The aim of this work was to evaluate the effect of prolonged administration of the pineal hormone melatonin and short-term fasting on metabolic variables in male and female Wistar:Han rats. Melatonin (MEL, 4 μg/ml of tap water) was administered daily since the 5th week of age. The control group drank tap water. Rats were fed a standard type of diet ad libitum and were kept in the light regimen L:D - 12:12 h. The experiment was terminated after 11 (variant B) or 12 (variant A) weeks of MEL administration. The animals were sacrificed by quick decapitation following overnight fasting (variant A) or 48-h fasting (variant B). Selected organs and tissues were removed and weighed and selected metabolic variables in the serum and tissues were determined. MEL decreased body mass independent of food and water intake in both sexes. In males (variant A) MEL increased the weight of the heart muscle, spleen and adrenals; it decreased the absolute weight of epididymal fat and increased serum corticosterone and phospholipids concentration in comparison with controls. In females, serum glucose decrease and liver triacylglycerols increase were found. After 48-h fasting (variant B) liver, spleen and adrenal weight increase in MELdrinking females was found. In males MEL increased the thymus weight and decreased the epididymal fat weight. In both sexes MEL increased serum corticosterone and liver glycogen concentration; MEL increased serum glucose in males and serum cholesterol concentration in females. Changes in the evaluated variables were also related to fasting duration prior to decapitation. A 48-h fasting at the end of the prolonged MEL intake (variant B vs. A) decreased the absolute liver weight in both sexes and the epididymal/periovarial fat weight, and increased thymus weight in males. In females it decreased the absolute heart muscle weight and increased the spleen weight. In males, 48-h fasting increased serum corticosterone and phospholipids concentration; it decreased the liver triacylglycerols content in females and the liver cholesterol content in males and females. In both sexes 48-h fasting increased glucose concentration in the serum and glycogen concentration in the liver and heart muscle as well as triacylglycerols and cholesterol concentration in the serum, phospholipids concentration in the liver and bone marrow and decreased malondialdehyde concentration in the liver. Forty-eight hour fasting after prolonged MEL administration resulted in a wider range of carbohydrate and lipid metabolism alterations of young rats of both sexes.

AbstractThe association between early glycemic change and short-term mortality in non-diabetic patients with acute intracerebral hemorrhage (ICH) is unclear. We retrospectively investigated non-diabetic patients with lobar (n = 262) and non-lobar ICH (n = 370). Each patient had a random serum glucose test on hospital admission and a fasting serum glucose test within the following 48 h. Hyperglycemia was defined as serum glucose ≥ 7.8 mmol/l. Four patterns were determined: no hyperglycemia (reference category), persistent hyperglycemia, delayed hyperglycemia, and decreasing hyperglycemia. Associations with 30-day mortality were estimated using Cox models adjusted for major features of ICH severity. Persistent hyperglycemia was associated with 30-day mortality in both lobar (HR 3.00; 95% CI 1.28–7.02) and non-lobar ICH (HR 4.95; 95% CI 2.20–11.09). In lobar ICH, 30-day mortality was also associated with delayed (HR 4.10; 95% CI 1.77–9.49) and decreasing hyperglycemia (HR 2.01, 95% CI 1.09–3.70). These findings were confirmed in Cox models using glycemic change (fasting minus random serum glucose) as a continuous variable. Our study shows that, in non-diabetic patients with ICH, early persistent hyperglycemia is an independent predictor of short-term mortality regardless of hematoma location. Moreover, in non-diabetic patients with lobar ICH, both a positive and a negative glycemic change are associated with short-term mortality.

Short-term (2 weeks) effects of the consumption of a high-energy (2920 kJ (700 kcal)) or low-energy (418 kJ (100 kcal)) breakfast on dietary patterns, blood variables and energy expenditure (indirect calorimetry) were compared in ten free-living healthy young men in a crossover study. During the high-energy breakfast, total energy intake was increased, the intake of protein and lipids was unchanged but the intake of carbohydrates was increased. Thus, 48 (SD 4) % OF ENERGY CAME FROM CARBOHYDRATES IN THE HIGH-ENERGY BREAKFAST COMPARED WITH 42 (sd 5) % in the low-energy breakfast. Excluding breakfast, the macronutrient composition of the diet remained identical in the two situations. After the high-energy breakfast, fasting serum triacylglycerol concentration was higher and HDL-cholesterol concentration was lower than after the low-energy breakfast. A high glycaemic response was observed in the morning after the high-energy breakfast period, while there was a peak of free fatty acids after the low-energy breakfast. The high-energy breakfast induced a strong inhibition of fat oxidation throughout the day. Although long-term adaptation to a high-energy breakfast cannot be excluded, the high-energy breakfast in this study did not appear to be favourable to health. Our results do not support the current advice to consume more energy at breakfast.

AbstractChemopreventive effect of non-steroidal antiinflammatory drugs (NSAIDs) in mammary carcinogenesis was reported in several studies. In this study, the effect of a nonselective cyclooxygenase inhibitor diclofenac (DICLO) in the prevention of N-methyl-N-nitrosourea (NMU)-induced mammary carcinogenesis in Sprague-Dawley female rats was evaluated. NMU was administered to animals intraperitoneally in two doses of 50 mg kg−1 b.w. within postnatal days 42-48. In experiment A (short-term administration), DICLO was administrated intramuscularly (5 mg kg−1 b.w.) every other day, starting 3 days before and for subsequent 25 days after first NMU injection. In experiment B (long-term administration), DICLO was administered in tap water (0.01 mg ml−1) continually, starting 7 days before and for subsequent 22 weeks after first NMU dose. The study was terminated 22 weeks after the first dose of NMU in both experiments. After DICLO treatment, tumor frequency per group was reduced in both variants of drug administration: in experiment A by 38% and in experiment B by 39.5%. Moreover, DICLO decreased tumor incidence by 11.5% and delayed tumor latency by 14 days in experiment B. In our preventive-curative experiments DICLO decreased some parameters of NMU-induced rat mammary carcinogenesis, mainly the tumor frequency.

AbstractThe question of effects of long-term melatonin (MEL) administration have not yet been explained sufficiently, especially its metabolic consequences in young persons and animals. The aim of the present study was to analyze the effects of MEL given during prolonged time (for 3 months) and chronically (for 6 months) at the dose of 4 µg/mL of tap water, on the selected metabolic and hormonal parameters in young female and male Wistar:Han (WH) rats. The weights of selected organs, tissues, body weight gains and food and water intake were registered. Six weeks aged rats were adapted to standard housing conditions and light regimen L:D=12:12 h, fed standard laboratory diet and drank tap water (controls) or MEL solution ad libitum; finally they were sacrificed after overnight fasting. Prolonged MEL administration decreased serum glucose concentration and increased triacylglycerol and malondialdehyde concentration/content in the liver in females. In males MEL increased concentrations of serum phospholipids, corticosterone and liver malondialdehyde. MEL treatment reduced the body weight in both sexes and weight of epididymal fat in males, without any alterations of food and water intake. Chronic MEL administration reduced serum glucose concentration and increased concentration/content of glycogen, triacylglycerol and cholesterol in the liver and glycogen concentration/content in heart muscle in males. In females, the significant rise of serum corticosterone concentration and liver malondialdehyde content was recorded. MEL significantly increased liver weight and decreased thymus weight in males. MEL administration increased temporarily water intake in males, body and epididymal fat weights were similar to that in controls. Body weight of MEL drinking females was reduced in the 1st half of experiment only; the food and water intake did not differ from control group. The response in WH rats on MEL was more prominent as in the Sprague-Dawley strain (our previous studies). Male rats were generally more affected, probably due to higher daily and total consumption of melatonin.

Flushing tap water is often promoted as a simple and low cost approach to reducing water lead exposures. This study evaluated lead reduction when prevailing flush guidelines (30 seconds-2 minutes) are implemented in a city compliant with lead-associated water regulations (New Orleans, LA). Water samples (n=1,497) collected from a convenience sample of 376 residential sites (2015-2017) were analyzed for lead in samples collected: at 1) first draw (n=375), and after incremental flushes of 2) 30-45 seconds (n=375), 3) 2.5-3 minutes (n=373), and 4) 5.5-6 minutes (n=218). There was no significant reduction when compared to the first draw lead level, until the 6 minute flush (p<0.05); but most sites (52%) still had detectable lead (≥1 ppb) after 6 minutes. Older homes (pre-1950) and low occupancy sites had significantly higher WLLs (p<0.05).Each sample type had health-based standard exceedances at over 50% of sites sampled (max: 58 ppb). While flushing is an effective short-term approach to remediate high lead, prevailing flush recommendations are an inconsistently effective exposure prevention measure that can often inadvertently increase exposures. Public health messages should be modified to ensure appropriate application of flushing for specific cities, while acknowledging its short-comings and practical limitations.

This experiment were conducted to compare the effect of aqueous leaves extract of Melia azedarach and Anastatica hierochuntic with the antibiotic (Enrofloxaccin) for 49 days, 270 chicks one day old (Ross 308) were randomly distributed into 6 treatments with 3 replicates per treatment and 15 chicks per replicate (45 chicks/treatment). The first treatment were the control, antibiotic (enrofloxaccin) were used in the second treatment at rate of 0.5 ml per liter of tap water, in the 3rd and 4th treatment the aqueous leaves extract of Anastatica hierochuntic were used at rate of 10 and 15 mg/L respectively, and in the 5th and 6th treatment the aqueous leaves extract of Melia azedarach were used at rate of 10 and 15 mg / ml resp. At the end of experiment, blood samples were collected for whole blood tests; Red and White Blood Cells count, biochemical serum tests were also involved ; the concentrate of total cholesterol, glucose, and total protein. The Data showed that using aqueous leaves extract of Anastatica hierochuntic at rate of 10 and 15 mg/L leading to significant increase in serum cholesterol concentration , while , protein serum concentration were significantly decreased in control and in 6th treatment where the aqueous extract of Melia azedarach were used at rate of 15 mg /ml , and there is no significant differences between the treatments in RBC , WBC count , and glucose concentration
