Inflammation is a reaction of a living vascularised tissue to an injury. Conventional or synthetic drugs used in the treatment of inflammatory diseases are inadequate, it sometimes have serious side effects. So, number of herbal medicines is recommended for the treatment of inflammation that has no side effects. Hence our study focused to investigate the physicochemical, qualitative phytochemical analysis of bioactive compounds and In-vitro and In-vivo anti-inflammatory activity of Plumeria indica Linn (P. Indica) flowers extract which has boundless medicinal properties. The physicochemical evaluations carried out in terms of loss on drying, ash value, extractive values and acid insoluble ash value ect. Qualitative analysis of various phytochemical constituents was determined by the well-known test protocol available in the literature. The aqueous and ethanolic extract of P. Indica flowers was screened for in-vivo anti-inflammatory activity by carrageenan induced paw edema in rat model and in-vitro anti-inflammatory activity by human red blood cell membrane stabilization method. Phytochemical analysis revealed the presence of phenols, flavonoids, tannins, saponins, alkaloids ect. Ethanolic extract showed best in vitro anti-inflammatory activity was screened for in vivo anti-inflammatory activity at the dose level of 250 and 500mg/kg. Indomethacin at the dose level of 10 mg/kg was used as reference standard drug. Both the extracts showed a dose dependent anti-inflammatory potential which provide scientific basis for the traditional claims of P. Indica flowers as an anti-inflammatory drug. Physalis peruviana calyces are used extensively in folk medicine. The crude ethanolic extract and some fractions of calyces were evaluated in order to explore antioxidant and anti-inflammatory activities. The anti-inflammatory activity was evaluated by the TPA-induced ear edema model. The antioxidant in vitro activity was measured by means of the superoxide and nitric oxide scavenging activity of the extracts and fractions. The butanolic fraction was found to be promising due to its anti-inflammatory and antioxidant activities. Therefore, a bio-assay guided approach was employed to isolate and identify rutin (1) and nicotoflorin (2) from their NMR spectroscopic and MS data. The identification of rutin in calyces of P. peruviana supports the possible use of this waste material for phytotherapeutic, nutraceutical and cosmetic preparations.

In the last decades, it has been intensively studied natural alternatives to synthetic products. Chassalia kolly is one of the medicinal plants used traditionally for treatment of skin infections. This study aimed at identifying the diversity secondary metabolites, to investigate the antioxidant and anti-inflammatory activities of ethanolic extract of Chassalia kolly leaves. Total phenols were determined by Folin Ciocalteu method. The aluminum trichloride method has been used to quantify total flavonoids, while the determination of condensed tannins was carried out by the hydrochloric vanillin method. The antioxidant capacity was evaluated by 2, 2-diphenyl-l-picrylhydrazyl (DPPH), 2, 2'-Azino-bis- (3-ethylbenzothiazolin-6-sulfonic acid) (ABTS) and phosphomolybdenum methods. In vitro anti-inflammatory activity of ethanolic extract of Chassalia kolly leaves was evaluated by method membrane stabilization. In this results, Chassalia kolly leaves contain flavonoid, mucilages, anthocyans, sterols and terpenes, saponosides, alkaloids and reducing compounds. The contents of total phenols, total flavonoids and total tannins are respectively (20.41±2.11) µgGAE/mgDM; (30.29±2.18) µgQE/mgDM and (1.211±0.122)µgCE/mgDM. The results obtained during this work allowed us to assert that all extracts of the studied plant have very good antioxidant properties (IC50=0.05µg/µL). The ethanolic extract of the leaves of Chassalia kolly showed an anti-inflammatory activity more interesting than aspirin which is the reference compound used in this study.

 Objectives: A persistent and chronic inflammation may act as an underlying definition for many of the long-term illness such as diabetic foot ulcer, Alzheimer’s disease and rheumatoid arthritis. Folklore medicinal plants provide immense health benefits with new remedies. Active compounds from plants containing alkaloids and phenolic compounds (flavonoids and tannins) possess antioxidant and anti-inflammatory activity. The leaves of Adhatoda vasica are most known for its effectiveness in treating respiratory diseases such as asthma, chronic bronchitis, and other respiratory conditions. In this report, we investigated the possible action of A. vasica against inflammation.Methods: Solvents with varying polarity (Hexane, dichloromethane, ethyl acetate, and methanol) were used for the sequential extraction of leaves of A. vasica Linn (Family: Acanthaceae). The anti‐inflammatory activity was assessed using simple in vitro methods. Phenols, flavonoids, phytosterols, saponins, alkaloids, and tannins were analyzed qualitatively using standard protocols. Anti‐inflammatory activity was evaluated using albumin denaturation assay, membrane stabilization assay and proteinase inhibitory activity. Aspirin (diclofenac) was used as a standard drug for the study of anti-inflammatory activity.Results: Our results substantiated that methanolic extract of A. vasica gave a maximum yield of 12.5% and apparently all bioactive phytochemical constituents were at a higher concentration in the methanolic extract. Methanolic extract of A. vasica showed a significant inhibition of the heat-induced albumin denaturation, red blood cells membrane stabilization, and proteinase inhibitory effects with 86±1.46, 88±1.24, and 96±0.93% for 1000 μg/ml, respectively.Conclusion: The above results delineate that the bioactive components were extracted using high polar solvent are mostly flavonoids and related polyphenols. These compounds may be the active constituents to render the anti-inflammatory activity to A. vasica.

Diabetes mellitus (DM) is most significant health problem in various developed and developing countries due to alteration of various clinical and pathological factors. Current work was intended for examine in vitro and in vivo anti diabetic competence of Cassia Auriculata flowers and its phytochemical analysis. The inhibitory effects on carbohydrate digestive enzyme α- amylase interaction with various extracts of Cassia Auriculata were contrast with acarbose. Hypoglycemic activity was executed with standard as a glibenclamide. Study indicated ethanolic extract showed higher action on α- amylase inhibition with assessment of IC50 value; 43.6%. Based on the above results of in vitro studies were used to selected ethanolic extract of Cassia Auriculata flower used for further study. Results of animal studies indicated that the ethanolic extract of C. Auriculata has shown dose dependent action (200 mg/ kg (1.20±0.91↓) and 400 mg / kg (4 ± 0.01↓) when compared to control and standard drug treated groups. Our study confirmed to ethanolic extract work through the α- amylase inhibition mechanism. Our view bioactive constituents confirm anti diabetic capacity and afford methodical source for validation of Cassia Auriculata flowers in ayurvedic formulations on diminution of DM prevalence.

Abstract Co-crystal is a promising class of solids that may provide options for improved properties. In the present study, ketoprofen- p-aminobenzoic acid (KP-PABA) co-crystal were prepared to sought enhanced solubility and dissolution rate of drug. KP-PABA co-crystal were prepared by solvent evaporation technique employing central composite experimental design, selecting independent variables as concentration of drug and PABA whereas dependent variables were assumed to be solubility and % drug release. The optimized batch as suggested by the experimental design was characterized by FTIR, DSC, XRD, SEM and NMR studies and further, evaluated for in-vitro and in-vivo anti-inflammatory and analgesic activities. The solubility and % drug release of different batches of co-crystal was found to be between 34.20-60.11 µg/ml and 68.11–93.45%, respectively. Co-crystal containing ketoprofen and PABA in molar ratio (1:1) was found to be optimized formulation batch. Physical characterization by X-ray diffraction spectra and differential scanning calorimetric studies confirms the crystallinity of prepared co-crystal. The half maximal inhibitory concentration (IC50) values for in-vitro anti-inflammatory activity comes out to be 34.04 µM for ketoprofen and 4.373µM for optimized formulation, exhibiting almost 8-fold amplification indicating higher anti-inflammatory effect of optimized batch as compared to drug ketoprofen. The results of in-vivo anti-inflammatory activity carried out by rat paw edema method revealed that the optimized batch of co-crystal preparation provided a significant % inhibition in paw volume in contrast to standard drug in wistar rats. In this case, a crystalline molecular complex of drug Ketoprofen, that demonstrate poor aqueous solubility, with p-aminobenzoic acid was recognized that further set out an improvement in solubility and also in anti-inflammatory activity of the drug in wistar rats.

In this study, we report the design, synthesis and the biological evaluation of 19 analogues of 2-mercapto benzoxazole coupled benzyl triazoles (BOTs) based on analysis of the binding site and literature of chemical space. These BOTs were evaluated both in vitro and in vivo for their anti-inflammatory activity. Eleven compounds showed less than 10 μM in vitro COX-2 enzyme activities. The most potent analogue among the BOT analogues were BOT15, BOT3 and BOT19 with IC50 3.40 μM, 4.50 μM and 4.57 μM respectively against COX-2. The in vivo anti-inflammatory activity of two BOTs has significantly higher than that of standard drug, ibuprofen. 2-Mercapto benzoxazole coupled benzyl triazoles (BOTs) were also tested for their antioxidant capacity and proved to be an as active scavenger, better than ascorbic acid.

Background: Inflammation is part of the complex biological response of vascular tissues to harmful stimuli. Though there are standard anti-inflammatory drugs like aspirin, diclofenac, etc., these are not free of side effects. This has led to an increase in demand for natural products with anti-inflammatory activity having less side effects. Hence the study was conducted to evaluate the anti-inflammatory activity of ethanolic extract of Ficus racemose (EEFR) in albino rats.Methods: Healthy albino rats of either sex were divided into 4 groups of 6 animals each. Group1-control, group 2-diclofenac sodium 2 mg/kg and group 3 and 4 EEFR (200 and 400 mg/kg respectively), anti-inflammatory activity was evaluated by Carrageenan induced paw oedema: formalin induced-peritonitis and cotton pellet induced granuloma model for in vivo activity and protein denaturation test for in vitro activity.Results: EEFR exhibited significant in vitro (p<0.001) anti-inflammatory effect at the dose of 200 and 400 mg/kg. EEFR produced 61.37% inhibition at the dose of 400 mg/kg and diclofenac (standard drug) produced 62.95% of inhibition after 3 hours of drug treatment in carrageenan induced paw oedema. The exudate volume was decreased in formalin induced peritonitis by EEFR and diclofenac significantly (p<0.001). In cotton pellet induced granuloma EEFR (400 mg/kg) and diclofenac showed decreased formation of granuloma by 28.36% and 28.00% (p<0.001) respectively.Conclusions: EEFR has significant anti-inflammatory activity in both acute and chronic model in a dose dependant manner in comparison with standard drug.

The present research work involves formulation of herbal gel containing stem bark extracts of Mesua ferrea Linn and its evaluation for in vitro anti-inflammatory activity. The gel formulations were prepared using ethanolic extracts along with different polymer. The physiochemical parameters of formulations like; pH, viscosity and spreadability etc. also evaluated. Phytochemical analysis revealed presence of phenols, flavonoids and alkaloids, etc. The ethanolic extracts showed appreciable anti-inflammatory activity compared to the standard drug. Study confirmed that potential anti-inflammatory formulation can be developed from bark extract of Mesua ferrea Linn.   Keywords: Mesua ferrea Linn., Anti-inflammatory, Herbal Gel, Phytochemical, Albumin denaturation.
