Objective: In this study, the leaf powder of Aegle marmelos (L.) Correa. was subjected to preliminary phytochemical and in vitro antioxidant analysis. Methods: The freshly prepared plant leaf extract was subjected to preliminary phytochemical screening, which revealed the presence of alkaloids, tannins, saponins, flavonoids, glycosides, phenolic compounds, terpenoids, and steroids. Reducing power, superoxide (SO) anion radical, nitric oxide (NO) radical, and hydroxyl radical scavenging assays were carried out to evaluate the antioxidant potential of the methanolic leaf extract of this plant. The amounts of total phenolic and flavonoid compounds were also determined. Results: This study has revealed that the A. marmelos (L.) Correa. leaf extract showed considerably high amounts of most of the phytochemicals, total antioxidant capacity, total phenolic, and flavonoid content. The study also indicated that the A. marmelos (L.) Correa. showed comparatively good scavenging activity, i.e., inhibition of hydroxyl radical, NO and SO anion scavenging and reducing power activities when compared with the respective standards. Conclusion: The leaf powder of A. marmelos (L.) Correa. can be used as easily accessible source of natural antioxidant and as a possible food supplement or in pharmaceutical industry.

Objectives: The work aims to screen the antioxidative potentials of different crude extracts of the fronds of two medicinally important pteridophytes of Southern Assam, India, namely, Cyclosorus interruptus (Willd.) H. Itô and Pronephrium nudatum (Roxb. ex Griff.) Holttum. Methods: Frond extracts of the pteridophytes were prepared by Soxhlet hot extraction method. Total phenolic content (TPC) and total flavonoid content (TFC) of the hexane, ethyl acetate, acetone, and methanol extracts of the fronds of the plants were done by following standard protocol. In vitro assessment of the antioxidative behavior of the extracts was performed using standard 2,2-diphenyl-1-picrylhydrazyl scavenging assay, reducing power assay, metal chelating assay, hydroxyl, superoxide, and 2,2’- azino-bis (3-ethylbenzothiazoline - 6 – sulfonic acid) radical scavenging methods in different in vitro systems. Results: Preliminary phytochemical analysis implicated the presence of phenolic and flavonoid compounds in all the frond extracts. The methanol extract of the fronds of both the plants showed maximum phenolic and flavonoid contents in comparison to the other extracts, however, that of C. interruptus was found to be higher than P. nudatum. Antioxidative potentials of the said extracts were also found to be impressive and noteworthy. The decreasing order of the antioxidative efficacies of the extracts was found to be same as that of TPC and TFC of the extracts. Conclusion: It is pertinent to comment that the methanol extract of the fronds of both the plants may be treated as a potential source of natural antioxidants.

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.

Pterygota alata is a large deciduous tree of Malvaceae family. The present study was designed to evaluate in vitro antioxidant activity of stem bark of the plant. Ferric reducing power (FRP) test, 1, 1- diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging test and total antioxidant capacity (TAC) assay were used to detect the antioxidant activity. Total phenolic and flavonoid contents were also determined as they are well known phytochemicals with antioxidant property. The bark of the plant showed efficient reducing power as well as free radical scavenging property (IC50 values 52.25 - 172.05). The bark also found rich in total phenolic and flavonoid content. The highest amount of total phenolic content was found in chloroform soluble fraction (29.898 µg/mL) followed by ethyl acetate soluble fraction (15.88 µg/mL). The highest content of total flavonoid also detected in chloroform soluble fraction (107.56 µg/mL) but followed by crude ethanol extract (98.66 µg/mL). Overall, the bark of the plant possesses significant antioxidant activity, therefore can be used as a good natural source of antioxidant. © 2014 JSR Publications. Objective: To evaluate the preliminary phytochemical content and antioxidant potential of the hydroalcoholic leaf extracts of Hemionitis arifolia. Methods: Total phenolic, flavonoid and alkaloid contents were evaluated using spectrophotometric methods. The free radical scavenging activity of the leaf hydroalcoholic extract were evaluated against DPPH+, ABTS+, Reducing power assay and nitric oxide assay were determined. Results: The hydroalcoholic concentrate of H. arifolia uncovered the most elevated polyphenol content when contrasted and the other phytoconstituents. Absolute phenol content of the hydroalcoholic separate was observed to be 31.78%, flavonoid content is 1.02% and Alkaloid content is 30.40% individually. The Solvent concentrates showed huge cell reinforcement movement, with hydroalcoholic extract. ABTS Assay, DPPH assay, Reducing power assay and Nitric oxide assay where the Inhibition concentration were 667.75µg/ml, 734.25 µg/ml, 791.58 µg/ml and 899.67 µg/ml. Conclusion: This study suggests that hydroalcoholic leaf extracts of H. arifolia could be a potential source of natural antioxidant and justifies its use in ethno-medicine.

Butanol and hexane leaves extracts ofParkinsonia aculeataL. (Fabaceae) were assessed for its antioxidant potential byin vitromethods. Phytochemical analysis and antioxidant activity of plant extracts were studied using differentin vitroassays. UPLC analysis of extracts was carried out for the identification of chemical constituents. The total phenolic contents of the butanol and hexane leaf extract were 42 mgGAE/g and 34 mgGAE/g whereas flavonoid contents of these extracts were found to be 0.044 mgRE/g and 0.005 mgRE/g, respectively. Among both extracts, butanol extract shows maximum inhibition (%) of 93.88%, 80.02%, 52.06%, 94.68%, and 69.37% in DPPH, non-site-specific and site-specific, FTC, and TBA assays and absorbance of 0.852 and 0.522 in reducing power and CUPRAC assay at the highest concentration tested. The FRAP and TAC values of butanol extract were found to be 678 μM Fe(II)/g and 36 mgAAE/100 mg. UPLC analysis of extracts revealed the presence of various polyphenols. The tested plant extracts were found to possess potent antioxidant and free radical scavenging activity which may be due to the presence of flavonoids and polyphenols.

Objective: The study was carried out to evaluate the preliminary phytochemical screening and antioxidant activity of ethanolic extract of Spinacia oleracea (SO).Methods: The leaves of SO were shade dried, and the extract was prepared using solvent ethanol by Soxhlet extraction method. The preliminary phytochemical screening was carried out on the leaf extract of the plant. The total phenolic content and total flavonoids were estimated using Folin- Ciocalteu’s and aluminum chloride reagents, respectively. Antioxidant activities were studied using 1,1-diphenyl-2-picrylhydrazyl, nitric oxide, hydrogen radical, lipid peroxidation, and phosphomolybdenum radical scavenging assays.Results: The preliminary phytochemical analysis revealed the presence of bioactive constituents such as phenols, alkaloids, flavonoids, saponins, and glycosides. As SO is a rich source of different bioactive component, it contains a considerable amount of flavonoids and phenols. The different antioxidant assays proved that spinach is one of the best antioxidants with its ability to scavenge different radicals that generate oxidative stress.Conclusion: The observed activity may be associated with bioactive components such as phenols and flavonoids present in the leaf extracts and could have greater importance as nootropic plant in oxidative stress-related degenerative diseases such as Alzheimer and dementia.

Background. Quezon province has been one of the primary agricultural sources of vegetables and crops in southern Luzon due to its diverse agricultural topography. Having increased dietary awareness, consumption of antioxidant-rich foods has become relevant. Objectives. Thirteen (13) methanolic extracts of endemic Quezon plant foods were evaluated for phytochemical constituents and antioxidant potential.Methods. The plant extracts were subjected to in vitro antioxidant assays, which include DPPH [2,2-diphenyl-1picrylhydrazyl], FRAP [Fluorescence recovery after photobleaching], metal chelation, superoxide, nitric oxide, hydroxyl radical scavenging activities and MTT [ (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide)] reduction. Qualitative phytochemical analysis were also employed. Results. P. umbellatum, locally known as kamamba and C. longa, locally known as luyang dilaw showed high antioxidant activity using DPPH and MTT assays by 82.46±0.75% and 82.35±1.79% and 78.46±0.71% and 60.97±2.60% at 66.67 μg/mL, respectively. FRAP assay revealed a comparable reducing power with BHT (butylated hydroxytoluene) (93.61±0.56%) at 72.15 μg/mL in C. longa (92.49±1.32%), P. umbellatum (92.38±0.64%), and Z. officinale (90.33±2.06%) (luya), but found with low metal chelating activity. The highest activity against hydroxyl radical were observed in S. edule (sayote) (34.30±1.88%), P. edulis (pasyonaryo) (33.48±1.34%) and D. philippinensis (katmon) (34.71±0.85%) at 45.5 μg/mL. On the other hand, Z. officinale, C. esculenta (gabi), V. unguiculata (sitaw) fruit and D. philippinensis showed dual action, as antioxidant and as pro-oxidant, in Superoxide Scavenging and Nitric Oxide assays. Quezon province plant foods contain flavonoids, phenolic compounds, glycosides and coumarins and quinones which may explain their behavior as antioxidant.Conclusion. The study revealed that different plant foods showed different capacity to scavenge particular oxidants. However, P. umbellatum and C. longa may be considered promising sources of natural antioxidants.

In-vitro antioxidant action of hydroalcoholic leaf extract of Grewia hirsuta (HAEGH) has been examined using one, “1-diphenyl-2-picryl-hydrazil (DPPH) free from radical scavenging” actions. The plant collected from the forest of the Western Ghats region of Karnataka province. The motive for plant collection from a specific location is the plant of forests exhibits the variation in growth, quantity, and quality of their active ingredients and secondary metabolites due to influence ecological factors like effect changes in location, soil, climate, etc. The work corresponds to preliminary phytochemical investigation for diverse phytoconstituents and quantitative phytochemical analysis of total phenolic, flavonoids & alkaloids content (TPC, TFC & TAC respectively) was evaluated with advanced methods. “HPTLC (High performance thin-layer” chromatography) fingerprint investigation was achieved for qualitative determination of the likely number of elements present in the hydroalcoholic extract. In-vitro antioxidant activity of HAEGH has been determined through hydroxyl radical scavenging assay that exhibited strong dose-dependent antioxidant activity as compared with standards compound, ascorbic acid. The IC50 value of HAEGH found, 25.90 % inhibition and for ascorbic acid, it was 17.68%. The Preliminary phytochemical estimation found presence of flavonoids, alkaloids, glycosides, phenol, proteins, diterpins and quantitative phytochemical analysis estimation of TPC, TFC & TAC found to be 3.627%, 4.059% & 5.671% respectively. HPTLC analysis of HAEGH at 354nm reveals the presence of a compound with Rf value 0.44 compare with Rf value 0.46 of quercetin. These outcomes indicated that the hydroalcoholic leaf extract of Grewia hirsuta plant contains phytoconstituents that exhibit antioxidant activity possible because of the existence of bioactive compounds.

Abstract Background The objective of the study is to evaluate the hepatoprotective activity of methanolic extract fractions of Lindernia ciliata (LC) and development of qualitative analytical profile of the bioactive fraction using HPLC fingerprinting analysis. All the fractions of methanolic extract of Lindernia ciliata (LCME) are assessed for their total phenolic, flavonoid contents and in vitro antioxidant properties by using DPPH, superoxide, nitric oxide, hydroxyl radical scavenging activities and reducing power assay. Acute toxicity study was conducted for all the fractions and the two test doses 50 and 100 mg/kg were selected for the hepatoprotective study. Liver damage was induced in different groups of rats by administering 3 g/kg.b.w.p.o. paracetamol and the effect of fractions were tested for hepatoprotective potential by evaluating serum biochemical parameters and histology of liver of rats. The effective fraction was evaluated for its antihepatotoxic activity against D-Galactosamine (400 mg/kg b.w. i.p.) and in vivo antioxidant parameters viz., Glutathione (GSH), Melondialdehyde (MDA) and Catalase (CAT) levels are estimated using liver homogenate. Results Among all the fractions, butanone fraction of LCME, (BNF-LCME) has shown better hepatoprotective activity and hence it is selected to evaluate the antihepatotoxicity against D-GaIN. The activity of BNF-LCME is well supported in in vitro and in vivo antioxidant studies and may be attributed to flavonoidal, phenolic compounds present in the fraction. Hence, BNF-LCME was subjected to the development of qualitative analytical profile using HPLC finger printing analysis. Conclusions All the fractions of LCME exhibited significant hepatoprotective activity and BNF-LCME (50 mg/kg) was identified as the most effective fraction.

Triterpene acid and phenolic constituents from nine ancient varieties of apple (Malus domestica) fruits cultivated in Fanna, Friuli Venezia Giulia region, northeast Italy, were analyzed and compared with four commercial apples (‘Golden Delicious’, ‘Red Delicious’, ‘Granny Smith’ and ‘Royal Gala’). Total phenolic and flavonoid contents were measured by spectrophotometric assays. The quali-quantitative fingerprint of secondary metabolites including triterpene acid was obtained by LC-DAD-(ESI)-MS and LC-(APCI)-MS, respectively. Based on the two LC-MS datasets, multivariate analysis was used to compare the composition of ancient fruit varieties with those of four commercial apples. Significant differences related mainly to the pattern of triterpene acids were found. Pomolic, euscaphyc, maslinic and ursolic acids are the most abundant triterpene in ancient varieties pulps and peels, while ursolic and oleanolic acids were prevalent in the commercial fruits. Also, the content of the phenolic compounds phloretin-2-O-xyloglucoside and quercetin-3-O-arabinoside was greater in ancient apple varieties. The antioxidant (radical scavenging, reducing power, metal chelating and phosphomolybdenum assays) and enzyme inhibitory effects (against cholinesterase, tyrosinase, amylase and glucosidase) of the samples were investigated in vitro. Antioxidant assays showed that the peels were more active than pulps. However, all the samples exhibited similar enzyme inhibitory effects. Ancient Friuli Venezia Giulia apple cultivars can be a source of chlorogenic acid and various triterpene acids, which are known for their potential anti-inflammatory activity and beneficial effects on lipid and glucose metabolism. Our results make these ancient varieties suitable for the development of new nutraceutical ingredients.
