Strong antihemolytic and antioxidant properties of aqueous extract from Algerian Hammada elegans (Bge.) Botsch (Chenopodiaceae)

Background: Research on medicinal plant antioxidants has emerged as a potential therapeutic to prevent free radical generated damage in the human body. Hammada elegans Botsch (popularly known as “Ajram”) is a xerophytic plant widely found in Laghouat region, but there are only a few reports about the biological or chemical properties of these species. Hence, the aim of this study is to investigate the antioxidant and the antihemolytic activities of hexanic, acetonic, methanolic and aqueous extracts of aerial parts of Algerian Hammada elegans Botsch by employing different in vitro assay systems. Methods: The total phenolic content, the flavonoid content and the condensed tannin amount were analyzed using Folin-Ciocalteu, aluminum chloride and vanillin assays, respectively. The in vitro antioxidant capacity of extracts was assessed by CUPRAC, iron chelating, ABTS•+and antihemolytic assays, and was expressed as EC50 values. Results: Among the analyzed extracts, the aqueous extract had the highest phenolic, flavonoid and tannin contents. Also, this extract displayed the highest antioxidant capacities compared to the other extracts and standards. Its EC50 value for ABTS radical-scavenging activity was 0.265 ± 0.003 mg/L. Moreover, this extract showed high iron (II) chelating ability (EC50 = 0.958 ± 0.001 mg/L), and good antioxidant activity in the cupric ion reducing activity (CUPRAC) in a concentration dependent manner (EC50 were 0.709 ± 0.002 mg/L). Additionally, this extract had the best antihemolytic activity against AAPH-induced hemolysis (EC50=0.090 ± 0.004 mg/L). Conclusion: Our study revealed that the aqueous extract of Hammada elegans Botsch, is a potential source of antioxidants which possess a high protective effect of membrane against free radical.

Aim: This study investigated the phytochemical constituents and in vitro antioxidant properties of methanol and aqueous leaf extracts of Geophila obvallata using standard methods. Materials and Methods: The in vitro antioxidant assays carried out were 1, 1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging ability, Nitric oxide (NO•) radical scavenging activity assay, 2, 2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS•+) radical cation scavenging assay, ferric reducing properties and hydroxyl radical scavenging assays. Results: Phytochemical analysis revealed the presence of alkaloids, flavonoids, phenolic compounds, steroids, saponins, terpernoids and cardiac glycosides in both extracts. Relative to the aqueous extract, the methanol extract contained a higher amount of the secondary metabolites. However, both extracts exhibited appreciable and dose-dependent capacities for quenching DPPH, ABTS•+ and NO• free radicals, and potent ferric reducing ability to levels comparable to those of ascorbic acid. The crude methanol extract showed significantly increased (P<0.05) antioxidant activity than the aqueous extract. Conclusion: It was concluded that the extract possesses strong antioxidant properties due to its content of phytochemicals, and provides scientific basis for its ethno medicinal applications.

Introduction: Diabetes mellitus (DM) has been recognized as the seventh leading cause of global mortality; however, researchers seek alternative means to manage the menace. The current study sought to investigate antioxidant potentials, α-amylase, and α-glucosidase inhibitory activities of ethanolic extract of Moringa oleifera flower in vitro. Methods: Antioxidant properties of the extract were appraised by assessing its inhibition against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl (OH•), and hydrogen peroxide (H2O2) free radicals, as well as ferric reducing antioxidant power (FRAP), the antidiabetic activity was evaluated by α-amylase and α-glucosidase inhibition.Results: In this study, ethanolic extract of M. oleifera flower demonstrated a significant (P < 0.05) inhibition against DPPH free radical (43.57–83.56%) in a concentration-dependent manner, while FRAP (101.76 ± 1.63 mg/100 g), OH• scavenging ability (71.62 ± 0.95 mg/100 g), and H2O2 free radical scavenging capacity (15.33 ± 1.20 mg/100 g) were also observed. In the same manner, ethanolic extract of M. oleifera flower revealed a significant (P < 0.05) inhibition against α-amylase (IC50= 37.63 mg/mL) and α-glucosidase activities (IC50= 38.30 mg/mL) in the presence of their respective substrates in a concentration-dependent manner in comparison with acarbose. Conclusion: Ethanoic extract of M. oleifera flower could be useful as an alternative phytotherapy in the management of DM, having shown a strong antioxidative capacity and substantial inhibition against the activities of key enzymes involved in carbohydrate hydrolysis in vitro.

The flowers of Bombax ceiba were investigated for their chemical composition, antioxidant effects and antiproliferative activity against seven human cancer cell lines. The antiproliferative responses of diethyl ether (DE) and light petroleum (PE) extracts were evaluated by sulforhodamine B (SRB) assay against MCF-7, HeLa, COR-L23, C32, A375, ACHN, and LNCaP cells in comparison with a human normal cell line, 142BR. Moreover, extracts were characterized by GC-MS analysis and tested for their antioxidant properties by different in vitro systems, namely DPPH, Fe-chelating activity and β-carotene bleaching test. Both PE and DE extracts showed the highest antiproliferative activity against human renal adenocarcinoma (ACHN) in a concentration-dependent manner. PE extract showed the highest radical scavenging activity against the DPPH radical, while DE extract was more active in the β-carotene bleaching test. The presence of β-sitosterol and some fatty acids may contribute to the bioactivity of B. ceiba flower extracts.

In order to facilitate the quality control of Trifolium repens flowers and leaves, a RP-HPLC method with UV detection was developed for the simultaneous quantitative determination of flavonols and isoflavones. The total flavonoid and phenolic (TPC) contents were determined spectrophotometrically in a visible part of the light spectrum at 425 and 760 nm, respectively. Additionally, evaluation of the antioxidant properties of the plant materials was performed using the DPPH in vitro test. The results showed that the flowers are the richest source of phenolics ranging from 28.7 to 38.8 mg GAE/g, and flavonoids, calculated for hyperoside, up to 20 mg HP/g, which hydrolyzed mainly to flavonols (the quercetin level greater than 6 mg/g). T. repens is poor in isoflavones; similar quantities of ca. 0.2 mg/g were detected in the flowers and leaves. The flower and leaf extracts showed antioxidant activity towards DPPH with EC50 values ranging from 72.3 to 179.3 μg/mL. Significant linear correlations were found between antioxidant potentials of the studied plant materials and total phenolic and flavonoid contents determined by HPLC and spectrophotometric methods ( R2 in the range of 0.97 - 0.99).

In this study, the effects of different fruit parts and extraction conditions on the antioxidant properties of jujube (Ziziphus jujuba Mill.) fruit were investigated. Five in vitro antioxidant models and statistical analyses were performed. The results revealed that jujube peel with pulp (peel pulp) exhibited better antioxidant capacity than did seeds. Overall, jujube peel pulp extracted using 50% ethanol at 60 °C exhibited the best antioxidant capacity in terms of 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (0.3 ± 0 mg/mL), 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity (0.5 ± 0 mg/mL), total phenolic content (38.3 ± 0.4 mg gallic acid equivalent per gram dry weight), total flavonoid content (43.8 ± 0.2 mg quercetin equivalent per gram dry weight), and reducing power (41.9 ± 2.2 mg ascorbic acid equivalent per gram dry weight). The results indicated that jujube peel pulp is a more potential natural antioxidant than seeds.

The aim of this study was to evaluate the antioxidant capacities of Cleome arabica leaves’ extract collected on two seasons in the town of Laghouat steppe region of Algeria. Five common tests for measuring antioxidant capacities were used to compare with three standard antioxidants: measurement of free radical scavenging activity with 1,1-diphenyl-2-picrylhydrazyl radical (DPPH·) and 2,2ʹ- azinobis(3-ethylbenzo-thiazo-line-6-sulfonate) (ABTS·+) radical cation, measurement of total antioxidant capacities with phosphomolybdenum, ferric reducing, and cupric reducing methods. The amount of the phenolic compounds was carried out by the quantification of total phenolic, total flavonoid, and condensed tannin contents in three organic solvents with varying polarities. The results show that the ethyl acetate is the best extractor solvent of flavonoids, while petroleum ether has the ability to extract more of terpenes. The values of quantification ranged from 0.341 to 0.751 mg of gallic acid equivalent/g of dry matter, from 0.172 to 0.682 mg of quercetin equivalent/g of dry matter and from 0.172 to 0.332 mg of catechin equivalent/g of dry matter for the total phenolic, flavonoids, and the condensed tannins, respectively. All the extract shows strong antioxidant activity, whose best are found in the ABTS and DPPH assay with IC50 values of 0.01 and 0.017 mg/ml, respectively, in a different season. These results suggest that the level of antioxidant activity in this plant varies to a great extent. They also suggest that phenolics in this plant provide substantial antioxidant activity. Upon achievement of this survey, an extra benefit of this medicinal plant may be found.

The present study was designed to evaluate the antioxidant, antimicrobial, antidiarrheal and analgesic activities of methanol extract of Diospyros malabarica (Desr.) Kostel buds growing in Bangladesh. The total phenolic content of methanol extract of D. malabarica and its Kupchan fractions was determined and expressed in gallic acid equivalent (GAE). In the DPPH free radical scavenging assay, the aqueous soluble fraction of the methanol extract of D. malabarica revealed maximum free radical scavenging activity having IC50 value of 6.17 μg/ml. In antimicrobial assay, all the test samples displayed prominent antimicrobial activity against the test organisms under in vitro conditions. Among these, the carbon tetrachloride soluble fraction was found to exhibit the highest activity against Bacillus cereus, Salmonella Typhi, Salmonella Paratyphi, and Candida albicans with the zone of inhibition as 32, 30, 28 and 30 mm, respectively. In evaluation of antidiarrheal activity, the D. malabarica extracts showed significant anti-diarrheal potential in a dose dependent manner. During the evaluation of analgesic activity by radiant heat tail-flick method, the plant extract at 400 mg/kg b.w. exhibited highest elongation (373.04%) as compared to morphine (472.48%). On the other hand, in acetic acid-induced writhing test, the extract at 200- and 400-mg/kg b.w. showed 61.11 and 66.67% inhibition of writhing in mice model, respectively as compared to 68.06% inhibition produced by the standard diclofenac-Na. The findings of this study justify some of the traditional uses of D. malabarica and reveal the bioactivity of the plants. In this study, the antioxidant activities and biochemical characteristics of Ulva rigida (Ulvophyceae) and Arthrospira platensis (Cyanophyceae) were determined. The extracts from two seaweed species were evaluated for their free radical scavenging activity, using the 1,1-diphenyl-2-picrylhydrazyl hydrate (DPPH) method, their total phenolic, flavonoid, and condensed tannin contents, through Folin–Ciocalteu, Quettier-Deleu, and Price methods, respectively. The extracts of two seaweeds showed a low free radical scavenging capacity in comparison with commercial antioxidant BHT (butylated hydroxytoluene), and vitamin C. The extract of U. rigida demonstrated greater antioxidant potential with a low IC50 (3.76±0.02 mg/g Ext.). The total phenolic contents were ranged from 2.21±0.08 (U. rigida) to 8.59±0.62 (A. platensis) mg GAE/g of extract. The highest flavonoid content was found in A. platensis as 22.70±0.65 mg rutin/g of extract. The contents of condensed tannin were measured 3.01±0.11 mg CE/g of extract for A. platensis and 3.76±0.06 mg CE/g of extract for U. rigida. According to results obtained, U. rigida and A. maxima possess antioxidant activity and could be used in for future applications in medicine, functional foods, and agriculture.

Purpose: The investigation of Equisetum ramosissimum Desf. (Equisetaceae) extracts for total phenolic content, potential antioxidant properties, and antibacterial activity against Propioni bacterium acne and antityrosinase effects.Methods: The aerial parts of the E. ramosissimum from Jordan were extracted by maceration and Soxhlet methods, using solvents of different polarities. The composition of the extracts were qualitatively screened using standard phytochemical tests. Quantitatively, total phenolic content (mg/mL, equivalent to gallic acid), ABTS free radical scavenging activity (IC50), anti-P. acne (MIC and MBC), and tyrosinase inhibitory effects (IC50) were also determined.Results: The aqueous-methanol Soxhlet extract contained the highest total phenolic content (0.675 mg/mL gallic acid equivalents). Besides, phytochemical screening tests revealed the presence of phenols, flavonoids, tannins, alkaloids and saponins in the aqueous methanol Soxhlet extract, contributing to the antioxidant (IC50 = 0.125) and antityrosinase (IC50 = 1.125) effects. This extract also showed potent antimicrobial effects against P. acne (MIC = 6.250 mg/mL; MBC = 12.500 mg/mL). Other extracts, including ethanol, water and ethyl acetate, showed lower total phenolic content with moderate and weak biological activity.Conclusion: E. ramosissimum is a promising plant species to be considered for antioxidant, antiacne, and antityrosinase effects. However, further testing (including invivo, histological examination, and highperformance liquid chromatography (HPLC) analysis) is necessary to understand more about its mechanisms of action. We have investigated effect ofMoringa oleiferaleaf and fruit extracts on markers of oxidative stress, its toxicity evaluation, and correlation with antioxidant properties usingin vitroandin vitroassays. The aqueous extract of leaf was able to increase the GSH and reduce MDA level in a concentration-dependent manner. The ethanolic extract of fruit showed highest phenolic content, strong reducing power and free radical scavenging capacity. The antioxidant capacity of ethanolic extract of both fruit and leaf was higher in thein vitroassay compared to aqueous extract which showed higher potentialin vivo. Safety evaluation studies showed no toxicity of the extracts up to a dose of 100 mg/kg body weight. Our results support the potent antioxidant activity of aqueous and ethanolic extract ofMoringa oleiferawhich adds one more positive attribute to its known pharmacological importance.
