Mushrooms contain a variety of secondary metabolites, including various phenolic compounds, which have been shown to act as excellent antioxidants. In this study, the contents of total phenolic, flavonoids and tannins of mushrooms methanolic extracts were evaluated by colorimetric assays to ranges of 277.36±0.66 to 420.86±0.90 mg (GAE)/100g DW; 31.99±0.90 to 90.90±0.07mg (QE)/100g DW and 150.61±0.16 to 220.47±1.01mg (TAE)/100g DW respectively. HPLC-profiles of methanolic extracts indicated that the individual phenolic compounds found to the samples of the species of mushrooms that are Lentinus squarrosulus and Auricularia politrich revealed us that gallic acid, catechin, acid ρ-hydroxybenzoїc, acid ellagique and naringerin were presented to the level of the two species. With respect to organic acids, the results showed that citric acid and fumaric acid was the major organic acid in all the samples of both mushrooms species. The methanolic extracts of the two mushrooms exhibited the high DPPH radical scavenging activities ranging from 34.10±1.12 to 58.95±0.52%. These data indicated that these mushrooms could constitute a potential good source of natural antioxidant for the local population.

The antibacterial and antifungal activities of the bulb and flower of Allium staticiforme and Allium subhirsutum were investigated. In addition, DPPH radical scavenging activity and total phenolic contents were determined. The results show that methanolic extracts of A. staticiforme and A. subhirsutum had antifungal activities against Candida albicans, together with a less activity level against Escherichia coli, Staphylococcus epidermidis, S. aureus, Enterecoccus faecalis, Salmonella typhimurium and Pseudomanas aeruginosa. The total phenolic contents of A. staticiforme leaf and bulb were determined as 17 and 2.4 mg of GAE/100 g, respectively. The IC50 of methanolic extracts of A. staticiforme and A. subhirsutum were also determined. In conclusion, both A. staticiforme and A. subhirsutum have antifungal activities with weak antibacterial activities. These plants have DPPH radical scavenging activities. H. perforatum, which is widely used in traditional medicine due to its bioactive compounds was extracted with ethanol-water (3:7). The extract was encapsulated with maltodextrin and gum arabic in a spray dryer in order to protect the phenolic compounds in its structure. Different amounts of microcapsules were added to our traditional drink, i.e. ayran (drinking yoghurt). The total phenolic content and DPPH radical scavenging activity of the microcapsules, extract of HP, and ayran samples were determined. The amount of total phenolic compounds in the microcapsule provided a superior effect than the extract. The ayran samples were supplemented with 2%, 3%, 4%, 5% and 6% of H. perforatum (HP) microcapsules and it is observed that total phenolic content (TPC) and DPPH radical scavenging activity indicated an increase with concentration. TPC and DPPH activity were determined as 256.94 mg GAE / 100mL and 78.05% for 6% of HP microcapsules supplemented samples. As a result of the sensory analysis, ayran samples which supplemented with 4% of HS microcapsule gained the highest scores by the panellists and received more appreciation than the control group. According to sensory analysis, HP4 (ayran produced with 4% HP added microcapsule) sample was determined as the best sample, while the HP6 (Ayran produced with 6% HP added microcapsule) sample had the highest scores in terms of DPPH scavenging activity and TPC results. The overall results of this study revealed that 4% HP supplemented ayran can be produced with its increased health benefits and desirable properties. In this study, the use of H. perforatum microcapsules in ayran, its effect on antioxidant and phenolic components, the usage rates and acceptability of microcapsules were investigated.

Grape canes are the main agro-wastes from vineyards. This work studied the antioxidant activities of the defatted methanolic extracts (ME) of canes from 11 genotypes: 5 Vitis vinifera widely known cultivars and 6 Chinese wild varieties from three species (V. amurensis, V. davidii, and V. pentagona) and the antioxidant activities of the ME’s chloroform fractions (CF), ethyl acetate fractions (EAF) and water fractions (WF). Among ME and its three fractions, EAF’s total phenolic contents (TPC) and total flavonoid contents (TFC) were the highest, at 586 mg/g of gallic acid equivalent and 320 mg/g of quercetin equivalent, respectively. The antioxidant power of the fractions/extracts was in the order EAF > ME > WF > CF, based on the DPPH radical-scavenging power and ferric-reducing antioxidant activity, while the order was EAF > CF > WF >ME based on the β-carotene-linoleic acid bleaching activity. Methanolic extracts demonstrated the strongest Fe2+-chelating activity. The antioxidant activities of the extracts/fractions generally correlated with the TPC and TFC in all assays, except with the Fe2+-chelating test. Grape canes from V. davidii had the highest TPC, TFC and antioxidant activities compared with those from other grape species. Catechin, epicatechin and trans-resveratrol were the predominant phenolic components of fractions/extracts. In light of these valuable bioactivities, grape canes from annual pruning practice considered as waste material have good commercial potential for utilization as a promising natural antioxidant in the food, pharmaceutical and cosmetic industries, given its low cost and availability in large amounts.

The present study focuses on the optimization of the parameters to extract phenolic products by decoction and the quantification of these compounds from grape pomace, using a central experimental design. The antioxidant activity of methanolic extracts from grape skin grown in Morocco was evaluated. The grapes variety is “Michael Paleiri”, it is a black variety with pips, and they are from the region of Benslimane. The total phenolic compounds contents were determined by the Folin-Ciocalteu method. The optimization suggested that extraction with methanol for 29 min, at 60°C were the best solutions for this combination of variables.The largest amount obtained was 1042.06 mg EGA/g DW. The antioxidant activity is carried out by the radical scavenging method 1,1-diphenyl-2-picryl hydrazyl (DPPH°) and the ferric iron reduction capacity (FRAP). The DDPH inhibition capacity reached 20.78%, compared with 15.22% of a BHT solution at 0.001 g/L. A significant relationship between antioxidant capacity and total phenolic content is evident (R2=0.994). These results demonstrate that methanol extracts from the waste from grapes grown in Morocco could be used as potential sources of natural extracts rich in phenolic compounds and endowed with significant antioxidant activity.

Background: Plants are new sources of antibacterial agents, hence the need to determine and evaluate the antibacterial properties, antioxidant activity and gas chromatography – mass spectrometer (GC-MS) profile of medicinal plants. Methodology: In this study, sequential extraction of Prunus africana and Harrisonia abyssinica was used to obtain ethyl acetate and methanol extracts. Antioxidant activity was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH), whereas the total phenolic and total flavonoid contents were estimated using Folin-Ciocalteu and aluminium chloride, respectively. Antibacterial properties of the extracts against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Escherichia coliand Candida albicans were estimated using the disc diffusion method and compared against streptomycin. Results: Screening of crude methanolic extracts revealed the presence of secondary metabolites which was further confirmed by Fourier transform infrared (FT-IR) characterisation that revealed the presence of H-bonded OH functional groups. The extracts revealed that P. africana had a higher total phenolic and total flavonoid contents compared to H. abyssinica. Methanolic extracts of both plants had moderate activity against selected microorganisms and both inhibited DPPH radical scavenging activity. GC-MS analysis of P. africana and H. abyssinica extracts revealed the presence of several phytochemicals that have been reported to have medicinal uses. Total phenolic and flavonoid contents showed positive correlations with the DPPH radical scavenging activity and negative correlations with EC50. Conclusion: Prunus africana and H. abyssinica extracts had moderate antimicrobial properties against the selected microorganisms because of the presence of secondary metabolites.

Bangia atropurpurea is a freshwater red filamentous alga. In this study, we report on the recovery yield of purified phycobiliproteins, total phenolic content (TPC) and their antioxidant capacity. B. atropurpurea suspended in phosphate buffer (pH 7.2), saturated with ammonium sulphate and further isolated R-phycoerythrin and phycocyanin from phycobilisome structure by gel filtration with Sephadex G-200. Phenolic compounds extracted by variable polarity solvents to exhibit optimum yield. Folin–Ciocalteu method was used to determine the TPC, while 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and ferric-reducing antioxidant power (FRAP) assays were used to quantitate the antioxidant activity of phycobiliproteins and phenolic compounds.  B. atropurpurea predominantly contains R-phycoerythrin 54.80 % of the total protein extracted at purity ratio A562 to A280 of 4.45 while, phycocyanin displayed 16 % recovery yield at purity ratio A615 to A280 of 2.18. The purified R-phycoerythrin exhibited better antioxidant activity than phycocyanin extract and BHT synthetic antioxidant by exhibiting (IC50%=7.66 ±0.81µg/mL) and 54.74 ± 0.11 mg GAE/1g extract for, DPPH scavenging and FRAP assay, respectively. The sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) of purified R-phycoerythrin and phycocyanin proteins have showed apparent single band at molecular weight of 19.6 kDa and 17.5 kDa, respectively. The methanol-soluble extract displayed the utmost TPC, 80.97±0.53 mg GAE/1g extract compared to phenolic content in other organic solvents used for extraction. In accordance with TPC, methanol-soluble extract exhibited as the most effective DPPH radical scavenging and ferric reduction, (IC50%=14.63±0.79 µg/mL) and 37.79±0.27 mg GAE/1g extract, respectively. The presence of R-phycoerythrin, phycocyanin and phenolic compounds in an array, amplifies the antioxidant property of B. atropurpurea.

Plum wines made from two cultivars of Prunus domestica L. (Pozegaca and Crvena ranka) and one cultivar of Prunus insititia L. (Trnovaca) were evaluated for their total phenolic and anthocyanin contents. LC?MS/MS analysis based on specific MS transitions in the multiple reaction monitoring (MRM) mode was used for the identification and quantification of selected phenolic compounds. Catechin, chlorogenic and caffeic acids, as well as quercetin, were identified as the main polyphenols in plum wines. The total amount of phenolic compounds ranged from 1.24 to 1.58 g gallic acid equivalent per L. Among the examined wines, the Crvena ranka wine had a higher content of anthocyanins (12.31 mg cyanidin-3-glucoside equivalent per L). The antioxidant capacity of the wines was determined using the DPPH assay. The variations in the physicochemical characteristics, phenolic composition and DPPH radical scavenging activity of these wines are related to differences due to the different plum cultivars used in the preparation of each wine.

Plant extracts are an important alternative to antibiotics, which are ever more restricted because of their developing microbial resistance and some adverse effects that have been observed following frequent application. The aim of the present study was to determine the antifungal and antioxidant activity of the methanolic extracts of Acorus calamus, Chlorella vulgaris, Lemna minuta and Scenedesmus dimorphus. The antifungal activity of the extracts against strains of Aspergillus flavus, Aspergillus parasiticus, Aspergillus ochraceus, Aspergillus niger, Aspergillus carbonarius, Fusarium graminearum, Fusarium oxysporum, Penicillium chrysogenum and Alternaria alternata was evaluated via the agar well diffusion method. The antioxidant activity of the extracts was measured through the determination of three parameters—total phenolic content, total flavonoid content and radical scavenging potential (determined through UV/Vis analysis). A. calamus extracts had the highest antimicrobial activity against eight fungal strains, followed by the C. vulgaris, L. minuta and S. dimorphus extracts, which were inhibitory against two to three strains. Among the extracts from the species studied, the extract from S. dimorphus showed the highest antioxidant potential, as determined via the DPPH (1,1’-diphenyl-2-picrylhydrazil-radical) method. This correlated to its high total phenolic and flavonoid content. From A. calamus and L. minuta, methanolic extracts were obtained that exhibited similar values of the aforementioned parameters, followed by C. vulgaris extracts, which showed the lowest antioxidant activity. Based on the Pearson correlation coefficients, the impacts of the total phenolic content and the total flavonoid content on radical scavenging capacity are similar, and flavonoids were a significant part of the total phenolic compounds extracted from the plant materials studied.

Sideritis niveotomentosa Hub. -Mor. is a local endemic species belonging to the Lamiaceae family. In this study, GC/MS analysis, total antioxidant capacity and anticancer effects of different extracts obtained from S. niveotomentosa were investigated comparatively. Total phenolic contents of extracts were determined by the Folin–Ciocalteu method, total flavonoid contents by aluminum chloride method, and also the free radical scavenging activities of the extracts by DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay. The cytotoxic effect of the extracts was studied via MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) assay on DLD1, HL60 and ARH77 cell lines. Pro-apoptotic gene expression levels were also tested in the most sensitive cell line ARH77 by Real-Time PCR. The expression levels of 4 pro-apoptotic genes, APAF, BAX, CASP3, and HRK were found to be upregulated in ARH77 cells that were treated extracts. Results showed that methanolic extracts contain more phenolic content than acetone extracts, consistent with DPPH results. As a result, Sideritis niveotomentosa extracts, especially methanolic extracts, are rich in phenolic content and have a strong radical scavenging effect. In addition, the extracts showed selective effects on cell lines. This study is pioneering in terms of future studies, and the findings provide hope for future experimentation.

Nonalcoholic fatty liver disease (NAFLD) is a major cause of morbidity and mortality worldwide. Additional therapies using functional foods and dietary supplements have been investigated and used in clinical practice, showing them to be beneficial. Honeybee pollen from Chile has shown a large concentration of phenolic compounds and high antioxidant activity. In this work, we characterized twenty-eight bee pollen extracts from the central zone of Chile according to botanical origin, phenolic profile, quercetin concentration, and antioxidant activity (FRAP and ORAC-FL). Our results show a statistically significant positive correlation between total phenolic content and antioxidant capacity. Selected samples were evaluated on the ability to reverse the steatosis in an in vitro cell model using Hepa1-6 cells. The pollen extracts protected Hepa1-6 cells against oxidative damage triggered by 2,2′-azo-bis(2-amidinopropane) dihydrochloride (AAPH)derived free radicals. This effect can be credited to the ability of the phenolic compounds present in the extract to protect the liver cells from chemical-induced injury, which might be correlated to their free radical scavenging potential. Additionally, bee pollen extracts reduce lipid accumulation in a cellular model of steatosis. In summary, our results support the antioxidant, hepatoprotective, and anti-steatosis effect of bee pollen in an in vitro model.
