GENETIC DIFFERENCES IN THE CORTISOL CONCENTRATION OF THE ADRENAL AND OTHER TISSUES OF WHITE AND COLOURED GUINEA-PIGS

SUMMARY The effect of genetic difference (white v. coloured) on the cortisol content of ten different organs and tissues, including the adrenals, of white and coloured guinea-pigs of both sexes and of different ages was studied by protein-binding techniques. Tissue cortisol concentration was significantly higher in all the tissues and adrenals of white animals of both sexes compared with coloured animals. The difference in tissue cortisol content between white and coloured animals was higher before (range 41·5–134·%) than after puberty (range 10·1–82·5%). The age-dependent decrease of cortisol concentration in different tissues and adrenals in adults was more marked in white guinea-pigs of both sexes (mean 30·6%, range 0·0–71·5%) than in coloured (mean 16·9%, range 0·0–63·1%). In adult animals the liver contained significantly 11·8–36·4%) more cortisol than in prepubertal animals. This pattern was the same in white and coloured guinea-pigs of both sexes.

Abstract Background Glucocorticoids (e.g. cortisol) are associated with variation in social behavior, and previous studies have linked baseline as well as challenge-induced glucocorticoid concentrations to dominance status. It is known that cortisol response to an acute challenge is repeatable and correlates to social behavior in males of many mammal species. However, it is unclear whether these patterns are also consistent for females. The aim of this study was to investigate whether baseline and response cortisol concentrations are repeatable in female guinea pigs (Cavia aperea f. porcellus) and whether dominance rank is stable and correlated to baseline cortisol concentration and/or cortisol responsiveness. Results Our results show that cortisol responsiveness (after 1 h: R = 0.635, 95% CI = 0.229, 0.927; after 2 h: R = 0.764, 95% CI = 0.433, 0.951) and dominance rank (R = 0.709, 95% CI = 0.316, 0.935) of females were significantly repeatable after six weeks but not correlated. Baseline cortisol was not repeatable (R = 0, 95% CI = 0, 0.690) and also did not correlate to dominance rank. Furthermore, the difference in repeatability estimates of baseline and response values was due to high within-individual variance of baseline cortisol concentration; the amount of between-individual variance was similar for baseline cortisol and the two measures of cortisol responsiveness. Conclusions Females occupying different dominance ranks did not have long-term differences in cortisol concentrations, and cortisol responsiveness does not seem to be significantly involved in the maintenance of dominance rank. Overall, this study reveals the remarkable stability of cortisol responsiveness and dominance rank in a female rodent, and it remains an open question whether the magnitude of cortisol responsiveness is adaptive in social contexts for females.

Young (12 to 15 day old) mice are approximately as susceptible to the virus of equine encephalomyelitis, Eastern or Western strain, when it is given intraperitoneally as are adult mice when the virus is injected intracerebrally. With this susceptibility by the intraperitoneal route as a basis, the injection of immune serum-virus mixtures intraperitoneally was found to result in protection in dilutions which give rise to infection after intracerebral inoculation. The difference of protective power by the two indicated routes was shown not to depend on the amount of inoculum nor on the age of the intracerebrally injected mice. Incubation at 37°C. for 2½ hours neither increases nor diminishes the protective action of immune serum when the intraperitoneal method is employed. The phenomenon of selective protection in different tissues is elicited by the sera of hyperimmunized mice, guinea pigs, and rabbits and by sera derived from horses infected with the disease in nature or exposed to it by contact. Of four horses recovered from the malady, all showed antibody in their sera; of others exposed by contact, four of nine animals revealed antiviral bodies, when the intraperitoneal technique was employed. These tests on horse sera have pointed to the potential value of this procedure for epidemiological studies. Finally, the reaction itself has significance through its bearing on the mechanism of immunity.

ABSTRACT Guinea pigs, kept on a iodine-sufficient diet, were injected with Na131I and the thyroids excised from 45 seconds to 5 days later. The thyroid tissue was homogenized and separated into a combined nuclear-mitochondrial-microsomal fraction and a supernatant fraction by centrifugation at 140 000 g for one hour. Protein bound 131iodine (PB131I) and free 131iodide were determined in the fractions and the PB131I was analysed for monoiodotyrosine (MIT), diiodotyrosine (DIT) and thyroxine after hydrolysis of PB131I. As early as only 20 minutes after the Na131I-injection almost 100% of the particulate fraction 131I was protein bound. In the supernatant fraction the protein binding was somewhat less rapid and PB131I values above 90% of total supernatant 131I were not found until 3 hours after the injection. In all experiments the total amount of PB131I was higher in the supernatant than in the corresponding particulate fraction. The ratio between supernatant PB131I and pellet PB131I was lower in experiments up to 3 minutes and from 2 to 5 days than in experiments of 6 minutes to 20 hours. Hydrolysis of PB131I yielded, even in the shortest experiments, both MIT and DIT. The DIT/MIT ratio was lower in the experiments up to 2 hours than in those of 3 hours and over.

This study's purpose was to determine if a correlation exists between cochlear antioxidant activity changes and auditory function after induction of aminoglycoside (AG) ototoxicity. Two groups of five 250-350 g albino guinea pigs served as subjects. For 28 days, albino guinea pigs were administered either 200 mg/kg/day amikacin, or saline subcutaneously. Auditory brainstem response testing was performed prior to the first injection and again before sacrifice, 28 days later. Cochleae were harvested and superoxide dismutase, catalase, glutathione peroxidase, glutathione-S-transferase, glutathione reductase activities and malondialdehyde levels were measured. All antioxidant enzymes had significantly lower activity in the amikacin group (p ≤ 0.05) than in the control group. The difference in cochlear antioxidant enzyme activity between groups inversely correlated significantly with the change in ABR thresholds. The greatest correlation was for the high frequencies, which are most affected by aminoglycosides. This study demonstrates that antioxidant enzyme activity and amikacin-induced hearing loss significantly covary.

Under conditions closely simulating the natural modes of tuberculous infection in man normal guinea pigs have acquired tuberculosis by being exposed under two degrees of crowding to tuberculous cage mates in ordinary cages, where the food became soiled with excreta, bearing tubercle bacilli, and in special cages, with wire-mesh floors, where this source of infection was almost entirely eliminated. Guinea pigs were also exposed in the same room but not in the same cage with tuberculous animals. It was found that the relative tuberculous involvement of the mesenteric and tracheobronchial nodes showed a gradation of change from an almost completely alimentary infection to a completely respiratory infection. The disease involved the mesenteric nodes predominantly in the crowded ordinary cages, with much less or no affection of the tracheobronchial nodes. It was similarly, but less markedly, enteric in origin in the less crowded ordinary cages, the mesenteric nodes again being larger than the tracheobronchial nodes, but the difference in size was not so great. In the more crowded special cages the relative affection of these two groups of nodes alternated, so that in some the mesenteric, in some the tracheobronchial nodes were more extensively tuberculous. A disease characterized by less or no affection of the mesenteric nodes and by extensive lesions of the tracheobronchial nodes was seen in the less crowded special cages. Finally there was a massive tuberculosis of the tracheobronchial nodes with usually no affection of the mesenteric nodes in the frankly air-borne tuberculosis acquired by guinea pigs exposed in the same room but not to tuberculous cage mates. This gradation in the rô1e played by the enteric and respiratory routes of infection, as first the one and then the other becomes the more frequent channel of entrance for tuberculosis, would indicate that the penetration of tubercle bacilli by the one portal of entry inhibits the engrafting of tuberculosis in the tissues by way of the other portal of entry. It is apparent that in the special cages the opportunities for inhaling tubercle bacilli are at most equal to if not much less than in the ordinary cages; for in the latter dust from the bedding, laden with tubercle bacilli, is stirred up almost constantly by the animals, whereas in the special cages there is no bedding at all, and therefore, presumably, no more tubercle bacilli in the air than may occur in any part of the room. Nevertheless the route of infection was predominantly the respiratory tract in the special cages, especially in the less crowded, apparently because the enteric route had been largely eliminated. The greater predominance of the respiratory route amongst guinea pigs that acquired tuberculosis in the less crowded ordinary cages as compared to the lesser significance of this route in the more crowded ordinary cages would point in the same direction. These observations are in harmony with our knowledge that tuberculosis once implanted in an organism confers a certain degree of immunity to the disease. It is noteworthy that in a study of human autopsy material Opie (3) has found that when healed lesions are present in the mesentery focal tuberculosis in the lungs is seldom found, and that when first infection occurs by way of the lungs it tends to prevent the engrafting of the disease by way of the intestinal tract.

This study examined longitudinal changes in the pulmonary oxygen uptake (pV̇O2) kinetic response to heavy-intensity exercise in 14–16 yr old boys. Fourteen healthy boys (age 14.1 ± 0.2 yr) completed exercise testing on two occasions with a 2-yr interval. Each participant completed a minimum of three ‘step’ exercise transitions, from unloaded pedalling to a constant work rate corresponding to 40% of the difference between the pV̇O2 at the gas exchange threshold and peak pV̇O2 (40% Δ). Over the 2-yr period a significant increase in the phase II time constant (25 ± 5 vs. 30 ± 5 s; p = .002, ω2 = 0.34), the relative amplitude of the pV̇O2 slow component (9 ± 5 vs. 13 ± 4%; p = .036, ω2 = 0.14) and the pV̇O2 gain at end-exercise (11.6 ± 0.6 vs. 12.4 ± 0.7 mL·min−1·W−1; p < .001, ω2 = 0.42) were observed. These data indicate that the control of oxidative phosphorylation in response to heavy-intensity cycling exercise is age-dependent in teenage boys.

The gastrointestinal absorption of Fe and Co by rats and guinea pigs of different ages was measured by comparing the whole-body retention of 59Fe and 57Co after oral and intraperitoneal administrations. The age-groups studied included newborn, weanlings and adults. The absorption of both Fe and Co decreased markedly with age in both rats and guinea pigs. In the rat, absorption remained ele vated during the suckling period, while in the guinea pig absorption decreased markedly during suckling. In both species, Fe and Co absorption were similar, and remained elevated above adult values for some time after weaning. The generally greater absorption of Fe and Co by rats than by guinea pigs and the longer duration of maximal absorp tion in the rats may have involved differences in Fe status in the two species and differences in the timing of gut mat uration.

In a paper which was read before the Institute of Actuaries on the 31st May, 1852, “On the Uniform Action of the Human Will, as exhibited by its Mean Results in Social Statistics,” I drew attention to the remarkable regularity with which marriages are contracted in any country, and the very small limits of difference from the average number which appear from year to year. The observations made by M. Quetelet in Belgium, from 1825 to 1845, showed that the extreme difference in the total number of marriages was little more than half the difference of the extremes in the number of deaths in the same period. Such a conclusion seemed to imply that the subject was worthy of more research. If the law of mortality can be so accurately defined at different ages, that pecuniary interests, amounting to some hundreds of millions sterling, can be valued and adjusted with the greatest nicety, it is reasonable to conclude that the labour of a statistical inquiry into the proportion of marriages at different ages would be rewarded with the discovery of some equally defined law, since the variations from year to year in a given number of facts appear, from a large number of observations, to be even less than in the former case. It is true that, as human life must fail at some time, from the natural decay of the powers of life, every interval of age after man has once attained maturity may be expected, under ordinary circumstances, to show a steady and progressive increase in the liability to disease and death. On the other hand, it may be, argued that marriage is the exercise of the free will of man—that consequently, it does not depend on the age or period of life, but on the arbitrary exertion of those feelings or mental and moral qualities which are not subject to natural laws, or at least not to such laws as we are able to express numerically in the same manner we can the law of mortality in any given population. If we consider, however, marriage as, in one sense, the natural provision for the preservation or increase of the species, and the counteraction to the law of mortality by which the species would perish, we should not be surprised to find that, however imperceptibly to individuals, there is a tendency to obey some unknown law of nature which at the period of maturity would lead to the maximum of marriages, and gradually diminish with age in the same manner as the tendency to disease and death increases with age. The motives and caprices of individuals would only have the same effect on the general results which the different habits of individuals may have in increasing or diminishing the rate of mortality. Accordingly, M. Quetelet, in a comparison of the number of marriages in Belgium for each five years of age after 21, for both sexes, for a period of five years consecutively, showed that the average results in each period scarcely differed at all from year to year. The table is so remarkable, that I have reduced the proportion to 100 of the total marriages in each year, and present it to show the small differences which will then be seen to prevail.

Aggregation tests were performed in 345 patients with peripheral occlusive arterial disease (PAD), 274 patients with coronary heart disease (CHD), and 502 diabetics (D). 263 healthy volunteers (N) served as controls. As in previous studies an age dependent increase of strongly enhanced spontaneous platelet aggregation in obviously healthy subjects can be demonstrated, reaching 28% in the 50-59 years’age group. In the various patient groups the incidence of enhanced aggregation is much higher throughout all age groups than in the control s.The overall incidence of missing spontaneous platelet aggregation vs. strongly enhanced aggregation is summarized below. The difference between normals and the various patient groups is significant (X2-test, α<0.0001).A prospective investigation has been started to establish whether enhanced spontaneous platelet aggregation is a risk factor and an early indicator of progressive atherosclerosis.

Abstract By using of the double antibody sandwich method of ELISA, the activities of five cytokines including IL-2, IL-4, IFN-α, IFN-β and TNF-α from the blood serum, liver, intestine and spleen at two developmental stages of Chinese giant salamander (Andrias davidianus) were determined to analyze the distribution of the cytokines. The result indicated that five cytokines were found in these four tissues, while their activities were different in different tissues and different ages. The highest activity of IL-2 and IL-4 was all present in blood serum of two different ages. The activity of IFN-α was the highest in blood serum of 1-year-old and in spleen of 2-year-old, respectively. The activity of IFN-β was also highest in blood serum of two different ages. The activity of TNF-α was highest in liver of two different ages. Thus, this study provides convincing reference for blood serum and liver as the most important distribution area of Chinese giant salamander.
