This work presents a novel and rapid approach to predict fat content in butter products based on nuclear magnetic resonance longitudinal (T1) relaxation measurements and multi-block chemometric methods. The potential of using simultaneously liquid (T1L) and solid phase (T1S) signals of fifty samples of margarine, butter and concentrated fat by Sequential and Orthogonalized Partial Least Squares (SO-PLS) and Sequential and Orthogonalized Selective Covariance Selection (SO-CovSel) methods was investigated. The two signals (T1L and T1S) were also used separately with PLS and CovSel regressions. The models were compared in term of prediction errors (RMSEP) and repeatability error (σrep). The results obtained from liquid phase (RMSEP ≈ 1.33% and σrep≈ 0.73%) are better than those obtained with solid phase (RMSEP ≈ 5.27% and σrep≈ 0.69%). Multiblock methodologies present better performance (RMSEP ≈ 1.00% and σrep≈ 0.47%) and illustrate their power in the quantitative analysis of butter products. Moreover, SO-Covsel results allow for proposing a measurement protocol based on a limited number of NMR acquisitions, which opens a new way to quantify fat content in butter products with reduced analysis times.

Background: Patients positive for three types of antiphospholipid antibodies (aPLs) (triple positivity) have been identified at a high risk for thrombotic events. However, the clinical significance of isolated lupus anticoagulant (LAC) positivity is debated. Objectives: To investigate the clinical relevance of isolated LAC. Patients/Methods 456 patients were enrolled in this study; 66 antiphospholipid syndrome patients and 390 control patients. The control group existed of autoimmune patients (n=91), patients with thrombosis but without aPLs (n=127) and normal controls (n=172). The criteria LAC, anti-cardiolipin (anti-CL) and anti-beta2glycoprotein I (anti-β2GPI) IgG and IgM and the non-criteria IgA anti-CL and anti-β2GPI, anti-domain I (anti-DI) of β2GPI IgG and anti-phosphatidylserine/prothrombin (anti-PS/PT) IgG and IgM were detected according to the ISTH guidelines for solid phase assays. Results: 70 patients were positive for LAC, of which 44 were negative for both anti-β2GPI and anti-CL. We found that isolated LAC proved to be strongly associated with vascular thrombosis (Odds ratio (OR) (95% CI) 7.3 (3.3-16.1)), even better than triple positive samples (OR 4.3 (1.6-12.2)). The titers of the anti-PS/PT IgG and IgM were significantly higher in triple positivity samples compared to samples with isolated LAC positivity. The majority of single LAC positives were anti-PS/PT negative. We observed that LAC positivity was weaker in isolated LAC positive patients compared to LAC activity in triple positive patients. Conclusions: Isolated LAC was highly associated with thrombosis. The presence of anti-PS/PT could not explain LAC positivity in isolated LAC. Isolated LAC showed a weaker LAC activity compared to triple positive patients.

This research aimed at investigating the effect using molasses and cornmeal as additives with Lactobacillus sp. and Streptomyces sp. as inoculums during fermentation on the quality of silage flour made from dead chickens. The study was conducted using a completely randomized factorial design, consisting of 2 factors with 5 replications. The materials used were the newly dead chickens which were chopped and mixed thoroughly with all ingredients; then transferred into a 5 liters plastic box for fermentation. Observations were made after 3 weeks fermentation, including: a) physical characteristics, b) microbial contents, and c) nutritional contents. The data were calculated using variance analysis utilizing computer program of SPSS version 21.0. It was found that based on the protein contents the Lactobacillus sp. (19.0%) was better than the Streptomyces sp. (17.8%) if combined with molasses and corn meal as the accelerators. However, the fat contents produced were relatively similar for both of the inoculums (mean of 37.8%). It can be concluded that in order to obtain a best fermented product in terms of the protein and fat content, the dead chicken should be fermented using molasses and cornmeal as the accelerator and Lactobacillus sp. as the inoculum.

A series of solid-phase syntheses of the protected precursor II of DDAVP was carried out. Experimental conditions were developed under which practically pure II can reproducibly be obtained in yields better than 60%. The protected precursors of DDAVP obtained by liquid- and solid-phase synthesis and DDAVP samples obtained from these precursors were undistinguishable by conventional analytical or pharmacological assays.

Food bar is high caloric food made from blended food, enriched with nutrients, then formed into a compact bar shape. It is usually consumed either as breakfast or as snack food and is better than cakes or chocolates. This study was aimed to characterize the effect of canarium nut addition on the making of banana puree food bar. A completely randomized experimental design with four levels of treatments, i.e. 0%, 5%, 10% and 15% was applied in this research. Result showed that banana puree food bar with canarium nut addition caused not only a decrease in moisture, ash, and carbohydrate contents of 3.96%, 0.1%, and 2.6% but also an increase in protein content and fat content of 1.99% and 4.68%, respectively. Keywords: canarium nut, food bar, Musa troglodytarum, puree, tongka langit banana   ABSTRAK Food bar merupakan produk pangan berkalori tinggi yang dibuat dari campuran bahan pangan (blended food), diperkaya dengan nutrisi, kemudian dibentuk menjadi bentuk padat dan kompak (a food bar form). Food bar biasanya digunakan untuk sarapan atau sebagai makanan ringan (camilan), food bar merupakan makanan yang lebih baik dibandingkan dengan kue ataupun coklat. Penelitian ini bertujuan untuk mengkarakterisasi pengaruh penambahan kenari dalam pebuatan food bar puree pisang tongka langit. Penelitian ini menggunakan rancangan acak lengkap faktor tunggal dengan perlakuan penambahan kenari 0%, 5%, 10%, 15%. Hasil penelitian menunjukkan bahwa food bar puree pisang tongka langit dengan penambahan kenari menyebabkan terjadinya penuruanan kandungan air, abu, karbohidrat yaitu 3,96%, 0,1%, 2,6% dan peningkatan kadar protein sebesar 1,99% dan lemak 4,68%. Abstract Two immunoglobulins secreted by hybridoma cell lines have been systematically investigated to determine if they could be used in solid-phase assays to give results comparable with those obtained by conventional liquid-phase radioimmunoassay (RIA). The antibodies, BQ.1 and BQ.2, bind with high specificity to the steroid hormone progesterone. The affinity constants, Ka, to 125I-labeled progesterone derivatives are 1.1 X 10(11) L/mol and 9.1 X 10(9) L/mol, respectively. Progesterone inhibited the binding of radioiodinated derivatives (amides of tyramine, histamine, and tyrosine methylester with 11 alpha-progesterone hemisuccinate) equally well. For solid-phase assays, we immobilized antibody BQ.1 via Protein A to different polystyrene surfaces (about 30 pg per tube at 50% inhibition of radiolabeled tracer). Under these conditions, the performance of this antibody for the quantification of progesterone was equivalent to that obtained in RIA. For the immobilized antibody BQ.2, only 1/10 of the amount used for optimal results in RIA was required in solid-phase assays. Binding of either antibody to the antigen was undiminished after several freezing-thawing cycles. When immobilized on solid matrices, both antibodies retained up to 95% of their binding properties for one year. Thus high-affinity, high-specificity monoclonal antibodies can be obtained for haptens and, when suitably immobilized, can be used in solid-phase assays with results equal to or better than those obtained with liquid RIA.

ABSTRACTA model has been developed to predict the true metabolizable energy (ME,) concentration in forages given to ruminants.The chemical description of forages is based on the concentrations of ash, crude protein, ether extract, sugars and α-glycans, β-glycans and lignin, volatile fatty acids and lactic acid. The model assumes complete fermentation of sugars, α-glycans and lactic acid. The extent of fermentation of β-glycans and crude protein is determined in part by rumen solid-phase outflow rate, which can itself be predicted from dry-matter intake.The model was tested using 121 graminaceous forages whose chemical composition and concentration of metabolizable energy had been measured in the Feedingstuffs Evaluation Unit at the Rowett Research Institute. The agreement between observed and predicted ME, for all classes of forage was nearly always as good as, or better than, the best prediction from single attributes of food chemistry which could only be determined retrospectively and were not consistent even within classes of forage. The model predicts a decline in ME, with increasing rumen solid-phase outflow rate which is determined mainly by the extent of fermentation of β-glycans.

Abstract BALB/c mice were immunized with human factor V. The immunogen was a mixture of procofactor (factor V) and thrombin-activated cofactor (factor Va). Spleen cells were obtained from an immunized animal and fused with NS-1 murine myeloma cells. Hybrid cell cultures were assayed for the production of antibodies to human factor V and factor Va by a solid-phase radioimmunoassay. Factor V and/or factor-Va-specific antibodies were detected in 38 of the 96 cultures assayed. The cells from 10 of these positive cultures were subcloned by limiting dilution and grown as ascites tumors in BALB/c mice. Ascitic fluids were obtained and characterized with respect to their binding interaction with human factor V and factor Va. Three hybridoma cell lines produce monoclonal antibodies that react equally well with factor V and factor Va. Another antibody reacts with both antigens, but the reactivity with factor V is better than with factor Va. An additional two antibodies react with factor Va better than factor V in the radioimmunoassay (RIA). The remaining four antibodies react exclusively with factor V. A previously described murine monoclonal antibody to human factor V (alpha HFV-1) has been used to study the peptides produced during the thrombin-catalyzed activation of human factor V. This antibody binds both factor V and factor Va, releases them at high ionic strength, and has an apparent dissociation constant for factor Va of 3 x 10(-9)M. When human factor V (mol wt 330,000) is activated by thrombin and passed over an alpha HFV-1-Sepharose affinity resin, factor Va binds and subsequently can be eluted. The eluate in 1.2 M NaCl contains two fragments of apparent mol wt 93,000 and 70,000. EDTA, which inactivates factor Va, promotes release of the mol wt 93,000 fragment from factor Va bound to the antibody. Subsequent elution with 1.2 M NaCl releases the mol wt 70,000 fragment. These observations indicate that human factor Va is a two subunit protein and that the epitope for alpha HFV-1 is on the mol wt 70,000 fragment.
