Abstract Hymenaea martiana is a species popularly known in Northeastern Brazil as “jatobá” and used in folk medicine to treat pain and inflammation. The aim of this work was to evaluate the antinociceptive and anti-inflammatory activity of H. martiana. In the present study, we carried out an investigation about the effects of the crude ethanolic extract (Hm-EtOH) and the ethyl acetate fraction (Hm-AcOEt) in models of nociception and inflammation in mice. Chemical (acetic acid-induced writhing and formalin) and thermal stimuli (hot plate) were used for the evaluation of antinociceptive activity, while for the anti-inflammatory profile paw edema induced by carrageenan was used, along with leukocyte migration to the peritoneal cavity. The presence of the flavonoid astilbin in the samples was characterized through HPLC-DAD-MS analysis. Hm-EtOH and Hm-AcOEt (100, 200 and 400 mg.kg-1, i.p.) significantly reduced the number of abdominal contortions and decreased the paw licking time in the formalin test. In the hot plate, the extract increased the latency time of animals. Hm-EtOH and Hm-AcOEt inhibited significantly the increase in the edema after the administration of carrageenan. Hm-EtOH and Hm-AcOEt inhibited leukocyte migration in the peritonitis test. HPLC-DAD-MS analysis of Hm-EtOH and Hm-AcOEt revealed the presence of the flavonoid astilbin in the samples. According to the results of this study, both Hm-EtOH and Hm-AcOEt have antinociceptive and anti-inflammatory activities, which could be related with the presence of flavonoid in the extracts. The results reinforce the popular use of this plant.

Inflammatory skin disorders, such as psoriasis and atopic dermatitis, are very common in the population; however, the treatments currently available are not well tolerated and are often ineffective.Averrhoa carambolaL. (Oxalidaceae) is an Asian tree that has been used in traditional folk medicine in the treatment of several skin disorders. The present study evaluates the topical anti-inflammatory effects of the crude ethanolic extract ofA. carambolaleaves, its hexane, ethyl acetate, and butanol fractions and two isolated flavonoids on skin inflammation. Anti-inflammatory activity was measured using a croton oil-induced ear edema model of inflammation in mice. Topically applied ethanolic extract reduced edema in a dose-dependent manner, resulting in a maximum inhibition of 73 ± 3% and an ID50value of 0.05 (range: 0.02–0.13) mg/ear. Myeloperoxidase (MPO) activity was also inhibited by the extract, resulting in a maximum inhibition of 60 ± 6% (0.6 mg/ear). All of the fractions tested caused inhibition of edema formation and of MPO activity. Treatment with the ethyl acetate fraction was the most effective, resulting in inhibition levels of 75 ± 5 and 54 ± 8% for edema formation and MPO activity, respectively. However, treatment of mice with isolated compounds [apigenin-6-C-β-l-fucopyranoside and apigenin-6-C-(2″-O-α-l-rhamnopyranosyl)-β-l-fucopyranoside] did not yield successful results. Apigenin-6-C-(2″-O-α-l-rhamnopyranosyl)-β-l-fucopyranoside caused only a mild reduction in edema formation (28 ± 11%). Taken together, these preliminary results support the popular use ofA. carambolaas an anti-inflammatory agent and open up new possibilities for its use in skin disorders.

Physalis peruviana calyces are used extensively in folk medicine. The crude ethanolic extract and some fractions of calyces were evaluated in order to explore antioxidant and anti-inflammatory activities. The anti-inflammatory activity was evaluated by the TPA-induced ear edema model. The antioxidant in vitro activity was measured by means of the superoxide and nitric oxide scavenging activity of the extracts and fractions. The butanolic fraction was found to be promising due to its anti-inflammatory and antioxidant activities. Therefore, a bio-assay guided approach was employed to isolate and identify rutin (1) and nicotoflorin (2) from their NMR spectroscopic and MS data. The identification of rutin in calyces of P. peruviana supports the possible use of this waste material for phytotherapeutic, nutraceutical and cosmetic preparations.

ABSTRACT The present study was designed to investigate the antioxidant, antinociceptive and anti-inflammatory activities of the ethanol extract from Ageratum fastigiatum branches. Phytochemical screening and total phenol and flavonoid contents were determined. The antioxidant activity was assessed by 2,2-diphenyl-1-pycrilhydrazin (DPPH) and iron reducing power methods. The antinociceptive effect was evaluated using the acetic acid-induced writhing, formalin, hot plate and tail immersion assays; while the carrageenan-induced paw edema and pleurisy tests were performed to examine the anti-inflammatory activity against acute inflammation. The extract revealed the presence of flavonoids, tannins, coumarins, terpenes, sterols and saponins. Expressive levels of total phenols and flavonoids and a promising antioxidant effect were quantified. At the doses of 50, 100 and 200 mg/kg, the extract inhibited the writhing, reduced both phases of paw licking time and increased the reaction time on the hot plate. In the tail immersion test, the extract (50, 100 and 200 mg/kg) caused a significant inhibition of pain. In these doses, the paw edema, exudate volume and leucocyte mobilization were significantly reduced. These results suggest that A. fastigiatum can be an active source of substances with antioxidant, antinociceptive and anti-inflammatory activities, adding scientific support to the appropriate use in the Brazilian folk medicine.

Enhydra fluctuations an edible semi-aquatic vegetable plant are widely used in the traditional system of medicine. Ethanolic extract of Enhydra fluctuans was screened for analgesic & anti-inflammatory activity. Analgesic activity was evaluated by the hot plate method and anti-inflammatory activity was evaluated by formalin induce paw edema in Wistar rats (male). Ethanolic extract dose of 300 mg/kg reduce formalin induce paw inflammation and also increased the pain threshold in rats evidenced by hot plate method. The experimental results concluded that Enhydra fluctuans have significant analgesic and anti-inflammatory activity may due to flavonoid and phenolic compound content.

Non-communicable diseases like diabetes and cancer are the major cause of death worldwide. Various drugs are used for the treatment of these diseases. However, they cause lots of side effects. There is a need for alternate drugs with fewer side effects. Medicinal plants serve as a good source for alternate form of treatment. Therefore, in this study, ethanolic and aqueous extracts of D. regia flowers were evaluated for their antioxidant, antidiabetic, anti-inflammatory and cytotoxic activity to justify its use as a medicinal plant. Total phenol and flavonoid content of the extracts were measured. GC-MS analysis of the extracts were done to investigate the presence of various bioactive compounds. Antioxidant activity was assessed by radical scavenging and reduction assays. Antidiabetic activity was assessed by the ability of extracts to inhibit enzyme alpha amylase. Anti-inflammatory activity was evaluated by membrane stabilization activity. Anticancer activity against MCF-7 and A549 cell lines were measured by the MTT assay.The ethanolic extract contained more phenols (282.940.80 mgGAE/g) and flavonoids (140.912.27 mgQE/g). GC-MS analysis showed the presence of compounds belonging to fatty acids, alkanes, phenols and organic alcohols. The aqueous extract showed strong superoxide radical scavenging activity with a low IC50 of 39.35±0.74 µg/mL. The ethanolic extract showed higher ferric reducing power with an IC50 of 59.65±0.28µg/mL. Ethanolic extract was more potent in inhibiting alpha amylase with a low IC50 value of 47.14±0.6 µg/mL. Ethanolic extract also showed maximum inhibition of 88.86±0.1% against heat induced lysis of cell membrane. Both extracts affected the proliferation of MCF-7 and A549 cell lines at 160 µg/mL. The results of the present study support the use of D. regia flower as a potential source of bioactive phytochemicals and can be used as a plant-based antioxidant, antidiabetic, anti-inflammatory and anticancer agent.

Catunaregam spinosa leaves have been ethnopharmacologically accounted for acquiring various pharmacological properties. The present study was undertaken to evaluate anti-inflammatory, analgesic and antipyretic potential of leaves of C. spinosa.The ethanolic extract was selected for this purpose based on phytochemical screening. Inflammation was inhibited at the dose of 200 mg/kg with percent inhibition of inflammation 32.06, 37.28 and 43.16 %, respectively, at 1, 3 and 5 h, while in egg albumin model % inhibition was found to be 47.81%. There was no significant analgesic activity seen in acetic acid induced writhing response method while significant effects were observed in the doses of 25 and 100 mg/kg on hot plate test. No antipyretic activity was shown by ethanolic extracts(25, 100 and 200 mg/kg) against Brewer’s yeast induced pyrexia in rats. Background: The antipyretic, analgesic and anti-inflammatory activities of two concentrations (100 and 200 mg/kg) of ethanolic extract of leaf, bark, flower and fruit pulp of C. fistula were determined in male wistar albino rats. Methods: Antipyretic activity was assessed by E. coli endotoxin induced pyrexia. Analgesic activity was assessed by hot plate, tail immersion and acetic acid induced writhing test. Anti-inflammatory activity was evaluated by carrageenan-induced rat paw edema assay. Result: Significant (p less than 0.05) antipyretic activity was exhibited from 2h onwards by bark extract @ 200 mg/kg and from 3h onwards by bark extract @100 mg/kg and leaves extract @ 200mg/kg as compared to control group. Significant (p less than 0.05) analgesic activity was shown by extract of bark @ 200 mg/kg as it is evident by increase in reflex time in hot plate (90,120,180 min), tail immersion test (120,180 min) and inhibition of writhing (32.12%). Significant (p less than 0.05) anti-inflammatory activity was exhibited from 3h post administration by bark @ 200 and leaves @ 100 and 200 mg/kg.

Lobelia chinensis Lour (LcL) is a popular herb that has been widely used as folk medicine in China for the treatment of fever, lung cancer, and inflammation for hundreds of years. Recently, several studies have shown that the anti-inflammatory properties were correlated with the inhibition of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) from the NF-κB pathway. The aim of this study was to evaluate the anti-oxidative and anti-inflammatory activities of L. chinensis. Both suppressive activities on LPS-induced nitric oxide production in RAW264.7 macrophages in vitro and the acute rat lung injury model in vivo were studied. The results showed that the methanol extract of LcL and its fractions within the range of 62.5–250 μg/mL did not induce cytotoxicity (p < 0.001). The ethyl acetate fraction of LcL showed better NO inhibition activity than other fractions. On the other hand, the Lc-EA (62.5, 125, 250 mg/kg) pretreated rats showed a decrease in the pro-inflammatory cytokines (TNF-α, IL-β, IL-6) and inhibited iNOS, COX-2 expression through the NF-κB pathway. These results suggested that L. chinensis exhibited an anti-inflammatory effect through the NF-κB pathways.

Plectranthus amboinicus (Lour.) Spreng. are generally used in Chinese folk medicine for treating fever, cough, mumps, sore throats, and mosquito bite. The current study was therefore carried out to provide vital pharmacological details about the leaf. This study investigated the anti-inflammatory properties of ethanolic extract from Plectranthus amboinicus. The leaves of Plectranthus amboinicus were rinsed, air dried and then powdered using machine. The ethanol extract of the plant was used for phytochemical analysis to identify the constituents present in the plants. HPTLC analysis was carried out to investigate its secondary metabolite profile by using the solvent mixture. The anti-inflammatory effect was analysed by inhibition of protein denaturation assay. The hypotonicity induced haemolysis was observed from 100µg/ml to 500µg/ml of extract concentration. The results proved that the ethanolic leaf extract of P.amboinicus has antiinflammatory activity. The presence of flavonoid, quercetin in plant extract was confirmed by HPTLC analysis and these flavonoids may be responsible for the antiinflammatory activity. Further studies are needed to the development of potential drug that may be used for various pharmacological activities.

Ximenia americana or “ameixa-do-sertão” is a plant used in the folk medicine of Northeast-Brazil in inflammatory and painful processes. The literature has shown anti-inflammatory and antinociceptive activities for the aqueous or hydroalcoholic extracts and polysaccharide fractions of X. americana stem barks. This study investigated the anti-inflammatory effect of polysaccharide fractions and tea prepared from X. americana stem barks in mice models of paw edema and peritonitis induced by zymosan. Tea (50,100, 150 mg/kg; p.o.) and polysaccharide fractions (FI, FII: 1 mg/kg; i.v.), containing carbohydrate (44.8, 25.0, 44.4%) and polyphenols (28.3, 0.78, 1.50%), respectively, were administered 1 h before zymosan (500 mg; s.c. or i.p). FT-IR analysis of the tea showed bands indicating C=O and C-O groups vibration, typical of phenolic compounds. Zymosan-induced paw edema (AUC: 355.0 ± 38.1) was inhibited by the tea by 38% (100 mg/kg) and 36% (150 mg/kg). FI and FII reduced the edema by 47% (933.8 ± 158.1 AUC) and 30% (1263 ± 124.8 AUC), respectively. Zymosan i.p. induced hypernociception (4.7 ± 0.3 g) and increased the peritoneal leukocyte migration (10979 ± 1475 cells/ml). FII increased by 66% the nociceptive threshold (13.87 ±1.59 g) and reduced by 63% leukocyte migration (3993 ± 439.1 cells/ml). FII decreased plasma protein by 64% (92.6 ± 4.5 vs. zymosan: 119.6 ± 5.4 mg/ml) and MDA by 63% (57.5 ± 3.5 vs. zymosan: 88.9 ± 6.3 mg/ml). The tea, containing polysaccharides and polyphenols, and polysaccharide fractions FI/FII of X. americana barks possess antiinflammatory effect in mice models of acute inflammation induced by zymosan.
