Abstract Background The health benefits of n-3 polyunsaturated fatty acids (PUFA), such as cardioprotective, anti-inflammatory and hypo-triglyceridemic properties, have been well documented. However, current dietary modification can easily lead to excessive concentrations of n-6 PUFA and an imbalance in the n-6:n-3 PUFA ratio, resulting in the pathogenesis of obesity and related diseases. This study investigated the effects of various dietary n-6:n-3 PUFA ratios on the growth performance, blood lipid and cytokine profiles, tissue fatty acid composition, and gene expression involved in lipid metabolism in finishing pigs. Seventy-two crossbred [(Duroc × Landrace) × Yorkshire] finishing pigs (68.5 ± 1·8 kg) were fed one of four isoenergetic diets with n-6:n-3 PUFA ratios of 2:1, 3:1, 5:1 and 8:1. Results Pigs fed diets with n-6:n-3 PUFA ratios of 3:1 and 5:1 had a higher average daily gain (ADG) and average daily feed intake (ADFI) (P < 0.05) and a lower feed conversion ratio (FCR) (P < 0.05). The concentrations of triglyceride (TG) and total cholesterol (TC) of pigs fed diets with n-6:n-3 PUFA ratios of 1:1 to 5:1 decreased (P < 0·05), whereas the ratio of concentrations of high-density to low-density lipoprotein cholesterol (HDL-C:LDL-C) in pigs fed a diet with an n-6:n-3 PUFA ratio of 2:1 increased (P < 0·05) compared with those fed a diet with an n-6:n-3 PUFA ratio of 8:1. Pigs fed a diet with n-6:n-3 PUFA ratios of 2:1 and 3:1 had lower concentrations of leptin (LEP) and interleukin 6 (IL-6) (P < 0·05), and higher concentrations of adiponectin (APN) (P < 0·05) than those fed diets with n-6:n-3 PUFA ratios of 5:1 and 8:1. Diets with n-6:n-3 PUFA ratios of 2:1 and 3:1 markedly down-regulated the expression levels of peroxisome proliferator-activated receptor-γ (PPARγ) and lipoprotein lipase (LPL) in both the longissimus dorsi muscle (LM) and subcutaneous adipose tissue (SCAT), but up-regulated the expression level of adipocyte fatty acid binding protein (aP2) in the LM. The expression levels of aP2 and hormone- sensitive lipase were similar to those of PPARγ in the SCAT. Conclusions This study demonstrated that dietary n-6:n-3 PUFA ratios between 3:1 and 5:1 beneficially affected growth performance, optimize blood lipid and adipocytokine levels, enhanced absorption and deposition of n-3 PUFA and regulated lipid metabolism-related genes.

The reliability of qRT-PCR results depend on the stability of reference genes used for normalization, suggesting the necessity of identification of reference genes before gene expression analysis. Morels are edible mushrooms well-known across the world and highly prized by many culinary kitchens. Here, several candidate genes were selected and designed according to the Morchella importuna transcriptome data. The stability of the candidate genes was evaluated with geNorm and NormFinder under three different experimental conditions, and several genes with excellent stability were selected. The extensive adaptability of the selected genes was tested in ten Morchella species. Results from the three experimental conditions revealed that ACT1 and INTF7 were the most prominent genes in Morchella, CYC3 was the most stable gene in different development stages, INTF4/AEF3 were the top-ranked genes across carbon sources, while INTF3/CYC3 pair showed the robust stability for temperature stress treatment. We suggest using ACT1, AEF3, CYC3, INTF3, INTF4 and INTF7 as reference genes for gene expression analysis studies for any of the 10 Morchella strains tested in this study. The stability and practicality of the gene, vacuolar protein sorting (INTF3), vacuolar ATP synthase (INTF4) and14-3-3 protein (INTF7) involving the basic biological processes were validated for the first time as the candidate reference genes for quantitative PCR. Furthermore, the stability of the reference genes was found to vary under the three different experimental conditions, indicating the importance of identifying specific reference genes for particular conditions.

Intramuscular fat (IMF) and fatty acid composition are characteristics that are used as important indicators of evaluating high quality pork and contribute to the economic benefits of the pig farming industry. In this study, quantitative trait loci (QTL) fine mapping of chromosome 12 was performed in a population of F2 intercross between Yorkshire (YS) and Korean native pigs (KNPs) by adopting combined linkage and linkage disequilibrium method using high-density SNP chips. QTLs for IMF (H3GA0034813 to H3GA0034965) and oleic acid (C18:1) (ASGA0054380 to ALGA0066299) were located at 120 cM (54.112–57.610 kb) and 85 cM (36.097–38.601 kb), respectively, within chromosome 12 (Sscrofa11.1 genomic reference). In addition, 31 candidate genes present within the IMF QTL region and 28 candidate genes existing within C18:1 QTL region were chosen. In order to understand the function of these candidate genes at the molecular level, these candidate genes were functionally categorized by studying gene ontology and analyzing network and pathway. Among the 59 candidate genes within the region of IMF QTL and C18:1 QTL, five (MYH1, MYH2, MYH4, ACACA, and RPS6KB1) directly interacting candidate genes were found. Furthermore, the RPS6KB1 gene was assumed to be an important candidate gene that is involved in leptin and insulin signaling pathway and participates in controlling adipogenic differentiation, fat deposition, and fatty acid composition, which is related to obesity of pigs.
