This study was carried out in the Tissue Culture Laboratory and the Experiment Field of the Dept. of Horticulture, Assiut Univ. Two garlic cultivars i.e., El-Balady (local cultivar) and the Chinese were used. Two media, MS and B5, were supplemented with 4%, 8%, 12% and 16% sucrose. The 8% sucrose gave best number of roots, root length and leaf length as compared to the other sucrose concentrations used in this study. Four percent or 16% sucrose gave the lowest values of these characters. The MS gave higher values in most of the studied characters than that of the B5. In another experiment three growth regulators i.e. IBA, BA and GA3 or used at 0, 5, 10, 20 ppm in two media, MS and B5 supplemented with 3% sucrose. All the studied growth regulators gave higher values than the control in respect of the all studied characters. The IBA was better than BA and GA3 in root formation, while GA3 was better than BA and IBA in leaf formation. It is found that 5 ppm of IBA formed the highest number of roots, root length and percentage of root formation. On the other hand, the 20 ppm of GA3 gave the highest number of leaves and leaf length. The garlic plantlets were successfully acclimatized in the laboratory giving 67.8% survival for the Balady cv. and about 62.6% for Chinese cv. The survival rate of these plantlets were 74.5% for the Balady cv. and 78.5% for the Chinese cv. on transfer to the open-field. No chromosomal aberrations was noticed.

To improve the growth of Grammatophyllum scriptum (Lindl.) Bl. in vitro plantlet, a photoautotrophic micropropagation system (PMS) was developed by growing in vitro plantlet on VW medium with varying concentration of sucrose (0, 5, 10, and 20 g/L) and additional carbon dioxide from the air (bottle covered with cap or filter). The result showed that the leaf length would increase up to 6.5 cm with PMS and it would keep growing by the adding of 5 g/L sucrose. Average number of leaves increased by 6.7 strands with PMS and the addition of sucrose increased the average quantity of leaves up to 7.7 strands. Average number and root length would increase with PMS and would even increase more with 5 g/L sucrose addition. PMS with 5 g/L sucrose can increase chlorophyll a and b concentration. The number of stomata per unit area in PMS was lower than closed culture. This shows that PMS can increase the growth of G. scriptum in vitro plantlet and the growth increase would be effective if it is combined with sucrose addition.

In vitro cultivation is a highly important biotechnological method widely used for the production of orchid seedlings, but it is necessary to study the suitability of the nutrients used in different kinds of formulation, as the nutritional requirement varies according to the species. The objective was to evaluate different concentrations of iron in the in vitro cultivation of Schomburgkia crispa Lindl seedlings. Seedlings were obtained from seeds germinated in vitro. Modified MS culture medium was used with half of the macronutrient concentration. The micronutrients were added according to the original formulation, except for the iron which was added from a stock solution of FeEDTA (FeSO4.7H2O: 5.6 g L-1 and EDTA: 7.48 g L-1) at 0.0; 2.5; 5.0; 7.5; 10.0 and 12.5mL L-1. At 200 days after seedling transplantation, shoot height, root length, number of leaves, shoot number, leaf length, leaf width, aerial and root dry mass, chlorophyll a, b and carotenoids content were evaluated. A completely randomized design was used, with six treatments and ten replicates of five seedlings. Regression analysis was performed at 5% of significance. The increase in iron concentration caused a reduction in root length and an increase in the number of leaves and shoots. The concentration of 4.13 mL L-1 of FeEDTA was the one that provided the best in vitro growth of S. crispa plants. High concentrations of iron caused a reduction of initial development, but stimulated an increase in the number of shoots.

The experiment was conducted to investigate the combined effect of different plant growth regulators with and without charcoal supplementation for root formation and plantlet development from protocorm like bodies (PLBS) of orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations were studied. It revealed that the highest number of roots was obtained from 1.0 mg/L each of IAA + IBA combination (6.667) and the highest root length was recorded from 2.0 mg/L BAP + 1.0 mg/L IBA with charcoal supplementation. The treatment combinations, 1.0 mg/L each of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA were found best for producing more rooted plantlets with charcoal supplementation. The study was conducted to mass micropropagation of big sage (Lantana camara L.) plant by shoot multiplication technique. The treatments 2.22 and 2.66 µmol·L–1 BA gave the highest significant increase in the percentage of response to shoot multiplication and number of shoots per explant compared to the other treatments as reached 96.70% and 100.00% and 4.33 and 6.00 shoots, respectively. The results showed that these two treatments did not differ significantly between them. While the 1.33 µmol·L–1 BA gave the lowest values in the percentage of response to shoot multiplication and number of shoots per explant were 80.00% and 2.00 shoots per explant, respectively. The MS medium supplemented with 4.30 or 5.37 µmol·L–1 NAA gave a high response to root formation, number of roots per shoot and root length. While the MS medium supplemented with 6.44 or 7.52 µmol·L–1 NAA gave low values in these characteristics. The MS medium with 2.22 or 2.66 µmol·L–1 concentration of BA or 7.52 µmol·L–1 concentration of NAA recorded the highest significant increase in the percentage of response to callus formation. While the MS medium supplemented with 1.33 µmol·L–1 BA or 4.30 µmol·L–1 NAA gave less response to the callus formation.

The in vitro organogenesis of embryos of type-variety (‘Albescens’, ‘Nigrescens’, ‘Virescens’ and ‘Idolatrica’) of the oil palm (Elaeis guineensis Jacq.) with growth regulators was studied. Murashige and Skoog (MS) medium, supplemented with nine treatment concentrations of Naphthalene acetic acid (NAA) and 6-Benzylaminopurine (BAP), alone and in combinations, was used to initiate embryo cultures of the type-variety. Data were collected on time of root production, root length, plant height, number of leaves and number of roots.  The results obtained indicated that there were swelling and expansion of the matured zygotic explants in cultures within 7 and 14 days after culture initiation (DCI). Time of root production did not vary significantly among type-variety, but differed significantly (p < 0.001) due to growth regulators, whereas their interaction effect was non-significant. Root length and plantlet height significantly varied (p < 0.05) due to type-variety and growth regulators, as well as their interaction. There were no significant variations among type-variety on number of leaves and roots produced at 84 DCI, but there was significant (p < 0.05) variations on number of leaves and non-significant variance on number of roots due to growth regulators. The type-variety combined with growth regulators effects on number of leaves and roots were not significant.  In the present study, the responses of the various type-variety in the regeneration of plantlets in vitro varied with respect to media supplementation and that low concentration of NAA and BAP, alone and in different combinations, favoured root, leaf and plantlet production in MS medium.

Background: The present investigation was carried out during the cropping season of year 2019-20 to find out the most suitable growing condition as well as variety of Asiatic lily for the production of bulbs/bulblets under Hisar (Haryana) conditions. Methods: The experiment was laid out in RBD (factorial) with three replications and twelve treatment combinations, comprising of three growing conditions (polyhouse of 200 µ, green shade-net of 50% shade and open field) and four varieties (Courier White, Tresor, Nova Lux and Red Stone). Result: These varieties of Asiatic lily when grown under polyhouse performed better in some parameters resulting in early bulb sprouting, maximum plant height and number of leaves/plant and the minimum days upto harvesting of bulbs, however, in other parameters viz. leaf length and width, weight of bulbs/plant, diameter of bulb and number of bulblets/plant, the shade-net condition was observed to be better than polyhouse. Overall, Tresor performed better, resulting in early bulb sprouting (5.11 days), minimum days (236.56) upto harvesting of bulbs and the maximum plant height (89.01 cm), number of leaves/plant (64.08), leaf length (9.50 cm), diameter of bulb (3.91 cm) and number of bulblets/plant (2.99) and it was followed by Red Stone which produced maximum weight of bulbs/plant (59.03 g) and observed next to Tresor in most of the parameters. Tresor grown under polyhouse took minimum days (231) upto the harvesting of bulbs, produced maximum plant height (115.13 cm) and number of leaves/plant (69.67) and it was closely followed by Red Stone grown under same condition. Further, under the shade-net, Red Stone produced maximum weight of bulbs/plant (65.30 g) followed by Tresor (63.93 g), while the maximum number of bulblets/plant (3.90) were produced by Tresor followed by Red Stone (3.44). Henceforth, Tresor and Red Stone when grown under polyhouse performed better in vegetative parameters and when grown under shade-net performed better in bulb parameters than the other combinations of growing conditions and varieties.

Background: Rooting of pineapple from Sipahutar in vitro is an alternative to obtain superior seeds in large quantities to meet the needs of farmers and consumers.Objective: The purpose of this study was to investigate the effect of Myo-inositol and IBA on pineapple rooting (Ananas comosus L.) from Sipahutar in vitro.Methods: The research method used a completely randomized factorial design with two factors: Factor I: Myo-inositol consisted of [4] treatment concentrations: M0 = 0 g/l, M0.02 = 0.02 g/l, M0.04 = 0.04 gr/l, M0.06 = 0.06 gr/l. Factor II: IBA growth regulators consisted of [3] treatment concentrations: IB0 = 0 ppm, IB1.5 = 1.5 ppm, IB3 = 3 ppm. The parameters observed were time of root emergence, number of roots, number of leaves, time of leaf emergence, number of tillers, and time of its emergence observed each week, while the height of tillers, shoots, and whole shoots, root length, leaf width and leaf length were observed 12 weeks after planting (WAP).Results: The result of the research showed that root time appeared on [3] WAP. The increasing number of leaves and timing of the puppiesin 1 WAP. Myo-inositol had significant effects on root counts in the 0.06 gr/l and root length on Myo-inositol treatment in the 0.04 g/l. IBA did not have a real effect on the entire observation parameters. The interactions of Myo-inositol and IBA had significant effects on root number, number of leaves, leaf length and tiller height.Conclusions: Myo-inositol and IBA had significant effects on root number, leaf number, leaf length and shoot height of pineapple’s explant from Sipahutar.

Abstract. This research was conducted to determine the effect of concentration of Ethyl Methyl Sulfonate (EMS) to performance of three types of Phalaenopsis Planlets and to know the optimum concentration of EMS on any type of Phalaenopsis orchids. The experiment was conduc-ted at Tissue Culture Laboratory of the Faculty of Agriculture, Winaya Mukti University, Sumedang. It used a completely randomized design (CRD) with factorial pattern, that consisted of two factors and replicated twice.The first factor was the type of Phalaenopsis that resulted form hybridization which consisted of v1 (Phalaenopsis 717 X Phalae-nopsis Fire Bird), v2 (Phalaenopsis Tianong Rose X Sibling), and v3 (Phalaenopsis Luchia Pink X Phalae-nopsis Chain Xen Mammon). The second factor was the concentration of EMS that consisted of e0 (0% EMS/control), e1 (0,05% EMS), e2 (0,10% EMS), e3 (0,15% EMS), e4 (0,20% EMS), and e5 (0,25% EMS). Explant protocorm of three types of Phalae-nopsis soaked in a solution of EMS by each treat-ment for 3 hours, and cultured on MS medium Basal Modified Multiplication Shoot for 10 weeks. The experimental results showed that there are interaction between the three types of Phalae-nopsis result of a cross with a concentration of EMS to variable number of roots. Orchids P. 717 X P. Fire Bird had higher  number of leaves, number of roots, leaf leang, and root length than the other. The result showed that there were interaction between three species of Phalaenopsis orchid from the crossing with EMS concentration on root variables. Orchid P. 717 X P. Fire Bird has the number of leaves, the number of roots, the number of shoots, leaf length and root length better than other types. EMS concentrations independently at all levels of treatment can not increase the number of leaves, the number of roots, fresh weight of planlet, leaf length, and root length.Keywords: Phalaenopsis, the concentration of EMS, in vitro cultureSari. Penelitian ini mempelajari dan mengetahui pengaruh konsentrasi Ethyl Methyl Sulfonate (EMS) terhadap keragaan planlet tiga jenis anggrek Phalaenopsis asal protocorm dan mencari konsentrasi optimum EMS untuk setiap jenis anggrek Phalae-nopsis secara in vitro.Percobaan dilaksanakan di Laboratorium Kultur Jaringan Fakultas Pertanian Universitas Winaya Mukti Sumedang. Eksperimen menggunakan Rancangan Acak Lengkap (RAL) Pola Faktorial, yang terdiri atas dua faktor dan diulang sebanyak dua kali.Faktor pertama adalah jenis anggrek Phalaenopsis hasil persilangan yang terdiri atas tiga jenis, yaitu v1 (Phalaenopsis 717 X Phalaenopsis Fire Bird), v2 (Phalaenopsis Tianong Rose X Sibling), dan v3 (Phalaenopsis Luchia Pink X Phalaenopsis Chain Xen Mammon). Faktor kedua adalah EMS yang terdiri atas enam taraf perlakuan, yaitu e0 (0% EMS/kontrol), e1 (0,05% EMS), e2 (0,10% EMS), e3 (0,15% EMS), e4 (0,20% EMS), dan e5 (0,25% EMS). Eksplan berupa protocorm dari tiga jenis anggrek Phalaenopsis hasil persilangan yang direndam dalam larutan EMS berdasarkan masing-masing perlakuan selama 3 jam, dan dikulturkan pada media MS Modified Multiplication Shoot Basal selama 10 minggu.Hasil penelitian menunjukkan terjadi interaksi antara tiga jenis anggrek Phalaenopsis hasil persilangan dengan konsentrasi EMS terhadap variabel jumlah akar.Anggrek P. 717 X P.Fire Bird memiliki jumlah daun, jumlah akar, jumlah tunas, panjang daun dan panjang akar yang lebih baik dibandingkan jenis lainnya. Konsentrasi EMS secara mandiri pada semua taraf perlakuan tidak dapat meningkatkan jumlah daun, jumlah akar, bobot segar planlet, panjang daun, dan panjang akar.Kata Kunci: Phalaenopsis, Konsentrasi EMS, dan Budidaya In Vitro

Abstract: The objective of this work was to evaluate how salt concentrations in media used for in vitro cultures affect the early development of “barueiro” (Dipteryx alata) seedlings obtained from the shoot apex. Two different in vitro culture media - Murashige & Skoog (MS) and wood plant medium (WPM) - in different salt concentrations (25, 50, 75, and 100%) were used. At 120 days, the D. alata seedlings in the MS medium, in its original concentration (100%), developed better than in other concentrations or in the WPM. Therefore, the MS medium was considered the most suitable for ensuring in vitro establishment because it provided greater root length (27.65 cm) and number of leaves per plant (26 leaves) than the other medium.

The success in micropropagation of Dendrobium phalaenopsis Deang Suree is high, but when transplanted into the greenhouse, their survival is minimal. To increase survival in production in the present study it was evaluated the effect of intermediate acclimatization for 30 days in a grow room utilizing the following luminosity conditions: 1- white fluorescent light (B) (18.9µmol m-2 s-1); 2- white fluorescent light + red fluorescent light (GRO-LUX(r)) (BV) (14.85µmol m-2 s-1); 3- red fluorescent light (GRO-LUX(r)) (V) (9.45µmol m-2 s-1) and the control plants were accommodated directly in a greenhouse (162.0µmol m-2 s-1). After this the leaves were characterized anatomically and the plants transferred to the control greenhouse. It was evaluated survival percentage and final number of roots, and calculated the relations between the final and initial values of fresh weight, number of leaves, length and diameter of the largest pseudo bulb, number of pseudo bulbs and longest root length. Only plants submitted to red light, were statistically better than the control in relation to the survival percentage and in relation to fresh weight, while the control showed a higher number of roots that plants acclimatized in this luminosity conditions. Intermediate acclimatization, using red light or red + white light, is recommended for D. phalaenopsis Deang Suree.
