AbstrakSabun cair cuci tangan terbukti dapat menghambat pertumbuhan bakteri Escherichia coli dan Staphylococcus aureus. Sebagian besar restoran waralaba di Kota Padang menyediakan sabun cair cuci tangan yang telah diencerkan. Proses pengenceran mengubah kemampuan sabun dalam menghambat pertumbuhan bakteri. Penelitian bertujuan untuk menguji daya hambat sabun cair cuci tangan pada restoran waralaba di Kota Padang terhadap pertumbuhan E. coli dan S. aureus. Sampel diambil dari empat restoran waralaba dan diuji dengan metode difusi. Hasil penelitian menunjukkan bahwa keempat sabun dapat menghambat pertumbuhan S. aureus namun hanya sebagian yang memiliki daya hambat terhadap E. coli. Median daya hambat pertumbuhan E. coli dan S. aureus pada setiap periode yaitu 0, 7.4, 7.3 dan 0, 26.1, 23.3. Nilai maksimum daya hambat pertumbuhan E.coli dalam tiga periode yaitu 19.5, 35.4, 27.1 dan 20.7, 40.2, 36.6 untukS. aureus. Daya hambat minimum terhadap kedua bakteri adalah 0.00. Hal tersebut dapat dipengaruhi komposisi dan konsentrasi antiseptik, antibakteri, pH sabun, pengenceran dan struktur dinding sel bakteri. Disimpulkan bahwa sabun cair cuci tangan yang diuji memiliki kemampuan lebih besar dalam menghambat pertumbuhan S.aureus daripada E.coli.Kata kunci: sabun cair cuci tangan, restoran, pengenceran, uji daya hambat, pertumbuhan bakteriAbstractLiquid hand soaps proved to inhibit Escherichia coli and Staphylococcus aureus bacterial growth. In Padang, most of Restaurants provide diluted liquid hand soaps. Research found dilution changed soap ability to inhibit bacterial growth. The purpose of this study was to examine the ability of Padang City Restaurants’ liquid hand soaps to inhibit E. coli and S. aureus bacterial growth. The samples were taken from four restaurants and examined by using diffusion method. The results showed all of them could inhibit S. aureus but only a half inhibited Escherichia coli bacterial growth. Median for E. coli and S. aureus bacterial inhibition growth for each period were 0, 7.4, 7.3 and 0, 26.1, 23.3. Maximum inhibition value for E. coli growth in the first, second, and third periods were 19.5, 35.4, 27.1 and 20.7, 40.2, and 36.6 for S. aureus. In addition, minimum inhibition in both bacteria were 0.00. It could be influenced by soap antiseptic and antibacterial composition and concentration, pH, dilution, and structure of bacterial cell wall. In conclusion, the liquid hand soaps has greater ability to inhibit S. aureus than E. coli.Keywords: liquid hand soaps, restaurant, dilution, inhibitory test, bacterial growth

Penelitian ini bertujuan untuk menguji daya hambat dari ekstrak metanol beberapa jenis porifera terhadap pertumbuhan bakteri E. coli dan S. aureus. Pengujian aktivitas antibakteri menggunakan metode Kirby-Bauer, yaitu dilakukan dengan mengukur zona hambat di sekeliling cakram kertas. Ekstrak Haliclona sp dapat menghambat pertumbuhan S. aureus pada konsentrasi ekstrak 30%, 60% dan 90% dengan hasil pengukuran diameter zona hambat 13.50 mm, 20.67 mm dan 27.33 mm; serta menghambat pertumbuhan E. coli dengan diameter zona hambat 10.08 mm, 12.83 mm dan 14.17 mm. Daya hambat ekstrak Agelas sp terhadap S. aureus menunjukkan diameter zona hambat 8.33 mm dan hanya menunjukkan pada konsentrasi ekstrak 90%, sementara daya hambat ekstrak Agelas sp pada konsentrasi 30%, 60% dan 90% terhadap E. coli menunjukkan diameter zona hambat sebesar 7.67 mm, 10.17 mm dan 14.17 mm. Daya hambat Spheciospongia sp terhadap S. aureus dan E. coli hanya terlihat pada konsentrasi ekstrak sebesar 90% dengan diameter zona hambat adalah 8.42 mm dan 8.75 mm. Berdasarkan hasil yang diperoleh, dapat disimpulkan bahwa ekstrak Haliclona sp memiliki potensi aktivitas antibakteri yang dapat digunakan sebagai bahan dasar pembuatan obat antibiotik.This research aimed to test the inhibition capabilities of methanol extract from several kinds of Porifera on Escherichia coli and Staphylococcus aureus growth. The antibacterial activity test using the Kirby-Bauer method, which delivered by measuring the inhibition zone around paper disc. The extract of Haliclona sp can inhibit the S. aureus growth at 30%, 60% and 90% of extracts concentration with the measurement of inhibition zone diameters are 13.50 mm, 20.67 mm and 27.33 mm; also inhibit the E. coli growth with inhibition zone diameters are 10.08 mm, 12.83 mm and 14.17 mm. The inhibition capability of Agelas sp extract on S. aureus shows that the inhibition zone diameters is 8.83 mm and only appear at 90% of extracts concentration, meanwhile the inhibition capability of Agelas sp extract at concentration 30%, 60% and 90% on E. coli shows diameters of inhibition zone are 7.67 mm, 10.17 mm and 14.17 mm. The inhibition capability of Spheciospongia sp on S. aureus and E. coli only occurred at 90% of extracts concentration with inhibition zone diameters 8.42 mm and 8.75 mm. Based on the results, it can be assumed that extracts of Haliclona sp has a potential antibacterial activity that can be used as a basic ingredients for antibiotic medicine.

AbstractThe aim of this study is to test two different methods for evaluating the in vitro antibacterial effect of Thymus fontanesii Boiss. et Reut. essential oil against standard and clinical bacterial strains responsible for bovine mastitis: the disc diffusion method or the aromatogram which allows the demonstration of the antibacterial power of essential oils on the bacterial strains tested, Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923 and two strains isolated from bovine mastitis milk S. aureus and E. coli. The inhibition activity of the essential oil of T. fontanesii on bacterial strains by the two methods shows that the antimicrobial power of this oil is very important and is characterized by bactericidal and bacteriostatic action against gram negative and gram positive bacteria. The antimicrobial evaluation by the aromatogram showed good antibacterial activity against all the strains tested, the zones of inhibition of the bacteria were between 23,33±1,527mm and 37,5±3,535 mm. The search for minimum inhibitory concentrations MIC and bactericides CMB made it possible to quantitatively assess the antimicrobial power of this essential oil. In this work, the MIC was 0,625 µl/ml for all strains tested, and the lowest CMB was that of T. fontanesii against E. coli ATCC 25922 was 0,625 µl/ml.

Devido ao aumento da resistência de patógenos humanos e animais aos medicamentos atuais, pesquisas por novos antimicrobianos têm sido incentivadas, principalmente por meio de plantas medicinais. Eugenia uniflora L., conhecida como pitangueira, é popularmente utilizada com diversas funções terapêuticas, justificando estudos nesta espécie. Assim, os objetivos deste estudo foram avaliar a atividade antibacteriana “in vitro” de extratos de folhas e cascas de Eugenia uniflora L. sobre Escherichia coli e Staphylococcus aureus. Para as etapas de triagem fitoquímica e microbiologia, foram utilizadas as metodologias usuais nessas áreas para obtenção dos extratos e avaliação da atividade antibacteriana com uso de controle positivo e negativo. Os resultados obtidos mostraram que no extrato das folhas foi verificada a presença de saponinas, flavonóides, alcalóides e taninos, enquanto na casca foram obtidos apenas saponinas e alcalóides. Os extratos da folha e da casca foram capazes de inibir o crescimento de Stapylococcus aureus apenas, promovendo halos de inibição de meio de 19,4 mm e 14,4 mm, respectivamente. As análises permitiram concluir que os extratos apresentam metabólitos secundários interessantes e de grande potencial antimicrobiano e que esses extratos foram capazes de inibir apenas as cepas de S. aureus, sendo resistentes às cepas de E. Coli.

A atividade antimicrobiana de uma cultura comercial probiótica  deLactobacillus  sakei  (ATCC  1521)  foi avaliada em termos de concentração celular (30, 42, 60, 78 e 90 mcg / ml, 105 - 107UFC/mL), temperatura de incubação (4, 10, 20, 30, 37 ° C) e pH inicial do meio de cultura (3.5, 4.0, 4.5, 5.0 e 5.5) contra três patógenos alimentares, Escherichia coli, Listeria monocytogenes e Staphylococcus aureus. O efeito antagônico da cultura probiótica in vitro foi realizada através da aplicação de método quantitativo espectrofotométrico. Os resultados indicaram que uma substância inibitória presente em caldo de cultura de 24 horas poderia ser vantajoso quando mantendo a cultura dominante durante fermentações mais prolongadas. Para a maior produção de ácido láctico (1,04%v/v), o período de incubação da bactéria em caldo MRS foi de 24 horas em condições aeróbicas e a 37 ºC, com um valor de pH mínimo do sobrenadante (3,5). Os dados sugerem que o sobrenadante pode ter atividade bacteriostática significativa contra E. coli,L. monocytogenes e S. aureus, e pode fornecer carnes curadas com um grau de protecção contra este microorganismo, particularmente se empregada com uma combinação de pH ácido, e de refrigeração adequada.

The problem of resistance is increasing along with the need for antimicrobials as an alternative in overcoming resistance problems. One solution is to utilize the content of secondary metabolite compounds in rhizome plants such as ginger. The content of compounds that are usually the most dominant in ginger such as essential oils, flavonoids, terpenoids and phenols. This study aims to determine the effect of ginger rhizome extract as an antibacterial on the growth of Staphylococcus aureus and Escherichia coli in vitro. This study was a laboratory experimental study with a completely randomized design (CRD) consisting of 7 treatments with 3 replications. The results showed that the ethanol extract of ginger rhizome had antibacterial activity against S. aureus and E. coli. At concentrations of 20%, 40% and 80% which are more effective in inhibiting the growth of these two bacteria. The higher the extract concentration level, the larger the diameter of the inhibition zone from bacterial growth. This inhibition of bacterial growth is thought to have an effect on the content of the ginger root.   Keywords: Ginger rhizome, Staphylococcus aureus, Escherichia coli, antibacterial ABSTRAK   Permasalahan tentang resistensi semakin meningkat seiring dengan kebutuhan antimikroba sebagai alternatif dalam mengatasi masalah resistensi. Salah satu solusinya yaitu dengan memanfaatkan kandungan dari senyawa metabolit sekunder pada tanaman rimpang-rimpangan seperti rimpang jahe. Kandungan senyawa yang biasanya paling dominan pada jahe seperti minyak atsiri, flavonoid, terpenoid dan fenol. Penelitian ini bertujuan untuk mengetahui pengaruh dari ekstrak rimpang jahe sebagai antbakteri terhadap pertumbuhan Staphylococcus aureus dan Escherichia coli secara in vitro. Penelitian ini termasuk penelitian eksperimental laboratorium dengan rancangan acak lengkap (RAL) yang terdiri dari 7 perlakuan dengan 3 kali ulangan. Hasil penelitian diperoleh bahwa ekstrak etanol rimpang jahe memiliki aktivitas antibakteri terhadap S. Aureus dan E. coli. Pada konsenrasi 20%, 40% dan 80% yang lebih efktif dalam menghambat pertumbuhan kedua bakteri tersebut. Semakin tinggi tingkatan konsentrasi ekstrak maka diameter zona hambat dari pertumbuhan bakteri juga semakin besar. Terhambatnya pertumbuhan bakteri ini diduga adanya pengaruh dari kandungan pada rimpang jahe.   Kata Kunci: Rimpang Jahe, Staphylococcus aureus, Escherichia coli, antibakteri

Aegle marmelos L. is one of the tropical plants that has many benefits and also has the potential to be used as herbal medicines. It caused in some areas in Indonesia, A. marmelos have been widely used in traditional medicine. This research was a qualitatively descriptive study to see the effectiveness of A. marmelos leaf extract in inhibiting the growth of Escherichia coli and Staphylococcus aureus bacteria. Stages of A. marmelos leaf extract inhibition test on bacterial growth included the stages: extraction by maceration method, phytochemical test, chromatography test, anti-microbial test. Data were analyzed qualitatively. The results showed that A. marmelos leaf extract was more effective in inhibiting the growth of E. coli bacteria than in S. Aureus bacteria. This can be information that Maja leaf extract can be used as a phytopharmaca material for diseases caused by E.coli

AbstrakPendahuluan: Madu digunakan sebagai agen makanan dan obat tradisional, mengandung nektar atau gulaeksudat dari tanaman yang dikumpulkan oleh lebah madu serta merupakan salah satu obat tradisional yangdigunakan oleh masyarakat. Madu diketahui memiliki kemampuan sebagai efek antibakteri, seperti Escherichia colidan Stahpylococcus aureus. Kedua bakteri ini memiliki sifat yang berbeda tetapi memilki kesamaan dari penyakityang disebabkannya. Kualitas dan jenis madu juga sesuai dengan tempat dan lokasi lebah berkembang biak.Tujuan penelitian ini adalah untuk melihat perbandingan efek antibakteri madu asli Sikabu dan Lubuk Minturunterhadap Escherichia coli dan Staphylococcus aureus secara in vitro. Metode Penelitian: Madu yang diuji untukpenelitian ini adalah madu yang berasal dari Sikabu dan Lubuk Minturun. Penelitian ini dilaksanakan pada bulanDesember 2011 sampai Mei 2012 di laboratorium Mikrobiologi Fakultas Kedokteran Universitas Andalas. Jenispenelitian yang digunakan adalah eksperimental dengan menggunakan metode difusi (metode cakram) dananalitik dengan menghubungkan perbedaan yang dimiliki kedua madu sebagai efek antibakteri. Both of these bacteriahave different properties but have the common of the diseases it causes. The quality and type of honey is also inaccordance with the breeding places and their location. The purpose of this study was to see a comparison of theantibacterial effect of honey Sikabu and Lubuk Minturun against Escherichia coli and Staphylococcus aureus by invitro. Methods: Honey to be tested for this study were derived from Sikabu and Lubuk Minturun. This study wasconducted in December 2011 to May 2012 in the laboratory of Microbiology, Faculty of Medicine, AndalasUniversity. Type of research is experimental diffusion method (disc method) and analytical distinction owned bylinking the two of honey as an antibacterial effect. Results: These results indicate that the honey Sikabu and LubukMinturun have no antibacterial effect against Escherichia coli, both of honey have differences antibacterial effectagainst Staphylococcus aureus. Sponge samples collected around Manado waters were obtained 30 species and their crude extracted have been tested in vitro for their activity in inhibiting bacterial growth. Based on the results of antibacterial screening in 30 sponge extracts, there were 23 sponge extracts which had bioactivity in inhibiting the growth of S. aureus, E. coli, S. saprophyticus and P. aeruginosa, Sponge extract No. 43 (of 30 sponge extracts tested) was the most active in inhibiting bacterial growth and had the widest inhibition zone diameter.Keywords: screening, sponge, crude extract, antibacterial, ManadoABSTRAKSampel spons dikoleksi di sekitar Perairan Manado sebanyak 30 jenis/spesies, dimana ekstrak kasarnya telah diuji secara in vitro aktivitasnya dalam menghambat pertumbuhan bakteri. The germicidal efficacy of four common disinfectants used for different purposes was tested against two common pathogens namely Escherichia coli and Staphylococcus aureus . The disinfectants (Dettol, Jik, Izal and Purit) were diluted with sterile distilled water to achieve different concentrations (100%, 50%, 25%, 12.5% and 6.25%) and they were tested on the two organisms using Agar well diffusion method. The plates were allowed to incubate for 24 hours at 37°C.The germicidal efficiency of the disinfectants was evaluated by measuring the zone of inhibition of each disinfectant based on their varying concentrations. Dettol was observed to have the highest inhibition at 100% concentration and was more germicidal on E. coli than S. aureus with Minimal Inhibitory Concentration (MIC) value of 1:16 dilution against the two test organisms. This was closely followed by Jik which also showed more inhibitory activity against E. coli at the initial concentration recording the second highest inhibition rate compared to other disinfectants, but its efficacy decreased as the concentration dropped. Jik exhibited MIC value of 1:8 and 1:2 dilutions for Staphylococcus aureus and Escherichia coli respectively. Purit showed more inhibitory activity on S. aureus with 3.8mm zone of inhibition as against 1.3mm at 100% concentration, recording MIC value of 1:8 dilution on S. aureus and 1:16 dilution for E. coli . The disinfectant Izal was observed to be the lowest as it did not inhibit the growth of any of the organisms, indicating resistance of the organisms to the disinfectant at all concentrations. All the disinfectants apart from Izal inhibited the two test isolates. Only Dettol proved to be best among them. Therefore, Dettol is encouraged to be used in homes to prevent pathogenic infection.

SummaryThe following experiment evaluated the inhibitory activity of a resin acids-based product (RAP) to bacterial pathogens. Clostridium perfringens isolated from chickens, turkeys and pigs, Staphylococcus aureus from chickens, pigs and cattle, and Escherichia coli O149 isolated from pigs were tested. Two different methods were used, a broth dilution method (BDM) using 0.01%, 0.1% and 0.5% resin acid, and an agar diffusion method (ADM) using 0.01%, 0.1%, 0.5%, 1% and 5% resin acid. For the BDM, C. perfringens was inhibited completely at all concentrations. S. aureus was inhibited completely at 0.5%, but only slightly at 0.1% and not at all at 0.01%. The E. coli strains showed no or little inhibition at 0.5%. For the ADM, narrow inhibition zones evolved around the concentration of 0.5% (8–10 mm), 1% (8.0–12.0 mm), and 5% (9.0–19.5 mm) on the C. perfringens strains, while the inhibition zones for S. aureus were smaller and E. coli developed no inhibition zones. Overall, the RAP inhibited C. perfringens at all concentrations of the product, S. aureus at 0.1%, 0.5%, 1% and 5% concentrations, and E. coli O149 only at 0.5% concentrations, although some strain variation was recorded.
