Ice creams have great acceptance and demand by consumers around the world. They are foods obtained through an emulsion of fats and proteins, these ingredients if stored or handled incorrectly can be the source of multiplication of several microorganisms. This work focuses on the microbiological analysis of artesanal and industrial ice cream in two times of the year (winter and summer), performed using the methodology adapted from Silva et al (2010) and the legislative microbiological standards used from RDC No 12 from Anvisa (Brasil, 2001). The main focus was the detection of microorganisms of importance in the food industry, as they are precursors of foodborne diseases epidemics, such as Escherichia Coli, Salmonella, total coliforms, thermotolerant coliforms, molds, yeasts and aerobic mesophilic bacteria. It was observed in the analysis results that all 30 samples showed growth of aerobic mesophilic bacteria, molds and yeasts, that only four (13.33%) did not have total coliforms and none had Salmonella. There was no great difference between the results of the analyzes of samples collected in the summer and winter, the level of contamination in general was high in samples from both seasons.

Quantitative microbiological analysis by dilution plating method of pollen and additional male and female catkins, leaves of Betula verrucosa Ehrh. and its two cultivars: ‘Purpurea’ and ‘Youngii’ relatively of flowering period of Betula has been realized with the aim to provide new knowledge of the microbiological quality of anemophilous pollen for processing and its further application. Qualitative microbiological analysis with MALDI-TOF MS Biotyper was used in the identification of aerobic, anaerobic mesophilic bacteria and coliforms. Mixed microbiota was determined, consisting of aerobic (4.68–4.89 log cfu/g) and anaerobic (3.30–3.48 log cfu/g) mesophilic bacteria, lactobacilli (0–3.48 log cfu/g), coliform bacteria (0–4.57 log cfu/g), fungi and yeast (3.78–3.95 log cfu/g) on the pollen grains, that indicates acceptable quality in comparison with the microbiological quality parameters for bee pollen. Pantoea agglomerans was found associated with pollen of Betula verrucosa Ehrh. Recommendations on the collection of anemophilous pollen were established.

Resumo Este trabalho teve como objetivo identificar e avaliar a qualidade ambiental e microbiológica de nascentes presentes na área urbana de Santo Antônio do Monte -Minas Gerais. Os impactos ambientais nas nascentes foram avaliados a partir da interpretação do Índice de Impacto Ambiental em Nascentes – IIAN. A análise microbiológica da água foi realizada através do Teste do Substrato Cromogênico (Colitest®), que mostra a presença ou ausência de coliformes totais e termotolerantes. Foram identificadas 14 nascentes, porém em quatro destas não foi possível a aplicação dos testes (nascentes 11, 12, 13 e 14), pois encontravam-se secas, soterradas ou tomadas pelo despejo de esgoto. Das nascentes avaliadas, nenhuma atingiu classificação "Ótima" (Classe A) ou "Boa" (Classe B). Apenas uma foi classificada como "Razoável" (Nascente 1; Classe C), cinco como "Ruim" (Nascentes 2, 3, 6, 7 e 8, Classe D) e quatro como "Péssima" (Nascentes 4, 5, 9 e 10, Classe E). Todas apresentaram resultados positivos para presença de coliformes totais e termotolerantes (Escherichia coli). Palavras-chave: Mananciais. Hidrologia. Poluição. Degradação. Abstract This work aimed to identify and evaluate the environmental and microbiological quality of springs present in the urban area of Santo Antônio do Monte -Minas Gerais. The environmental impacts on the springs were assessed based on the interpretation of the Environmental Impact Index on Springs - IIAN. The microbiological analysis of the water was performed using the Chromogenic Substrate Test (Colitest®), which shows either the presence or absence of total and thermotolerant coliforms. Fourteen springs were identified, but in four of them, the application of the test was not possible (springs 11, 12, 13 and 14), as they were dry, buried or taken over by sewage discharge. Out of the springs evaluated, none reached a classification of "Excellent" (Class A) or "Good" (Class B). Only one was classified as "Reasonable" (Source 1; Class C), five as "Bad" (Source 2, 3, 6, 7 and 8, Class D) and four as "Terrible" (Source 4, 5, 9 and 10, Class E). El análisis microbiológico del agua se realizó por medio de la Prueba del Sustrato Cromogénico (Colitest®), que muestra la presencia o ausencia de coliformes totales y termotolerantes. Se identificaron 14 manantiales, pero en cuatro de ellos no fue posible aplicar las pruebas (Manantiales 11, 12, 13 y 14), ya que estaban secos, enterrados o absorbidos por la descarga de aguas residuales. De los manantiales evaluados, ninguno alcanzó clasificación de "Excelente" (Clase A) o "Bueno" (Clase B). Solo uno fue clasificado como "Aceptable" (Manantial 1; Clase C), cinco como "Malo" (Manantiales 2, 3, 6, 7 y 8, Clase D) y cuatro como "Muy malo" (Manantiales 4, 5, 9 y 10, clase E). Ice creams are susceptible to contamination by handling and bad hygiene conditions during the storage process and the fraction for sale, once those can be means of diseases. This survey’s objective was to evaluate the microbiological quality of ice creams sold in bulk, of the pasty and soft types, offered for consuming. Thirty samples of pasty ice cream, sold in bulk, and thirty samples of soft ice cream were analyzed by counting thermotolerant coliforms, coagulase-positive Staphylococcus, and researching the presence of Salmonella. During the survey, were found ten (33%) samples of pasty ice cream and five (16%) samples of express ice cream out of the limits established by the Brazilian law. Salmonella was found in four samples. These results are an alert to the need of a bigger attention to the microbiological quality of ice creams, in order to ensure the consumer’s security.

The aims of this study were to evaluate the microbiological quality and the occurrence of nontuberculous mycobacteria (NTM) in a variety of salads and sprouts from supermarkets and street vendors in Mexico City. Aerobic-mesophilic bacteria (AMB) were present in 100% of RTE-salads samples; 59% of samples were outside guidelines range (>5.17 log10CFU per g). Although fecal coliforms (FC) were present in 32% of samples, only 8% of them exceeded the permissible limit (100 MPN/g). Regarding the 100 RTE-sprouts, all samples were also positive for AMB and total coliforms (TC) and 69% for FC. Seven NTM species were recovered from 7 salad samples; they included threeM. fortuitum, twoM. chelonae, oneM. mucogenicum, and oneM. sp.Twelve RTE-sprouts samples harbored NTM, which were identified asM. porcinum(five),M. abscessus(two),M. gordonae(two),M. mucogenicum(two), andM. aviumcomplex (one). Most RTE-salads and RTE-sprouts had unsatisfactory microbiological quality and some harbored NTM associated with illness. No correlation between the presence of coliforms and NTM was found. Overall, these results suggest that RTE-salads and RTE-sprouts might function as vehicles for NTM transmission in humans; hence, proper handling and treatment before consumption of such products might be recommendable.

Tilapia (Oreochromis niloticus) is the most cultivated and consumed freshwater fish in Brazil. The present study aimed to evaluate the microbiological quality of ice and fresh tilapia samples commercialized in the Federal District. Tilapia samples were tested for counts of mesophilic and psychrotrophic bacteria, determination of total and thermotolerant coliforms, Staphylococcus aureus counts and presence of Salmonella. Ice samples were analyzed for determination of total coliforms and thermotolerant coliforms and presence of Escherichia coli. Of the 20 samples of fresh tilapia analyzed, ten samples (50%) presented Salmonella (genetically confirmed through the presence of the invA gene) and, therefore, were unfit for consumption. S. aureus was found in 11 samples (55%), and one sample of fillet presented S. aureus counts (3.15 CFU/g) above the limit allowed by Brazilian legislation (3 log CFU/g). S. aureus colonies were confirmed by detection of CoA gene in molecular analysis. Of the 14 ice samples analyzed, 12 samples (85.7%) were unfit for use in fish conservation due to the presence of total coliforms and 9 ice samples (64.3%) were also contaminated with thermotolerant coliforms. E. coli was isolated from 6 ice samples (42.9%) and confirmed in the molecular analysis through the amplification of the MalB gene. In conclusion, the high contamination of tilapia samples with Salmonella and of the ice used for its conservation with coliforms and E. coli indicates the need for better hygienic practices in the tilapia production chain, to increase its quality and microbiological safety.

Traditional concoctions sold in marketplaces are always assumed to be safe and efficient; however, they can be potentially toxic because of poor hygienic practices in plant processing or storage. The present study aimed to assess for the microbial quality of market-sold vegetable drugs in Lomé. Thus, a total of 209 plant remedies were collected in marketplaces and analysed for the presence of total aerobic bacteria, total coliforms, thermotolerant coliforms, Staphylococcus aureus, sulphite reducing bacteria, and yeast and moulds according to the French Association of Normalisation (AFNOR) guidelines. The results revealed that all formulations were contaminated by several microorganisms, excepted alcohol-based mixtures. According to AFNOR limits nonconform drugs were according to total aerobic bacteria (86.96% powders, 81.82% capsules, 66.67% tisanes, and 42.11% decoctions); to total coliforms (9.10% capsules, 8.70% powders and 1.75% decoction); to yeasts and moulds (77.78% ointments, 40% calcined powders, 36.36% capsules, and 23.91% powders). The microbiological analysis revealed that the majority of contaminating bacteria were gram positive catalase positive and oxidase positive bacilli. Quality control studies on market-sold remedies are currently needed to evaluate the microbial risk in consuming these products and they may allow the standardisation of plant processing and storage.

Monitoring the microbiological quality of water used for recreational activities is very important to human public health. Although the sanitary quality of recreational marine waters could be evaluated by standard methods, they are time-consuming and need confirmation. For these reasons, faster and more sensitive methods, such as the defined substrate-based technology, have been developed. In the present work, we have compared the standard method of membrane filtration using Tergitol-TTC agar for total coliforms and Escherichia coli, and Slanetz and Bartley agar for enterococci, and the IDEXX defined substrate technology for these faecal pollution indicators to determine the microbiological quality of natural recreational waters. ISO 17994:2004 standard was used to compare these methods. The IDEXX for total coliforms and E. coli, Colilert®, showed higher values than those obtained by the standard method. Enterolert® test, for the enumeration of enterococci, showed lower values when compared with the standard method. It may be concluded that more studies to evaluate the precision and accuracy of the rapid tests are required in order to apply them for routine monitoring of marine and freshwater recreational bathing areas. The main advantages of these methods are that they are more specific, feasible and simpler than the standard methodology.

The consumption of lamb meat in Brazil has increased in the last years but little information about the microbiological quality of this product is available. To evaluate the hygienic-sanitary conditions of lamb carcasses, the quantification of microorganism populations indicators (mesophiles and psychrotrophs; total and thermotolerant coliforms; Escherichia coli; moulds and yeasts) and the pathogenic microorganisms indentification (Salmonella sp. and Listeria spp.) were performed. A total of 60 lamb carcasses were sampled from one abattoir in São Paulo. Swab samples were collected from three points (forequarter, back and hindquarter) on the muscle surface after carcasses final washing. Statistical analysis consisted of descriptive evaluation of the results whose counts were grouped by intervals of microorganism populations. Counts ranged from 1.0 x 10¹ to 8.0 x 10(4) colony-forming unit cm-2 (CFU cm-2) for mesophiles; 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for psychrotrophs; < 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for moulds and yeasts; < 0.3 to > 32.0 most probable number/cm² (MPN cm-2) for total and thermotolerant coliforms and Escherichia coli. Salmonella sp. and Listeria spp. were not found in any of the carcasses. Most carcasses presented low counts for all microorganisms. Overall results may be explained by the small size of the industry where the study was taken. Results suggest that good microbiological quality lamb meat is possible to be obtained, but improvement in hygienic-sanitary conditions is still required.

A sanitation profile scoring form for evaluating sanitation in retail food stores was designed. The profile was tested in 10 Oregon retail markets to evaluate its ability to reflect sanitary conditions. At the time of inspection, samples of meat processed in-store were purchased for microbiological analysis to explore the feasibility of bacterial quality as a measurement of sanitary conditions. Microbiological tests performed included total aerobic plate count (A PC), coliform, Escherichia coli, Staphylococcus aureus, Clostridium perfringens, and Salmonella. Certain deficiencies were noted in the profile designed; however, it provides a means for objective, uniform measurement of sanitary conditions. Data show no correlation exists between microbiological quality of products processed in the store and total store profile sanitary conditions. Fifty percent of the products sampled exceeded bacterial load guidelines currently enforced in Oregon. These “high” counts appear to be directly related to poor temperature control.

The market of ready-to-eat leafy green salads is experiencing a noticeable growth in Europe. Since they are intended to be consumed without additional treatments, these ready-to-eat products are associated with a high microbiological risk. The aim of this work was to evaluate the microbiological quality and safety of ready-to-eat leafy green salads sold in widespread supermarket chains in Lazio, Italy, on the packaging date during shelf-life and during home-refrigeration. The study also aimed to determine the differences between low-, medium-, and high-cost products. Salmonella spp. and L. monocytogenes were chosen as safety indicators as specified by European regulations while total aerobic mesophilic bacteria and Escherichia coli were chosen as quality indicators as suggested by national guidelines. Analyses were performed following the ISO standards and in parallel for the evaluation of total aerobic mesophilic bacteria, with an alternative colorimetric system, the Micro Biological Survey method, in order to propose a simple, affordable and accurate alternative for testing the microbiological quality of products, especially suitable for small and medium enterprises and on-site analyses. The study revealed high, unsatisfactory, total bacterial loads in all analyzed samples on the packaging date and expiry date and a very high prevalence of Salmonella spp. (67%) regardless of the selected varieties and cost categories; L. monocytogenes was not recovered aligning with the results obtained in other studies.
