Variability in expression of male fertility in triticale (Xtriticosecalewittmack) withTriticum timopheevicytoplasmWe present a reliable, visual method for evaluation of the level of male fertility during flowering, that is indispensable for breeding of hybrid cultivars of winter triticale based on the cms-T.timopheevisystem. Detailed observations of anther development were performed on 20 F2and BC1plants derived from crosses between male-sterile and fertility restoring lines. Variation of anther development within florets, spikelets, spikes, and among spikes of the same plant was examined. Hierarchic analysis showed significant role of these factors in determination of anther development, irrespective of the level of plant fertility. The sterilizing effect of cytoplasm was always better visible in the tip and base spikelets of the spike, in the third floret, and the anther adjacent to the floret axis. Our data indicate that during selection toward male-sterile plants at anthesis, at least 5 spikes should be evaluated. Special attention should be paid to the development of the anthers at the 2ndand 7thspikelet of the spike along with the variability in anther development within spikelet and floret. The anthers in the tip and base spikelets of the spike must be precisely evaluated during selection toward restorer lines.

Abstract Pollen development is critical to the reproductive success of flowering plants, but how it is regulated is not well understood. Here, we isolated two allelic male-sterile mutants of OsMYB80 and investigated how OsMYB80 regulates male fertility in rice. OsMYB80 was barely expressed in tissues other than anthers, where it initiated the expression during meiosis, reached the peak at the tetrad-releasing stage and then quickly declined afterward. The osmyb80 mutants exhibited premature tapetum cell death, lack of Ubisch bodies, no exine and microspore degeneration. To understand how OsMYB80 regulates anther development, RNA-seq analysis was conducted to identify genes differentially regulated by OsMYB80 in rice anthers. In addition, DNA affinity purification sequencing (DAP-seq) analysis was performed to identify DNA fragments interacting with OsMYB80 in vitro. Overlap of the genes identified by RNA-seq and DAP-seq revealed 188 genes that were differentially regulated by OsMYB80 and also carried an OsMYB80-interacting DNA element in the promoter. Ten of these promoter elements were randomly selected for gel shift assay and yeast one-hybrid assay, and all showed OsMYB80 binding. The 10 promoters also showed OsMYB80-dependent induction when co-expressed in rice protoplast. Functional annotation of the 188 genes suggested that OsMYB80 regulates male fertility by directly targeting multiple biological processes. The identification of these genes significantly enriched the gene networks governing anther development and provided much new information for the understanding of pollen development and male fertility.

The article is dedicated to the determination of the peculiarity of stages of legal support of academic virtue in Ukraine.It has been proven that the first stage for development of legal support to academic virtue has started in 2004, when the all-European processes of increasing the role of universities within a society have been initiated as well as the transformation ofrequirements to the participants of educational activity. This stage has been characterized by the acceptance of a Bucharestdeclaration on ethical values and principles of higher education in the European region (2-5 September 2004) and its result has been the establishment of an overall European discourse of academic virtue which included pedagogical, ethical and legalaspects of the phenomenon under research.It has been pointed out, that the third stage of legal support to academic virtue is related with stipulating in 2011 within aNational framework, the qualifications of adherence to proper academic virtue as the one that is considered to be necessary toinitiate, plan, implement and correct the subsequent process of a thorough scientific research of the level of basic skills andabilities. From this moment on there’s an active development of the terminological core dedicated to the issue of academicvirtue, there’s a preparation of suggestions to be implemented to the legislation on education, particularly, by determining thelegal definition of academic virtue, the forms and types of academic responsibility, and also the active reaffirming of ethicalrequirements on the academic virtues of teachers, scientists and individuals obtaining education in the codes of academicvirtues, approved in the course of gatherings of working teams or the scientific councils of universities.

Several different expressions of male sterility were observed in the plants sampled for the study of pollen and anther development in the third substitution backcross generation to intermediate wheatgrass. These included (1) pollen abortion following engorgement with starch, (2) abortion of microspores with exine abnormalities (found most often in plants with a high degree of meiotic irregularities), (3) abortion of microspores with normal exines, (4) release at anther dehiscence of normal-appearing late vacuolate microspores, (5) enlargement of one of the parietal layers of the anther, and (6) tapetal abnormalities, including orbicular wall malformations. Anthers from plants which were male sterile or partially male sterile and meiotically irregular were shorter than anthers from fertile or partially male-sterile, meiotically regular plants.Definite statements about specific causes of the male sterility could not be made. Meiotic irregularities may be involved in some of the male sterility, and exine malformations appeared to be directly related to sterile microspores. Tapetal disturbances were observed infrequently. Probably the consequences of several factors were observed in the backcross material.

The study is devoted to the analysis of Western research on aspects of social security of the Russian-Chinese border. The article provides the allocation social security aspects in the Western descriptions of the Russian-Chinese border in the XVII-XIX centuries. The degree of political conjuncture influence on the interpretation of the social security of the Russian-Chinese border is determined; Modern transformations of the critical concept of social development of border Russian-Chinese territories in the XXI century are analyzed. The novelty of the study is determined by the fact that it is the first analysis of the factors and patterns assessments of social security formation in the territory of the Russian-Chinese border in the works of European and American authors. The result is the determination of the development vector of the Western Social Security methodology of the Russian-Chinese border area from early idealizations to positivist descriptions and a “critical concept”, where Russia is assigned the role of a social threat. The second result is to identify the content of the critical concept of the social security of the Russian-Chinese border, which is that Russian-Chinese relations are perceived by Western researchers as the factor in the lagging of regional society development from the global level. The third result is to analyze the transformation of the critical concept of the social security of the Russian-Chinese border, which is recognized by the regional framework independence and its dependence not so much from the Western trends of globalism but on the influence of East Asian players, the leading among which is China

Majority of the genes expressed during spermatogenesis are autosomal. Mice with different deletions of Yq show sub-fertility, sterility and sperm abnormalities. The connection between Yq deletion and autosomal gene regulation is not well understood. We describe a novel mouse Yq-derived long noncoding RNA, Pirmy, which shows unprecedented number of splice variants in testis. Further, Pirmy transcript variants act as templates for several piRNAs. We identified ten differentially expressed autosome-encoded sperm proteins in mutant mice. Pirmy transcript variants have homology to &5acute/&3acuteUTRs of these deregulated autosomal genes. Thus, subfertility in Y-deleted mice appears to be a polygenic phenomenon that is partially regulated epistatically by the Y-chromosome. Our study provides novel insights into possible role of MSY-derived ncRNAs in male fertility and reproduction. Finally, sperm phenotypes from the Y-deleted mice seem to be similar to that reported in inter-specific male-sterile hybrids. Taken together, this study provides novel insights into possible role of Y-derived ncRNAs in male sterility and speciation.

Male fertility in maize is controlled by development and genetic programming and is directly impacted by environmental factors such as light, temperature, water, and nutrient availability; the control of this trait has substantial agronomic utility. Maize anthers emerge from male florets, which are clustered to form the tassel at the top of the plant separated from the female ear. Quantification of anther extrusion is one important aspect in the determination of male fertility. To address the lack of an automated method to measure anther extrusion on a large scale, we developed 'Tasselyzer', a quantitative, image-based color trait analysis pipeline for tassel image segmentation, based on the existing PlantCV platform, and we applied it to determine the proportion of anther extrusion. We evaluated Tasselyzer in maize during the seven-day period of pollen shedding as well as in the temperature-sensitive male sterile mutant dcl5. With tassel images obtained with a smart phone camera, we show that the anther scores positively correlate with anther extrusion, and such methods can be used to measure environmental impacts on the dcl5 mutant. Altogether, this work establishes an automated and inexpensive method to quantify anther extrusion in maize, which would be useful for research and breeding.

The role of glycoconjugates in tumor cell differentiation has been well documented. We have examined the expression of the two anomers of the Thomsen-Friedenreich antigen on the surface of human, canine and murine tumor cell membranes both in vitro and in vivo. This has been accomplished through the synthesis of the disaccharide terminal residues in both a and ß configuration. Both entities were used to generate murine monoclonal antibodies which recognized the carbohydrate determinants. The determination of fine specificities of these antibodies was effected by means of cellular uptake, immunohistopathology and immunoscintigraphy. Examination of pathological specimens of human and canine tumor tissue indicated that the expressed antigen was in the β configuration. More than 89% of all human carcinomas tested expressed the antigen in the above anomeric form. The combination of synthetic antigens and monoclonal antibodies raised specifically against them provide us with invaluable tools for the study of tumor marker expression in humans and their respective animal tumor models.

Summary Aim of the study was to find out wether there is a common stop of growth of mandibular bone, so that no individual determination of the optimal time for surgery in patients with asymmetric mandibular bone growth is needed. As there are no epiphyseal plates in the mandibular bone, stop of growth cannot be determined on X-ray films. Methods: Bone scans of 731 patients [687 patients (324 male, 363 female) under 39 y for exact determination of end of growth and 44 (21 male, 23 female) patients over 40 y for evaluation of nongrowth dependant differences in tracer uptake] were reviewed for the study. All the patients were examined 3 hours after injection of 99mTc-DPD. Tracer uptake was measured by region of interest technique in different points of the mandibular bone and in several epiphyseal plates of extremities. Results: Tracer uptake in different epiphyseal plates of the extremities shows strong variation with age and good correlation with reported data of bone growth and closure of the epiphyseal plates. The relative maximum of bone activity is smaller in mandibular bone than in epiphyseal plates, which show well defined peaks, ending at 15-18 years in females and at 18-21 years in males. In contrast, mandibular bone shows no well defined end of growing but a gradually reduction of bone activity which remains higher than bone activity in epiphyseal plates over several years. Conclusion: No well defined end of growth of mandibular bone exists. The optimal age for surgery of asymmetric mandibular bone growth is not before the middle of the third decade of life, bone scans performed earlier for determination of bone growth can be omitted. Bone scans performed at the middle of the third decade of life help to optimize the time of surgical intervention.

It has been suggested that heparin caused potentiation of aggregation induced by ADP or epinephrine. The exact mechanism of heparin-induced platelet activation, however, remained unknown. In this paper, we have investigated the role of anti-thrombin III ( AT ) in heparin-induced platelet activation using purified AT and AT depleted plasma. When ADP or epinephrine was added to citrated PRP one minute after addition of heparin ( 1 u/ml, porcine intestinal mucosal heparin, Sigma Co. USA ), marked enhancement of platelet aggregation was observed, compared with the degree of aggregation in the absence of heparin. However, in platelet suspensions prepared in modified Tyrode’s solution, heparin exhibited no potentiating effect on platelet aggregation induced by epinephrine or ADP. Potentiation of epinephrine- or ADP-induced platelet aggregation by heparin was demonstrated when purified AT was added to platelet suspensions at a concentration of 20 μg/ml. AT depleted plasma, which was prepared by immunosorption using matrix-bound antibodies to AT, retained no AT, while determination of α1-antitrypsinα2- macroglobulin and fibrinogen in AT depleted plasma produced values which corresponded to those of the original plasma when dilution factor was taken into account. The activities of coagulation factors were also comparable to those of the original plasma. Heparin exhibited potentiating effect on ADP- or epinephrine-induced aggregation of platelets in original plasma, but no effect in AT depleted plasma. When purified AT was added back to AT depleted plasma at a concentration of 20 μg/ml, potentiation of platelet aggregation by heparin was clearly demonstrated.Our results suggest that effect of heparin on platelet aggregation is also mediated by anti-thrombin III.
