The main objective of the study was to determine the influence of breed on the traits of boar ejaculate: ejaculate volume (VOL, ml), sperm concentration (CON, x106 sperm/ml), total sperm count in ejaculate (TNS, x109 spermatozoa), sperm motility in native ejaculate (MON, %), sperm motility after dilution (MOD, %) and number of doses produced (NPD). The aim was also to evaluate the correlation of the boar performance test traits: average life daily gain (g), backfat thickness measured in two locations (mm), depth of longissimus dorsi muscle (mm) and carcass meat content (%) with ejaculate traits. Total of 931 ejaculates of 36 boars during reproductive exploitation were analysed (16 Landrace boars and 20 Large White boars). The effect was assessed using the procedure of the general linear model of the statistical package SAS 9.1.3 (SAS Inst. Inc., 2002- 2003). The model for analysis included the influence of breed, season and the linear regression influence of body weight at the end of the performance test. The correlation of the traits was determined by applying the Pearson?s correlation coefficient. Most of the examined ejaculate traits (VOL, CON, MOD and NPD) varied under the influence of boar breed (p<0.01; p<0.001). Weight at the end of the test (p<0.05; p<0.01; p<0.001) affected all examined traits, except CON and TNS. A weak association was found between production performance and ejaculate traits.

In the present study, the aim was to determine the impact of the following factors: age, farm, and gilt genotype, as well as the regression impact of body weight at the end of the performance test on the following tested properties: age at the end of the test/final age (FA), lifetime daily gain (LDG), the backfat thickness measured in two places (according to the Main Breeding program for Central Serbia), the depth of the long back muscle (BM) and the estimated lean meat content/meatiness (M). The study included two farms of pigs (farm 1 and farm 2), for three consecutive years (year 1, year 2 and year 3). The number of tested heads per year was 974 (year 1), 1311 (year 2) and 757 (year 3). The tested gilts were of Swedish Landrace, Large White and Duroc breeds. The gilts originated from 97 sires, while the number of daughters per sires ranged from 10 to 100. The results show that the Duroc animals were the oldest (245 days), which had the highest values for both measures of backfat thickness, but the lowest values for meatiness. In the third study year, the lowest average values were determined for the properties of the LDG, BM and M. The female animals from the farm 1 showed less growth/gain and had lower values for the estimated meatiness. As a result of the study, it was established that all included factors had a very high statistically significant influence on the variation of the tested properties (P <0.001), only the genotype of gilts showed a high statistically significant effect on the BM property (P <0.01).

At present great attention is paid to studying genetic regulation of farm animal adaptations to environmental conditions. This problem is very important due to a wide expansion of highly productive cattle breeds created in Europe and North America. However, until the present no investigation of changing semen quality in bulls of imported breeds during their adaptations to environmental conditions of Western Siberia has been conducted. The aim of this study was to investigate semen quality peculiarities and the diversity of morphological sperm abnormalities in bulls of imported and local breeds kept in the environmental conditions of the southern part of Western Siberia. We determined sperm concentration, sperm count, and rate of sperm with progressive motility and percentage of morphologically normal spermatozoa. The rate of sperm abnormalities according to Blome’s classifcation was determined too. It was found that the mean values of sperm concentration, sperm motility and percentage of morphologically normal spermatozoa in the bulls investigated were similar to those in bulls kept in European countries. Inter­breed differences in these parameters were not found. However, bulls of the Red Danish, Angler, and Simmental breeds had a higher percentage of misshapen sperm head and pyriform sperm head than bulls of the Black­White breed. An inter­strain difference in sperm motility in bulls of the Black­White breed was observed. It was found that bulls of Reflection Sovereign 198998 strain had lover sperm motility than bulls of Wis Burke Ideal 1013415 strain. No inter­strain differences in sperm production, percentage of morphologically normal spermatozoa and rate of main sperm abnormalities were found. Thus, it has been found that the environmental conditions of the southern part of Western Siberia do not seriously affect the sperm production, sperm motility or percentage of morphologically normal spermatozoa in bulls. However, the increased rate of misshapen and pyriform sperm heads in the bulls of the foreign breeds points to a need to study sperm DNA fragmentation.

The aim of this experimental study was to evaluate whether infertile patients may benefit from the evaluation of bio-functional sperm parameters in addition to the conventional semen analysis. To accomplish this, we evaluated the correlation between conventional and bio-functional sperm parameters based on their percentile distribution in search of a potential threshold of these latter that associates with conventional sperm parameter abnormalities. The study was conducted on 577 unselected patients with infertility lasting at least 12 months. We identified cut-off values according to the median of the population for mitochondrial membrane potential (MMP), number of alive spermatozoa, and chromatin abnormality. High MMP (HMMP) (≥46.25%) was associated with sperm concentration, sperm count, progressive motility, and normal form. Low MMP (LMMP) (≥36.5%) was found to be associated with semen volume, sperm concentration, total sperm count, progressive motility, total motility, and normal form. The number of alive spermatozoa (≥71.7%) was associated with sperm concentration and progressive motility whereas abnormal chromatin compactness (≥21.10%) was associated with sperm concentration, total sperm count, and progressive motility. The data would suggest that, for every increase in the percentile category of sperm concentration, the risk of finding an HMMP≤46.25 is reduced by 0.4 and by 0.66 for a total sperm count. This risk is also reduced by 0.60 for every increase in the percentile category of sperm progressive motility and by 0.71 for total sperm motility. Each increment of percentile category of the following sperm parameter was followed by a decrease in the risk of finding an LMMP≤36.5: sperm concentration 1.66, total sperm count 1.28, sperm progressive motility 1.27, total sperm motility 1.76, and normal form 1.73. Lastly, the data showed that, for every increase in the percentile category of total sperm count, the risk of finding an abnormal chromatin compactness ≤21.10 is reduced by 1.25 (1.04–1.51, p < 0.05) and an increase of total sperm motility is associated with a reduced risk by 1.44 (1.12–1.85, p < 0.05). Results suggest a correlation between bio-functional and conventional sperm parameters that impact the sperm fertilizing potential. Therefore, the evaluation of bio-functional sperm parameters by flow cytometry may be useful to explain some cases of idiopathic male infertility.

ABSTRACT Background It is unknown which compounds in spermatozoa or seminal plasma may be involved in the regulation of sperm motility. Objectives The aim of this study was to investigate the effects of DHA (22:6n–3), vitamin E, and their probable interactions in men with asthenozoospermia. Methods A factorial, randomized, double-blind, placebo-controlled trial was conducted in infertility clinics in Tehran, Iran. The participants were idiopathic asthenozoospermic men aged 20–45 y, with normal endocrine function. Their concentration of spermatozoa and percentage of morphologically normal spermatozoa were equal to or above the lower reference limits, according to the fifth edition of the WHO guideline. Out of 717 men referred to the infertility clinics, 180 asthenozoospermic men were randomly assigned to 1 of 4 groups according to stratified blocked randomization by age and sperm concentration. Participants took daily 465 mg DHA plus 600 IU vitamin E (DE), 465 mg DHA plus placebo (DP), 600 IU vitamin E plus placebo (EP), or both placebo capsules (PP) for 12 wk. Sperm characteristics, oxidative stress of seminal plasma, serum and sperm membrane fatty acids, dietary intakes, anthropometric measurements, and physical activity were measured at baseline and after 12 wk. Results After the intervention, mean ± SD sperm progressive motility was greater in the DE group (27.9 ± 2.8) than in the DP (25.7 ± 3.4), EP (26.1 ± 2.8), and PP (25.8 ± 2.6) groups (P &lt; 0.05). Sperm count (P = 0.001) and concentration (P = 0.044) increased significantly in the DE group compared with the other 3 groups, whereas other semen parameters were not significantly different between the groups after the intervention. Serum concentrations of n–3 PUFAs were significantly higher in the DE and DP groups than in the EP and PP groups. Conclusions Combined DHA and vitamin E supplements led to increased sperm motility; however, no significant changes occurred in sperm morphology and vitality in asthenozoospermic men. Visfatin encoded by the <I>NAMPT</I> gene <I>(nicotinamide phosphoribosyltransferase,</I> formerly also known as <I>PBEF – pre-B cell colony-enhancing factor</I>) is suggested to play a role in lipid metabolism and pathophysiology of diabetes mellitus type 2. A new microsatellite <I>SCZ004</I> was detected within intron 9 of the <I>NAMPT</I> gene. In Czech Large White (<I>n</I> = 95) frequencies of alleles <I>282, 286, 287, 299</I>, and <I>304</I> were 0.02, 0.39, 0.07, 0.04, and 0.48, respectively. Allele <I>286</I> was predominant also in Landrace (<I>n</I> = 11) and Black Pied Prestice (<I>n</I> = 11) breeds. Association analysis was carried out between previously reported SNP AM999341:g.669T>C in intron 9 of the <I>NAMPT</I> gene and backfat thickness, average daily gain and lean meat content in Czech Large White (<I>n</I> = 215), Black Pied Prestice (<I>n</I> = 96) and Landrace (<I>n</I> = 105). The <I>CC</I> genotype was associated with higher backfat thickness (<I ≤ 0.01) in Black Pied Prestice, however in Czech Large White <I>CC</I> was associated with lower backfat thickness when compared to <I>TT</I> and <I>CT</I> genotypes (<I ≤ 0.05). In Czech Large White, <I>CC</I> genotype was associated with higher lean meat content when compared to <I>CT</I> (not significant) and <I>TT</I> genotype (<I ≤ 0.05) while in Landrace <I>CC</I> had the lowest lean meat content when compared to <I>CT</I> and <I>TT</I> genotypes but only the difference between <I>CC</I> and <I>CT</I>T was statistically significant (<I ≤ 0.05). In Black Pied Prestice no association with lean meat content was found. Average daily gain was not associated with the SNP in any breed.

Perilipin, encoded by the <I>PLIN</I> gene, is a lipid droplet-associated phosphoprotein that functions as a key regulator of triacylglycerol storage and hydrolysis in adipocytes. In this study, structure and variability of the porcine <I>PLIN</I> gene were characterised. PCR fragments encompassing exons 1 to 9 and interspersed introns were sequenced and the obtained sequence was deposited in EMBL/GenBank/DDBJ databases (AM931171). Seven silent polymorphisms and one nonsynonymous polymorphism were detected in the coding sequence. The nonsynonymous polymorphism g.627G>A causing an amino acid substitution p.Val3Ile was found only in Meishan and Meishan × Piétrain cross. Associations were studied between loci g.4119A>G and g.7966T>C, and average daily gain (ADG), backfat thickness (BFT) or lean meat content (LMC) in 166 gilts from two herds. Due to supposed complete linkage disequilibrium between loci g.4119A>G and g.7966T>C only genotype combinations <I>AA-TT, AG-TC</I> and <I>GG-CC</I> were detected. A significant difference (<I>P</I> = 0.0290) between <I>GG-CC</I> and <I>AG-TC</I> genotype combinations for BFT in herd 2 and a suggestive difference (<I>P</I> = 0.0516) between <I>GG-CC</I> and <I>AA-TT</I> genotypes for ADG in herd 1 were detected.

AbstractThe data sets consisted of field performance data from 54 848 purebred and 16 175 crossbred animals (Czech data set, CZ) and 16 610 purebred and 9 228 crossbred animals (Slovak data set, SK). Animals from the following breeds were included: Duroc, Hampshire, Piétrain, sire line of Large White (CZ) or Yorkshire (SK), Czech Meat pig (CZ) or Slovak Meat pig (SK), Belgian Landrace (SK). Two-trait animal models were calculated for average daily gain from birth to the end of the field test (ADG) and lean meat content (LM, only in CZ) or backfat thickness (BF, only in SK). The models included additive breed and breed heterotic effects. Piétrain was the breed with the highest LM and the lowest BF. The additive genetic breed effect was about 1·5% LM (CZ) in comparison with Large White or −0·4 mm BF (SK) in comparison to Yorkshire. The sire line of Large White (CZ) or the Yorkshire breed (SK) clearly exceeded all the remaining sire breeds in ADG (on average by 30 to 50 g/day). There was a clear tendency to negative heterosis in LM in all crossbred combinations (CZ). In BF, heterotic effects between −0·2 mm and +0·3 mm were estimated, mostly not being significant (SK). The estimates of the heterotic effects for ADG were positive throughout. Higher values up to 40 g/day (7%) were observed in the Czech data set. Though heterotic breed effects are of some importance especially for ADG, their inclusion in the equations for breeding value estimation will have only a minor impact on the predicted breeding values.

Abstract Background Identification of relevant in vivo biomarkers for fertility remains a challenge for the livestock industry. Concentrations of the small peptide hormone oxytocin (OXT), involved in male reproductive function and present in the seminal plasma (SP) of several species could be a robust one. This study characterized concentrations of SP-OXT in ejaculates from boars used in artificial insemination (AI) programs aiming to evaluate its relationship with sperm quality variables and in vivo fertility of their liquid-stored AI-semen. Seminal OXT concentrations (ng/mL) were measured in 169 ejaculates from 61 boars of the Duroc, Pietrain, Landrace and Large White breeds using a direct competitive immunoassay test based on AlphaLISA® technology. Ejaculate (ejaculate volume, sperm concentration, total sperm count) and sperm parameters (motility, viability, intracellular generation of reactive oxygen species, plasma membrane fluidity) were assessed at 0 h and 72 h in AI-semen samples stored at 17 °C. In vivo fertility included only 18 Large White and Landrace boars whose AI-semen was used to inseminated > 100 sows and evaluated both farrowing rate and litter size of 3,167 sows. Results The results showed that SP-OXT differed between boars and between ejaculates within boar (P < 0.05) but not between breeds (Duroc, Pietrain, Landrace and Large White). Ejaculates with higher SP-OXT concentration/mL (hierarchically grouped; P < 0.001) had larger volume and came from younger boars (P < 0.05). Ejaculates of boars showing positive farrowing rate deviation exhibited higher (P < 0.05) SP-OXT concentration/mL than those with negative farrowing rate deviation. Conclusion The SP concentrations of OXT are boar, ejaculate and age dependent, and positively related with ejaculate volume and farrowing rates of liquid-stored semen AI-doses.

&nbsp;In this study association analyses were performed between genes tagged SNP (IGF2, NAMPT, DGAT1, MYF4, MC3R, MC4R and MYOD1), performance traits (backfat thickness, lean meat content, average daily gain from birth to the end of the test, average daily gain in test) and estimated breeding values (EBVs) in a population of Czech Large White sows (n = 101). Genotyping of all SNPs was performed by SNapShot with the exception of SNP within NAMPT gene for which HpaII PCR-RFLP assay was used. The following significant associations between genes tagged SNPs and traits or EBVs were found out: DGAT1 &ndash; lean meat content (AG &gt; AA, P &le; 0.05), MC4R &ndash; EBV for lean meat content (GG &gt; AA, P &le; 0.05; GG &gt; AG, P &le; 0.05), IGF2 &ndash; EBV for reproduction (piglets born alive in the second and subsequent parity) (AG &gt; AA, P &le; 0.05) and total EBV (AG &gt; AA, P &le; 0.01) and MC3R &ndash; EBV for average daily gain (CT &gt; TT, P &le; 0.05).

The aim of the research was to determine the dependence of physical properties of boar ejaculates on the time interval between successive ejaculate collections. The material for the study comprised 1954 ejaculates collected manually from boars used for artificial insemination. For all ejaculates, the ejaculate volume, sperm concentration, percentage of motile spermatozoa, and number of spermatozoa in the ejaculate were determined, and the number of insemination doses per ejaculate was calculated. The boars’ suitability for artificial insemination was evaluated taking into account the effect of the time passed between ejaculate collections. The ejaculate volume and the concentration and number of sperm in the ejaculate were shown to increase with the time elapsed since the previous ejaculate. The number of insemination doses obtained per ejaculate increased as well. Ejaculates with a high sperm count were obtained when the ejaculation frequency was no more than every 4 days. The percentage of spermatozoa with progressive motility increased with the time elapsed since the collection of the previous ejaculate, but only up to 4 days. Further decreases in ejaculation frequency no longer resulted in increased sperm motility.
