus and every alternate day until the onset of the next estrus in eight cycling dairy heifers. The mean level of progesterone was < 1 ng/ml during the first 2 days of the cycle, increased rapidly over the 4th–12th day period and reached a peak level value of 5.2 ± 1.1 ng/ml on day 14. Thereafter, the level declined rapidly to 2.6 ± 0.6 ng/ml on day 16 and then more gradually to 0.4 ± 0.1 ng/ml on day 21. In the second experiment, eight cycling heifers at diestrus were treated with gonadotrophin (2,000 IU PMSG or 1,000 IU PMSG + 1,000 IU HCG) followed 48 h later by 15 mg prostaglandin (PGF2α). Mid-ventral laparotomies were performed 4 days after the onset of estrus to observe ovarian activity. Progesterone levels were considerably higher in some animals and were slightly higher on the average after gonadotrophin treatments. The number of corpora lutea (CL) in these heifers ranged from 1 to 17. Progesterone levels of three heifers with 4–9 CL did not differ (P > 0.05) from those of three heifers with single CL. Two heifers each with 17 CL had peak progesterone levels of 38.4 and 27.8 ng/ml which were still high (9.6 and 26.5 ng/ml) by day 21. The remaining six heifers had low progesterone levels (< 1 ng/ml) by days 8–14, indicating premature regression of the CL. Thus, progesterone levels were not correlated with the number of CL.
