The prevalence of house mice (Mus musculus) Hymenolepiasis was determined in Baghdad, Iraq to study the effects of location, sex, and months on the infection rate of Hymenolepis spp. in house mice. Fifty house mice were captured from Abu Ghraib and Al-Ameriya, Baghdad, Iraq and examined for detecting parasites in laboratory in College of Veterinary Medicine, University of Baghdad. The total infection prevalence of intestinal parasites was 11 (22%) out of 50 samples, the higher prevalence was 28.57% and found in Abu Ghraib area, while lower was (13.63%) and recorded in Al-Ameriya area. The study revealed that the house mice were infected with two species of Hymenolepis: Hymenolepis nana 4 (8%) and Hymenolepis diminuta 7(14%). The intestinal parasites revealed a significance prevalence value (P<0.05). There was statistical difference in between males and females in parasites infection, in which the higher rate was 8 (27.58%) and recorded in males and the lower was 3 (14.28%) and observed in females. The monthly distribution of confirmed cases over a 9-month period revealed that reported cases of house mice and Hymenolepiasis increased significantly (P<0.01) in autumn (65.44%), followed by winter (15.38%), and summer (13.33%). The findings showed that house mice play an important role in the spread of zoonotic parasitic illnesses to people, as well as attention must pay to public health.

Abstract Tapeworms of zoonotic importance have been described as a leading public health problem. Current research was aim to assess the prevalence of tapeworms among 5-12years school children residing in district Lower Dir, Pakistan from January 2019-December 2019. The wet mount preparation in saline/iodine/methods were used for stool examination. Data was analyzed using appropriate descriptive, static methods. Of the 400 children studied 71.7% were infected with one or more species of intestinal parasites. Single infection of cestode species was found in 69 individuals with 17.2% prevalence and multiple parasitic infections were identified in 19.7% (n=79/400) individuals. The multiple infection were comprised as 10% (n=40) double, 6.75% (n=27) triple and 3% (n=12) quadruple. A total of 9 species of helminths and one species of protozoan infection. Among the helminths Ascaris lumbricoides was the most prevalent 33.1% (n=95), Taenia saginata 22.6% (n=65), hookworm 19.8% (n=57), Hymenolepis nana 18.8% (n=54), Enterobius vermicularis and Hymenolepis diminuta 1.39% (n=4each), Trichuris trichura 1.04% (n=3), Toxocara spp 0.69% (n=2) and Schistosoma japonicum 0.34% (n=1) were reported. One protozoan species was Cryptosporidium spp 0.69% (n=2) in current study. In case of A.lumbricoides, hookworm, E.vermicularis, T.trichura, T.saginata, H.nana and H.diminuta the male children of below 8 years of age were highly infected. Other infections are reported in the same prevalence with slight difference if any. We conclude that there is a need for mass scale campaigns to create awareness regarding health and hygiene in children and the need for development of effective poverty control programs because deworming alone is not adequate to control parasitic infections.

Abstract The study reveals the prevalence of a potential rodent-borne zoonotic helminth species, Hymenolepis diminuta in commensal rodents caught from irrigated and rain-fed areas of Swat, Pakistan. Three hundred and fifty rodents (269 rats and 81 mice) trapped during vegetative, flowering/fruiting and mature/harvesting stages of crops were studied from 2011-2013. Hymenolepisdiminuta eggs were identified on the basis of their shape, size, colour and markings on the surface of the egg shell and three pairs of embryonic hook-lets. Overall prevalence of H. diminuta was 3.14% (n=11/350). The highest prevalence 3.49% (n=5/143) of H. diminuta was noted at harvesting stages of the crops whereas the lowest 2.59% (n=2/77) during vegetative stage. Infection was higher in males 3.25% (n=7/215) than females 2.96% (n=4/135). Adult rodents were highly infected while no sub-adult was found infected. Infection was higher in mice 3.70% (3/81) than rats 2.69% (8/269) while no significance (p=1.0000:0.1250 to 32.00 CI). Rats and mice appears to show the most suitable reservoirs by hosting H. diminuta a zoonotic helminth. The presence of these rodents in all possible habitats can act as a main channel of transferring parasites through various habitats and can pose a hazard to humans in the area.

Summary Tapeworms parasitize at sites that are important for the management of micronutrients, including zinc. Therefore, it has been hypothesized that tapeworms will significantly affect the excretion of zinc in the feces of a host. The aim of this work was to evaluate the effects that tapeworms have on the excretion of zinc in the feces of the host. Rats were divided into 4 groups: groups 0T and MT (infected with Hymenolepis diminuta (Rudolphi, 1819)) and groups 00 and M0 (uninfected). The experimental groups (M0 and MT) were fed a standard rodent compound feed (ST-1) with added zinc lactate; the daily zinc intake was 20.5 mg. The control groups (00 and 0T) were fed only ST-1 with 1.75 mg of added Zn per day. For six weeks, the amount of consumed feed was recorded and fecal samples were taken. The samples were then analyzed by optical emission spectrometry (ICP-OES), and levels of excreted zinc were subsequently calculated as a percentage. The most signifi cant difference in zinc excretion levels between the experimental groups was observed in the third week, when rats infected with tapeworms (MT) excreted substantially lower levels of zinc than did uninfected rats (M0). This difference amounted to 28.36 % (p <0.01). In the control groups, tapeworms affected the excretion of zinc in the feces to a lesser extent, and the most substantial difference in zinc levels was seen in the fifth week (8.46 %). However, there was no signifi cant difference in zinc excretion levels between the control groups during any of the monitored weeks. Tapeworms in the host affect levels of zinc excreted in the feces. However, this is dependent on the amount or form of zinc ingested.

RESUMO A concepção dos sistemas de drenagem de águas pluviais como separador absoluto é uma característica marcante na gestão das águas urbanas, no entanto não é o reflexo da realidade de tal sistema na maioria das cidades brasileiras, onde frequentemente esgotos e águas pluviais escoam pelos mesmos condutos. Diante disso, o objetivo do presente trabalho foi investigar as contribuições de contaminação fecal dos canais de drenagem afluentes ao Canal do Prado, pertencentes ao sistema de macrodrenagem de águas pluviais da cidade de Campina Grande, na Paraíba. Para a realização da pesquisa de campo, foram definidos sete pontos de monitoramento ao longo do canal e, como indicadores de contaminação fecal, coliformes termotolerantes, Escherichia coli e os ovos dos helmintos Ascaris lumbricoides, Taenia sp, Hymenolepis nana, Hymenolepis diminuta, Enterobius vermicularis, Ancilostomideo sp e Trichuris sp. Os resultados apontaram que ovos de Ascaris lumbricoides foram os que apresentaram maior frequência nos pontos de monitoramento, relacionados à maior descarga de esgotos sanitários. Tais descargas também foram verificadas por elevados valores médios de coliformes termotolerantes que atingiram até 1,6 x 107 UFC.100 mL-1, valor típico de esgoto sanitário. Escherichia coli ocorreu em mais de 90% das amostras coletadas em todos os pontos de monitoramento.

Abstract Background Studies on the inhibition of inflammation by infection with helminth parasites have, until recently, overlooked a key determinant of health: the gut microbiota. Infection with helminths evokes changes in the composition of their host’s microbiota: one outcome of which is an altered metabolome (e.g., levels of short-chain fatty acids (SCFAs)) in the gut lumen. The functional implications of helminth-evoked changes in the enteric microbiome (composition and metabolites) are poorly understood and are explored with respect to controlling enteric inflammation. Methods Antibiotic-treated wild-type, germ-free (GF) and free fatty-acid receptor-2 (ffar2) deficient mice were infected with the tapeworm Hymenolepis diminuta, then challenged with DNBS-colitis and disease severity and gut expression of the il-10 receptor-α and SCFA receptors/transporters assessed 3 days later. Gut bacteria composition was assessed by 16 s rRNA sequencing and SCFAs were measured. Other studies assessed the ability of feces or a bacteria-free fecal filtrate from H. diminuta-infected mice to inhibit colitis. Results Protection against disease by infection with H. diminuta was abrogated by antibiotic treatment and was not observed in GF-mice. Bacterial community profiling revealed an increase in variants belonging to the families Lachnospiraceae and Clostridium cluster XIVa in mice 8 days post-infection with H. diminuta, and the transfer of feces from these mice suppressed DNBS-colitis in GF-mice. Mice treated with a bacteria-free filtrate of feces from H. diminuta-infected mice were protected from DNBS-colitis. Metabolomic analysis revealed increased acetate and butyrate (both or which can reduce colitis) in feces from H. diminuta-infected mice, but not from antibiotic-treated H. diminuta-infected mice. H. diminuta-induced protection against DNBS-colitis was not observed in ffar2−/− mice. Immunologically, anti-il-10 antibodies inhibited the anti-colitic effect of H. diminuta-infection. Analyses of epithelial cell lines, colonoids, and colon segments uncovered reciprocity between butyrate and il-10 in the induction of the il-10-receptor and butyrate transporters. Conclusion Having defined a feed-forward signaling loop between il-10 and butyrate following infection with H. diminuta, this study identifies the gut microbiome as a critical component of the anti-colitic effect of this helminth therapy. We suggest that any intention-to-treat with helminth therapy should be based on the characterization of the patient’s immunological and microbiological response to the helminth.

Summary This first comprehensive report from Punjab province of India relates to patho-physiological alterations alongwith morpho-molecular characterisation and risk assessment of natural infections of Hymenolepis diminuta and Hymenolepis nana in 291commensal rodents including house rat, Rattus rattus (n=201) and lesser bandicoot rat, Bandicota bangalensis (n=90). Small intestine of 53.61 and 64.95 % rats was found infected with H. diminuta and H. nana, respectively with a concurrent infection rate of 50.86 %. There was no association between male and female rats and H. diminuta and H. nana infections (ᵡ2 = 0.016 and 0.08, respectively, d.f.= 1, P>0.05), while the host age had significant effect on prevalence of H. diminuta and H. nana (ᵡ2 = 28.12 and 7.18, respectively, d.f.= 1, P≤0.05) infection. Examination of faecal samples and intestinal contents revealed globular shaped eggs of H. diminuta without polar filaments (76.50 ± 3.01μm x 67.62 ± 2.42 μm), while smaller sized oval eggs of H. nana were with 4 – 8 polar filaments (47.87 ± 1.95 μm x 36.12 ± 3.05 μm). Cestode infection caused enteritis, sloughing of intestinal mucosa, necrosis of villi and inflammatory reaction with infiltration of mononuclear cells in the mucosa and submucosa. Morphometric identification of the adult cestodes recovered from the intestinal lumen was confirmed by molecular characterisation based on nuclear ITS-2 loci which showed a single band of 269 bp and 242 bp for H. diminuta and H. nana, respectively. Pairwise alignment of the ITS-2 regions showed 99.46 % similarity with sequences of H. diminuta from USA and 100 % similarity with sequences of H. nana from Slovakia, Kosice.

Background and Aim: Some rat cestodes are zoonotic and are capable of parasitizing humans and animals, raising serious concerns regarding human and veterinary health. The study aimed to determine the prevalence and risk factors for cestodes in Egyptian house rats and to characterize the cestodes molecularly. Materials and Methods: The current survey examined 115 house rats (Rattus rattus) in two cities (Edfu and Aswan) in Egypt's Aswan Governorate for cestode infection using integrated molecular approaches (polymerase chain reaction, sequencing, and phylogenetic analysis) and morphological/morphometrical approaches. Results: The cestodes identified in this study exhibited the typical morphological characteristics of Hymenolepis diminuta (Rudolphi, 1819), Hymenolepis nana (Siebold, 1852) (from rat intestine), and Hydatigera taeniaeformis (from rat liver). The species prevalence rates from these three studies were reported to be 8.7%, 10.4%, and 20.9%, respectively. The ribosomal DNA (ITS1, 18S, and complete ITS) sequences revealed that the hymenolepid sequences were highly distinct but were related to other sequences in the GenBank database, with some sequences showing high similarities to those of H. nana and H. diminuta. In addition, the H. taeniaeformis sequences (ITS2 and mitochondrial cytochrome c oxidase subunit 1 [mtCOX1]) obtained in this study were highly similar to some Taenia taeniaeformis GenBank sequences. The constructed phylogram revealed that the hymenolepidid tapeworms examined in this study were classified into four major branches (the majority of which were hybrids of the two species) and belonged to the genus Hymenolepis. In addition, the phylogram of H. taeniaeformis assigned this species to T. taeniaeformis. Conclusion: When typical hymenolepid morphology is combined with molecular and phylogenetic divergence, it may indicate the existence of possible cryptic species. In addition, on the basis of the phylogenetic analysis, genetic diversity within T. taeniaeformis may exist as determined by comparing the metacestode mtCOX1 sequences. The current study presents the prevalence values of zoonotic cestodes and contributes to the body of knowledge, including identification keys and the use of molecular tools for species confirmation.

The study is dedicated to the investigation of serotonin (5-hydroxytryptamine, 5-HT) and 5-HT7 type serotonin receptor of localisation in larvae of two parasitic flatworms Opisthorchis felineus (Rivolta, 1884) Blanchard, 1895 and Hymenolepis diminuta Rudolphi, 1819, performed using the immunocytochemical method and confocal laser scanning microscopy (CLSM). Using whole mount preparations and specific antibodies, a microscopic analysis of the spatial distribution of 5-HT7-immunoreactivity(-IR) was revealed in worm tissue. In metacercariae of O. felineus 5-HT7-IR was observed in the main nerve cords and in the head commissure connecting the head ganglia. The presence of 5-HT7-IR was also found in several structures located on the oral sucker. 5-HT7-IR was evident in the round glandular cells scattered throughout the larva body. In cysticercoids of H. diminuta immunostaining to 5-НТ7 was found in flame cells of the excretory system. Weak staining to 5-HT7 was observed along the longitudinal and transverse muscle fibres comprising the body wall and musculature of suckers, in thin longitudinal nerve cords and a connective commissure of the central nervous system. Available publications on serotonin action in flatworms and serotonin receptors identification were reviewed. Own results and the published data indicate that the muscular structures of flatworms are deeply supplied by 5-HT7-IR elements. It suggests that the 5-HT7 type receptor can mediate the serotonin action in the investigated species and is an important component of the flatworm motor control system. The study of the neurochemical basis of parasitic flatworms can play an important role in the solution of fundamental problems in early development of the nervous system and the evolution of neuronal signalling components.
