Impact of soil sterilization and irrigation intervals on P and K acquisition by mycorrhizal onion (Allium cepa)

AbstractDrought is a world-spread problem seriously influencing crop production. Arbuscular mycorrhizal (AM) association and soil microorganisms can help plant growth under water stress condition by improvement of its nutrient and water uptake. In this experiment, onion plants (Allium cepa L. cv. Red Azar Shahr) were inoculated with three AM fungi species (Glomus versiforme, G. intraradices, G. etunicatum) or left un-inoculated as non-mycorrhizal plants, in a sterile or non-sterile sandy loam soil. Plants were irrigated at 7, 9 or 11-day intervals to keep the soil moisture content to field capacity at the irrigation time. Mycorrhizal root colonization decreased (p < 0.05) with an increase in irrigation interval, and the highest root colonization was achieved at 7-day irrigated onions in symbiosis with G. versiforme. Phosphorus content in plant tissue was significantly increased in mycorrhizal than non-mycorrhizal onions. Plants inoculated with G. versiforme at 9-day interval treatment had the highest leaf P content, while the lowest P was observed in non-mycorrhizal plants at all irrigation intervals. Onions inoculated by G. versiforme or G. etunicatum at 9-day irrigation interval had the highest K content. Results revealed that the inoculation of onion plant with G. versiforme or G. etunicatum and increasing irrigation interval up to 9 days, could improve P and K uptake.

This study aimed to investigate the effects of arbuscular mycorrhizal (AM) fungi with different levels of NPK fertilizers on yield production of common bean plants which common bean plants were subjected to five levels of NPK fertilizers (0, 25, 50, 75, 100 %). Application of AMF significantly increased the growth and yield components of common beans with minimized the levels of NPK comparing to equivalents non-mycorrhizal ones. The results obtained revealed that inoculation with AMF and the concentrations 50% and 75% of NPK with AMF are the greater than other concentrations and non-mycorrhizal plants. Mycorrhizal Common bean plants had significantly higher number of pods, length of one pod, pods weight, 100 seeds weight, weight of seed/plant and intensity of mycorrhizal colonization(M%) . Concentrations of nutrients (N, P, K, Ca and Mg) and total carbohydrates, crude protein and mycorrhizal dependency of some yield parameters were significantly increased in mycorrhizal plants at different NPK levels when comparing to those of non-mycorrhizal plants paticularly at (50% and 75%) concentration of NPK, but lower Na concentration in mycorrhizal common bean seeds than those of non-mycorrhizal.Int J Appl Sci Biotechnol, Vol 4(2): 191-197

Clitoria ternatea L. is an excellent herbal medicinal plant. Arbusular Mycorrhizal fungal (AMF) colonization and biomass of three different tested conditions of Clitoria ternatea plant was investigated. Inoculums of indigenous AMF and Trichoderma harzianum was tested greenhouse experiment and compared with natural condition after 60th days of treatment. Percentage of Arbuscular mycorrhizal (AM) infection, number of resting spores and AM fungi species varies in different land. Among three different conditions, natural conditions showed maximum root colonization (75.89%) than treated one but minimum spore density (358.8/100gsoil). Highest spore density (481.6/100g soil) was found in T. harzianum treated condition followed by indigenous AMF treatment Acaulospora Glomus and Sclerocystis these three genera were found frequently. AMF inoculums and T.harzianum treatments conditions were observed promising biomass data of 60th days after treatment (DAT).When AMF are more colonized to plants then enhanced the biomass productivity.

Araucaria angustifolia (Bert.) O. Ktze. is an endangered Brazilian coniferous tree that has been almost exterminated in the native areas because of uncontrolled wood exploitation. This tree has been shown to be highly dependent on arbuscular mycorrhizal fungi (AMF) and, therefore, AMF may be essential for forest sustainability and biological diversity. Root colonization, density and diversity of AMF spores were assessed in two Araucaria forest stands at the State Park of Alto Ribeira (PETAR), at two sampling dates: May and October. A comparison was made between a mature native stand composed of Araucaria trees mixed into a variety of tropical trees and shrubs, without any sign of anthropogenic interference (FN) and an Araucaria stand planted in 1987 (R), which has been used as a pasture. Assessments included percent root colonization, AMF spore numbers and species richness, Simpson's dominance index (Is), and Shannon's diversity index (H). Mycorrhizal root colonization did not differ between ecosystems in May. In October, however, the native stand (FN) presented a higher colonization than the planted forest (R), and the root colonization was more intense than in May. When considering both sampling periods and forests, 27 species of AM fungi, with higher numbers of spores in FN than in R were found. Canonical discriminant analysis (CDA) indicated Shannon's diversity index as the ecological attribute that contributed the most to distinguish between forest ecosystems, with higher value of H in FN in relation to R. CDA showed to be a useful tool for the study of ecological attributes.

Pyrolyzed biomass waste, commonly called biochar, has attracted interest as a soil amendment. A commercial prototype biochar produced by fast pyrolysis of hardwood dust was examined in soils to determine if it could reduce the damaging effect of allelopathy on arbuscular mycorrhizal (AM) root colonization and on Fusarium crown and root rot of asparagus. In greenhouse studies, biochar added at 1.5 and 3.0% (wt/wt) to asparagus field soil caused proportional increases in root weights and linear reductions in the percentage of root lesions caused by Fusarium oxysporum f. sp. asparagi and F. proliferatum compared with a control. Concomitant with these effects was a 100% increase in root colonization by AM fungi at the 3.0% rate. Addition of aromatic acids (cinnamic, coumaric, and ferulic) that are known allelopathic agents affecting asparagus reduced AM colonization but the deleterious effects were not observed following the application of biochar at the higher rate. When dried, ground, asparagus root and crown tissues infested with Fusarium spp. were added to soilless potting mix at 0, 1, or 5 g/liter of potting mix and then planted with asparagus, there was a decrease in asparagus root weight and increase in disease at 1 g/liter of potting mix but results were inconsistent at the higher residue rate. However, when biochar was added at 35 g/liter of potting mix (roughly 10%, vol/vol), these adverse effects on root weight and disease were equal to the nontreated controls. A small demonstration was conducted in field microplots. Those plots amended with biochar (3.5% [wt/wt] soil) produced asparagus plants with more AM colonization in the first year of growth but, in the subsequent year, biochar-treated plants were reduced in size, possibly due to greater than average precipitation and the ability of biochar to retain moisture that, in turn, may have created conditions conducive to root rot. These studies provide evidence that biochar may be useful in overcoming the deleterious effects of allelopathic residues in replant soils on asparagus.

Leguminous plants have a genetic system that provides interaction with different beneficial soil microorganisms (BSM). The system has been formed on the basis of the genetic mechanisms that had arisen during the co-evolution of plants with arbuscular-mycorrhizal (AM) fungi and appeared to provide pre-adaptations for further evolution of interaction with various BSM. A concept of the use of BSM in sustainable agriculture is proposed, which postulates an establishment of the multi-component beneficial plant-microbe communities based on varieties of legumes with high potential for interaction with the BSM.

Drought is a major constraint in drylands for crop production. Plant associated microbes can help plants in acquisition of soil nutrients to enhance productivity in stressful conditions. The current study was designed to illuminate the effectiveness of desert rhizobacterial strains on growth and net-return of chickpeas grown in pots by using sandy loam soil of Thal Pakistan desert. A total of 125 rhizobacterial strains were isolated, out of which 72 strains were inoculated with chickpeas in the growth chamber for 75 days to screen most efficient isolates. Amongst all, six bacterial strains (two rhizobia and four plant growth promoting rhizobacterial strains) significantly enhanced nodulation and shoot-root length as compared to other treatments. These promising strains were morphologically and biochemically characterized and identified through 16sRNA sequencing. Then, eight consortia of the identified isolates were formulated to evaluate the growth and development of chickpea at three moisture levels (55%, 75% and 95% of field capacity) in a glass house experiment. The trend for best performing consortia in terms of growth and development of chickpea remained T2 at moisture level 1 > T7 at moisture level 2 > T4 at moisture level 3. The present study indicates the vital role of co-inoculated bacterial strains in growth enhancement of chickpea under low moisture availability. It is concluded from the results that the consortium T2 (Mesorhizobium ciceri RZ-11 + Bacillus subtilis RP-01 + Bacillus mojavensis RS-14) can perform best in drought conditions (55% field capacity) and T4 (Mesorhizobium ciceri RZ-11 + Enterobacter Cloacae RP-08 + Providencia vermicola RS-15) can be adopted in irrigated areas (95% field capacity) for maximum productivity of chickpea.

Industrialized tomato production faces a decrease in flavors and nutritional value due to conventional breeding. Moreover, tomato production heavily relies on nitrogen and phosphate fertilization. Phosphate uptake and improvement of fruit quality by arbuscular mycorrhizal (AM) fungi are well-studied. We addressed the question of whether commercially used tomato cultivars grown in a hydroponic system can be mycorrhizal, leading to improved fruit quality. Tomato plants inoculated with Rhizophagus irregularis were grown under different phosphate concentrations and in substrates used in industrial tomato production. Changes in fruit gene expression and metabolite levels were checked by RNAseq and metabolite determination, respectively. The tests revealed that reduction of phosphate to 80% and use of mixed substrate allow AM establishment without affecting yield. By comparing green fruits from non-mycorrhizal and mycorrhizal plants, differentially expressed genes (DEGs) were found to possibly be involved in processes regulating fruit maturation and nutrition. Red fruits from mycorrhizal plants showed a trend of higher BRIX values and increased levels of carotenoids in comparison to those from non-mycorrhizal plants. Free amino acids exhibited up to four times higher levels in red fruits due to AM, showing the potential of mycorrhization to increase the nutritional value of tomatoes in industrialized production.

Little attention has been paid to the effect of arbuscular mycorrhizal (AM) fungi on the uptake of nutrients that move mainly by mass flow. The objective of this study was to assess the possible contribution of indigenous AM fungi to the K, Ca and Mg nutrition of maize (Zea mays L.) as influenced by soil P levels and its impact on plant dry mass. The field experiment had a split plot design with four replicates. Treatments included soil fumigation status (fumigation and non-fumigation) and three levels of P fertilization (0, 60 and 120 kg P2O5 ha-1) in a loamy sand soil in 1997 and a fine sandy loam soil in 1998. Soil fumigati on with Basamid® was used to suppress indigenous AM fungi. Plants were sampled at four different growth stages (6-leaf stage, 10-leaf stage, tasseling and silking). Soil fumigation decreased shoot dry weight, but P fertilization increased shoot dry weight at most sampling times. When no P fertilizer was added, fumigation in the loamy sand soil reduced shoot K and Ca concentrations while, in contrast, in the fine sandy loam soil only Mg concentration was reduced by soil fumigation. The concentration of K in maize shoots was positively correlated (P < 0.05) with extraradicular hyphal length in both soils. The correlation between the abundance of extraradicular hyphae and the concentrations of Ca and Mg in maize shoots was significant only for soils where available Ca or Mg was relatively low. Arbuscular mycorrhizal fungi could increase corn biomass production and K, Ca and Mg uptake in soil low in these elements and low in P. These results indicate that the contribution of mycorrhizae to maize K, Ca and Mg nutrition can be significant in a field situation and that the extent of this contribution depends on the availability of these nutrients and of P in soils. The study was conducted to determine the effect of arbuscular mycorrhizal (AM) fungi inoculation on growth of pea grown under water stressed pot culture conditions. Water stress was given to the pea plants after 30 days at the interval of 4, 8 and 12 days. The data was collected at an interval of 15 days. Three replicates of each set were maintained. . The mixture of AM fungi used for current experiment included the species of Acaulospora denticulata, A. gerdemannii, Glomus macrocarpum, G. maculosum, G. fasciculatum and Scutellospora minuta. The mycorrhizal plants have shown more shoot and root length as compared to the control plants. The height of shoot and root was significantly decreased with the increase in drought stress. Mycorrhizal plants with low water stress showed enhanced shoot and root length than high water stress. The mycorrhizal plants have shown more number of leaves than control plants during drought stress. The number of leaves significantly reduced with the increase in drought stress. The leaves produced by the control plants were comparatively smaller than those of mycorrhizal plants. The dry weight of root and shoot of both control and mycorrhizal plants decreased with the increase in water stress. Mycorrhizal plants showed more dry weight of shoot and root as compared to control plants.  Plants inoculated with AM fungi produce more dry weight than the control plants. The fresh weight of both control and mycorrhizal plants has been decreased with the increase in water stress interval and also the fresh weight of root and shoot was observed higher in mycorrhizal plants as compared to those of control plants.
