Justification of technical parameters of an ultrasonic device for improvement of meat salting efficiency

The results of a study of the effect of wet ultrasonic lamb meat salting on the muscle tissue microstructure are presented, and the technical parameters of the ultrasonic device are justified. It has been established that significant destruction and swelling of muscle fibers, local destruction of the sarcolemma with its pronounced rugosity are observed at ultrasonic salting with a frequency of 35 kHz, the cross-striation is poorly expressed, the tissue structure is disturbed. An ultrasonic salting with a frequency of 26 kHz was accompanied an increase in the number of transverse microcracks and crevices, loosening of muscle fibers, the formation of cavities between them while retention of the tissue structure, which contributes to the appearance of a brine between muscle fibers and accelerates its penetration into the fiber. It allows us to recommend ultrasound at a frequency of 26 kHz for cavitation activation of the brine to intensify the technological process of lamb meat salting.

Modern methods of differentiating chilled and thawed meat are based on the development of organoleptic indicators and morphological changes in tissues defined in histological preparations. We conducted studies to assess the thermal state of 69 prototypes of meat and fish using a histological method to evaluate the structure of muscle tissue. Microscopic examination of histological preparations of chilled meat and fish showed a clear and even tissue structure, whole muscle fibers in the transverse section of muscle fibers, and indicators such as the structure of muscle tissue, voids within and between muscle fibers, and the presence of thickened muscle fibers were recorded in defrosted samples fibers, while in similar preparations made from chilled products, similar morphological changes were found in isolated cases or were not detected in the material. The above information allows us to consider these criteria as identification and to differentiate the thermal state of meat and fish raw materials.

Choline-deficiency was induced in Holtzman young rats of both sexes by feeding them a high fat - low protein diet.Preliminary studies of the ultrastructural changes in the myocardium of these animals have been recently reported from this laboratory. Myocardial lesions first appeared in the form of intraventricular mural thrombi, loss of cross striation of muscle fibers and focal necrosis of muscle cells associated with interstitial myocarditis. Prolonged choline-deficiency induced cardiomegaly associated with pericardial edema.During the early phase of this nutritional disorder, heart mitochondria - despite of not showing any swelling similar to that usually encountered in liver mitochondria of the same animal - ware the most ubiquitous site of marked structural abnormalities. Early changes in mitochondria appeared as vacuolation, disorganization, disruption and loss of cristae. Degenerating mitochondria were often seen quite enlarged and their matrix was replaced by whorls of myelin figures resembling lysosomal structures especially where muscle fibers were undergoing necrosis. In some areas, mitochondria appeared to be unusually clumped together where some contained membranelined vacuoles and others enclosed dense bodies and granular inclusions.

Purpose of the study. To investigate the efficacy of focal destruction tissues of local liver diseases, chole- and hemostasis of created endoscopic cryoapplicator in operations on the liver and gallbladder.Material and methods. Review the results of surgical treatment of 121 patients with various diseases of the liver (tumors, alveococcosis, parasitic and non-parasitic cysts) and gallbladder (chronic and acute cholecystitis) using a new endoscopic porous TiNi cryoapplicator. 49 patients were operated by an open method, 72 - laparoscopically. Hemo-and cholestatic control of the instrument was performed intraoperative and in postoperative period - by clinical condition of patient and by ultrasonography. Efficiency of destruction of focal liver diseases was investigated by histological examinations of surgical materials, is confirmed by the absence of relapses in the period from 4 to 6 years.The results. Cryoapplicator is simple in use, in sterilization, no energy consumption, can be used both in open surgery, particularly in arduous regions of the liver and laparoscopic procedures and, most importantly, patients with artificial cardiac pacemakers.Cryodestruction of the liver stump after its resection and the bed of the gallbladder after cholecystectomy reduces blood loss by 30-40%, and also helps to reduce the number of relapses of the disease. There were not observed bleeding and bile leakage in postoperative period. Histological examination proved picture of avascular necrosis, a violation of tissue structure of subjected to destruction liver focal diseases, death of epithelial lining of non-parasitic cysts and membranes of echinococcal cysts. There were not diseases recurrences in the period from 4 to 6 years.Conclusion. Endoscopic porous TiNi cryoapplicator is simple and convenient in use, it has a good hemo- and cholestatic effects, pronounced effect to local destruction of liver focal diseases, can be recommended for open and laparoscopic operations on liver.

The specificity of post-mortem redox processes in pork with the development of PSS (Porcine Stress Syndrome), which is the syndrome of pig stress, is considered. Pre-slaughter stress leads to the increased breakdown of glycogen, a slight decrease in the pH of muscle tissue during autolysis and a significant pH shift to the acidic side. To study the physicochemical parameters of muscle tissue after deboning, samples of the longest muscle were taken at the level of 9–12 thoracic vertebrae, 400 g of each half-carcass. Changes in the pH of meat in the experimental groups correlated with the indicators of volatile fatty acids (VFA), which considered the indicator of meat quality. The level of VFA in the group with a dosage of 10 mg/kg of lithium ascorbate was 3.07 % lower than in the control. We also recorded the higher pH values in the first experimental group (by 3 % relative to the control group).When the pH balance shifted to the acidic side, the acidosis occurs, which negatively affected muscle contraction in living organisms. In post-slaughter period the acidosis prevented the contraction of muscle fibers, which increased the organoleptic quality of meat products. The lithium ascorbate prevented the activation of metabolic processes under the action of catecholamines and reduced the level of organic acids in muscle fibers with increasing the stress resistance. The use of lithium ascorbate as a stress protector could reduce these negative effects and increase the organoleptic quality. We suggested to include lithium ascorbate in a standard diet in a dosage of 10 mg/kg of the body weight during the entire feeding period. The presented scheme of lithium ascorbate usage will allow achieving the best organoleptic and physicochemical qualities of pig meat products.

On the basis of histological and micromorphometric studies, general patterns and breed features of the structural organization of the superficial pectoral muscle in meat (Smena-8, Cobb-500) and egg (Hisex white, Andalusian blue) productivity directions are presented. The age of the examined individuals is 29 days. The study evaluated the structural and morphometric parameters of not only muscle fibers, but also the connective tissue component of the muscle under study. It is shown that representatives of meat breeds surpass representatives of egg breeds in terms of muscle fiber thickness and perimisia. At the same time, the indicators of the thickness of the endomysium, bundles of muscle fibers and the area of muscle tissue had similar values in all studied birds. The data obtained indicate the presence of both common morphological traits of muscle tissue, genetically determined and stable regardless of productivity, and variable, which can change under the influence of selection. The research results are important for biology, comparative, species and breed morphology, as well as for veterinary and sanitary examination of chicken meat and in poultry farming to assess the purity of the breed.

Abstract: There are three main components of skeletal muscle: connective tissue, muscle tissue, and membrane system. The connective tissue protects the muscle fibers and separate them into fasicles. The skeletal muscle consists of paralel muscle fibers with their myofibrils which are composed by smaller contractile units, thick myofilaments and thin myofilaments. The membrane system consists of sarcolemma, transverse tubules (TT), foot structure, and sarcoplasmic reticulum (SR) with its cisternae. Depolarization of the sarcolemma spreads to TT, foot structure, and SR, resulting in the release of Ca2+ ions from SR. These ions trigger the formation of cross bridges to begin a contraction.Keywords: sarcolemma, T tubule, sarcoplasmic reticulum, thick myofilament, thin myofilamentAbstrak: Terdapat tiga komponen utama jaringan otot rangka, yaitu: jaringan ikat, jaringan otot seran lintang, dan sistem membran. Sistem membran terdiri atas sarkolema dimana terjadinya depolarisasi yang paling awal dan dihantarkan ke dalam serat otot melalui tubulus T, struktur kaki pada daerah triad, dan sisterna terminalis yang selanjutnya memicu pelepasan ion Ca2+ dari retikulum sarkoplasma.
