High in vitro Antioxidant Capacities of Algerian Cleome arabica Leaves’ Extracts

The aim of this study was to evaluate the antioxidant capacities of Cleome arabica leaves’ extract collected on two seasons in the town of Laghouat steppe region of Algeria. Five common tests for measuring antioxidant capacities were used to compare with three standard antioxidants: measurement of free radical scavenging activity with 1,1-diphenyl-2-picrylhydrazyl radical (DPPH·) and 2,2ʹ- azinobis(3-ethylbenzo-thiazo-line-6-sulfonate) (ABTS·+) radical cation, measurement of total antioxidant capacities with phosphomolybdenum, ferric reducing, and cupric reducing methods. The amount of the phenolic compounds was carried out by the quantification of total phenolic, total flavonoid, and condensed tannin contents in three organic solvents with varying polarities. The results show that the ethyl acetate is the best extractor solvent of flavonoids, while petroleum ether has the ability to extract more of terpenes. The values of quantification ranged from 0.341 to 0.751 mg of gallic acid equivalent/g of dry matter, from 0.172 to 0.682 mg of quercetin equivalent/g of dry matter and from 0.172 to 0.332 mg of catechin equivalent/g of dry matter for the total phenolic, flavonoids, and the condensed tannins, respectively. All the extract shows strong antioxidant activity, whose best are found in the ABTS and DPPH assay with IC50 values of 0.01 and 0.017 mg/ml, respectively, in a different season. These results suggest that the level of antioxidant activity in this plant varies to a great extent. They also suggest that phenolics in this plant provide substantial antioxidant activity. Upon achievement of this survey, an extra benefit of this medicinal plant may be found.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

Antidesma bunius (L.) spreng (Mamao) is widely distributed in Northeastern Thailand. Antidesma bunius has been reported to contain anthocyanins, which possess antioxidant and antihypertensive actions. However, the antidiabetic and antiglycation activity of Antidesma bunius fruit extract has not yet been reported. In this study, we investigated the inhibitory activity of anthocyanin-enriched fraction of Antidesma bunius fruit extract (ABE) against pancreatic α-amylase, intestinal α-glucosidase (maltase and sucrase), protein glycation, as well as antioxidant activity. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) chromatogram revealed that ABE contained phytochemical compounds such as cyanidin-3-glucoside, delphinidin-3-glucoside, ellagic acid, and myricetin-3-galactoside. ABE inhibited intestinal maltase and sucrase activity with the IC50 values of 0.76 ± 0.02 mg/mL and 1.33 ± 0.03 mg/mL, respectively. Furthermore, ABE (0.25 mg/mL) reduced the formation of fluorescent AGEs and the level of Nε-carboxymethyllysine (Nε-CML) in fructose and glucose-induced protein glycation during four weeks of incubation. During the glycation process, the protein carbonyl and β-amyloid cross structure were decreased by ABE (0.25 mg/mL). In addition, ABE exhibited antioxidant activity through DPPH radical scavenging activity and Trolox equivalent antioxidant capacity (TEAC) with the IC50 values 15.84 ± 0.06 µg/mL and 166.1 ± 2.40 µg/mL, respectively. Meanwhile, ferric reducing antioxidant power (FRAP) showed an EC50 value of 182.22 ± 0.64 µg/mL. The findings suggest that ABE may be a promising agent for inhibiting carbohydrate digestive enzyme activity, reducing monosaccharide-induced protein glycation, and antioxidant activity.

Background: Research on medicinal plant antioxidants has emerged as a potential therapeutic to prevent free radical generated damage in the human body. Hammada elegans Botsch (popularly known as “Ajram”) is a xerophytic plant widely found in Laghouat region, but there are only a few reports about the biological or chemical properties of these species. Hence, the aim of this study is to investigate the antioxidant and the antihemolytic activities of hexanic, acetonic, methanolic and aqueous extracts of aerial parts of Algerian Hammada elegans Botsch by employing different in vitro assay systems. Methods: The total phenolic content, the flavonoid content and the condensed tannin amount were analyzed using Folin-Ciocalteu, aluminum chloride and vanillin assays, respectively. The in vitro antioxidant capacity of extracts was assessed by CUPRAC, iron chelating, ABTS•+and antihemolytic assays, and was expressed as EC50 values. Results: Among the analyzed extracts, the aqueous extract had the highest phenolic, flavonoid and tannin contents. Also, this extract displayed the highest antioxidant capacities compared to the other extracts and standards. Its EC50 value for ABTS radical-scavenging activity was 0.265 ± 0.003 mg/L. Moreover, this extract showed high iron (II) chelating ability (EC50 = 0.958 ± 0.001 mg/L), and good antioxidant activity in the cupric ion reducing activity (CUPRAC) in a concentration dependent manner (EC50 were 0.709 ± 0.002 mg/L). Additionally, this extract had the best antihemolytic activity against AAPH-induced hemolysis (EC50=0.090 ± 0.004 mg/L). Conclusion: Our study revealed that the aqueous extract of Hammada elegans Botsch, is a potential source of antioxidants which possess a high protective effect of membrane against free radical.

AbstractWe conducted this study to determine and compare the content of phenolic compounds and flavonoids in the argan and olive leaves as well as their antioxidant capacity in aqueous, methanolic, and ethyl acetate extracted fractions. In vitro antioxidant activity was evaluated in comparison with synthetic antioxidants by assessing DPPH• radical scavenging capacity, ferric reducing antioxidant power, scavenging ability by inhibiting the β-carotene/linoleic acid emulsion oxidation, and by the ABTS radical scavenging activity assay. Total phenolic content in argan samples ranged from 221.69 ± 2.07 to 1.32 ± 0.01 mg GAE/g DW and in olive samples from 144.61 ± 0.82 to 1.21 ± 0.02 mg GAE/g DW. Total flavonoids content in argan samples varied from 267.37 ± 1.12 to 25.48 ± 0.02 mg QE/g DW, while in olives from 96.06 ± 0.78 to 10.63 ± 0.05 mg QE/g DW. In vitro antioxidant studies strongly confirmed the antioxidant potency of argan and olive leaves and their richness in secondary metabolites that are effective in free radicals scavenging and metal chelating capacities, indicating their antioxidant power.

This study evaluated the potential antioxidant and antidiabetic properties in vitro of four millet grain varieties cultivated in South Korea. The free fractions were tested for their total antioxidant capacity using 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS+) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assays, followed by α-glucosidase, α-amylase, and advanced glycation endproducts (AGEs) formation inhibition assays. The total phenolics, flavonoids, and condensed tannins in the free fractions ranged from 107.8 to 136.4 mg ferulic acid equivalent (FAE)/100 g, 101.3 to 115.8 mg catechin equivalent (CE)/100 g, and 17.65 to 59.54 mg catechin equivalent (CE)/100 g, respectively. Finger Italian millet had the highest total phenolic content (136.4 mg FAE/100 g) and flavonoid content (115.8 mg CE/100 g). Barnyard and finger Italian millet showed the highest DPPH (IC50 = 359.6 µg/mL and 436.25 µg/mL, respectively) and ABTS radical scavenging activity (IC50 = 362.40 µg/mL and 381.65 µg/mL, respectively). Similarly, finger Italian millet also exhibited significantly lower IC50 values for the percentage inhibition of α-glucosidase (18.07 µg/mL) and α-amylase (10.56 µg/mL) as compared with acarbose (IC50 = 59.34 µg/mL and 27.73 µg/mL, respectively) and AGEs formation (33.68 µg/mL) as compared with aminoguanidine (AG) (52.30 µg/mL). All eight phenolic compounds identified in finger Italian millet were flavonoids, with flavanols being the predominant subclass. Taken together, millet flavonoids play important roles in the prevention and management of type 2 diabetes, and hence finger Italian millet has the potential to be developed as a functional food.

Abstract Objectives Inhibition of carbohydrate absorption from intestine is an efficient way to prevent type 2 diabetes. Some edible plants have shown the potential as anti-diabetic agents by inhibiting carbohydrate-digestive enzymes in intestine. In terms of edible plant's health effect, bioactive compounds’ stability during the process of digestive enzymes has been commonly questioned. Therefore, the objectives of this study were to 1) examine inhibitory effect of garlic scape (GS), the green flower head of garlic on carbohydrate-degrading enzymes and 2) investigate antioxidant activity and total phenolic content (TPC) of GS treated with intestinal digestive enzymes. Methods GS extract (GSE) was prepared with 80% methanol (v/v) by homogenization, sonication, and filtration, and evaporation. α-glucosidase, maltase, glucoamylase, sucrase, α-amylase inhibitory activities were determined by measuring glucose hydrolyzed from nitrophenyl-glucopyranoside, maltose, starch, sucrose, and starch, respectively. To prepare gastrointestinal enzyme-digested GSE, GSE was digested with pepsin and a pancreatin-bile solution. GSE and gastrointestinal enzyme-digested GSE were assessed for TPC using a Folin-Ciocalteu reagent and for total antioxidant and radical scavenging activities by the following methods, phosphomolybdeum, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods, respectively. Results GSE (20 mg/ml) showed 30%, 50%, and 42% inhibition on α-glucosidase, maltase, and glucoamylase enzyme activities, respectively. GSE (50 mg/ml) showed 55.1% DPPH radical and 5782 mg vitamin C equivalent (VCE)/g ABTS radical scavenging activities. Total antioxidant activity of the GSE (1.5 mg/ml) exhibited 2.76 mg VCE/g. Consistent with its antioxidant property, the GSE showed a dose-dependent increase in TPC. Gastrointestinal enzymatic treatment resulted in approximately 46% reduction in DPPH and ABTS radical scavenging activity, total antioxidant activity, and TPC of the GSE. Conclusions Garlic scape has the potential as a natural anti-diabetic agent. Garlic scape keeps beneficial effects after the process of intestinal digestive system. Antioxidant potential of Rosa L. leaves methanolic extracts was evaluated in vitro using a spectrophotometric method based on measuring the radical scavenging effect on 2,2-diphenyl- 1-picrylhydrazyl (DPPH) radicals. The contents of ellagic acid, quercetin and kaempferol in the extracts from leaves of seventeen rose species were determined using SPE-RPHPLC methods. Additionally, total phenolic content was determined spectrophotometrically according to the Folin-Ciocalteu procedure and calculated as gallic acid equivalents (GAE). Remarkable high antioxidant activity and high total phenolic content (5.7% < GAE < 15.2%), large ellagic acid (EA) content from 9.37 to 19.42 mg/g of dry weight, a quercetin content ranging from 3.68 to 15.81 mg/g of dry weight and kaempferol content from 1.25 to 9.41mg/g of dry weight were found in rose leaves. Significant correlation between EA (r2 = 0.6131), quercetin (r2= 0.5158), total phenolic content (r2= 0.8485) and antioxidant activity was observed. Basing on the studies conducted one may assume that the extracts of rose leaves are a rich source of natural antioxidants and could be used to prevent free-radical-induced deleterious effects.

This study was designed to examine the in vitro antioxidant DPPH free radical-scavenging activity, and the total phenolic and flavonoid content of ethanol and methanol extracts of Salvia amplexicaulis Lam. in the whole plant and different parts, leaves, stems and flowers. The largest amounts of extract yield were obtained from the flowers, 14.14% and 12.00 % (w/w) in the ethanol of methanol extracts, respectively. The ethanol extract of leaves (16.07 ?g/ml) and methanol extract of the whole plant (21.28 ?g/ml) showed the highest activity against the DPPH radical. The ethanol extract of the leaves was the richest in phenols (222.40 mg GAE/g) and flavonoids (49.81 mg QE/g), whereas the methanol extract of the whole plant contained the highest amount of phenolics (180.89 mg GAE/g) and flavonoids (38.15 mg QE/g). A very strong linear correlation between antioxidant activity and the phenolic content of the extracts was established. The obtained results suggest that S. amplexicaulis could be regarded as an important source of natural antioxidants.

Echinocystis lobata (Mich.) Torr. et Gray extracts were investigated for their bioactive compounds and their potential for natural antioxidant properties. In vitro antioxidant activity of methanol, ethanol, and water extracts were evaluated by 1,1-diphenyl-picrylhydrazyl (DPPH) radical scavenging activity. Total phenolic content (TPC) and total flavonoid content (TFC) of fruits and seeds were determined. TPC is a fairly constant value and varies weakly both among the different organs of Echinocystis lobata and among the geographical locations of the populations studied. Like other invasive species, Echinocystis lobata is superior to many other plants in terms of the total amount of flavonoids contained in its fruits and seeds. The dried fruits contain more flavonoids than the seeds. Seeds from Slovak populations have higher flavonoid content than those from Russian populations. The results demonstrated that Echinocystis lobata can be a good source of natural antioxidants and have a great potential for commercialization, especially in the pharmaceutical industry.

Germander (Teucrium, Lamiaceae) is a genus commonly used as a medicinal herb in the middle east. The aim of this study was to estimate total polyphenols and antioxidant activity of different extracts from two Syrian Teucrium species (Teucrium polium and Teucrium creticum). Total polyphenols content was measured using Folin-Ciocalteu assay and the antioxidant activity was evaluated in vitro by DPPH assay (free radical scavenging activity against 1, 1 –diphenyl-2-picrylhydrazyl). The total polyphenols content and antioxidant activity of T. creticum extracts were higher than T. polium ones. Phenols contents ranged from 16.388 to 105.672 mg GA E/ g dry weight extract. The extracts showed significant scavenging activity against DPPH, with IC50 values ranging between 0.195 and 0.710 mg/ml. The methanolic extracts show higher antioxidant activity from the two species. A good correlation between DPPH scavenging activities and contents of polyphenols was found. These results show that Syrian Teucrium species, especially T. creticum least Known among all, is a rich source of phenols and natural antioxidant compounds, that can be used as a natural food preservative.
