Two compounds namely lupeol (1) and epilupeol (2) were isolated from n-hexane soluble fraction of crude methanol extract of the powdered whole plant of Samanea saman Merr. for the first time. The structures of the isolated compounds were established by extensive analyses of their high resolution NMR spectral data as well as co- TLC with authentic samples. In our preliminary screening, the n-hexane, carbon tetrachloride, and dichloromethane soluble fractions of the crude methanolic extract of S. saman were subjected to antimicrobial activity and brine shrimp lethality bioassay. The carbon tetrachloride soluble partitionate of the methanol extract exhibited mild to moderate antimicrobial activity and strong cytotoxicity having LC50 of 0.831μg/ml. In the present investigation the n-hexane, carbon tetrachloride and choloroform soluble fractions of crude methanolic extract of Samanea saman bark were tested for antioxidant, antimicrobial and cytotoxic potential. Antioxidant activity of the extracts was evaluated by DPPH radical scavenging assay and total antioxidant activity test. Antimicrobial activity was tested using disc diffusion method against thirteen bacteria and three fungi and cytotoxicity was tested by brine shrimp lethality bioassay. Chloroform and hexane soluble fraction showed IC50 value of 12μg/ml and 14μg/ml respectively in scavenging DPPH radical while the reference Butylated hydroxytoluene showed an IC50 value of 10μg/ml. The carbon tetrachloride fraction showed the highest total antioxidant capacity. The carbon tetrachloride fraction was also found to possess mild to moderate microbial growth inhibitory capacity. In the brine shrimp lethality bioassay, the n-hexane, carbon tetrachloride, chloroform soluble fractions showed LC50 value of 14.94μg/ml, 0.831μg/ml and 3.288μg/ml respectively. The results suggest good antioxidant and cytotoxic potential of chloroform and hexane soluble fractions and antimicrobial activity of carbon tetrachloride fraction of Samanea saman bark extract. In this study, GC-MS analysis has shown that whole plant butanol fraction of rheum ribes (WBFRR) comprises of 21 compounds which exhibited anticancer (MCF-7) activity having IC50 value of 36.01± 0.26. MTT assay (MCF-7), Oxidative Burst assay using chemiluminescence technique, and B-Hatching techniques were the methods used for anticancer MCF-7, anti-inflammatory, and Brine Shrimp Lethality Assay (BSLA). GC-MS was used for structural elucidation. Whole plant methanol extract of rheum ribes (WMERR), whole plant n-hexane fraction of rheum ribes (WHFRR), and whole plant aqueous fraction of rheum ribes (WAFRR) were inactive against anticancer (MCF-7) cell line. Whole plant methanol extract of rheum ribes (WMERR), whole plant aqueous fraction of rheum ribes (WAFRR) and whole plant butanol fraction of rheum ribes (WBFRR) showed anti-inflammatory activity on ROS having IC50 value of 23.2±1.9, 24.2±2.7 and 12.0±0.6. Whole plant butanol fraction of rheum ribes (WBFRR) showed Brine Shrimp Lethality with LD50 693.302 while whole plant methanol extract of rheum ribes (WMERR) and whole plant aqueous fraction of rheum ribes (WAFRR) showed high lethality at highest concentration. This study revealed that whole plant butanol fraction of rheum ribes (WBFRR) exhibited significant anticancer (MCF-7) activity. In the near future, the constituent of whole plant butanol fraction of rheum ribes (WBFRR) can be the alternative drug against MCF-7 cell line with least toxicity and side effects.

Bangladesh possesses a rich flora of medicinal plant. Out of the estimated 5000 species of phanerogams and pteridophytes growing in this country, more than a thousand are regarded as having medicinal properties. The aim of this study was to evaluate the in vitro antimicrobial activity and brine shrimp lethality of the whole plant extracts of Spilanthes paniculata, a plant belonging to the family Asteraceae. The crude extract, n-Hexane, chloroform and ethyl acetate soluble fraction of crude extract showed significant to moderate antimicrobial activity against three Gram positive and three Gram negative microorganisms. The zones of inhibition produced by the crude extract (methanolic extract), n-hexane, choloform and ethyl acetate soluble fractions were found to be 14.89 mm-19.40 mm, 14 mm-19.40 mm, 10.66 mm-13.50 mm and 9.50 mm-13.26 mm, consecutively at a concentration of 30 µg/disc. N-Hexane, chloroform and ethyl acetate soluble fractions of methanolic extract of Spilanthes paniculata were screened for antitumor properties using brine shrimp lethality bioassay. From the results of the brine shrimp lethality bioassay, it can be well predicted that n-hexane, chloroform and ethyl acetate soluble fractions of methanolic extract possess mild cytotoxicity on shrimp naupalii. The median lethal concentration (LC50, the concentration at which 50% mortality of brine shrimp nauplii occurred) of n-hexane, chloroform and ethyl acetate were 48.978 mg/ml, 92.61 mg/ml and 216.770 mg/ml, respectively, comparison with positive control vincristine sulphate with 0.839 mg/ml. Cryptocoryne ciliata belonging to the Araceae family has been investigated for isolation of its secondary metabolites and evaluation of biological activities of the crude extractives with special emphasis to the antioxidant activity and brine shrimp lethality bioassay. The whole plant was extracted with methanol and concentrated extract was partitioned using petroleum ether, carbon tetrachloride and ethyl acetate. Aqueous soluble fraction of the methanolic extract showed the highest antioxidant activity. The carbon tetrachloride soluble fraction of the methanolic extract and the ethyl acetate soluble fraction of the methanolic extract showed moderate antioxidant activity as compared to free antioxidant activity of tert-butyl-1-hydroxytolunene. In the brine shrimp lethality bioassay, among all extracts of whole plant of C. ciliata, the carbon tetrachloride soluble fraction of the methanolic extract showed strong cytotoxic activity. Aqueous soluble fraction of the methanolic extract, methanolic crude and ethyl acetate soluble fraction of the methanolic extract showed mild cytotoxity as compared to that of vincristine sulphate. The study confirms the mild to moderate antioxidant and moderate potent cytotoxic activities of C. ciliata plant extract as compared to reference standards and therefore demands the isolation of active principles and thorough bioassay.DOI: http://dx.doi.org/10.3329/icpj.v2i2.13196 International Current Pharmaceutical Journal 2013, 2(2): 38-41
