Background: Mangrove plants produce polyisoprenoid compounds. Polyisoprenoids have been proven to have anticancer properties. This study investigated the inhibitory activity of polyisoprenoids derived from the leaves of mangrove plants Avicennia alba and Avicennia lanata regarding the expression of PI3K, Akt1, mTOR, P53, and EGFR genes against human colorectal adenocarcinoma WiDr cells. Methods: Anticancer activity was tested through the MTT assay method performed on WiDr cells. The cell cycle and apoptosis were analysed by flow cytometry and double staining. Gene expression of PI3K, Akt1, mTOR, P53, and EGFR was observed using the RT-PCR method. Results: Cytotoxic activity against WiDr cells showed that the IC50 for A. alba and A. lanata was 258.14 ug/mL and 243.32 ug/mL, respectively. This observation indicated the possibility to develop moderate anticancer agents. The cell cycle showed that inhibition of A. alba and A. lanata occurred in the late phase of apoptosis S (10.60 and 10.51%) and G2-M1 (22.05 and 23.84%), which was higher than negative and positive control cells. Furthermore, the polyisoprenoids derived from A. alba and A. lanata leaves exhibited anticancer activity in WiDr cells through the downregulated gene expression of PI3K, Akt1, mTOR, and EGFR as well as the upregulated gene expression of P53. Conclusion: This study demonstrated that polyisoprenoids obtained from A. alba and A. lanata leaves are promising chemopreventive agents for colon cancer.

BACKGROUND: Mangrove plants distributed in the intertidal of the tropical and subtropical region including in North Sumatra, Indonesia. The production of secondary metabolite compounds is well known to mangroves. Characterisation of prominent compounds from mangrove plants such as genus of Avicennia is required to explore for their biological and pharmacological properties of these compounds. AIM: The purpose of this research was to analyse the prominent secondary metabolites through the characterisation of phytochemical, physicochemical, and microscopic of the mangrove genus Avicennia leaves, particularly Avicennia alba, A. lanata, A. marina, and A. officinalis. METHODS: Phytochemical screening was carried out on Avicennia spp leaves to the established process. Physicochemical characters of mangrove leaves were investigated by simplicial powder consisting of moisture content, water-soluble, ethanol-soluble, ash content and ash soluble acid according to the WHO formula. Microscopic analysis on the simplicial powder was carried out based on the WHO procedure. RESULTS: The result showed that physicochemical feature displays diversity among the species and important findings on the water concentration was less than 10% as a prerequisite for the drug. The phytochemical search of simplified grain also depicted divergence among the species, only alkaloid, saponin, and triterpenoid or phytosterol were found entirely in Avicennia spp leaves. Microscopic search found a similar type of stoma in Avicennia spp leaves, namely diacytic. CONCLUSION: The prominent secondary metabolites in Avicennia spp leaves consisting of alkaloid and saponin in simplicial and triterpenoid/sterol was either in simplicial or hexane extract. The present study may provide significant pharmacological properties from mangrove Avicennia genus green foliages, which could accelerate another prospect for non-wood mangrove utilisation. nus green foliages, which could accelerate another prospect for non-wood mangrove utilization.

ABSTRACTThe coastal area of Wonorejo is the mangrove area rehabilitated to become an ecotourism area. The research aims to analyze the patterns of formation of mangrove vegetation zoning that have been rehabilitated. The observation locations are at 3 stations, i.e. station 1 is located at the upper limit of supratidal zone, station 2 is in the middle of intertidal zone, and station 3 is in the subtidal zone. The structure of the mangrove vegetation were analyzed based on the species density (K), dominance (D), and important value index (IVI); while the distribution of mangrove vegetation based on environmental characteristics was analyzed using Principal Component Analysis (PCA). The results showed that the dominant species at Station 1 was Nypa fruticans (84.2%), whereas in Station 2 the dominant species was Excoecaria agallocha (40.9%), and at Station 3 was Avicennia alba (83.4%) The level of damage to mangrove vegetation in Wonorejo is categorized as medium, with tree densities between ≥ 1000 - <1500 per hectare. Diversity index at all stations is also relatively low because it is worth less than 1.5. This ecosystem begins to show the existence of an ecosystem succession, as evidenced by the start of other species of mangroves that are not planted intentionally. The results of PCA analysis showed a positive correlation between the parameters of vegetation types with high tide baths, salinity, and pH, which contributed to forming a positive F2 axis. This means that the three parameters are the main factors that determine whether the ecosystem is suitable for the growth of certain species of mangroves.

The biomass of three dominant mangrove species (Sonneratia apetala, Avicennia alba and Excoecaria agallocha) in the Indian Sundarbans, the designated World Heritage Site was evaluated to understand whether the biomass vary with spatial locations (western region vs. central region) and with seasons (pre-monsoon, monsoon and post-monsoon). The reasons for selecting these two regions and seasons are the contrasting variation in salinity. Among the three studied species, Sonneratia apetala showed the maximum biomass followed by Avicennia alba and Excoecaria agallocha. We also observed that the biomass varied significantly with spatial locations (p<0.05), but not with seasons. The variation may be attributed to different environmental conditions to which these forest patches are exposed to.

Mangrove species, viz., Avicennia officinalis, Avicennia marina var. acutissima, Avicennia marina (dwarf), Rhizophora mucronata, Sonneratia alba, Aegiceras  corniculatum, Kandelia  candel were chosen for measurement of height of the tree and girth or circumference. The sampling was random and at least 50 records were made. The girth is measured by the tape. The measurement of the height is made with the help of abny level. The Tables 1 to 8 records the values for girth, height and canopy cover as well as for correlation coefficient (r). There correlation between girth and canopy in all the species studied however in case of Avicennia officinalis and Aegiceras corniculatum girth and height show more co-relation than girth and C. cover. The positive co-relation observed between girth and canopy is more or less 0.7 except Avicennia marina (dwarf) Excoecaria agallocha and Aegiceras corniculatum. The co-relation is observed in girth and height is difficult to explain. This case is observed in Avicennia officinalis and Aegiceras corniculatum.
