With respect to given contradictory data in radiobiology, that according to some authors, irradiation of cells of plant and animal origin initiates DNA changes and according to others, repeated irradiation of cells in small doses has a stimulating effect on metabolism without increasing the purity of mutations, we conducted the present study, with the aim to study the stability of the genome of MDBK cell line against a background of single and double (fractional) irradiation. The object of the study was a continuous MDBK cell line (Madin-Darby Bovire Kidney Cell, cattle, kidney), which was grown in medium 199, manufactured by Federal State Budget Institution FCTRB-ARRVI (Kazan) with 10% bovine serum and the addition of antibiotics (benzylpenicillin sodium salt, kanamycin at 100 units/ml) (streptomycin sulfate at 100 μg/ml) according to standard procedure. A monolayer culture of MDBK cells in the logarithmic growth phase (3-4 days after culture) served as contacting cells. As a result, a radio-modified cell line, called MDBK-2, was obtained. Moreover, in an unirradiated MDBK cell culture, the level of chromosomal breakdowns was in the range of 1.03-3.1% with respect to the total number of studied anaphases. Based on the obtained data, it can be concluded, that during prolonged cultivation of MDBK cells, irradiated in a small (adaptive) dose and re-irradiated in 3-minute higher dose (5.95 Gy), there is a significant decrease in the yield of aberrant cells, induced by a test dose of γ-rays.

The Madin–Darby bovine kidney (MDBK) cell line is currently used for the production of bovine alphaherpesvirus-1 (BoHV-1) vaccine. For the purpose of vaccine manufacturing, suspension cells are preferred over adherent ones due to simplified sub-cultivation and an easier scale-up process, both of which could significantly reduce production cost. This study aimed to establish a procedure for the culture of BoHV-1 in the suspended MDBK cell line in serum-free medium. We screened several commercially available serum-free media and chose ST503 for subsequent experiments. We successfully adapted the adherent MDBK cells to suspended growth in ST503 in the absence of serum. The maximum density of suspension-adapted MDBK cells could reach 2.5 × 107 cells/mL in ST503 medium with optimal conditions. The average size of suspension-adapted cells increased to 18 ± 1 µm from 16 ± 1 µm. Moreover, we examined tumorigenicity of the suspended cells and found no sign of tumorigenicity post adaptation. Next, we developed a protocol for the culture of BoHV-1 in the cell line described above and found that ultrasonic treatment could facilitate virus release and enhance virus yield by 11-fold, with the virus titer reaching 8.0 ± 0.2 log10TCID50/mL. Most importantly, the prototype inactivated BoHV-1 vaccine we generated using the suspension cultures of MDBK cells induced neutralizing antibodies to a titer comparable to that of the commercial inactivated BoHV-1 vaccine. Overall, we established and optimized a protocol for the production of inactivated BoHV-1 vaccine in MDBK cells adapted for suspension culture, which provides insights for future large-scale manufacturing of BoHV-1 vaccine.

Neutral oligomannosides possessing one GlcNAc (OS-Gn1) and two GlcNAc (Os-Gn2) at the reducing end have been reported to be released during the N-glycosylation process in various biological models. To investigate which enzyme is responsible for OS-Gn1 formation, we used the Madin-Darby bovine kidney (MDBK) cell line which exhibits neither lysosomal chitobiase nor endoglucosaminidase activities. However, these cells produced OS-Gn1 and we showed that a neutral chitobiase is responsible for the transformation of OS-Gn2 into OS-Gn1. Using streptolysin O-permeabilized MDBK cells, we demonstrated that this neutral chitobiase activity is located in the cytosolic compartment and is active on oligomannoside species released during the N-glycosylation process.

The development of the pharmaceutical industry is one of the priority tasks of our state, aimed at providing the population of the Russian Federation with modern safe and effective medicines. The solution to this problem is impossible without the formation of a highly qualified personnel potential that meets the demand and expectations of the pharmaceutical market and society as a whole. In this regard, in the system of training of pharmacists in recent years, quite dynamic and flexible transformations have been taking place, dictated by the urgent needs of domestic health care. It should be noted that in the process of implementing this educational standard, the competency-based approach to organizing the process of training modern pharmacists comes to the fore. One of the effective tools for the formation of professional competencies in various educational fields is the case study method. Case study is a training method based on the analysis of real situations from various areas of professional activity and contributing to the development of specialist competency. The competency-based orientation of the case study method is in line with modern ideas about the organization of the educational process for the training of pharmacists. The case study method is actively used in the process of teaching disciplines of the curriculum of the Federal State Budget Educational Establishment of Higher Education KubGMU of the Ministry of Health of Russia, specialty 33.05.01 Pharmacy. Examples of case study tasks as educational technology are presented in the work programs of the curriculum disciplines of the specialty 33.05.01 Pharmacy developed at the Department of Pharmacy. Depending on the content of the taught discipline, these tasks simulate a particular situation from the professional activities of pharmacists, offered to students for a comprehensive analysis and evaluation. The use of this educational technology contributes to the integration of knowledge, skills acquired in the learning process and their competency-based profiling in accordance with the current level of development of domestic health care.

The information resources of the Federal State Budget Scientific Institution “Marine Hydrophysical Institute of RAS” (FSBSI MHI) oceanographic data bank (MHI BOD), which contains about 115,000 oceanographic and more than 27,000 hydrochemical stations accomplished in the Black Sea coastal zone, as well as experience accumulated while providing information support of the coastal zone research, main directions of activities, and short-term plans are considered.


The article analyzes the dynamics of the indicators of target enrollment and education at the Federal State Budget Educational Institution of Higher Education of the Far Eastern State Medical University of the Ministry of Health of Russia for the period from 2013 to 2018, a comparative assessment of the passing score for the places of targeted enrollment and the main places depending on the specialty (training direction) was made, average mark of a unified state exam (USE) was analyzed; the agreements on targeted training with the assessment of measures of social support were analyzed, as well as proposals were formulated to improve the activities of targeted training of medical personnel.

Bovine viral diarrhea virus (BVDV) entry into bovine cells involves attachment of virions to cellular receptors, internalization, and pH-dependent fusion with endosomal membranes. The primary host receptor for BVDV is CD46; however, the complete set of host factors required for virus entry is unknown. The Madin-Darby bovine kidney (MDBK) cell line is susceptible to BVDV infection, while a derivative cell line (CRIB) is resistant at the level of virus entry. We performed complete genome sequencing of each to identify genomic variation underlying the resistant phenotype with the aim of identifying host factors essential for BVDV entry. Three large compound deletions in the BVDV-resistant CRIB cell line were identified and predicted to disrupt the function or expression of the genesPTPN12,GRID2, andRABGAP1L. However, CRISPR/Cas9 mediated knockout of these genes, individually or in combination, in the parental MDBK cell line did not impact virus entry or replication. Therefore, resistance to BVDV in the CRIB cell line is not due to the apparent spontaneous loss ofPTPN12,GRID2, orRABGAP1Lgene function. Identifying the functional cause of BVDV resistance in the CRIB cell line may require more detailed comparisons of the genomes and epigenomes.

The development of the table grapes market is directly related to the renewal of the assort-ment and technological modernization of pro-duction, taking into account the growing re-quirements of consumers. Equally important is the discrepancy between the tastes of con-sumers of fresh grapes and the quality indica-tors of the grapes supplied by the industry, therefore, an important task is to improve the assortment. Recently, a large number of pri-vate breeders have joined the breeding work on the breeding of table varieties, the results of whose work often have no scientific as-sessment. The article presents the results of a variety study of five selected promising table hybrid forms of grapes obtained by amateur winegrowers. Variety studies were conducted in 2018–2020 at the experimental vineyard of the experimental field All-Russian Research Institute for Viticulture and Winemaking – Branch of Federal State Budget Scientific In-stitution «FRARC». The methods used for the assessment were generally accepted in vit-iculture. Vineyards grafted, covering, non-irrigated, planting scheme 3×1.5 m. The agro-biological characteristics are given, the yield is estimated, the uvological, tasting and eco-nomic assessment of hybrid forms is given. The studied varieties have desirable character-istics: high quality, attractive appearance, large and very large bunches and berries, dense pulp consistency, different tastes, bright color of berries.

Abstract. The article discusses the issues of the efficiency of using a system of machines for growing fiber flax using intensive technology. Attention is focused on the need to form highly mechanized, automated and robotic systems of machines adapted to the specific conditions of farms to minimize labor costs. The purpose of this study is to correct the assessment of the efficiency of the machine system when using new technical means using a universal methodology for calculating the costs of performing technological operations in the cultivation of fiber flax. Methodology and methods: the research used methods of comparative, abstract-logical, functional-cost analysis, and expert evaluation. In the course of the research, we relied on the methodological approaches of leading scientists engaged in the study and development of a system of machines and technologies for complex mechanization of crop production, as well as domestic and foreign scientists who addressed the problems of technical and technological modernization of the flax subcomplex. The leading scientists of the Federal State Budget Research Institution “Federal Research Center of Bast Fiber Crops” (CBFC) in the field of mechanization and agricultural technologies for the cultivation of fiber flax, as well as in the field of improving the efficiency of flax production. Also, the paper presented for the first time a methodology for calculating the costs of performing technological operations in the cultivation of fiber flax, proposed by the doctor of economic sciences, a leading scientist in the field of flax growing B. A. Pozdnyakov Results. As a result of the study, a refined methodological approach to calculating the reduced costs based on element-by-element costs was proposed, based on technological maps of growing crops, taking into account the support coefficient, and reserves for increasing the efficiency of using machine systems in the process of growing fiber flax at individual technological operations were revealed. The scientific novelty lies in the proposed method for calculating the costs of performing technological operations in the cultivation of fiber flax, the dependence of the change in the costs of cultivation with intensive technologies is also presented, alternative measures to optimize costs in agricultural organizations are shown.
