Abstract This study aimed at investigating the levels and distribution of progesterone in receiving waters and wastewaters in Arusha, a fast-growing urban area and the third largest city in Tanzania. Specifically, the study was conducted along the Themi River and the adjacent waste stabilization ponds (WSPs). Progesterone was detected and quantified by using an enzyme-linked immunosorbent assay (ELISA) kit. For Themi River samples, the level of progesterone obtained ranged from ‘no detection’ to 439 ng/L with a mean value of 120.3 ng/L. The levels detected were significantly higher in the midstream than the upstream and downstream sections of the river (P<0.05). The higher values at the midstream were attributable to livestock, WSPs and household effluents; agricultural activities; and sewage infiltration. Progesterone levels in the present study, although not extremely high, correspond to those associated with harmful effects in other studies. Results for samples collected from the WSPs indicated a release of 215 ng progesterone per L of the receiving waters. Although progesterone removal efficiency by the WSPs was 75%, the amount released was still high enough to cause harm to aquatic organisms. Thus, more efficient techniques are required to adequately remove progesterone to the recommended levels.

The optical character of waste stabilization ponds (WSPs) is of concern for several reasons. Algal photosynthesis, which produces oxygen for waste oxidation in WSPs, is influenced by attenuation of sunlight in ponds. Disinfection in WSPs is influenced by optical characteristics because solar UV exposure usually dominates inactivation. The optical nature of WSPs effluent also affects assimilation by receiving waters. Despite the importance of light behaviour in WSPs, few studies have been made of their optical characteristics. We discuss simple optical measures suitable for routine monitoring of WSPs (including at sites remote from laboratories): optical density of filtrates – an index of dissolved coloured organic (humic) matter, visual clarity – to provide an estimate of the beam attenuation coefficient (a fundamental quantity needed for optical modelling) colour (hue) – as an indicator of general WSP ‘condition’ and irradiance attenuation quantifying depth of light penetration. The value of optical characterisation of WSPs is illustrated with reference to optical data for WSPs in NZ (including high-rate algal ponds) treating dairy cattle wastewater versus domestic sewage. We encourage increased research on optical characteristics of WSPs and the incorporation of optical measures in monitoring and modelling of WSP performance

Abstract Background and aims: Gestational diabetes represents a form of diabetes diagnosed during pregnancy that is not clearly overt diabetes. In the last trimester of gestation the growth of fetoplacental unit takes place, thus maternal hyperglycemia will determine an increased transplacental passage, hyperinsulinemia and fetal macrosomia. The aim of our study was that o analyzing the effect of maternal glycemia from the last trimester of pregnancy over fetal weight. Material and method: We run an observational study on a group of 46 pregnant women taken into evidence from the first trimester of pregnancy, separated in two groups according to blood glucose determined in the third trimester (before birth): group I normoglycemic and group II with hyperglycemia (>92mg/dl). Results: The mean value of third trimester glycemia for the entire group was of 87.13±22.03. The mean value of the glycemia determined in the third trimester of pregnancy was higher in the second group (109.17 mg/dl) in comparison to the first group (74.,21 mg/dl). The ROC curve for third trimester glycemia as fetal macrosomia appreciation test has an AUC of 0.517. Conclusions: Glycemia determined in the last trimester of pregnancy cannot be used alone as the predictive factor for fetal macrosomia.

Rotaviruses are generally excreted in large numbers in diarrhoeal stools, but in wastewaters their numbers are subject to variations. Detection and enumeration of these viruses involve a concentration step followed by an assay method. Enumeration in wastewater concentrates is complicated by the presence of toxic substances which are often concentrated with the viruses. These toxic substances often cause the destruction of cells during rotavirus assay, thus leading to underestimation of viral numbers. Such concentrates were detoxified by a simple and effective method using polyacrylamide (Biogel P-6DG) or dextran (Sephadex G50) beads. Concentrates (10 ml) were mixed with 0.5 g gel and the mixtures were allowed to stand for 2 h at room temperature during which time the beads swell by the passage of water into them along with inorganic ions and substances with molecular weights of less than 30,000. The supernatants were then decontaminated with antibiotics and assayed for rotaviruses by the indirect immunofluorescent technique. Most untreated ultrafiltrates of raw sewage and those from anaerobic ponds were found to be too toxic to MA104 and LLC MK2 cells, whereas the above treatment rendered over 90% of wastewater concentrates non-toxic to cells. This technique was used to study virus removal in samples from deep waste stabilization ponds in northeast Brazil.

Rotavirus removal in waste stabilization ponds is a relatively slow process: in a series of ten ponds (a 1-d anaerobic pond followed by nine 2-d ponds) its numbers were reduced from 1.4 × 105 per litre to zero, and in an “innovative” series (a 1-day anaerobic pond, 3-d facultative pond, 3.8-d, 3-d and 5-d maturation ponds) from 5.1 × 104 per litre to <5 per litre. Faecal coliforms were better indicators of rotaviruses than was Clostridium perfringens .

In 1992, a survey was conducted on the performance of waste stabilization ponds in France. The data selected come from a sample of 178 ponds, with an average capacity of 600 p.e., throughout France. For each plant, one or several input--output load measurements over a 24-h period are available. The average organic load level received is approximately 25 kg BOD/ha.d, representing 50% of the nominal load. The quality of the treated water is presented based on the type of sewerage system feeding the ponds. The results appear dispersed, however; in 70% of the cases the concentrations in COD and BOD on filtered samples are under 120 mg/l and 40 mg/l, respectively, and the concentration in TSS under 120 mg/l (discharge standards in France for waste stabilization ponds). The reductions in nitrogen and phosphorus nutrients are on average from 60% to 70%. The influence of different parameters (sewerage system type, organic load, season, age of plant, etc.) was studied. The results appear noticeably worse when the ponds receive wastewater from a strictly separate sewerage system.

Current study investigated the occurrence of aflatoxin contamination in poultry feed and feed materials in different poultry farms and feed factories in Bangladesh. A total of 100 samples of finished feed and raw feed materials were collected and tested through direct competitive Enzyme-Linked Immunosorbent Assay (ELISA) for total aflatoxin detection. Overall, 97% samples (n=97/100) in our study, were found positive for aflatoxin contamination. Among finished feed categories, layer grower feed contained highest level of aflatoxin with a mean value of 21.64 ppb whereas layer feed was less susceptible for aflatoxin contamination (mean value 9.49 ppb). Between raw feed materials, maize samples were highly contaminated (n=15/15, 100%) with aflatoxin while 86.67% soybean samples showed positive result. Twenty one percent (21%) of the samples in our study contained aflatoxin concentration more than the acceptable limit employed by USFDA and many other countries which might pose severe health risk to poultry and human consumer. Proper surveillance and immediate control measures should be taken to ensure safe poultry feed and feed materials. Bangladesh J Microbiol, Volume 35 Number 1 June 2018, pp 75-78

Studies over the past several decades have dramatically increased our understanding of the immune response to Mycobacterium tuberculosis infection, and advances in proteomics and genomics have led to a new class of immune-diagnostic tests, termed interferon-γ (IFN-γ) release assays (IGRAs), which appear to obviate many of the problems encountered with the tuberculin skin test (TST). Worldwide, 2 IGRAs are currently commercially available. QuantiFERON-TB Gold In-Tube (Cellestis) is a third-generation product that uses an enzyme-linked immunosorbent assay to measure IFN-γ generated in whole blood stimulated with M. tuberculosis–specific antigens. T-Spot-TB (Oxford Immunotec) employs enzyme-linked immunosorbent spot technology to enumerate the number of purified lymphocytes that respond to M. tuberculosis–specific antigens by producing IFN-γ. These in vitro tests measure the host immune response to M. tuberculosis–specific antigens, which virtually eliminates false-positive cross reactions caused by bacillus Calmette-Guérin vaccination and/or exposure to environmental nontuberculous mycobacteria that plague the interpretation and accuracy of the tuberculin skin test (TST). The high specificity of IGRAs, together with sensitivity commensurate with or better than that of the TST, promises an accurate diagnosis and the ability to focus tuberculosis-control activities on those who are actually infected with M. tuberculosis.
