Studies on 38 children and adults who ingested large amounts of salicylate revealed a poor correlation of salicylate level in serum with clinical severity of salicylate intoxication. Serial measurements of serum salicylate levels in dogs and in 17 previously well patients revealed that the disappearance of salicylate from circulation is a first-order reaction in the post-absorptive period. It is possible, therefore, to calculate the half-life of circulating salicylate. In the present study the mean salicylate half-life in 17 patients was 20 hours. It is possible to eliminate the variable of time since ingestion by utilizing the rate of removal (half-life) of salicylate to extrapolate from the observed salicylate level at any time following ingestion to a theoretical zero-time salicylate concentration (So). There was found to be an excellent correlation of So with the clinical severity of salicylate intoxication. A nomogram that eliminates the necessity for calculating So is presented. Calculation of So or use of the nomogram provide better means than were previously available for predicting the severity of salicylate intoxication on the basis of measurements of blood salicylate. Particularly noteworthy was the fact that four patients who died despite intensive conventional treatments had So values which greatly exceeded those found in patients who recovered. This observation suggests that an So of greater than 160 is essentially incompatible with life and that measures for rapidly reducing salicylate levels, such as hemodialysis or exchange transfusion, should be instituted. The data are not sufficient to permit conclusions to be drawn concerning the influence of various factors on salicylate removal and toxicity. However, the use of So and salicylate half-life should facilitate such studies.

BackgroundMunicipal drinking water contaminated with perfluorinated alkyl acids had been distributed to one-third of households in Ronneby, Sweden. The source was firefighting foam used in a nearby airfield since the mid-1980s. Clean water was provided from 16 December 2013.ObjectiveTo determine the rates of decline in serum perfluorohexane sulfonate (PFHxS), perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA), and their corresponding half-lives.MethodsUp to seven blood samples were collected between June 2014 and September 2016 from 106 participants (age 4–84 years, 53% female).ResultsMedian initial serum concentrations were PFHxS, 277 ng/mL (range 12–1660); PFOS, 345 ng/mL (range 24–1500); and PFOA, 18 ng/mL (range 2.4–92). The covariate-adjusted average rates of decrease in serum were PFHxS, 13% per year (95% CI 12% to 15%); PFOS, 20% per year (95% CI 19% to 22%); and PFOA, 26% per year (95% CI 24% to 28%). The observed data are consistent with a first-order elimination model. The mean estimated half-life was 5.3 years (95% CI 4.6 to 6.0) for PFHxS, 3.4 years (95% CI 3.1 to 3.7) for PFOS and 2.7 years (95% CI 2.5 to 2.9) for PFOA. The interindividual variation of half-life was around threefold when comparing the 5th and 95th percentiles. There was a marked sex difference with more rapid elimination in women for PFHxS and PFOS, but only marginally for PFOA.ConclusionsThe estimated half-life for PFHxS was considerably longer than for PFOS and PFOA. For PFHxS and PFOS, the average half-life is shorter than the previously published estimates. For PFOA the half-life is in line with the range of published estimates.

Aim: To elucidate the influence of non target Arbuscular Mycorrhizal (AM) fungi on glyphosate dissipation in okra cultivated sodic soil of Tamil Nadu. Methodology: A field experiment was carried out during Rabi 2017-18 to study the interactive effect of glyphosate and Arbuscular mycorrhizal fungi (AM) on glyphosate dissipation in sodic soil from okra (A. esculentus) grown field. Experiment was laid adopting Randomized Block Design (RBD) with different rates of glyphosate and with and without AM fungi. Glyphosate was applied as pre-plant herbicide 20 days before sowing, and AM biofertilizer was applied as basal before sowing of okra. Soil samples were collected from the field at different time interval after glyphosate application and analysed for glyphosate and Amino Methyl Phosphonic Acid (AMPA) residues. Results: Glyphosate persistence decreased with time and degraded with the mean half-life ranging from 5.09 to 10.35 days by following first order reaction kinetics. AMPA was witnessed from day 1 and increased up to 15th day and followed first order reaction kinetics dissipation with the mean half-life of 8.62 to 13.06 days. Glyphosate and AMPA residues were recorded below detection limit (0.01 mg kg-1) on 45th and 60th days, respectively, in soil. Interpretation: The higher rates of glyphosate showed enhanced persistence of glyphosate and its metabolite AMPA in soil, hence application of AM fungi 100 kg ha-1 with lower glyphosate rate of 6 ml/L can be recommended for okra cultivated sodic soil with the safer waiting period of after 45 days harvest.

SummaryThe use of plasma thrombospondin (TSP) concentration was investigated as an indicator of intravascular platelet activation. Patients (n = 20) with diseases that have known vasculitis were included in the study. The range and the mean of plasma TSP concentrations of patients with vasculitis were 117 ng/ml to 6500 ng/ml and 791±1412 ng/ml (mean ± SD); the range and the mean of plasma TSP concentrations of control individuals (n = 33) were 13 ng/ml to 137 ng/ml and 59±29 ng/ml. When plasma TSP concentrations were correlated with plasma concentrations of another platelet activation marker, β-thromboglobulin (P-TG), it was found that the TSP concentration inei eased exponentially as the plasma β-TG level rose. A positive correlation between plasma levels of plasma TSP and serum fibrin degradation products was also observed. The results suggest that platelets are the primary source of plasma TSP in patients with various vasculitis and that plasma TSP can be a better indicator than β-TG to assess intravascular platelet activation due to its longer circulation half life.

ABSTRACT Eight amenorrhoeic women were given 100 μg synthetic LRH (Hoechst) iv and im, respectively, at an interval of 2 weeks. Four of the women received the iv injection first and four the im injection. The urinary excretion of oestrogens and pregnanediol was low and unaltered throughout the test weeks. The effects of LRH were compared by serial measurements of the plasma LH and FSH during 8 h. The initial response of LH for up to 25 min and that of FSH for up to 60 min were equal whether LRH was given iv or im. The difference appeared later. Four hours after the injection the mean increase of LH to iv injection was 0.5 ng/ml (N. S.), while that to im injection was 1.9 ng/ml (P < 0.01). The corresponding values for FSH were 1.3 (P < 0.05) and 3.2 (P < 0.001). The effect of LRH administration im was thus found to be larger and more prolonged.

ABSTRACT 10 mg/kg of dibutyryl cyclic adenosine 3′,5′-monophosphate (Db-cAMP) was iv pulse injected into twelve healthy adult women. The plasma cortisol levels were determined as 11-OHCS at zero time and then at 2.5, 5, 7.5, 10, 15, 30, 60 and 180 min after the injection. The data were compared with those obtained at the corresponding times in two groups of eleven and seventeen healthy women after the injection of 250 ng and 250 μg of synthetic β-1-24 corticotrophin performed in the same manner as the injection of the nucleotide. The mean increments in plasma cortisol were significantly lower after Db-cAMP than after ACTH. Differences were noted by analyzing the time course of the responses. In the case of stimulation with Db-cAMP the 11-OHCS levels rose progressively to a maximum at 15–30 min. By contrast, a peak of plasma cortisol was evident in most cases within a few min after the injection of ACTH; after a fall, a later rise was then observed starting from 15 min. The differences in the plasma 11-OHCS responses after the two stimuli may also be of interest clinically for the investigation of some aspects of adrenal steroidogenesis.

Background: Prodrugs principle is widely used to improve the pharmacological and pharmacokinetic properties of some active drugs. Much effort was made to develop metronidazole prodrugs to enhance antibacterial activity and or to improve pharmacokinetic properties of the molecule or to lower the adverse effects of metronidazole. Objective: In this work, the pharmacokinetic properties of some of monoterpenes and eugenol pro metronidazole molecules that were developed earlier were evaluated in-vitro. The kinetic hydrolysis rate constants and half-life time estimation of the new metronidazole derivatives were calculated using the validated RP-HPLC method. Method: Chromatographic analysis was done using Zorbbax Eclipse eXtra Dense Bonding (XDB)-C18 column of dimensions (250 mm, 4.6 mm, 5 μm), at ambient column temperature. The mobile phase was a mixture of sodium dihydrogen phosphate buffer of pH 4.5 and methanol in gradient elution, at 1ml/min flow rate. The method was fully validated according to the International Council for Harmonization (ICH) guidelines. The hydrolysis process carried out in an acidic buffer pH 1.2 and in an alkaline buffer pH 7.4 in a thermostatic bath at 37ºC. Results: The results followed pseudo-first-order kinetics. All metronidazole prodrugs were stable in the acidic pH, while they were hydrolysed in the alkaline buffer within a few hours (6-8 hr). The rate constant and half-life values were calculated, and their values were found to be 0.082- 0.117 hr-1 and 5.9- 8.5 hr., respectively. Conclusion: The developed method was accurate, sensitive, and selective for the prodrugs. For most of the prodrugs, the hydrolysis followed pseudo-first-order kinetics; the method might be utilised to conduct an in-vivo study for the metronidazole derivatives with monoterpenes and eugenol.

The establishment of chemical equilibrium in a system with a reversible first order reaction is characterized in terms of the distribution of first passage times for the state of exact chemical equilibrium. The mean first passage time of this state is a linear function of the logarithm of the total number of particles in the system. The equilibrium fluctuations of composition in the system are characterized by the distribution of the recurrence times for the state of exact chemical equilibrium. The mean recurrence time is inversely proportional to the square root of the total number of particles in the system.

The final R was 0.050 for the unique set of 3103 observed reflections. The central 4H-thiopyran ring forms a boat conformation for both symmetrically independent molecules with average boat angles 4.4(3) and 6.8(3)° at S and C(sp3), respectively. The mean planes of phenyls at the position 2 and 6 are turned from the double plane of 4H-thiopyran by 42.5(5) and 35.8(3)°, respectively. The investigated material undergoes a photochromic change in the solid state after irradiation with UV light or X-rays. The maximum of the new absorption band is situated at 564 nm. The non-exponential time dependence of photochromic bleaching is analysed in terms of a dispersive first-order reaction.

Background Several studies have shown comparable early efficacy of mechanochemical ablation to endothermal techniques. The goal of this report was to show if early efficacy is maintained at 24 months. Methods This was a two-year analysis on the efficacy of mechanochemical ablation in patients with symptomatic C2 or more advanced chronic venous disease. Patients with reflux in the great saphenous vein involving the sapheno-femoral junction and no previous venous interventions were included. Demographic information, clinical, and procedural data were collected. The occlusion rate of treated veins was assessed with duplex ultrasound. Patient clinical improvement was assessed by Clinical-Etiology-Anatomy-Pathophysiology (CEAP) class and venous clinical severity score. Results Of the initial 126 patients, there were 65 patients with 24 month follow-up. Of these 65 patients, 70% were female, with a mean age of 70 ± 14 years and an average body mass index (BMI) of BMI of 30.5 ± 6. The mean great saphenous vein diameter in the upper thigh was 7.6 mm and the mean treatment length was 39 cm. Adjunctive treatment of the varicosities was performed in 14% of patients during the procedure. Closure rates were 100% at one week, 98% at three months, 95% at 12 months, and 92% at 24 months. There was one patient with complete and four with partial recanalization ranging from 7 to 12 cm (mean length 9 cm). There was significant improvement in CEAP and venous clinical severity score (P < .001) for all time intervals. Conclusion Early high occlusion rate with mechanochemical ablation is associated with significant clinical improvement which is maintained at 24 months, making it a very good option for the treatment of great saphenous vein incompetence.

Structural studies are largely performed without taking into account vibrational effects or with incorrectly taking them into account. The paper presents a first-order perturbation theory analysis of the problem. It is shown that vibrational effects introduce errors on the order of 0.02 Å or larger (sometimes, up to 0.1-0.2 Å) into the results of diffraction measurements. Methods for calculating the mean rotational constants, mean-square vibrational amplitudes, vibrational corrections to internuclear distances, and asymmetry parameters are described. Problems related to low-frequency motions, including torsional motions that transform into free rotation at low excitation levels, are discussed. The algorithms described are implemented in the program available from the author (free).
