The yellow pigment in potato (Solanum L. sp.) tuber flesh is caused by various carotenoids that may protect against cancer, cardiovascular disease, and macular eye degeneration. The objectives of this research were to 1) identify and quantify the carotenoids present in 11 diploid clones from a hybrid population of Solanum phureja ssp. phureja Juz. & Bukasov-S. stenotomum ssp. stenotomum Juz. & Bukasov and two tetraploid potato cultivars (the yellow-fleshed `Yukon Gold' and the white-fleshed `Superior'), and 2) determine the relationship between tuber yellow intensity and carotenoid content. Yellow intensity was measured by a colorimeter programmed to calculate a yellowness index, YI E-313. Carotenoid analyses were performed on an automated high-performance liquid chromatography system with software for integration and quantitation with detection at 450 nm using a diode array detector. Six major carotenoids were detected: neoxanthin, violaxanthin, lutein-5,6-epoxide, lutein, zeaxanthin, and an unknown carotenoid. Total carotenoid content in the yellow-fleshed diploid clones was 3 to 13 times higher than `Yukon Gold' and 4 to 22 times higher than `Superior'. Both total and individual carotenoid contents were positively correlated with tuber yellow intensity. There was an exponential relationship between total carotenoid content and tuber yellow intensity. This suggests that selecting for more intense yellow flesh will result in higher levels of carotenoids. These specific diploid clones were selected for this study because they produced at least five percent 2n pollen; they have the potential to make significant contributions to improving the nutritional status of tetraploid potatoes through 4x-2x hybridizations.

Consumption of carotenoid-containing foods can promote human health. Although yellow-fleshed potatoes (Solanum tuberosum) have a higher carotenoid content than white-fleshed potatoes, little is known about how growing environments may affect individual and total carotenoid content in different potato clones. The purposes of this study were to estimate the amount of genetic variability in potato for five xanthophyll carotenoids, their concentration, and to determine the stability of these carotenoids across environments. Nine white- or yellow-fleshed tetraploid clones were grown in Maine and Florida for 2 years. Carotenoids were extracted in acetone and analyzed by high-performance liquid chromatography. There were significant differences among clones for zeaxanthin, antheraxanthin, lutein, and total carotenoid content. There were significant clone × environment interactions for zeaxanthin, antheraxanthin, violaxanthin, neoxanthin, lutein, and total carotenoid. Broad-sense heritabilities (and their 95% confidence intervals) were 0.89 (0.79–0.98) for zeaxanthin, 0.93 (0.87–0.99) for antheraxanthin, 0.68 (0.14–0.92) for violaxanthin, 0.51 (0.00–0.88) for neoxanthin, 0.85 (0.70–0.97) for lutein, and 0.96 (0.89–0.99) for total carotenoid. Clonal mean total carotenoid content ranged from 101 to 511 μg/100 g fresh weight. A higher proportion of carotenoids were produced by the lycopene epsilon cyclase branch of the carotenoid biosynthetic pathway in white-fleshed than yellow-fleshed clones. Total carotenoid content in B2333-5 was significantly greater than in ‘Yukon Gold’. With genetic variation for individual and total carotenoid content in potatoes, improving the levels of carotenoids has been and should continue to be feasible; however, concentrations are likely to vary in different environments.

In this study the native carotenoids composition in kumquat (Fortunella margarita) (peel + pulp) from Brazil was determined for the first time by a HPLC-DAD-APCI/MS (high performance liquid chromatography-diode array detector-atmospheric pressure chemical ionization/mass spectrometry), methodology. Eleven carotenoids were successfully identified and quantified in kumquat: four carotenoids in the free form and seven carotenoids in the esterified form. β-citraurin-laurate was the carotenoid found in the highest content (607.33 µg/100 g fresh matter), followed by β-cryptoxanthin-laurate (552.59 µg/100 g). The different esterified forms of β-citraurin and β-cryptoxanthin represented 84.34% of the carotenoids found, which demonstrates the importance of esterification in natural fruits. β-carotene and free xanthophylls (β-cryptoxanthin, lutein and zeaxanthin) represented 5.50% and 14.96%, respectively, of total carotenoids in kumquat. The total carotenoid content of kumquat from Brazil was very high (2185.16 µg/100 g), suggesting that this fruit could contribute significantly to the intake of important bioactive compounds by the population.

Japanese knotweed (Fallopia japonica Houtt.) and Bohemian knotweed (Fallopia x bohemica) are invasive alien plant species, causing great global ecological and economic damage. Mechanical excavation of plant material represents an effective containment method, but it is not economically and environmentally sustainable as it produces an excessive amount of waste. Thus, practical uses of these plants are actively being sought. In this study, we explored the carotenoid profiles and carotenoid content of mature (green) and senescing leaves of both knotweeds. Both plants showed similar pigment profiles. By means of high performance thin-layer chromatography with densitometry and high performance liquid chromatography coupled to photodiode array and mass spectrometric detector, 11 carotenoids (and their derivatives) and 4 chlorophylls were identified in green leaves, whereas 16 distinct carotenoids (free carotenoids and xanthophyll esters) were found in senescing leaves. Total carotenoid content in green leaves of Japanese knotweed and Bohemian knotweed (378 and 260 mg of lutein equivalent (LE)/100 g dry weight (DW), respectively) was comparable to that of spinach (384 mg LE/100 g DW), a well-known rich source of carotenoids. A much lower total carotenoid content was found for senescing leaves of Japanese and Bohemian knotweed (67 and 70 mg LE/100 g DW, respectively). Thus, green leaves of both studied knotweeds represent a rich and sustainable natural source of bioactive carotenoids. Exploitation of these invaders for the production of high value-added products should consequently promote their mechanical control.

The β-carotene and total carotenoid content of either fresh or dried tissue of fruits of a total of 57 cultivars of six Capsicum species were analyzed using high performance liquid chromatography (HPLC). β-Carotene levels in ripe fruit varied from 0 to 166 μg·g-1 fresh weight, and carotenoid levels were from 1 to 896 μg·g-1 in ripe fruit in 1996. The range of values for β-carotene was similar in 1997, but that for total carotenoids was wider (4 to 1173 μg·g-1 fresh weight). Fresh fruit of the cultivars Greenleaf Tabasco, Pulla, Guajillo, NuMex Conquistador, Ring-O-Fire, and Thai Dragon contained greater amounts of β-carotene per 100 g fresh weight than the recommended dietary allowance (RDA) for vitamin A for the average adult. For dried Capsicum entries, New Mexican, aji, pasilla, and ancho types had the highest levels of β-carotene. In 1996, β-carotene levels among the dried Capsicum germplasm ranged from 2 to 739 μg·g-1 dry weight, and carotenoid levels from 111 to 6226 μg·g-1. Values were higher in 1997, ranging from 24 to 1198 μg·g-1 dry weight for β-carotene and from 187 to 10,121 μg·g-1 for total carotenoids. A pasilla type (C. annuum L.) had the highest total carotenoid content among the dried entries in both years.

<p>The influence of certain nitrogen compounds - components of glucose-peptone medium (GPM) on the accumulation of carotenoids by some strains was investigated by surface cultivating basidiomycetes. The total carotenoid content was set in acetone extracts of mycological material spectrophotometrically and calculated using the Vetshteyn formula.</p> <p>As the nitrogen-containing components used GPM with 9 compounds, such as peptone, DL-valine, L-asparagine, DL-serine, DL-tyrosine, L-proline, L-alanine, urea, NaNO<sub>3</sub>. The effect on the accumulation of specific compounds both in the mycelium and in the culture fluid of carotenoids by culturing certain strains of Basidiomycetes was identified.</p> <p>Adding to standard glucose-peptone medium peptone at 5 g/l causes an increase of carotenoid accumulation by strain <em>L. sulphureus</em> Ls-08, and in a concentration of 4 g/l by strains of <em>F. hepatica </em>Fh-18 and <em>F. fomentarius</em> Ff-1201.</p> <p>In order to increase the accumulation of carotenoids in the mycelium  we suggested to make a standard glucose-peptone medium with proline or valine for cultivating of <em>L. sulphureus</em> Ls- 08 strain; alanine for <em>F. fomentarius</em> Ff-1201 strain; proline, asparagine and serine - for strain Fh-18 of <em>F. hepatica</em>. The results can be implemented in further optimization of the composition of the nutrient medium for culturing strains of Basidiomycetes wich producing carotenoids.</p> <p><em>Keywords: nitrogen-containing substances, Basidiomycetes, mycelium</em><em>,</em><em> culture filtrate, carotenoids</em></p>

The aim of this study was to evaluate the effect of a supplementation diet for hens consisting of dried basil herb and flowers of calendula and dandelion for color, carotenoid content, iron-induced oxidative stability, and sensory properties of egg yolk compared with commercial pigment (control) and marigold flower. The plant parts were supplemented in diets at two levels: 1% and 3%. In response to dietary content, yolks from all diets differed in carotenoid profile (p < 0.001). The 3% supplementation level resulted in a similar total carotenoid content as the control (21.25 vs. 21.79 μg/g), but by 3-fold lower compared to the 3% marigold (66.95 μg/g). The tested plants did not achieve yolk color fan values as the control (13.47) or 3% marigold (11.47), and among them, calendula had the highest values (9.73). Despite the low carotenoid content in diets supplemented with basil herb, iron-induced malondialdehyde (MDA) concentration was low as for marigold (on average 106.83 vs. 92.68 ng/g after 250 min). The treatments differed in sensory color scores for fresh and hard-boiled yolks and flavor while other sensory properties were similar. In conclusion, the supplementation of plants in a hen diet may result in yolks containing carotenoids and other compounds showing a high antioxidant effect.

Proper diet is critical for laboratory-reared animals, as it may affect not only their welfare, but also experimental results. Amphipods (Crustacea: Amphipoda) play important roles in ecosystems and are often used in environmental research. Endemic amphipods from the ancient Lake Baikal are promising for laboratory bioassays; however, there are currently no laboratory cultures. In this work, we determine how different diets affect the color and metabolism of a laboratory-reared Baikal amphipod, Eulimnogammarus cyaneus. We found that in freshly collected blue-colored animals, body color correlated with total carotenoid content. Total carotenoid levels did not differ after long-term (two months) feeding with a close to natural carotenoid-enriched, or even a carotenoid-depleted diet. Nevertheless, antennae color was closer to red in the natural-like diet group. It is likely that the carotenoids from the commercial diet are not properly metabolized in E. cyaneus. The animals fed commercial diets had a higher glycogen content, which may signify a higher metabolic rate. Overall, we show that a carotenoid-enriched diet optimized for decapods is not optimal for amphipods, likely due to different carotenoid compositions, and the diet for long-term rearing of E. cyaneus and other Baikal amphipods requires supplementation.

Background. Barbary fig (Opuntia spp), called prickly pear, is a cultivated species belonging to family Cactaceae. It produces fruits one time per year during a short season. It is largely spreading in the Mediterranean countries. The aim of the study was to investigate the physicochemical and biochemical analysis of powder of prickly pear fruit peels of three Moroccan varieties (Aakria, Derbana, and Mles). Material and Methods. Both physicochemical analysis (humidity, water activity, Brix, ash content, pH, and total titratable acidity) and biochemical analysis (total carotenoid content, betalain content, total polyphenolic content, and ascorbic acid content) were effectuated according to protocols reported within the present research work. Results. Regarding the physiochemical analysis, the humidity of powder of prickly pear fruit peels ranged from 10.173 ± 0.002 to 15.27 ± 0.02%. The water activity (aw) ranged from 0.17 ± 0.002 to 0.336 ± 0.002. The values of Brix ranged from 68.67 ± 0.33 to 74.22 ± 0.51° Bx. pH values ranged from 5.41 ± 0.01 to 5.62 ± 0.01. The total titratable acidity values ranged from 0.644 ± 0.014 to 0.76 ± 0.007 g citric acid/100 g DM (dry matter). The ash content ranged from 1.02 ± 0.02 to 11.41 ± 0.03 g/100 g DM. Regarding the biochemical analysis, the total carotenoid content ranged from 5.14 ± 0.10 to 9.79 ± 0.06 mg/g of FM (Fresh matter). The total betalain content ranged from 2 ± 0.69 to 37.66 ± 2.65 mg/100 g of DM. The total polyphenolic content ranged from 1739.92 ± 13.69 to 2409.66 ± 43.65 mg gallic acid equivalent (GAE)/100 g of DM. The ascorbic content values ranged from 186.78 ± 13.23 to 294.04 ± 5.81 mg/100 g of DM. Conclusion. The results showed an interesting richness of the investigated powder in pigments (betacyanins, indicaxanthins, and carotenoids) and nutritional compounds including sugar, proteins, and vitamins that make this powder interesting for being used as a dye in food.
