Abstract The aim of this study was to emphasize the importance of conservation of the Lipe sheep, as a local endangered type of Zackel breed, and to perform a comprehensive phenotypic characterization in order to ensure a better use and preservation of this genetic resource. In addition, we compared the results of a detailed morphometric characterization of the modern Lipe sheep carried out in the present study with the morphometric parameters of the native form of this breed described in 1935. The comparative analysis revealed the development dynamics of the local Lipe sheep in the traditional habitat, over a period of nearly one century. Throughout this period, different factors affecting Lipe sheep management, such as biogeografic, agro-economic, sociocultural and others, led to significant population erosion, and to the current status of the Lipe sheep as an endangered genetic resource. Although a slight increase has been registered in some body measurements, the major body indexes of the Lipe sheep, such as body format, body compactness, body massiveness, body proportion, pelvic, and head index, remained without significant fluctuations over the last century (p>0.05). Body length of the modern type still exceeds the height at withers, which was also recorded in the native Lipe sheep nearly 100 years ago. A prominent sexual dimorphism is still evident. Therefore, our comparative analysis showed no significant differences between the native and modern form of Lipe sheep in their body format and growth potential. The slight increases in absolute body measurements of the modern Lipe type we recorded could be linked to better housing conditions and improved quality of feed, without application of strategic selection measures over the last 100 years. The importance of conservation of this ovine resource is less economical, but mostly cultural, historical and heritage oriented, and still essential for the survival of the breed.

Subject. The working time of workers at any stage of economic development is a value reflecting the level of labor productivity. Any progress in productivity contributes to changes in the volume of labor costs and the number of employed. Depending on the relationship between the total volume of labor costs and the number of employed, the duration of working time per one worker may change (it may increase, decrease, or remain unchanged). Objectives. The study aims to confirm the importance of such a macroeconomic indicator as the number of employed in varying working hours. Methods. The study rests on the comparative analysis of countries with developed economies based on some indicators like dynamics of the working time fund, dynamics of the number of employed, average number of hours worked during the year per employee, etc. The analyzed timespan is 25 years (from 1991 to 2016). Results. The comparative analysis revealed that in the non-production sphere and the economy as a whole the macroeconomic determinants correlate so that the length of working time per worker reduces. When considering the analysis results for the manufacturing sector, no single trend was identified. Conclusions. One of the key factors affecting the change in working hours is the number of employed. The relationship between the working time fund and the number of employed directly determines the dynamics of working time per worker.

The factors affecting the TBM tunnel behavior in jointed rock mass is investigated. In the numerical models the concrete segment lining of TBM tunnel is concerned, which is simulated as a tube neglecting the segment joint. And the TBM tunnel construction process is simulate considering the excavation and installing of the segment linings. Some cases are analyzed with different joint orientation, joint spacing, joint strength and tunnel depth. The results show that the shape and areas of loosing zones of the tunnel are influenced by the parameters of joint sets and in-situ stress significantly, such as dip angle, spacing, strength, and the in-situ stress statement. And the stress and deformation of the tunnel lining are influenced by the parameters of joint sets and in-situ stress, too.

ABSTRACT Low signal intensity due to poor probe hybridization efficiency is one of the major drawbacks of rRNA-targeted in situ hybridization. There are two major factors affecting the hybridization efficiency: probe accessibility and affinity to the targeted rRNA molecules. In this study, we demonstrate remarkable improvement in in situ hybridization efficiency by applying locked-nucleic-acid (LNA)-incorporated oligodeoxynucleotide probes (LNA/DNA probes) without compromising specificity. Fluorescently labeled LNA/DNA probes with two to four LNA substitutions exhibited strong fluorescence intensities equal to or greater than that of probe Eub338, although these probes did not show bright signals when they were synthesized as DNA probes; for example, the fluorescence intensity of probe Eco468 increased by 22-fold after three LNA bases were substituted for DNA bases. Dissociation profiles of the probes revealed that the dissociation temperature was directly related to the number of LNA substitutions and the fluorescence intensity. These results suggest that the introduction of LNA residues in DNA probes will be a useful approach for effectively enhancing probe hybridization efficiency.

Arginine-phenylalanine-amide (RFamide)-related peptide 3 (RFRP-3, encoded by the Rfrp gene) is the mammalian ortholog of gonadotropin-inhibiting hormone and can inhibit GnRH neuronal activity and LH release. However, the development and regulation of the RFRP-3 system in both sexes is poorly understood. Using in situ hybridization, we examined changes in Rfrp-expressing neurons in mice of both sexes during development and under different adulthood hormonal milieus. We found no sex differences in Rfrp expression or cell number in adult mice. Interestingly, we identified two interspersed subpopulations of Rfrp cells (high Rfrp-expressing, HE; low Rfrp-expressing, LE), which have unique developmental and steroidal regulation characteristics. The number of LE cells robustly decreases during postnatal development, whereas HE cell number increases significantly before puberty. Using Bax knockout mice, we determined that the dramatic developmental decrease in LE Rfrp cells is not due primarily to BAX-dependent apoptosis. In adults, we found that estradiol and testosterone moderately repress Rfrp expression in both HE and LE cells, whereas the nonaromatizable androgen dihydrotestosterone has no effect. Using double-label in situ hybridization, we determined that approximately 25% of Rfrp neurons coexpress estrogen receptor-α in each sex, whereas Rfrp cells do not readily express androgen receptor in either sex, regardless of hormonal milieu. Lastly, when we looked at RFRP-3 receptors, we detected some coexpression of Gpr147 but no coexpression of Gpr74 in GnRH neurons of both intact and gonadectomized males and females. Thus, RFRP-3 may exert its effects on reproduction either directly, via Gpr147 in a subset of GnRH neurons, and/or indirectly, via upstream regulators of GnRH.

PurposeThe purpose of this study is to examine the factors influencing the transaction costs (TCs) in megaprojects to provide a basis for controlling project costs.Design/methodology/approachThis study selects six factors influencing the TCs in megaprojects from the perspective of TC theory and relational contract theory (RCT) through literature review. On the basis of crisp-set qualitative comparative analysis (QCA), this study tests combined factors influencing the TCs and the interaction between them.FindingsResults show that in megaprojects, TCs are affected by combination factors. The combination of asset specificity, uncertainty, transaction frequency and trust and the combination of asset specificity, reputation and trust will control TCs in certain situations. In the configuration leading to high project TCs, the combination of environmental and behavioral uncertainties is a necessary condition.Originality/valueThis paper fills up the research gap in the field of megaproject TCs, and researchers can focus on this field in the future.
