In vivo antioxidant potential of Teucrium polium, as compared to α-tocopherol The present study was undertaken to explore antioxidant potential of Teucrium polium (Lamiaceae) in vivo. Antioxidant activity was measured by three tests including inhibition of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, total antioxidant power (TAP), and thiobarbituric acid reactive substances (TBARS) in serum. Rats received dry extract of T. polium in 80% ethanol by intragastric intubation at doses of 50, 100 and 200 mg kg-1 daily for 14 days. Treatment of rats with T. polium extract showed significant antioxidant activity in the DPPH test as compared to the control. T. polium extract at doses of 50 and 100 mg kg-1 significantly increased rats TAP and decreased TBARS compared to the control. Administration of T. polium at a dose of 200 mg kg-1 per day did not significantly alter serum TAP and TBARS. Antioxidant activities of T. polium at a doses of 50 and 100 mg kg-1 were in all experiments comparable to that of α-tocopherol (10 mg kg-1).

Rheum emodiWall has been reported to possess protective effect in many inflammatory diseases and oxidative stress-related injuries. This study aims to investigate antioxidant power of stilbenoids fromR. emodiand then explore the material basis for its antioxidant potential. The most abundant stilbenoid piceatannol-4′-O-β-D-glucopyranoside (PICG) and its aglycon piceatannol (PICE) were isolated fromR. emodirhizome. Using well-accepted antioxidant chemicals as reference, antioxidant activity of these stilbenoids was examined by measuring DPPH and superoxide anion radical scavenging, ferric reducing power, and inhibition of lipid peroxidationin vitro. Both PICG and PICE displayed promising antioxidant activity in all the four assays. Comparisons among the tested compounds indicated that PICE has the most potent antioxidant activity and the presence of 3′-hydroxyl group may enhance antioxidant activity of stilbenoids. The antioxidative effect of PICE at the cellular level was further demonstrated on the model of hydrogen-peroxide-induced H9c2 rat cardiomyoblasts injury. Taking into account the rapidin vivometabolic transformation of PICG into PICE it can be inferred that the most abundant stilbenoid PICG may be an important constituent responsible for the antioxidant potential ofR. emodiand promising to be developed as an antioxidant agent for supplementary or therapeutic use.

Fat-enriched diet is strongly associated with cataract development. Laurus nobilis shows antioxidant activity. Herein we evaluated the effect of Laurus nobilis oral administration on the blood and lenses antioxidant activity in rabbits under fat-enriched diet. Sixty rabbits divided into 4 groups were used. One group represented the control (N-CTR). The second group (P-CTR) fed a diet supplemented with 2.5 % of pig fat; the third group (EXP1) received a diet supplemented with 2.5 % of pig fat and 1 g/kg of dried-bay leaves; the fourth group (EXP2) was treated with dried-bay leaves at the rate of 1 g/kg of feed. At baseline and at the end of the study (56 days) the following blood parameters were determined: thiobarbituric acid reactive substances (TBARS), reactive oxygen metabolites (ROMs), total phenols, superoxide dismutase (SOD), oxygen radical absorbance capacity (ORACpca), ferric ion reducing antioxidant power (FRAP), retinol and alfa-tocopherol. At the end of the follow-up, the eyes were enucleated and the antioxidant profile, such as total antioxidant activity (TAC), TBARS, retinol and alfa-tocopherol of lenses was evaluated. Plasma ROMs and TBARS levels were statistically lower in the groups receiving bay leaves integration. A significant increase of plasma retinol, FRAP and ORACpca levels was found in EXP1 and EXP2 groups, whereas plasma alfa-tocopherol resulted statistically higher only in EXP2 group. Bay leaves supplementation enhanced TAC, retinol and alfa-tocopherol in rabbit lens, particularly in EXP2 group; whereas lenses TBARS levels significantly decreased in both treated groups. These findings demonstrate that Laurus nobilis oral administration exerts a protective effect on the risk of cataract development in rabbits under fat-enriched diet.

The antioxidant activity and bioactive compounds of different blackberry fruit genotypes from the major Brazilian producer region (three cultivars and four selections) were evaluated and compared to the Cherokee cultivar. Phenolic and anthocyanic extracts were obtained and evaluated for each fruit genotype. The phenolic extracts of selections 02/96 and 07/001 presented higher antioxidant activity than those of cultivars in most assays. This activity was partially correlated to the higher amount of total phenolics in these samples. Thus, the phenolic compounds are probably the major responsible for the antioxidant activity in the diphenyl-2-picrylhydrazyl radical scavenging assay (DPPH), ferric reducing antioxidant power (FRAP) assay and thiobarbituric acid reactive substances (TBARS) assay. Quercetin seems to be responsible for the antioxidant activity of blackberry phenolic extracts in the β-carotene bleaching assay. Concerning anthocyanic extracts, the selection 02/96 and Cherokee cultivar from harvest 2007 had higher antioxidant activity than the other genotypes in most assays. Anthocyanins appear to be the major responsible for the antioxidant activity of anthocyanic extracts in the DPPH and FRAP assays, although ascorbic acid also contributed to the DPPH antioxidant activity. Selection 02/96 appears to have higher antioxidant activity than the commercial cultivars cultivated in the southern Brazil and appears to be promising for nutritional and health purposes.

Abstract Background The present study was focused on the optimization of yield of the essential oil extraction from leaves of Lawsonia inermis, and the determination of chemical composition, antioxidant activities, and lipid peroxydation and antiproliferative effects. Methods Henna essential oil (HeEO) were extracted by hydrodistillation; the identification of the chemical composition were done by GC/MS method. HeEO was analyzed for antioxidant power in: (1) chemical system by the DPPH test, the ABTS test and the total antioxidant activity test; and (2) in biological system by lipid peroxydation tests (MDA and DC) in cells culture. The cytotoxicity effects of HeEO were assessed using MTT assay against Raji and HeLa cell lines. Results The optimal extraction yield was 6.8 g/100 g d.b. HeEO showed a remarkable anti-oxidant activities including DDPH (42%), ABTS (87%) and the power of ammonium phosphomolybdate (2992 ± 230 mg of HeEO by equivalent to 1 mg of vitamin C in terms of total antioxidant power). Conclusion Beyond notable antioxidant activities of the HeEo, our results showed a significant decrease in the production of ERO in the Raji cell line. The anti-tumor power of the Henna essential oil shows an interesting cytotoxicity effect (IC50 at 0.26 μg/mL for Raji and at 1.43 μg/mL for HeLa) with a total mortality percentage reaching 60%, for both.

The residue from commercial propolis extraction may have significant antioxidant power in food technology. However, among the challenges for using the propolis co-product as an inhibitor of lipid oxidation (LO) in baked goods is maintaining its bioactive compounds. Therefore, this study aimed to determine the propolis co-product extracts’ capability to reduce LO in starch biscuit formulated with canola oil and stored for 45 days at 25 °C. Two co-product extracts were prepared: microencapsulated propolis co-product (MECP) (with maltodextrin) and lyophilized propolis co-product (LFCP), which were subjected to analysis of their total phenolic content and antioxidant activity (AA). Relevant antioxidant activity was observed using the methods of analysis employed. The spray-drying microencapsulation process showed an efficiency of 63%. The LO in the biscuits was determined by the thiobarbituric acid reactive substances (TBARS) test and fatty acid composition by gas chromatography analysis. Palmitic, stearic, oleic, linoelaidic, linoleic, and α-linolenic acids were found in biscuits at constant concentrations throughout the storage period. In addition, there was a reduction in malondialdehyde values with the addition of both propolis co-product extracts. Therefore, the propolis co-product extracts could be utilized as a natural antioxidant to reduce lipid oxidation in fatty starch biscuit.

Background: Stilbene phytalexis (1,2-diphenyloethylen) and benzamide are beneficial for human health. To increase the stilbene ring activity, a new series of its derivatives containing benzamide structure was synthesized and evaluated for their in vitro antioxidant power. Methods: 1H nuclear magnetic resonance, mass spectroscopy, and chromatographic analyses were used to confirm the successful synthesis. The antioxidant properties were determined by the elimination of , HO , DPPH , ABTS+ radicals, total antioxidant status (TAS) and the ferric reducing antioxidant activities (TAC) measurements. Results: Stilbenebenzamide compounds showed a wide spectrum of antioxidant ability, however their total antioxidant power was weaker than those of butylated hydroxytoluene (BHT), ascorbic acid, and resveratrol. The highest antiradical activity towards and HO was shown by the compounds with structures containing amine group (SBEBA, SBA) ( : 37.7 – 38.0% and 40.8 – 43.5%, HO : 29.8%, 28.7% inhibition, respectively) at1.25 mM concentration. The antiradical power of SBEBA (0.29) in DPPH assay was lower than those of resveratrol (1.83), ascorbic acid (3.63) and BHT (4.09). The TAS values of the synthesized compounds ranged from 152.9±5.3 to 240.2±6.7µM trolox equivalent/gram (TE/g) and were much lower than those of BHT (1304±43.0), reservatrol (1360±29.0) and ascorbic acid (2782±39.7) µM TE/g. Similarly, the TAC values ranging from 29.7±0.9 to 41.5±1.6 µM TE were weaker than that of resveratrol (239.2 ±6.7 µM TE/g). Conclusion: The results suggest that the presence of hydroxyl group in stilbene ring should be considered in further design of stilbenebenzamide compounds to enhance their antioxidant activity.

AbstractThe aim of the present study was to examine the antioxidant activity of three Veronica species (Plantaginaceae). The antioxidant potential of various extracts obtained from aerial flowering parts was evaluated by DPPH-free (1,1-diphenyl-2-picrylhydrazyl-free) radical scavenging activity and ferric-reducing antioxidant power assays. Considerable antioxidant activity was observed in the plant samples (FRAP values ranged from 0.97 to 4.85 mmol Fe2+/g, and DPPH IC50 values from 12.58 to 66.34 µg/ml); however, these levels were lower than the activity of the control compound butylated hydroxytoluene (BHT) (FRAP: 10.58 mmol Fe2+/g; DPPH IC50: 9.57 µg/ml). Also, the in vivo antioxidant effects were evaluated in several hepatic antioxidant systems in rats (activities of glutathione peroxidase, glutathione reductase, peroxidase, catalase, xanthine oxidase, glutathione content and level of thiobarbituric acid reactive substances) after treatment with different Veronica extracts, or in combination with carbon tetrachloride (CCl4). Pretreatment with 100 mg/kg b.w. of Veronica extracts inhibited CCl4-induced liver injury by decreasing TBA-RS level, increasing GSH content, and bringing the activities of CAT and Px to control levels. The present study suggests that the extracts analyzed could protect the liver cells from CCl4-induced liver damage by their antioxidative effect on hepatocytes.

Rhodomyrtus tomentosa(Myrtaceae) has been employed in traditional Thai medicine to treat colic diarrhoea, dysentery, abscesses, haemorrhage, and gynaecopathy. In addition, it has been used to formulate skin-whitening, anti-aging and skin beautifying agents. Ethnomedical activities of this plant may be due its antioxidant property. Hence, the aim of this study was to evaluate bothin vitroandin vivoantioxidant activities ofR. tomentosaleaf extract.In vitroantioxidant activity of the extract was assessed by lipid peroxidation inhibition capacity, ferric reducing antioxidant power, and metal chelating activity.R. tomentosaextract demonstrated its free radical scavenging effects in concentration dependent manner.In vivoantioxidant activity of the extract was conducted in SwissAlbinomice. Levels of thio-barbituric acid reactive substances (TBARS), glutathione (GSH), and the activities of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) in blood, liver, and kidney were analyzed using microtitre plate photometer. Administration of CCl4caused significant increase in TBARS and decrease in GSH, SOD, CAT and GPx levels. In contrast,R. tomentosaextract (0.8 g/kg) effectively prevented these alterations and maintained the antioxidant status. The results suggest thatR. tomentosaextract can serve as a potent antioxidant.

Aerva javanica (Amaranthaceae) is a grey coloured woolly perennial tomentose shrub. Its traditional and folklore usage motivates further investigation on its pharmacognostic parameters and pharmacological potential. Hydro-alcoholic extract (AJCE) was prepared from flowering tops of A. javanica. In order to work further on activity guided fractions, ethyl acetate (AJEAF) fraction was prepared.  Therefore, in order to establish its antioxidant potential, in-vivo effect on LPO, GSH, SOD and catalase activity was determined. For comparison, silymarin and Centella asiatica extract (CAE) were used as standard antioxidant compound/extract. Lipid peroxidation in term of MDA content expressed as nM/mg, which was 82.18 and 67.39 for AJCE with increasing doses of complete hydro-alcoholic extract (AJCE represented as AJCE-1 and AJCE-2) and 51.65 for AJEAF in contrast to 40.64 nM/mg for standard silymarin and 46.81 nM/mg for standard CAE. GSH content was determined as 3.12, 3.82 and 4.56 μg/mg wet tissue in contrast to 5.59 for standard silymarin and 4.42 for standard CAE. Superoxide scavenging was expressed as SOD U/mg wet tissue, determined as 7.26, 9.16 and 9.91 U/mg wet tissue for AJCE-1 (250 mg/kg i.p. b.w), AJCE-2 (500 mg/kg i.p. b.w), and AJEAF respectively in comparison to silymarin (10.11) and CAE (46.81 U/mg wet tissue). Catalase activity expressed as μM of H2O2 decomposed / min / mg wet tissue was determined as 0.61, 0.72 and 0.78 repectively for AJCE-1 (250 mg/kg i.p. b.w), AJCE-2 (500 mg/kg i.p. b.w), and AJEAF. Results indicated the SOD values and total antioxidant power of DEE and EAF fractions even better than standard ascorbic acid which expressed the prospective potential of fractions (DEE and EAF) against metabolic disorders.

Leafy vegetables are an excellent source of bioactive factors, traditionally used as important medical ingredients. Recently, some leafy vegetables are domesticated without the use of fertilizer, as well as the assessment of the effect of fertilizer on their nutritional value. This study aims at testing the effect of three mineral and organic fertilizers (Cowpat, NPK and NPK + cowpat) on three traditional leafy vegetables: Ceratotheca sesamoïdes, Sesamum radiatum and Justicia tenella. Their antioxidant activities were assessed at different harvesting times varying from six (6) to fourteen (14) Weeks After Transplantation (WAT) using three in vitro methods: Ferric reducing antioxidant power (FRAP), 2,2’-Azinobis3-ethylbenzothiazoline-6- sulfonic acid (ABTS) and 2,2-diphenyl-1-picryl hydrazil (DPPH). The FRAP, DPPH, ABTS assays were consistent and positively correlated (p < 0.001). Total antioxidant activities of leafy vegetables depended on species, harvesting time and methods. They ranged from 32.0 to 45.7 µmol Fe/g DW (for FRAP), with non fertilized J. tenella cut at 12 WAT giving the highest antioxidant activity. Percentage of inhibition using DPPH assay ranged from 11.4 to 87.2 % and showed that J. tenella fertilized with NPK and cowpat, and harvested at 9 WAT had the highest antioxidant activity. Regarding ABTS, the range of 17.6 to 28.9 µmol TE/g DW was recorded, and the leaves of C. sesamoïdes harvested at 10 WAT and fertilized with cowpat showed the highest level. Compared to other species, those studied here may best contribute to improve human health related to degenerative diseases. Moreover, significant and positive correlations were observed between the total phenolic compounds content and antioxidant activities of leaves regardless the methods used. The positive and significant correlations between the three assays (FRAP, DPPH, ABTS) allow to suggest the use of only anyone of them to check factors in the study. This paper highlights the potential of antioxidant capability of the leafy vegetables even fertilized.
