Estimation of the polymorphism of matrix metal proteinase genes in patients with endometrioid ovarian cysts

Aim. To determine the association of the MMP-9 genetic polymorphism with the risk of developing endometrioid ovarian cysts (ECC). Materials and methods. 55 women aged 19 to 47 were examined. 27 women underwent surgery for ECC (group 1). The control (group 2) consisted of 28 patients without endometriosis and operated because of tubal infertility. Single nucleotide polymorphism was investigated. Genotyping was performed by restriction fragment length polymorphism analysis. Results. We did not find statistically significant differences between the group of patients with ECF and the control group in terms of age 29.0 (25.95; 33.1), 34.5 (29.3; 37.0); p0.05, body mass index 21.2 (19.8; 22.6), 21.95 (20.4; 23.9), p0.05, age of menarche onset 13.0 (12.95; 14.0), 13.0 (12.0; 14.0), p0.05, duration of menstrual bleeding 5.0 (5.0; 5.0), 5.0 (5.0; 6.0), p0.05 for the 1st and 2nd groups, respectively, also dysmenorrhea (2 0.019; p=0.8918), the number of births (2 3.441; p=0.3285) and abortions (2 2.822; p=0.0930) in anamnesis. The frequencies of all studied genotypes of metalloproteinase MMP-9 C (1562) T of the MMP9 gene in the group of patients with ECF and the control group are in HardyWeinberg equilibrium (p=0.99, p=0.43 for 1 and 2 group) which excludes differences in the distribution of genotype frequencies of polymorphic loci of MMP9 genes in the group of patients with ECF and the control group. However, the result could be influenced by the severity of the disease and the size of the study groups. Conclusion. Given the undoubted role of matrix metalloproteinases in the pathogenesis of genital endometriosis, further studies with large samples in various populations are needed.

Repairing bone defects in oral surgery often requires the use of bone regeneration techniques. Silicon is an element that has been employed as regeneration material in several studies. In our study, silicon was combined with autologous bone and platelet-rich fibrin (PRF) membranes to analyse the behaviour of this element in bone regeneration. Four circumferential defects were created in the cranial vault of five New Zealand rabbits. The following elements were applied to the regeneration of the defects: (P): PRF; (S): silicon and (B): autologous bone, with the following distribution of study groups: Group 1 (PSB); Group 2 (PS); Group 3 (SB) and Group 4 (CONTROL): unregenerate group. The animals were sacrificed after 3 weeks. Computed microtomography studies (μ-CT) were carried out, as well as histomorphometric ones. The ANOVA statistical test was used with a Bonferroni post-hoc test to compare the results (p ≤ 0.05). Radiologically, groups PSB and SB were better as far as quantity and percentage of healthy bone observed, but not significantly compared to the control group. The PS group was significantly worse. The histological test revealed that the PSB group was the one to present the largest area, percentage and perimeter of mineralised bone. On evaluating the forming bone (osteoid), no difference was observed across the groups with the exception of the bone perimeter, where the SB group was significantly better. The bone height variable showed no significant differences. In conclusion we can state that the combination of PRF, autologous bone and silicon provides good results at 3 weeks whilst the PS group shows the worst results. This highlights the importance of autologous bone forming part of the graft material in order for the bone to mineralise.

Bronchial asthma is a multifactorial disease, with both environmental factors and genetic predisposal affecting its development. A number of gene associations have been obtained between polymorphisms of cytokine genes produced by different types of immune cells and asthma development. Interleukin-13 is involved in allergic inflammation, increased bronchial hypersensitivity, regulation of eosinophil levels and IgE production by B cells, thus making it promising for studying IL13 gene polymorphisms in bronchial asthma coupled to development of the disease. The aim of this study was to investigate possible association between asthma and IL13 rs1800925 polymorphism in the children of Caucasian origin in Eastern Siberia. Four groups of patients with asthma were examined (mean age 12.8±1.2 years): with a controlled (n = 95) and uncontrolled course (n = 107), with severe (n = 71) and moderate severity (n = 131) diseases. The control group consisted of healthy individuals: children (n = 33) and adults (n = 102). DNA was isolated with sorbent method; genotyping was carried out using RT-PCR using specific oligonucleotide primers and fluorescent TaqMan probes. The allele and genotype frequencies were compared by the χ-square test using an online calculator. The odds ratio (OR) with a 95% confidence interval (CI) was performed to link genetic markers with pathological phenotypes. The CT IL13 rs1800925 genotype was shown to be associated with moderate asthma and cases of uncontrollable clinical course, whereas the TT genotype was associated with severe asthma. Thus, rs1800925 polymorphism of IL13 gene (the T* variant is known to be associated with increased IL-13 expression) may be associated with bronchial asthma in children. Our data are consistent with results of other authors. E.g., Liu Z. et al. revealed an association between rs1800925 IL13 and the risk of developing asthma in children, with CT and TT genotypes being more common in the patient group. Radhakrishnan A. et al., was studied rs1800925 IL13 in adult population of Malaysia and found that the T* allele frequency in the group of patients significantly exceeds the frequency of this allele in the control group. Thus, the results of our study showed that IL13 rs1800925 polymorphism is associated with bronchial asthma in children, especially, with level of its control and severity of the disease.

Abstract Background Zinc transporter 8 autoantibodies (ZnT8Abs) together with glutamic acid decarboxylase autoantibodies (GADAbs), insulinoma antigen 2 autoantibodies (IA-2Abs) and insulin autoantibodies (IAbs) are markers of type 1 diabetes mellitus (T1DM). We studied the prevalence of ZnT8Ab in children with autoimmune thyroid diseases (AITDs) to assess the association of AITDs and T1DM at the serological level. Methods The study groups consisted of 44 children with Graves’ disease (GD), 65 children with Hashimoto’s thyroiditis (HT), 199 children with T1DM with or without AITDs and 58 control children. ZnT8Ab, GADAb, IA-2Ab, IAb, 21-hydroxylase autoantibodies (21-OHAbs) and acetylcholine receptor autoantibodies (AChRAbs) were measured. Results ZnT8Abs were found in 4/44 (9.1%) patients with GD, and 4/44 (9.1%) patients with GD were positive for GADAb. Of the 65 HT patients, six (9.2%) were positive for ZnT8Ab, while four (6.2%) were positive for GADAb. In the T1DM group, 128/199 (64%) of the patients were positive for ZnT8Ab, 133/199 (67%) for GADAb and 109/199 (55%) for IA-2Ab. One GD patient and one HT patient were positive for all the four diabetes-associated autoantibodies. Two HT patients were positive for three diabetes autoantibodies. Two GD (4.5%) and five HT (7.7%) patients were positive for 21-OHAb only. None of the patients had AChRAb. In the control group, 2/58 (3.4%) were positive for GADAb and 2/58 (3.4%) were positive for ZnT8Ab. Conclusions Diabetes-associated autoantibodies including ZnT8Ab were found in children and adolescents with GD and HT.

The objective of the work: to study the changes of the functional activity of the endothelium and the plasma-coagulation link of hemostasis in women with chronic inflammatory processes of the internal genital organs (CIPIGO) with and without varicose veins of the small pelvis (VVSP).Materials and methods: 68 women with chronic inflammatory processes of the internal genital organs against the background of varicose veins of the small pelvis (group 1); 52 women with chronic inflammatory processes of the pelvic organs without varicose veins of the small pelvis (group 2); 30 practically healthy women (control group) were examined before and after the treatment. The age of women was between 18 and 45 years old. All the patients of the study groups received the treatment offered by us, which included a diosmine-containing phlebotropic preparation normoven, 500 mg twice a day in continuous mode and a donor of nitric oxide tivortin aspartatum, 5 ml 3 times per os for 14 days each month during six months.Determination of concentration of endothelin-1, prostacyclin was performed by immunoassay using a set of reagents from the firm “Biomediсa” (Austria); the determination of D-dimers in blood plasma was performed by latex-test of the firm “Organon-Tekhnika”.Results of workIn women with CIPIGO with and without VVSP, the increased production of endothelin-1, which has a pro-aggregate and pro-inflammatory effect, was determined. At the same time, the decrease of the synthesis of prostacyclin, which has anti-aggregant properties, was noted. In evaluating the indicators of coagulograms in patients of the studied groups, the propensity for hypercoagulation was revealed, as evidenced by an increase in the level of fibrinogen and a rise of index of the phospholipid-dependent coagulation tests. The decrease of the percentage of fibrinolytic blood activity and the increase in plasma concentration of the molecular marker of thrombophilia D-dimer can be considered as activation of intravascular coagulopathy and the presence of endothelial dysfunction.Conclusions: 1. Our studies found endothelial dysfunction and violations of the plasma-coagulation linkage of hemostasis in both studied groups. 2. After the performed treatment, the improvement of the functional activity of the endothelium and normalization of the main parameters of the plasma-coagulation linkage of hemostasis were observed.

<p><strong>OBJECTIVE:</strong> The objective of the study was to investigate the effect of immersion in water strategy during labor on postpartum bleeding by calculating the postpartum reduction rates of the hematocrit values of the patients.</p><p><strong>STUDY DESIGN:</strong> The study groups consisted of 84 women undergoing vaginal delivery with immersion in water during labor (group 1) and the control group (group 2) of 84 women undergoing normal vaginal delivery. Patients who have received additional medical and surgical interventions for alleviation of postpartum hemorrhage, patients who have undergone an episiotomy and/or perineal trauma were not included in the study. All data were taken from patients who have delivered with spontaneous vaginal delivery. Postpartum hematocrit fall rates of the groups have been compared and the effect of immersion in water on postpartum hemorrhage has been evaluated.</p><p><strong>RESULTS:</strong> The study groups consisted of 84 women undergoing vaginal delivery with immersion in water during labor (Group 1) and the control group (Group 2) of 84 women undergoing vaginal delivery at the hospital. The women in the two groups were matched with respect to age, parity, birth weight and gestational age. The mean age of the women was 29.8±4.8 and 30.5±4.9 respectively. The mean hematocrit difference in the first group was 2.08 ± 1.88 and in the second group was 3.81 ± 1.55. The mean percentage of hematocrit reduction in the first group was 5.71% and in the second group 10.23%.</p><p><strong>CONCLUSION:</strong> Our data showed that mean hematocrit level decreases among women following vaginal delivery more than women who give birth vaginally within immersion in water during labor. The percentage of hematocrit reduction in the water birth group was lower than in the control group. Water birth seems to facilitate uterine contractions more efficiently following vaginal delivery.</p>

Drugs induced nephrotoxicity now considered as an essential reason for kidney failure. Aminoglycoside anti-infection agents, for example, gentamicin, which causes ototoxicity and nephrtotoxicity as a side effect , this research is centered around the utilization of natural materials as an antioxidants against the lethal oxidative activity that applied on the kidney cells ,the most imperative one of these materials is the honey. This work aimed to assess the defensive impact of citrus honey against the histopathological and biochemical harms of gentamicin induced nephrotoxicity .24 locally breed rabbits (Oryctolagus cuniculus) were used and divided into 4 study groups (6 rabbits per each group), group 1 received I.P daily dose of normal saline (control), group 2 received (80 mg/kg/day) I.P dose of gentamicin, group 3 received (80mg/kg/day) of gentamicin I.P dose in combination with oral dose of Citrus Honey625 mg/kg/day for 14 days and group 4 received (80mg/kg/day) of gentamicin I.P dose in combination with extra dose of Citrus Honey orally for 14 days. All animals (at 15th day) were anesthetized by ether and sacrificed; blood samples were gathered for the subsequent measurement of the serum creatinine, urea and albumin  while an isolated kidney was kept in 10 % of formaldehyde for the histopathological examination. The results demonstrated that gentamicin causes nephrotoxicity showen by elevation of serum level of creatinine, urea and  a decrease in the serum albumin. While the administration of honey in combination with gentamicin reduced the nephro-toxic effect of gentamicin that represented by a reduction of the serum creatinine and urea with improvement of the kidney histological findings.This study concluded that, honey decreased nephrotoxic effect of  gentamicin   http://dx.doi.org/10.25130/tjps.25.2020.001

Different procedures of treatment of large follicular ovarian cysts in 177 sows using GnRH, hCG and PGF2α are evaluated in this study. Ovarian cysts were diagnosed by transcutaneous ultrasonography, which was a part of routine pregnancy diagnosis. No treatment was performed in the control group (Group 1, <i>n</i> = 29); the method of treatment used in the other groups immediately after the diagnosis was intramuscular administration of lecirelin in doses 50 µg (Group 2, <i>n</i> = 28), 100 µg (Group 3, <i>n</i> = 27) and 200 µg divided into 2 equal doses administered at a 12-hour interval (Group 4, <i>n<i> = 25) and of hCG in doses 1 500 IU (Group 5, <i>n</i> = 23), 3 000 IU (Group 6, <i>n</i> = 21), and 250 µg of cloprostenol (Group 7, <i>n</i> = 24). Insemination rate (IR), conception rate (CR) in inseminated sows, pregnancy rate (PR = recovery rate), treatment-insemination interval (TII) and treatment-pregnancy interval (TPI) within 42 days after the initial examination were evaluated. In addition PR in groups of sows divided according to parity (1–3, 4–6 and ≥ 7) were also evaluated. IR and PR were higher in Group 4 (84.0% and 44.0%) and lower in Group 1 (17.2% and 6.9%) in comparison with the other groups (<i>P</i> < 0.001 and <i>P</i> < 0.05). CR, TII and TPI did not differ between the experimental groups. PR were similar in sows with different parity. The study proved a positive response in sows with large follicular ovarian cysts to the treatment consisting of 2 administrations of 100 µg GnRH at a 12-hour interval.

Abstract Objective: To determine the correlation between spontaneous abortion and Chlamydia trachomatis infection. Methods: A cross-sectional study was conducted in women who experienced spontaneous abortion. Normal pregnancies with gestational age more than equal to 37 weeks as control. Detection of C. trachomatis in the product of conception or placenta from curettage using the PCR method. Results: Positive C. trachomatis was found 3 cases in the abortion group and 4 cases in control. In the abortion group, C. trachomatis found in 1 case with vaginal discharge history and 2 cases without this history. C. trachomatis also found in 3 cases without a history of abortion. This bacteria was not found in patients with an abortion history. In the control group, 2 cases of positive C. trachomatis were found in pregnant women with or without a history of vaginal discharge and abortion, respectively. There were no significant differences regarding the positive of C. trachomatis between the two study groups regarding history of vaginal discharge and abortion. Conclusions: Spontaneous abortion does not correlate with C. trachomatis infection. Keywords: chlamydia trachomatis, infection, spontaneous abortion. Abstrak Tujuan: Untuk mengetahui hubungan antara abortus spontan dan infeksi Chlamydia trachomatis. Metode: Penelitian potong lintang dilakukan pada perempuan yang mengalami abortus spontan. Kehamilan normal dengan usia kehamilan lebih dari sama dengan 37 minggu sebagai kontrol. Deteksi C. trachomatis pada produk konsepsi atau plasenta dari kuretase menggunakan metode PCR. Hasil: C. trachomatis positif ditemukan 3 kasus pada kelompok aborsi dan 4 kasus dalam kontrol. Pada kelompok abortus spontan, C. trachomatis ditemukan pada 1 kasus dengan riwayat keputihan dan 2 kasus tanpa riwayat keputihan. C. trachomatis juga ditemukan pada 3 kasus tanpa riwayat abortus. Bakteri ini tidak ditemukan pada pasien dengan riwayat abortus. Pada kelompok kontrol, 2 kasus positif C. trachomatis masing-masing ditemukan pada perempuan hamil dengan atau tanpa riwayat keputihan dan abortus. Tidak ada perbedaan signifikan C. trachomatis positif antara kedua kelompok penelitian berdasarkan riwayat keputihan dan abortus. Kesimpulan: Abortus spontan tidak berkorelasi dengan infeksi C. trachomatis. Kata kunci: abortus spontan, chlamydia trachomatis, infeksi.

The neuropeptides comprise an important part in the nervous system interacting with endocrine and immune systems. Peptide regulators are responsible for the continuity of communicating elements, which support homeostasis, however, despite abundant research examining neuropeptides, not all specific mechanisms and features of interacting proteins with cells and immune components have been uncovered. Objective: to perform a comprehensive assessment of neuropeptide system in patients with herpes zoster. Materials and methods: 106 in-hospital patients were examined diagnosed with herpes zoster within 2016–2019 period. Control group consisted of 30 healthy age- and sex-matched volunteers. Blood serum was collected after verifying diagnosis on day 1. After discharge, patients were monitored for signs of pain syndrome and overall state within 3 months. It allowed to divide patients into 3 groups retrospectively. Group 1 — patients with herpes zoster, accompanied by mild or moderate pain syndrome; group 2 — patients with herpes zoster, accompanied by severe pain; group 3 — patients with herpes zoster, complicated by postherpetic neuralgia. Level of serum protein s100B, myelin basic protein, nerve growth factor, brain-derived neurotrophic factor, neuron specific enolase was measured by using specific reagents purchased from “R&D Diagnostics Inc.” (США). Results. it was found that level of serum protein S100B in all groups was significantly increased compared to control group, showing no inter-group differences. Amount of myelin basic protein in all study groups vs. control was significantly higher. Moreover, level of these parameters in group 2 vs. group 1 and 3 was significantly elevated. In addition, level of nerve growth factor was significantly increased in group 1 vs. groups 2 and 3, whereas in group 3 it was significantly lower than in control and group 2. Brain-derived neurotrophic factor was significantly decreased in all the study groups compared to control, showing no significant intergroup differences. Level of neuron-specific enolase was significantly increased in group 3 vs. control as well as group 1 and 2. The data obtained allowed to identify two parameters for assessing a risk of postherpetic neuralgia in acute herpes zoster, as well as provided deeper insights into the pathogenesis of neuroimmune disorders accompanying herpes zoster.

Salmon skin extract contains high proline and hydroxyproline, and has been suggested as a potential topical agent for traumatic oral ulcer healing. The aim of this study was to evaluate the effect of salmon skin extract as traumatic oral ulcer healing. A total of 32 Wistar rats (200 g to 250 g) were distributed into four groups. Group 1 served as the control group (no treatment), Group 2 was topically treated with salmon skin extract agent 4%, Group 3 was topically treated with salmon skin extract agent 5%, and Group 4 was topically treated with salmon skin extract agent 6%. Traumatic ulcers at lip mucosa were performed in all rats and 0.1 ml salmon skin extract was applied on the ulcer twice daily for seven days. The animals were euthanised on the last day of treatment. Biopsy specimens were taken from the lip mucosa in all rats for epithelial thickness evaluation and the study for number of fibroblasts by histological analysis. Significant increase in epithelial thickness and the number of fibroblasts (p > 0.05) was observed in salmon skin extract treatment groups as compared to the control group. Salmon skin extract 6% treatment group had the highest epithelial thickness and the number of fibroblasts amongst the study groups. Salmon skin extract promises an innovative topical application treatment for traumatic oral ulcer healing. Salmon skin extract 6% was the most effective concentration for traumatic oral ulcer healing.
