Influence of gastrointestinal trichostrongylidosis on ram fertility A study on the influence of gastrointestinal trichostrongyles on ram fertility was performed. Two groups of semen donor rams (Sarda breed) were utilized; one was experimentally infected with Haemonchus contortus, Trichostrongylus colubriformis and Teladorsagia circumcincta and the other was used as a control group. In all animals, coprological, haematological and reproductive parameters were studied. The results suggest that the parasites had a limited effect with some changes in phosphorus, cholesterol and chlorine levels. In our experience the parasitic burden produced no significant change on ram fertility.

Abstract Loline, an alkaloid produced by Epichloë endophytes in pastoral grass species, is considered non-toxic with suggested antimicrobial and anthelmintic properties. In vivo metabolism of loline suggests ability to reach the abomasum or intestine, and anthelmintic benefit is indicated in vitro when parasites are in either a developmental stage close to the gastric mucus layers or adult stage consuming blood. Therefore, the objective of this project was to investigate the validation of using a loline seed extract as a natural anthelmintic in parasitized lambs. Sixteen Coopworth lambs were individually penned, offered ad-libitum lucerne pellets and water and randomly assigned to 3 treatment groups; CON; no treatment, or treatment with a Festuca pratensis seed extract with (LOL) or without loline (NIL). Lambs were orally infected with 20,0000 L3 Haemonchus contortus on day 0, and 10,000 L3 Teladorsagia circumcincta, and Trichostrongylus colubriformis on day 14. Parasites then matured to determine lolines efficacy on blood feeding adult Haemonchus contortus and L4 mucosal browsers Teladorsagia circumcincta, and Trichostrongylus colubriformis. Loline was orally dosed at 52.5 mg/kg LW starting on day 13, and every other day until slaughter for worm enumeration on day 28. Feed intake and refusals were recorded daily, and fecal samples were taken prior to infection, on loline dose days and at slaughter. Worm and fecal egg counts were transformed to log10 and all data were analyzed in Genstat (18th edition). Significance was declared at P < 0.05. Worm burdens of T. circumcincta (P = 0.96), T. colubriformis (P = 0.43), and H. contortus (P = 0.15) were not different between treatments. No treatment difference (P = 0.39) was observed in fecal egg counts, weight gain (P = 0.51), or feed intake (P = 0.18). Average growth efficiency (kg LWG/ kg DM intake) was 0.18 in CON which was less (P = 0.01) than LOL (0.24) or NIL (0.23). In conclusion, limited evidence supported an in vivo anti-parasitic effect of loline.

Este trabalho foi desenvolvido em três propriedades rurais nos municípios de Lages, São Joaquim e Campos Novos, estado de Santa Catarina, com os objetivos de determinar a prevalência, a intensidade e a variação sazonal de helmintos gastrintestinais e pulmonares em ovinos no Planalto Catarinense. Com base nos resultados aintenção é propor um esquema estratégico de controle. Para isso, foram utilizados mensalmente três cordeiros traçadores por propriedade, os quais, antes de serem conduzidos às mesmas, foram estabulados por 30 dias e executados tratamentos supressivos com anti-helmínticos de diferentes princípios ativos, com exames parasitológicos semanais para verificar a total eliminação de infecção parasitária. A seguir, foram encaminhados às três propriedades onde permaneceram em pastejo por 28 dias, sendo posteriormente recolhidos ao estábulo por mais 20 dias. Após foram sacrificados e realizada a coleta de alíquotas de 10% dos conteúdos do abomaso e intestino delgado, todos os helmintos do intestino grosso e pulmão. As maiores infecções por Haemonchus contortus ocorreram durante o período de outubro a março. O parasitismo por Trichostrongylus axei e Trichostrongylus colubriformis teve índices crescentes ao longo dos anos experimentais, apresentando maior pico de maio a outubro de 1999. Predominaram as espécies, no abomaso: Haemonchus contortus (100%); Trichostrongylus axei (98,7%); Trichostrongylus colubriformis (1,3%); Teladorsagia circumcincta (100%); Ostertagia ostertagi (100%). No intestino delgado: Trichostrongylus colubriformis (100%); Cooperia punctata (69,1%); Cooperia pectinata (18,4%); Cooperia curticei (6,9%); Cooperia oncophora (4,8%); Cooperia spatulata (0,8%) e Nematodirus spathiger (100%). No intestino grosso: Oesophagostomum venulosum (100%) e Trichuris ovis (100%). No pulmão, não foram encontrados parasitos.

SUMMARYThe amounts of acetylcholinesterase (AChE) in homogenates of infective-stage larvae of strains, from different countries, of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus colubriformis resistant or susceptible to benzi-midazole (BZ) anthelmintics, were compared using a rapid colorimetric assay. The levels of AChE were much greater in those strains which were BZ-resistant than in BZ-susceptible populations. The larvae of susceptible and resistant H. contortus contained less AChE than did the larvae of the other two species. One-way analysis of variance showed that the observed differences between susceptible and resistant strains were significant in each of the species studied. It is suggested that the extra enzyme may act as an anthelmintic-detoxifying enzyme, may inactivate the active site of the anthelmintic or may prevent binding of the anthelmintic to its receptor

1. The degree of nematode larvae infestation in goats fed malojillo (Pará grass) and Merker grass in Puerto Rico was investigated in an attempt to determine whether goats would become more significantly parasitized when eating either one of the two grasses. 2. The technique of raising and-maintaining parasite-free animals is described. 3. Blood samples for hemoglobin and packed-cell determinations and fecal samples for nematode ova counts were obtained every 15 days from the 10 experimental goats. 4. It was found that goats eating malojillo harbored a larger number of nematode parasites than those eating Merker grass. Ova of Strongyloides papillosus appeared in the feces of the goats even before the beginning of this work, but that infestation disappeared about a month before the end of the experiment. Haemonchus contortus ova were seen next, being followed chronologically by Trichostrongylus spp. and Oesophagostomum columbianum. The latter species was harbored only by the goats receiving malojillo. Goats eating Merker grass exhibited 8.9 ova per gram of fecal sample throughout the experiment, those receiving malojillo showed 48.2 eggs per sample, while those in the control group had only 0.57 eggs per sample. 5. All animals were sacrificed at the conclusion of this work and the digestive tracts thoroughly searched for helminths. More nematodes were secured from the goats eating malojillo than from the other two groups. A total of 1,028 nematodes was recovered from animals in the malojillo group, 191 from the Merker group, and only 3 from the control group receiving imported hay.

Abstract The effect of four diets on the performance of Dorper lambs that were artificially mix-infected with 1000 infective larvae (L3) of Haemonchus contortus and 1000 L3 of Trichostrongylus colubriformis every three days for 12 weeks was evaluated. For each diet, one infected group (n = 7) and one control group (n = 4) were set up. The four diets contained the following proportions of hay (H) and concentrate (C): Diet 1 - 100%H; Diet 2 - 75%H:25%C; Diet 3 - 50%H:50%C; Diet 4 - 25%H:75. All the infected groups showed decreases in mean packed cell volume (PCV) and total plasma protein, especially the Diet 1-infected, which was also the group with the highest fecal egg count. The diets and infection had significant effects (P < 0.05) on daily body weight gain (DBWG), cold carcass weight and daily food consumption. The control and infected groups had mean DBWG (in kg), respectively, of 0.048 and 0.025 with Diet 1; 0.082 and 0.067 with Diet 2; 0.188 and 0.189 with Diet 3; and 0.303 and 0.221 with Diet 4. In conclusion, the infection affected the productive performance regardless of the diet; however, with increased metabolizable protein and energy, the risk of occurrence of anemia and hypoproteinemia diminished.

Broad-spectrum anthelmintics were tested for their efficacy in either single active or combination formulations against multiple anthelmintic-resistant strains of Haemonchus contortus, Teladorsagia circumcincta and Trichostrongylus colubriformis. Three efficacy trials based on post mortem worm counts showed that the triple and quadruple combinations of benzimidazoles, levamisole, macrocyclic lactones and closantel were effective against most strains that were resistant to each of the components when drenched individually. Consequently, use of any one of these classes of anthelmintics by itself against multiple-resistant strains would lead to a rapid increase in the level of resistance. When used in combinations, the anthelmintics reduced the survival rate of most strains to <5%. However, because some strains of T. circumcincta and T. colubriformis were found to be resistant to all combinations currently marketed in Australia, it is clear that combinations should be used before resistance levels become too high.
