Os óleos essenciais são produtos do metabolismo secundário de plantas e são conhecidos por possuir diferentes propriedades biológicas, incluindo atividades antimicrobianas, podendo agir como antibacteriano, antifúngico e antiviral. Objetivou-se nesta pesquisa avaliar a atividade antibacteriana de óleos essenciais sobre cepas de Escherichia coli e Pseudomonas aeruginosa isoladas de alimentos. Foram utilizados os óleos de Eucalyptus globolus (eucalipto comum), Prunus amygdalus (amêndoa), Cymbopongon nardus (citronela), Rosmarinus officinalis (alecrim), Cinnamomum zeylanicum (caneleira), Cymbopogon citratus (capim limão), Citrus limon (limão), Caryophyllus aromaticus (cravo). Foram utilizadas dez linhagens de E. coli e dez de P. aeuriginosa. Para determinação da Concentração Inibitória Mínima (CIM) dos óleos essenciais foi utilizado o método de microdiluição em placas de 96 poços. As concentrações bactericidas mínimas (CBM) foram determinadas a partir dos resultados da CIM. Designou-se como CBM a concentração mínima em que não ocorreu crescimento bacteriano. Verificou-se  que todos os óleos essenciais apresentaram atividade antibacteriana, no entanto os óleos de E. gobulus e R. officinalis foram mais ativos frente a E. coli (CBM=3,13%), e menos eficazes frente a P. aeruginosa (CBM=25%), enquanto que o de C. nardus apresentou atividade biológica frente a P. aeruginosa na concentração de 6,25%. A atividade antimicrobiana dos óleos essenciais testados aponta a possibilidade de desenvolver agentes antimicrobianos eficientes e de baixo custo no controle de E. coli e P. aeruginosa.  Palavras-chave: Escherichia coli.  Pseudomonas aeruginosa. The objective of this research was to evaluate the essential oils antibacterial activityon strains of Escherichia coli and Pseudomonas aeruginosa isolated from food. Essential oils Eucalyptus globolus, Prunus amygdalus, Cymbopongon nardus, Rosmarinus officinalis, Cinnamomum zeylanicum, Cymbopogon citratus, Citrus limon, Caryophyllus aromaticus wereused. Ten strains of E. coli and ten of P. aeuriginosa were used. To determine the Minimum Inhibitory Concentration (MIC) of the essentialoils, the 96-well plate microdilution method was used. Minimum bactericidal concentrations (MBC) were determined from MIC results. CBM was the minimum concentration at which no bacterial growth occurred. It was verified that all the essential oils presented antibacterial activity, however the oils of Eucaliptus gobulus and Rosmarinus officinalis were more active against E. coli (MBC = 3.13%), and less effective against P. aeruginosa (CBM = 25 %), while that of Cymbopongon nardus showed biological activity against P. aeruginosa at 6.25% concentration. The antimicrobial activity of the tested essential oils indicates the possibility of developing efficient and low cost antimicrobial agents in the control of E. coli and P. aeruginosa.Keywords: Escherichia coli. The present study was carried out to determine, for the first time, the chemical composition and antibacterial activity of essential oils derived from the aerial parts of three aromatic plants Thymus algeriensis Boiss & Reut, Rosmarinus officinalis L., and Salvia officinalis L. growing under semiarid conditions. The essential oils were chemically analyzed and identified by gas chromatography (GC) and GC/ mass spectrometry (GC/MS) and their antimicrobial activity was individually evaluated against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa using both agar disk diffusion and agar dilution methods. The major constituents of Thymus algeriensis essential oil were identified as camphor (13.62%), 1,8-cineol (6.00%), borneol (5.74%), viridiflorol (4.00%), and linalool (3.93%). For Rosmarinus officinalis essential oil, 48 compounds were characterized, of which the main constituents were camphor (17.09%), Z-β-ocimene (10.88%), isoborneol (9.68%), α-bisabolol (7.89%), and borneol (5.11%). While, Salvia officinalis essential oil was characterized by β-thujone (16.44%), followed by viridiflorol (10.93%), camphor (8.99%), 1,8-cineol (8.11%), trans-caryophyllene (5.85%), and α-humulene (4.69%) as the major components. Notably, results from antibacterial screening indicated that Thymus algeriensis and Salvia officinalis essential oils exhibited a strong inhibitory effect against both Escherichia coli and Staphylococcus aureus compared to Rosmarinus officinalis essential oil. Further, less activity was recorded against Pseudomonas aeruginosa for the three tested essential oils.

Patógenos alimentares resistentes a antimicrobianos são uma preocupação de saúde pública ao redor do mundo. A resistência a antibióticos está sendo cada vez mais comum entre cepas isoladas de alimentos, assim, alternativas aos antibióticos estão sendo propostas. Óleos essenciais vêm sendo estudados para a aplicação na indústria de alimentos por possuírem atividade antimicrobiana e antioxidante. Desse modo, este trabalho teve como objetivo identificar o perfil de resistência antimicrobiana de Salmonella spp., Staphylococcus aureus e Escherichia coli isolados de frangos empregando diferentes antibióticos e os óleos essenciais de alecrim (Rosmarinus officinalis), capim-limão (Cymbopogon citratus) e pimenta preta (Piper nigrum). Métodos convencionais de microbiologia foram utilizados para a obtenção dos isolados bacterianos, método de disco-difusão foi empregado para identificar a resistência a antibióticos, ensaios de microplaca de resazurina foram realizados para identificar a concentração inibitória mínima (CIM) de óleos essenciais, e então a concentração bactericida mínima (CBM) foi estabelecida a partir da CIM desenvolvendo a técnica da micro-gota. Frequência, média, desvio-padrão, análise de variância e teste de Tukey foram calculados para a análise dos resultados. Destacam-se entre os resultados obtidos a frequência de carcaças de frangos contaminadas por Escherichia coli, Staphylococcus aureus e Salmonella spp., 70%, 40%, e 25%, respectivamente, sendo que a resistência dos isolados a um ou mais antibióticos foi detectada em 90,9%, 66,6% e 55,6% dos isolados de E. coli, Salmonella spp. e S. aureus, respectivamente. Em adição, a multiresistência foi amplamente identificada. Quanto aos resultados obtidos para as análises de Concentração Inibitória Mínima dos óleos essenciais analisados, foi possível observar um melhor desempenho dos óleos essenciais de alecrim e capim limão, respectivamente, contudo sem diferença significativa entre as amostras. Os resultados reforçam a preocupação com a disseminação de cepas resistentes a antimicrobianos e a necessidade do desenvolvimento ou melhoramento de alternativas ao uso de antibióticos.

Abstract This paper investigates the antimicrobial action of the extracts and essential oil of wildgrowing Tanacetum vulgare L on: Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacilus subtilis, using the diffusion disc method. The essential oils but also the ethanolic extracts tested exhibited moderate action on Staphilococcus aureus and Bacillus subtilis and low action on E. coli and Pseudomonas aeruginosa. The moderate antimicrobial activity is related to the amount of some chemical components of the essential oil of T. vulgare flos. Thus, this paper presents also the quantitative and qualitative analysis of the essential oils of T. vulgare harvested from two different habitats. The essential oils obtained by steam-distillation were analysed by gas-cromatography coupled with mass spectrometry (GC-MS).

The present study examined the volatile composition and antimicrobial and gastrointestinal activity of the essential oils of Elettaria cardamomum (L.) Maton harvested in India (EC-I) and Guatemala (EC-G). Monoterpene were present in higher concentration in EC-I (83.24%) than in EC-G (73.03%), whereas sesquiterpenes were present in higher concentration in EC-G (18.35%) than in EC-I (9.27%). Minimum inhibitory concentrations (MICs) of 0.5 and 0.1 mg/mL were demonstrated against Pseudomonas aeruginosa in EC-I and EC-G, respectively, whereas MICs of 0.125 and 1 mg/mL were demonstrated against Escherichia coli in EC-I and EC-G, respectively. The treatment with control had the highest kill-time potential, whereas the treatment with oils had shorter kill-time. EC-I was found to be more potent in the castor oil-induced diarrhoea model than EC-G. At 100 and 200 mg/kg, EC-I exhibited 40% and 80% protection, respectively, and EC-G exhibited 20% and 60% protection, respectively, in mice, whereas loperamide (positive control) exhibited 100% protection. In the in vitro experiments, EC-I inhibited both carbachol (CCh, 1 µM) and high K+ (80 mM)-induced contractions at significantly lower concentrations than EC-G. Thus, EC-I significantly inhibited P. aeruginosa and E. coli and exhibited more potent antidiarrheal and antispasmodic effects than EC-G.

Los ejemplares de las especies vegetales Cymbopogon citratus (hierba luisa) y Melissa officinalis (toronjil), se recolectaron al azar en la localidad de Machala, Ecuador. Las hojas fueron lavadas, secadas, molidas y extraídas por maceración con metanol; los filtrados concentrados por evaporación a presión reducida. A cada extracto, se le realizaron pruebas fitoquímicas y ensayos biológicos como: actividad antimicrobiana (antibiograma: difusión en agar) y letalidad con Artemia salina. Los extractos de C. citratus y M. officinalis revelaron la presencia de esteroles insaturados, triterpenos pentacíclicos, fenilpropanoides y catequinas. Mientras que en el de M. officinalis se detectaron, además, cumarinas y metilencetonas. Los extractos de ambas especies (soluciones de 20 y 40 mg/L) mostraron actividad antibacteriana contra las cepas Escherichia coli, Pseudomonas aeruginosa y Staphylococcus aureus, a excepción de los extractos metanólicos de M. officinalis (20 mg/L) y C. citratus (40 mg/L) que no exhibieron actividad frente a E. coli. La especie C. citratus mostró el mayor efecto antibacteriano contra las bacterias P. aeruginosa y S. aureus (halos de inhibición > 15mm). Se observó una actividad antifúngica alta del extracto de C. citratus, y una actividad moderada del extracto de M. officinalis, contra la cepa del hongo Candida albicans. C. citratus y M. officinalis mostraron una actividad letal significativa (CL50 <1000 μg/ml) frente a nauplios de A. salina a las 24 h de exposición: 358,03 y 72,25 μg/ml respectivamente. La especie M. officinalis presentó la mayor letalidad, considerado altamente tóxico según CYTED. A partir de los resultados obtenidos, se puede inferir que las plantas C. citratus y M. officinalis cultivadas en Ecuador son una fuente promisoria de metabolitos secundarios bioactivos con actividad farmacológica (antimicrobianos y citotóxicos).

O Cymbopogon citratus é uma planta originária da Índia, pertencente à família Poaceae e facilmente adaptada em regiões tropicais como o Brasil. Este trabalho avaliou a atividade antimicrobiana do óleo essencial (OE) obtido por hidrodestilação das folhas de C. citratus. A concentração inibitória mínima (CIM) foi determinada pela técnica de microdiluição em caldo para as bactérias Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa e a levedura Candida albicans. O menor CIM foi de 7,81 µg mL-1 para S. aureus, a média de 15,62µg mL-1 para P. aeruginosa e C. albicans e a maior de 62,50µg mL-1 para E. coli. O óleo essencial de C. citratus apresentou atividade antimicrobiana para todos os micro-organismos testados. O óleo mostrou potencial aplicabilidade no controle microbiano e sugerem-se novos estudos para auxiliar na ampliação das aplicações desta planta.

Мета роботи — вивчити резистентність до антибіотиків бактеріальних збудників інфекцій сечових шляхів (ІСШ), виділених у пацієнтів урологічного стаціонару в м. Києві. Матеріали і методи. Досліджено 1612 штамів бактерій, виділених із сечі хворих з ІСШ (цистит, уретрит, пієлонефрит), госпіталізованих в урологічне відділення ДУ «Інститут урології НАМН України» у м. Києві протягом 2016 р. Серед пацієнтів переважали жінки — 1201 (74,5 %). Вік хворих становив від 17 до 74 років. Для збору даних використано медичну документацію лікарні. Мікробіологічні дослідження виконано у лабораторії мікробіології ДУ «Інститут урології НАМН України». Аналізували результати культурального дослідження зразків сечі, зібраних за наявності клінічних ознак ІСШ. Дослідження клінічного матеріалу та інтерпретацію отриманих результатів проводили загальноприйнятими методами. Вивчено чутливість уропатогенів до 31 антибіотика дискодифузійним методом відповідно до рекомендацій Інституту клінічних та лабораторних стандартів США (Clinical and Laboratory Standards Institute (CLSI)). Результати та обговорення. Аналіз мікробного спектра сечі виявив домінування серед уропатогенів штамів Escherichia coli (32,0 %), Enterococcus faecalis (19,5 %), Klebsiella pneumoniae (10,9 %), Staphylococcus epidermidis (8,9 %), S. haemolyticus (6,5 %) та Pseudomonas aeruginosa (6,4 %). Частка Enterococcus faecium, Enterobacter aerogenes і Streptococcus viridans становила відповідно 2,5, 2,2 і 1,6 %, Enterobacter cloacae, Klebsiella oxytoca, Acinetobacter baumannii, Proteus vulgaris та Providencia rettgeri — менше 1,0 %. У більшості випадків (69,7 %) мікроорганізми виділено у монокультурі, у решті випадків — у мікробних асоціа- ціях. Високу резистентність до тестованих антибіотиків виявили штами E. aerogenes (45,1 %), E. cloacae (45,7 %), E. faecium (40,9 %), E. faecalis (40,7 %), E. coli (39,9 %), P. aeruginosa (34,0 %), K. pneumoniae (28,6 %). Найбільш активними до уропатогенів були іміпенем (E. coli — 87,6 %, P. aeruginosa — 75,7 %, E. cloacae — 67,3 %, E. aerogenes — 72,6 %, K. pneumoniae — 93,2 %), меропенем (E. coli — 89,1 %, P. aeruginosa — 76,7 %, K. pneumoniae — 82,6 %), лефлоцин (E. coli — 74,5 %, ентерококи — 78,7 %, P. aeruginosa — 76,7 %, E. cloacae — 73,9 %, E. aerogenes — 80,4 %, K. pneumoniae — 83,5 %), амоксицилін/клавуланат (ентерококи — 84,6 %), фурагін (ентерококи — 82,6 %), цефоперазон (K. pneumoniae — 89,2 %, P. aeruginosa — 73,8 %), цефтріаксон (K. pneumoniae — 80,1 %). Висновки. Антибіотикорезистентність збудників ІСШ — важлива терапевтична проблема. Найбільшою активністю до уропатогенів характеризуються іміпенем, меропенем, лефлоцин, амоксицилін/ клавуланат, фурагін, цефоперазон, цефтріаксон, які можна розглядати як препарат вибору для призначення стартової терапії ІСШ. Необхідно здійснювати постійний моніторинг за резистентністю до дії антибіотиків. Політику використання антибіотиків у кожному стаціонарі слід визначати залежно від локальних даних щодо резистентності до протимікробних препаратів.

Background: Escherichia coli O157:H7 is one of the most common causes of contamination in Lighvan cheese processing. Using from natural antimicrobial essential oils is applied method to decrease the rate of microbial contamination of dairy products. The present investigation was done to study the antimicrobial effects of Z. multiflora and O. basilicum essential oils on survival of E. coli O157:H7 during ripening of traditional Lighvan cheese. Methods: Leaves of the Z. multiflora and O. basilicum plants were subjected to the Clevenger apparatus. Concentrations of 0, 100 and 200 ppm of the Z. multiflora and 0, 50 and 100 ppm of O. basilicum essential oils and also 103 and 105 cfu/ml numbers of E. coli O157:H7 were used. The numbers of the E. coli O157:H7 bacteria were analyzed during the days 0, 30, 60 and 90 of the ripening period. Results: Z. multiflora and O. basilicum essential oils had considerable antimicrobial effects against E. coli O157:H7. Using the essential oils caused decrease in the numbers of E. coli O157:H7 bacteria in 90th days of ripening (P <0.05). Using from Z. multiflora at concentration of 200 ppm can reduce the survival of E. coli O157:H7 in Lighvan cheese. Conclusion: Using Z. multiflora and O. basilicum essential oils as good antimicrobial agents can reduce the risk of foodborne bacteria and especially E. coli O157:H7 in food products.

ABSTRACT The use of bacterial transposon mutant libraries in phenotypic screens is a well-established technique for determining which genes are essential or advantageous for growth in conditions of interest. Standard, inactivating, transposon libraries cannot give direct information about genes whose over-expression gives a selective advantage. We report the development of a system wherein outward-oriented promoters are included in mini-transposons, generation of transposon mutant libraries in Escherichia coli and Pseudomonas aeruginosa and their use to probe genes important for growth under selection with the antimicrobial fosfomycin, and a recently-developed leucyl-tRNA synthase inhibitor. In addition to the identification of known mechanisms of action and resistance, we identify the carbon–phosphorous lyase complex as a potential resistance liability for fosfomycin in E. coli and P. aeruginosa. The use of this technology can facilitate the development of novel mechanism-of-action antimicrobials that are urgently required to combat the increasing threat worldwide from antimicrobial-resistant pathogenic bacteria.

A novel AgBr/Ag2MoO4@InVO4 composite photocatalyst with different heterojunction structures was successfully constructed by compounding InVO4 with Ag2MoO4 and AgBr. According to the degradation, antibacterial and free radical trapping data, the photocatalytic antibacterial and antifouling activities of AgBr/Ag2MoO4@InVO4 composite were evaluated, and the corresponding photocatalytic reaction mechanism was proposed. Adding AgBr/Ag2MoO4@InVO4 composite, the degradation rate of ciprofloxacin (CIP) achieved 95.5% within 120 min. At the same time, the antibacterial rates of Escherichia coli (E. coli), Staphylococcus aureus (S. aureus) and Pseudomonas aeruginosa (P. aeruginosa) achieved 99.99%. The AgBr/Ag2MoO4@InVO4 composite photocatalyst showed promising usage in photocatalytic antibacterial and purification areas.
