Summary This first comprehensive report from Punjab province of India relates to patho-physiological alterations alongwith morpho-molecular characterisation and risk assessment of natural infections of Hymenolepis diminuta and Hymenolepis nana in 291commensal rodents including house rat, Rattus rattus (n=201) and lesser bandicoot rat, Bandicota bangalensis (n=90). Small intestine of 53.61 and 64.95 % rats was found infected with H. diminuta and H. nana, respectively with a concurrent infection rate of 50.86 %. There was no association between male and female rats and H. diminuta and H. nana infections (ᵡ2 = 0.016 and 0.08, respectively, d.f.= 1, P>0.05), while the host age had significant effect on prevalence of H. diminuta and H. nana (ᵡ2 = 28.12 and 7.18, respectively, d.f.= 1, P≤0.05) infection. Examination of faecal samples and intestinal contents revealed globular shaped eggs of H. diminuta without polar filaments (76.50 ± 3.01μm x 67.62 ± 2.42 μm), while smaller sized oval eggs of H. nana were with 4 – 8 polar filaments (47.87 ± 1.95 μm x 36.12 ± 3.05 μm). Cestode infection caused enteritis, sloughing of intestinal mucosa, necrosis of villi and inflammatory reaction with infiltration of mononuclear cells in the mucosa and submucosa. Morphometric identification of the adult cestodes recovered from the intestinal lumen was confirmed by molecular characterisation based on nuclear ITS-2 loci which showed a single band of 269 bp and 242 bp for H. diminuta and H. nana, respectively. Pairwise alignment of the ITS-2 regions showed 99.46 % similarity with sequences of H. diminuta from USA and 100 % similarity with sequences of H. nana from Slovakia, Kosice.

Data on the dry weight of 410 worms from both male and female rats is shown not to differ significantly from the normal (Gaussian) frequency distribution. This finding justified the use of statistics based on this function.Host body weight is shown to have a highly significant (P < 0.01) effect upon worm dry weight. The heavier the rat, the smaller the worms. An experimental design taking rat body weight into consideration is shown to be up to 36% more efficient in demonstrating differences between groups than one that ignores this source of variation. The point of scolex attachment behind the pyloric sphincter also has a significant effect (P < 0.05) upon worm dry weight. The nearer the scolex is to the stomach, the smaller the worm.The distribution of worm biomass in the intestine does not follow a normal (Gaussian) frequency, but is both asymmetrical (P < 0.001) and flattened (P < 0.001). Over 50% of the parasite biomass lies within the second quarter of the intestine. The distribution of the median points of worm strobilae in the rat intestine is also asymmetrical (P < 0.01) with a peak in the zone which represents a distance of 30–35% from the stomach.The migration of H. diminuta within the rat intestine results in the greater part of the parasite body lying in the second quarter of the intestine. The median points of the strobilae are concentrated at the junction of the first and second quarters of the intestine. This region of the rat intestine appears to offer the optimum site for the growth of H. diminuta.

AbstractA total of 179 urban rats were sampled in the city of Doha in Qatar across the winter seasons (February–April) of 2002 and 2003. Only two parasites were identified, with overall prevalences of 35.8% and 41.3% for the cestodeHymenolepis diminutaand the fleaXenopsylla astiarespectively. The prevalence ofH. diminutawas markedly influenced by both year of study and host age, being higher in 2003 and amongst older rats. The abundance of infection ofH. diminutawas influenced by the year of study, host age and sex. Worm burdens in adult rats were almost twice as heavy in males compared with females and adults of both sexes harboured heavier infections than juveniles. The prevalence ofX. astiawas influenced by both year and host age, being higher in juvenile rats in 2002 and in adults in 2003. The abundance ofX. astiawas significantly higher in 2003 and both male and female rats showed similar abundances, but in 2003 females were more heavily infested. Reasons for this are discussed in relation to the differing foraging strategies shown by male and female rats. The prevalence and abundance profiles for bothH. diminutaandX. astiawere higher overall in 2003 due to a significant increase in the rat population density, although this did not reflect in any increase in parasite species richness. Rats that were infected withH. diminutawere almost twice as likely to be infected withX. astiathan those without the cestode, but when controlled for the effects of year, host age and sex, no quantitative interactions were detected between the two parasite species.

Rodent repellents are chemicals which by taste or odour or possibly by both will prevent animal from feeding or gnawing. Such substances may be used in protecting an area from rodent infestation or in protecting packaged food, packing materials, electric cables, and other important vulnerable materials. Mature and healthy house rat,Rattus rattusof both sexes, was exposed to 5, 10, and 20% eucalyptus oil applied as spray in laboratory pens in bichoice tests. Each concentration was applied through three different modes of application, that is, daily, once, and alternatively in a week. Repellent effect of the oil was assessed based on food consumption from treated and untreated sides for four days. In overall, food consumption was significantly (P<0.0001) low from treatment side compared to the untreated side indicating significant repellent effect of the oil at all the three concentrations tested. Repellent effect of the oil was, however, not found to differ significantly between the two sexes. Percent repellency in both male and female rats was apparently more with daily application of 5 and 10% eucalyptus oil. Present studies reveal the potential of eucalyptus oil in repelling awayR. rattus; however, further studies may be conducted to enhance the persistence of repellent effect for longer period of time.

AbstractA field study was conducted to disclose the prevalence of different helminth parasites in the black rat, Rattus rattus, in some regions of Stara Zagora district, Bulgaria. Out of 67 examined rats, 53 (79.1%) were found to be positive. Helminths of three classes were detected such as Cestoda: Hymenolepis diminuta (47.8%), Hymenolepis nana (43.3%), Taenia taeniaeformis larvae (7.5%), Taenia polyacantha larvae (1.5%); Secernentea: Syphacia obvelata (4.5%), Aspiculuris tetraptera (3.0%) and Adenophorea: Capillaria hepatica (9.0%), Trichuris muris (3.0%). Of all the investigated rats, 27 (40.3%) were infected by one helminth species and 26 (38.8%) by more than one. Hymenolepis diminuta was a predominant species in cases of single infections (23.9%). The most frequent co-infections were observed by H. nana and H. diminuta (16.4%).

Blood clotting response (BCR) test is a faster method with fewer ethical constraints than feed-testing and is widely used to monitor resistance. The second-generation anticoagulant, bromadiolone is being used to control rodents all over India for the last several years. To investigate whether its use has resulted in development of resistance in house rat, Rattus rattus, BCR test was standardized. Plasma samples of 15 rats of both the sexes were pooled separately and diluted with phosphate-buffered saline (pH 7.4). Ten different dilutions of plasma i.e. 4, 5, 6, 8, 10, 20, 40, 60, 80, 100% were tested for Prothrombin Time (PT) and International Normalized Ratio (INR) using rabbit brain thromboplastin reagent in coagulation analyzer. Time to clotting was converted into percent clotting activity (PCA) relative to plasma dilutions, to plot a standard curve. The PT of 15.7 and 17.6 sec for 100% plasma corresponded to INR of 0.8 and 0.9 in male and female rats, respectively. PCA of 22.3% corresponded with PT and INR of 70.3 sec and 3.8, respectively in male rats and PCA of 27.6% corresponded with PT and INR of 63.6 sec and 3.5, respectively in female rats. Data for PT and INR and standard curve for PCA vs PT prepared during present study can be further used to determine current status of bromadiolone resistance in R. rattus populations based on discriminating dose of bromadiolone.

The pineal gland synthesizes melatonin exclusively at night, which gives melatonin the characteristic of a temporal synchronizer of the physiological systems. Melatonin is a regulator of insulin activities centrally and also peripherally and its synthesis is reduced in diabetes.  Since monosodium glutamate (MSG) is often used to induce the type 2 diabetic and metabolic syndrome in animal models, the purpose of this work is to evaluate the potential effects of MSG given to neonates on the pineal melatonin synthesis in different aged male and female rats. Wistar rats were subcutaneously injected with MSG (4mg/g/day) or saline solution (0.9%) from the second to eighth post-natal day. The circadian profiles both melatonin levels and AANAT activity were monitored at different ages. Body weight, naso-anal length, adipose tissues weight, GTT, ITT and serum insulin levels were also evaluated. Typical obesity with the neonatal MSG treatment was observed, indicated by a great increase in adipose depots without a concurrent increase in body weight. MSG treatment did not cause hyperglycemia or glucose intolerance, but induced insulin resistance. An increase of melatonin synthesis at ZT 15 with phase advance was observed in in some animals. The AANAT activity was positively parallel to the melatonin circadian profile. It seems that MSG causes hypothalamic obesity which may increase AANAT activity and melatonin production in pineal gland. These effects were not temporally correlated with insulin resistance and hyperinsulinemia indicating the hypothalamic lesions, particularly in arcuate nucleus induced by MSG in early age, as the principal cause of the increase in melatonin production.

Young female Black rat (Rattus rattus), were administered monthly long acting steroid contraceptive to induce hypertriglyceridemia. It was observed that by 3 weeks of the second injection of estrogen containing mixed type of contraceptive, female rats developed consistent and frank hyperglyceridemia . TG in the treated rats was 195.8 ± 7.44 mg /100 ml as compared to 91.5 ± 6.27 mg/100ml in plasma of the control group.

ABSTRACT Oestriol (oestra-1,3,5(10)-triene-3,16α,17β-triol) is a weakly oestrogenic substance which, however, in contrast to what was formerly believed, is of physiological significance. Its effect is localized largely to the uterine cervix and vagina. Clinical experience argues both for and against an effect on the pituitary gland. This investigation is concerned with the morphological changes in the pituitary gland and adrenal cortex of gonadectomized male and female rats after the injection of oestriol. It was found that oestriol has the same type of action on these glands as other oestrogens, but under the experimental conditions used, this effect proved much weaker than that produced by oestradiol (oestra-1,3,5(10)-triene-3,17β-diol).

ABSTRACT Male and female rats of varying ages were placad on a low iodine diet (LID) plus KClO4 or 6-propyl-2-thiouracil (PTU) or on the same diet supplemented with I (control rats). Goitrogenesis was also induced with LID plus PTU in gonadectomized animals of both sexes. The weight of the control and goitrogen treated animals, and the weight and iodine content of their thyroids were determined, as well as the plasma PBI, TSH, insulin and glucose levels. The pituitary GH-like protein content was assessed by disc electrophoresis on polyacrylamide gels. If goitrogenesis was induced in young rats of both sexes starting with rats of the same age, body weight (B.W.) and pituitary growth hormone (GH) content, it was found that both the males and females developed goitres of the same size. On the contrary, when goitrogenesis was induced in adult animals, it was found that male rats, that had larger B.W. and pituitary GH content than age-paired females, developed larger goitres. However, both male and female rats were in a hypothyroid condition of comparable degree as judged by the thyroidal iodine content and the plasma PBI and TSH levels. When all the data on the PTU or KClO4-treated male and female rats of varying age and B.W. were considered together, it was observed that the weights of the thyroids increased proportionally to B.W. However, a difference in the slope of the regression of the thyroid weight over B.W. was found between male and female rats, due to the fact that adult male rats develop larger goitres than female animals. In addition, in the male rats treated with PTU, gonadectomy decreased the B.W., pituitary content of GH-like protein and, concomitantly, the size of the goitre decreased; an opposite effect was induced by ovariectomy on the female animals. However, when goitrogenesis was induced in weight-paired adult rats of both sexes, the male animals still developed larger goitres than the females. Among all the parameters studied here, the only ones which appeared to bear a consistent relationship with the size of the goitres in rats of different sexes, treated with a given goitrogen, were the rate of body growth and the amount of a pituitary GH-like protein found before the onset of the goitrogen treatment. Moreover, though the pituitary content of the GH-like protein decreased as a consequence of goitrogen treatment, it was still somewhat higher in male that in female animals. The present results suggest that GH may somehow be involved in the mechanism by which male and female rats on goitrogens develop goitres of different sizes, despite equally high plasma TSH levels.
