The success in micropropagation of Dendrobium phalaenopsis Deang Suree is high, but when transplanted into the greenhouse, their survival is minimal. To increase survival in production in the present study it was evaluated the effect of intermediate acclimatization for 30 days in a grow room utilizing the following luminosity conditions: 1- white fluorescent light (B) (18.9µmol m-2 s-1); 2- white fluorescent light + red fluorescent light (GRO-LUX(r)) (BV) (14.85µmol m-2 s-1); 3- red fluorescent light (GRO-LUX(r)) (V) (9.45µmol m-2 s-1) and the control plants were accommodated directly in a greenhouse (162.0µmol m-2 s-1). After this the leaves were characterized anatomically and the plants transferred to the control greenhouse. It was evaluated survival percentage and final number of roots, and calculated the relations between the final and initial values of fresh weight, number of leaves, length and diameter of the largest pseudo bulb, number of pseudo bulbs and longest root length. Only plants submitted to red light, were statistically better than the control in relation to the survival percentage and in relation to fresh weight, while the control showed a higher number of roots that plants acclimatized in this luminosity conditions. Intermediate acclimatization, using red light or red + white light, is recommended for D. phalaenopsis Deang Suree.

The objetive of this study was to evaluate the effects of adding two types of banana pulp, combined with varying concentrations of sacarose on the growth of Cattleya sp. plantlets. Hybrid LCTV-01 seedlings (Cattleya labiata rubra x Cattleya labiata semi alba) made to germinate in vitro were inoculated in an MS culture medium with half the concentration of nutrients and supplemented with 60 g.L-1 'maçã' or 'terra' banana pulp in addition to different concentrations of sacarose (10, 20 and 30 g.L-1. The entirely randomized experimental design was chosen, implemented in seven treatments, ten repetitions and eight seedlings per repetition. After 160 days of in vitro cultivation, variables of fresh weight, number of leaves, number of roots and length of the longest root were evaluated. It was found that the addition of banana pulp of any of the analyzed cultivars promoted better seedling growth. Additionally, the 20 g.L-1 sacarose concentration yielded better results for the analyzed variables.

The effect of white, blue, green, red and UV + white light on the growth and development of shoots and roots of Gerbera jamesonii cv. Queen Rebecca in relation to the presence of kinetin or IAA were investigated. The highest number of axillary shoots was obtained in red and green light on the medium with 5 mg l<sup>-1</sup> kinetin. Also, green and red light markedly increased the number of leaves developed on the plantlets on the medium supplemented with kinetin. Light quality and IAA added to culture medium variously affected the development of root system: roots were regenerated under all light treatments, higher root number was recorded under red light when 5 mg l<sup>-1</sup> IAA was added to the media, the shortest roots were found in red light on the medium supplemented with IAA. The greatest fresh weight of shoots was found under white light on the medium with kinetin. Red light markedly decreased shoot fresh weight on hormone-free medium. Blue and white light caused increase in fresh weight of roots.

The aims of this research were to determine the lighting of incubation rooms and browning prevention agents on the growth of “Pisang Raja Bulu” planlets by in vitro and determine whether there is a best interaction between the incubation rooms lighting and the type of browning prevention agents in the growth of “Pisang Raja Bulu” planlets that plants in vitro. This research was conducted in the Biotechnology laboratory of the Faculty of Agriculture, UPN "Veteran" Yogyakarta on June - August 2018. The research method was a laboratory experiment method compiled with the Split Plot Design of two factors. The first factor as a main plot is the incubation room lighting, which is with light for 90 days, without light for 90 days, without light in the first 45 days, and without light in the last 45 days. The second factor as a sub plot is browning prevention agents, named thidiazuron, activated charcoal, and vitamin C. Each combination of treatments was repeated 3 times. The result showed that there was the best combinations of treatments that is all combination of lighting and vitamin C 0,88 mg / l in terms of browning. The first 45 days lighting treatment gave the best result on the percentage of life, plantlet height, number of shoots, number of leaves, root length, and fresh weight. The treatment of vitamin C 0.88 mg / l gave the best result on the percentage of life, plantlet height, number of shoots, root length, number of roots, fresh weight, and dry weight.Keyword: in vitro, lighting, browning, raja bulu banana.

This research was conducted at the plant tissue culture Lab. College of Agriclture, University of Baghdad from February to October 2015. The aim of the study was investigate the  influence of light quality and Naphthalene  Acetic Acid(NAA) on some properties of roots for two varieties Martinque and Yanara shoots of Gerbera jamesonii, cultured on MS medium supplemented with 0.3 mg.L -1 Spermidine. The results indicated that 1/2 MS medium with free auxin NAA gave the highest percentage of rooting (75 %) Which differed significantly from the other concentrations of NAA. White light gave the hightest percentage of rooting (89 %), that was not significantly different from the red  light (78%) . While the treatment of Martinque variety under White light gave the highest percentage of rooting (100%), that was not significantly different from Yanara variety that gave (78%). MS medium with free auxin NAA gave the highest number of roots (7.5 % root/shoot ) that was significantly different on all the other concentrations of NAA. MS medium with free auxin NAA under White light of roots (6.6 root/shoot) at Martinque variety . While MS medium with free auxin NAA under blue light gave the highest number of roots (10.10 roots/shoot) that was not different significantly from red light it gave (8.9 root/shoot) Martinque variety gave the highest rate of root length ( 1.62 cm) Which not differend  significantly from Yanara variety it gave (1.49 cm) While The treatment of Martinque variety under White light with NAA- free medium gave the highest rate of root length (3.40 cm) and was significantly different on all the other interactions

This study was carried out in the Tissue Culture Laboratory and the Experiment Field of the Dept. of Horticulture, Assiut Univ. Two garlic cultivars i.e., El-Balady (local cultivar) and the Chinese were used. Two media, MS and B5, were supplemented with 4%, 8%, 12% and 16% sucrose. The 8% sucrose gave best number of roots, root length and leaf length as compared to the other sucrose concentrations used in this study. Four percent or 16% sucrose gave the lowest values of these characters. The MS gave higher values in most of the studied characters than that of the B5. In another experiment three growth regulators i.e. IBA, BA and GA3 or used at 0, 5, 10, 20 ppm in two media, MS and B5 supplemented with 3% sucrose. All the studied growth regulators gave higher values than the control in respect of the all studied characters. The IBA was better than BA and GA3 in root formation, while GA3 was better than BA and IBA in leaf formation. It is found that 5 ppm of IBA formed the highest number of roots, root length and percentage of root formation. On the other hand, the 20 ppm of GA3 gave the highest number of leaves and leaf length. The garlic plantlets were successfully acclimatized in the laboratory giving 67.8% survival for the Balady cv. and about 62.6% for Chinese cv. The survival rate of these plantlets were 74.5% for the Balady cv. and 78.5% for the Chinese cv. on transfer to the open-field. No chromosomal aberrations was noticed.

The spectrum of light significantly influences the growth of plants cultivated in closed systems. Five lettuce cultivars with different leaf colours were grown under white light (W, 170 μmol m−2 s−1) and under white light with the addition of red (W + R) or blue light (W + B) (230 μmol m−2 s−1). The plants were grown until they reached the seedling phase (30 days). Each cultivar reacted differently to the light spectrum applied. The red-leaved cultivar exhibited the strongest plasticity in response to the spectrum. The blue light stimulated the growth of the leaf surface in all the plants. The red light negatively influenced the length of leaves in the cultivars, but it positively affected their number in red and dark-green lettuce. It also increased the relative chlorophyll content and fresh weight gain in the cultivars containing anthocyanins. When the cultivars were grown under white light, they had longer leaves and higher value of the leaf shape index. The light-green cultivars had a greater fresh weight. Both the addition of blue and red light significantly increased the relative chlorophyll content in the dark-green cultivar. The spectrum enhanced with blue light had positive influence on most of the parameters under analysis in butter lettuce cultivars. These cultivars were also characterised by the highest absorbance of blue light.

The purpose of this research is to determine the concentration of young coconut water that is appropriate for the growth of orchid plantlets (Dendrobium sp.) In vitro. This study used an experimental method, descriptive data analysis to compare several different concentrations of coconut water. The design of this study uses a completely randomized design (CRD). The treatments consist of 0% coconut water concentration (as a control), 15%, 30% and 60%. Each concentration was carried out 5 replications and each repetition consisted of 5 Dendrobium sp plantlets in each culture bottle conducted for 40 HST, for observing the root length carried out for 50 HST. The highest number of shoots and leaves were produced at the same concentration, namely 150 ml / L coconut water treatment (15% concentration) with an average of 2.8 shoots and the average number of leaves 10.8 leaves. The average number of roots and the longest root length was produced at a concentration of 600 ml / L coconut water (60% concentration) with an average of 6 roots, and the longest root length was 0.5 cm.Keywords: Young coconut water, (Cocos nucifera L.), Dendrobium sp., in vitro, growth.ABSTRAKTujuan penelitian ini ialah menentukan konsentrasi air kelapa muda yang tepat untuk pertumbuhan planlet anggrek (Dendrobium sp.) secara in vitro. Penelitian ini menggunakan metode eksperimen, analisis data secara deskriptif untuk membandingan beberapa konsentrasi air kelapa yang berbeda. Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Perlakukan terdiri dari konsentrasi air kelapa 0 % (sebagai kontrol), 15% , 30% dan 60%. Masing-masing konsentrasi dilakukan 5 kali ulangan dan setiap ulangan terdiri dari 5 planlet Dendrobium sp dalam setiap botol kultur yang dilakukan selama 40 HST, untuk pengamatan panjang akar dilakukan selama 50 HST. Jumlah tunas dan jumlah daun terbanyak dihasilkan pada konsentrasi yang sama, yaitu perlakuan air kelapa 150 ml/L (konsentrasi 15%)  dengan rata-rata jumlah tunas terbanyak 2,8 tunas dan rata-rata jumlah daun terbanyak 10,8 helai daun. Rata-rata jumlah akar terbanyak dan panjang akar terpanjang dihasilkan pada konsentrasi air kelapa 600 ml/L (Konsentrasi 60%) dengan rata-rata jumlah akar terbanyak sebanyak 6 akar, dan rata-rata panjang akar terpanjang 0,5 cm.Kata kunci : Air kelapa Muda (Cocos nucifera L.), Dendrobium sp., in vitro, pertumbuhan

<p>ABSTRACT</p><p>The objectives of this study were to study the effect of several concentrations of polyethylene glycol (PEG) on the in vitro growth of eggplant, to find the appropriate PEG concentration for in vitro selection to drought  tolerance  of eggplant  and the drought tolerant eggplant accessions. The experiment  was conducted  at  the  Laboratory  of  Tissue  Culture,  Department  of  Agronomy and Horticulture,  Bogor  Agricultural  University.  The  experiment  was arranged  in  a  completely randomized design with two factor. The first factor was concentration of PEG (0, 5, 10,  and  15%) while the second factor was eggplant accessions (Kania F1, 001, 007, 013, 016, 030, 034, 035, 055, 057, 069,  071,  072,  078,  085,  and  090).  The  results  showed  that  the addition  of PEG  to  in  vitro media significantly affected the survival percentage, the percentage of callus, developed the bud and the number of leaves of eggplant. Addition of PEG 10 and 15% in media can be used as the drought tolerance selective agent of eggplant in vitro. Penelitian ini dilaksanakan di laboratorium Kultur Jaringan,  Departemen Agronomi dan Hortikultura,  Institut  Pertanian  Bogor.  Penelitian  ini  disusun dalam  rancangan  acak  lengkap  dua faktor. Faktor pertama adalah konsentrasi PEG  terdiri atas  0, 5, 10, dan 15%.  Faktor kedua adalah nomor terung terdiri atas enam belas nomor (Kania F1, 001, 007, 013, 016, 030, 034, 035, 055, 057, 069,  071,  072,  078,  085,  dan  090).  Hasil  penelitian menunjukkan  bahwa  penambahan  PEG  pada media  in  vitro  memberikan pengaruh  nyata  dan  sangat  nyata  terhadap  persentase  hidup eksplan, persentase  eksplan  berkalus,  pertambahan  tinggi  tunas,  dan jumlah  daun  tanaman  terung.  Media PEG 10 dan 15% merupakan media yang dapat digunakan untuk seleksi kekeringan tanaman terung in vitro. Nomor terung Kania F1, 001, 007, 016, 034, 035, 055, 057, 069, 071, 072, 078, 085, dan 090 merupakan nomor-nomor terung yang toleran terhadap cekaman kekeringan.</p><p>Kata kunci: kultur jaringan, seleksi in vitro, solanaceae, toleran kekeringan</p>

The experiment was conducted to investigate the combined effect of different plant growth regulators and charcoal supplementation in MS medium on growth and development of plantlets regenerated from protocorm like bodies (PLBs) of hybrid orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations with charcoal supplementation was studied. The result revealed that the use of different growth regulators had significant effect on different parameters studied. The maximum weight of PLBs (5.123 g) was obtained from the combination of BAP + IBA at 1.0 mg/l each. The highest shoot height (3.239 cm) and maximum number of rooted plantlets (4.473) was obtained from 1.0 mg/l each of BAP + NAA combination. The maximum number of leaves (3.490) and the maximum length of leaves (1.946 cm) were obtained from 1.0 mg/l each of BAP + IBA and the highest leaf width (1.166 cm) was obtained from 0.5 mg/l BAP +1.0 mg/l IBA combination. The highest root length was obtained from 0.5 mg/l each of BAP + IAA and the maximum number of regenerated plantlets (20) was obtained from 0.5 mg/l IAA + 1.0 mg/l IBA combination. However, the maximum fresh weight of single shoot (0.219 g) and the maximum number of roots per plantlet (6.300) was obtained from 1.0 mg/I each of IAA + IBA combination. The first experiment involving different explants and concentrations of 2,4-D and kinetin showed highly significant differences for length and weight of callus formed except interaction of callus weight. Leaf explant appeared to be best of all for callus length and weight when 1.0 mg/L 2,4-D + 0.25 mg/L kinetin concentration was used. Similarly, different explants versus different concentrations of BAP/GA3/IAA showed significant differences for shoot length and leaf number per plantlet and also for root length. However, interaction term confirmed node and node/internode explants produced better results in shoot length and number of leaves per plantlet when concentrations 1 .0 mg/L BAP + 0.1 mg/L GA3 and 1.0 mg/L BAP + 0.2 mg/L GA3, 1.0 mg/L BAP + 0.4 mg/L GA3, respectively, were used. Similarly, internode explants produced better results for root length after 21 days plantlet-1 when concentration of 1.0 mg/L IAA + 0.25 mg/L GA3 was used. Shoot tip explants also produced better results in root length after 28 days plantlet-1 when concentrations 1.0 mg/L IAA + 0.25 mg/L GA3 were used.
