The work reflects the aspects of introducing two varieties of apple trees into the culture in vitro: Prikubanskoe and Kubanskoe Bagryanoe. The aims of study included testing a new disinfection protocol that accurately regulates the content of active chlorine in the disinfecting solution, as well as studying the regenerative ability of explants of apple trees of the Prikubanskoe and Kubanskoe Bagryanoe varieties on different culture medium: Murashige and Skoog (MS) ¼ NH4 NO3 , and Kvorin-Lepuavra (QL). The disinfection protocol used made it possible to obtain an insignificant level of explant contamination with fungal infection in passage zero, which indicates the effectiveness of the tested extraction protocol. The contamination of explants in passage zero by fungal infection was insignificant, which indicates the high efficiency of the sterilizing agent. The level of bacterial infection is higher in the Kubanskoe Bagryanoe variety (21.3 % of the total amount of explants). The influence of the component composition of nutrient media on the primary growth of apple micro-shoots was revealed. The following results were obtained: the yield of viable explants of the Prikubanskoe variety grown on nutrient medium MS ¼ NH4 NO3 was 44,4 %, which is 16,4 % higher than explants grown on QL medium (28 %). Explants of the Kuba nskoe bagryanoe variety, in turn, have a yield of vital microplants grown on MS ¼ NH4 NO3 16.7 %, which is 30.4 % lower than explants grown on QL medium (47.1 %). The combined effect of the genotype and the composition of the nutrient medium on the development of explants was statistically significant for the studied parameters. The share of the cumulative effect of factors of the variety x medium is 74.93 %, the influence of the genotype is 20.72 %. The data obtained indicate the variety specificity of the nutrient medium during clonal micropropagation of these apple varieties.

Quantitative and qualitative composition of essential oils of peppermint breeds Lebedinaya Pesnya, Lubenchanka, Lidiya, Ukrainskaya Perechnaya, Mama, Chornolista was investigated before and after clonal microreproduction by the method of isolated tissues and bodies culture in vitro. Methods of essential oil steam distillation, capillary gas chromatography and statistical analysis were used in the research. It is established that increase in essential oil quantity was observed for peppermint breeds on which reproduction and improvement іn vіtro technology was applied. As a result of clonal microreproduction of peppermint plants in culture іn vіtro on nutrient medium Murasige and Skug, in which the growth regulators 0.75 mg/l of 6-benzylaminopurine, 0.1 mg/l of adenine, 0.05 mg/l of indolil-3-acetic acid and 0.5 mg/l of gibberellins acid were added and virocide Ribavirin in concentration 10 mg/l, improvement was obtained in comparison with vegetatively reproduced plants; increase in essential oil quantity per hectare was established for the following breeds; Chornolista by 54.2%, Lebedinaya pesnya by 38.2%, Ukrainskaya Perechnaya by 36.7%, Mama by 28.5%, Lubenchanka by 17.1% and Lidiya by 11.6%. For oil content the highest indices were noted for Lubenchanka, Mama and Lebedinaya Pesnya peppermint breeds with product yield 4.02%, 3.98% and 3.84% respectively. It was established that the essential oil component composition in non-clonal peppermint plants raw materials and plants-regenerants after culture in vitro is variable depending on breed. Limonene, cineole, menthone, menthofuran, iso-menthone, menthyl acetate, β-caryophyllene, iso-menthol, menthol, pulegone, germacren, piperitone, carvone were identified in peppermint essential oil. High content of menthol, low content of carvone, piperitone, pulegone (except for Chornolista, Ukrainskaya Perechnaya breeds) and menthofuran (except for Chornolista, Ukrainskaya Perechnaya and Lubenchanka breeds) are characteristic for Ukrainian selection peppermint investigated breeds. A clear tendency to menthol and menthone content ratio increase is observed in plants which were improved in conditions іn vіtro. Pulegone was not detected in essential oil samples of Lebedinaya Pesnya, Lidiya and Mama breeds. Biochemical markers of Lebedinaya Pesnya, Lubenchanka, Mama breeds, which differentiate them within the group of investigated breeds, are higher limonene, piperitone and menthol pool; for Ukrainskaya Perechnaya and Chornolista breeds – pulegone, cineole and menthone; for Lidiya breed – iso-menthone.

The microclonal (in vitro) propagation of the original potato plants is important stage in the cultivation of high-quality seed potato. The appropriate microclimate parameters (illumination, temperature, air humidity) are to be maintained in phytotrons for optimal clones’ growth. It is necessary to consider the specific response of potato varieties and, according to it, carefully select the lighting spectral composition. The purpose of the work was to study the effect of various light sources on the morphological parameters of 22 varieties of potato microplants. 10 clones of every variety were selected for the experiment after the initial multi-varietal analysis. After 27 days of cultivation, growth parameters were measured: root length, plant height and the number of internodes. The light sources were light-emitting diode (LED) irradiators ECOLED-BIO-195W-D120 FMatrix IP66, ECOLED-BIO-112-185W-D120 UniversalLED, and 3-ECOLED-60-LX Fito IP65 (“SET” ltd). For the first time, data on the lighting effect on a large sample of potato varieties were obtained, demonstrating significant differences in the physiological response of plants to lighting conditions. According to the research results, it was shown that ECOLED-60-LX Fito IP65 was the optimal source of photosynthetically active radiation for plants among the studied LED-lamps, providing a positive effect on the growth parameters of potato plants, including plants height, length of roots and internodes amount.

Aim. The purpose of our research was to establish the possibility of initiation of potato and tomato culture plants using industrial growth regulators, which are legal for use in Ukraine, which include substances with pronounced auxin-cytokinin activity. Methods. In this work, we used varieties of tomato: Khoriv, Borivsky and Bozhedar, Povin potatos. Work in culture in vitro was performed according to conventional methods. Results. A phytohormone substitute was created in a known nutrient agar medium according to Murasige-Skuga. For the in vitro cultivation of tomato and potato plants, phytohormones were replaced by solutions of Ecostim and Ecostim 1, which exhibited auxin-cytokinin activity. The cost of these substitutes is much lower than that of commercial phytohormones. Conclusions. It is shown that optimal for growth in the MS medium in the callusogenesis of Solanacea cultures in vitro. That variant with the use of cytokinin substitutes Ecostim and Ecostim 1 with the rate of using of 35.0 and 40.0 mg/L. Keywords: modification of MS medium, potatoes, tomatoes, cell culture in vitro.

Aim. Determination the possibility of agar replacement with normal and waxy type starches produced from barley seeds in the nutrient media for spring barley haploid production. Methods. Anthers of spring barley line DH00-126 were inoculated on inductive media containing N6 macro-, MS micronutrients, organic supplements, maltose (9.0 %) and differed in solidifying agents (agar, starch from barley seeds or chemically modified starch D5a-1). Trophic capacities of barley starch were determined both in anther and embryo culture of barley. Results. It was determined that barley starch of waxy type was not suitable for solidifying of nutrient medium. Use of normal type barley starch (6.5 %) resulted in sufficient decrease in the frequency of green plant regeneration. At the same time, this modification had no effect on the number of morphogenic anthers. Barley starch was inferior to pea starch in the ability to support seedling growth in embryo culture in vitro. Conclusions. Barley starch of normal type has worse trophic capacities in comparison to pea starch in embryo culture. It was appeared barley starch to be a less suitable gelling agent of medium for spring barley haploid production in anther culture in vitro than agar and chemically modified starch D5a-1. Keywords: Hordeum vulgare L., anther culture in vitro, embryo culture in vitro, agar, barley starch.
