Fusarium verticillioides is the most predominant fungal phytopathogen of cereals and it is posing great concern from a global perspective. The fungus is mainly associated with maize, rice, sorghum, wheat, sugarcane, banana, and asparagus and causes cob, stalk, ear, root, crown, top, and foot rot. F. verticillioides produces fumonisins as the major secondary metabolite along with trace levels of beauvericin, fusaric acid, fusarin C, gibberiliformin, and moniliformin. Being a potential carcinogen, fumonisins continue to receive major attention as they are common contaminants in cereals and its processed food products. The importance of elimination of F. verticillioides growth and its associated fumonisin from cereals cannot be overemphasized considering the significant health hazards associated with its consumption. Physical and chemical approaches have been shown to reduce fumonisin B1 concentrations among feeds and food products but have proved to be ineffective during the production process. Hence, biological control methods using microorganisms, plant extracts, antioxidants, essential oils, phenolic compounds, and other advanced technologies such as growing disease-resistant crops by applying genetic engineering, have become an effective alternative for managing F. verticillioides and its toxin. The different methods, challenges, and concerns regarding the biocontrol of F. verticillioides and production of fumonisin B1 have been addressed in the present review.

The fungi Fusarium moniliforme Sheldon and Fusarium proliferatum (Matsushima) Nirenberg produce a series of toxins on corn which include the fumonisins of which fumonisin B1 and B2 are considered to have cancer promoting activity. Both fungi produce similar ratios of the fumonisins B1 to B2. Other mycotoxins produced include moniliformin, fusarin C and fusaric acid. Fumonisin B1 has been shown to be responsible for most of the toxicological affects observed from ingesting corn infected by toxigenic isolates of these fungi. The distribution of the two fungi is generally similar, although F. proliferatum is isolated more frequently from sorghum than corn. They occur worldwide on other food crops, such as rice, sorghum, millet, several fruits and vegetables. Both fungi are ear rot pathogens of corn, thus, mycotoxin production occurs under field conditions, although it also may occur in storage. One or both fungi may have a frequency of occurrence of 90% or higher in corn; 90% of the F. moniliforme isolates produce the fumonisins. On corn and corn products the range of concentrations reported is 0.3 to 330 μg/g of corn-based product. These concentrations include both corn-based animal feed and human foods.

Mycotoxins contaminate crops worldwide and play a role in animal health and performance. Multiple mycotoxins may co-occur which may increase the impact on the animal. To assess the multiple mycotoxin profile of corn (Zea mays), we conducted a 7-year survey of new crop corn grain and silage in the United States. A total of 711 grain and 1117 silage samples were collected between 2013 and 2019 and analyzed for the simultaneous presence of 35 mycotoxins using ultra-performance liquid chromatography–tandem mass spectrometry. The measured mean number of mycotoxins per sample were 4.8 (grain) and 5.2 (silage), ranging from 0 to 13. Fusaric acid (FA) was most frequently detected in 78.1 and 93.8% of grains and silages, respectively, followed by deoxynivalenol (DON) in 75.7 and 88.2% of samples. Fumonisin B1 (FB1), fumonisin B2 and 15-acetyl-deoxynivalenol (15ADON) followed. The greatest (p < 0.05) co-occurrence was between FA and DON in 59.1% of grains and 82.7% of silages, followed by FA with FB1, DON with 15ADON, and FA with 15ADON. Although many samples had lower mycotoxin concentrations, 1.6% (grain) and 7.9% (silage) of tested samples had DON ≥ 5000 µg/kg. Fumonisins were detected ≥ 10,000 µg/kg in 9.6 and 3.9% of grain and silage samples, respectively. Concentrations in grain varied by year for eight mycotoxin groups (p < 0.05), while all 10 groups showed yearly variations in silage. Our survey suggest that multiple mycotoxins frequently co-occur in corn grain and silage in the Unites States, and some of the more prevalent mycotoxins are those that may not be routinely analyzed (i.e., FA and 15ADON). Assessment of multiple mycotoxins should be considered when developing management programs.

AbstractPeroxisomes are ubiquitous organelles in eukaryotic cells that fulfill various important metabolic functions. In this study, we investigated the role of Docking/Translocation Module (DTM) peroxins, mainly FvPex8, FvPex13, FvPex14, and FvPex33, in Fusarium verticillioides virulence and fumonisin B1 (FB1) biosynthesis. Protein interaction experiments suggested that FvPex13 serves as the core subunit of F. verticillioides DTM. When we generated gene deletion mutants (ΔFvpex8, ΔFvpex13, ΔFvpex14, ΔFvpex33, ΔFvpex33/14) and examined whether the expression of other peroxin genes were affected in the DTM mutants, ΔFvpex8 strain showed most drastic changes to PEX gene expression profiles. Deletion mutants exhibited disparity in carbon source utilization and defect in cell wall integrity when stress agents were applied. Under nutrient starvation, mutants also showed higher levels of lipid droplet accumulation. Notably, ΔFvpex8 mutant showed significant FB1 reduction and altered expression of FUM1 and FUM19 genes. However, FvPex13 was primarily responsible for virulence, while ΔFvpex33/14 double mutant also showed virulence defect. In summary, our study suggests that FvPex13 is the core component of DTM, regulating peroxisome membrane biogenesis as well as PTS1- and PTS2-mediated transmembrane cargo transportation. Importantly, we predict FvPex8 as a key component in DTM that affects peroxisome function in FB1 biosynthesis in F. verticillioides.

The article discusses the possibility of expanding the range of strategically important consumer food products such as vegetable oil.The necessity of developing an innovative technology for obtaining non-traditional vegetable oils, which will improve the economic mechanism for the development of the food market, has been substantiated. The physical and chemical characteristics of oil seeds grown in Uzbekistan have been studied. It has been proven that the oils obtained from non-traditional oilseeds can be used for consumption, and will play an important role in expanding the range of non-traditional vegetable oils.

Climate change is primarily manifested by elevated temperature and carbon dioxide (CO2) levels and is projected to provide suitable cultivation grounds for pests and pathogens in the otherwise unsuitable regions. The impacts of climate change have been predicted in many parts of the world, which could threaten global food safety and food security. The aim of the present work was therefore to examine the interacting effects of water activity (aw) (0.92, 0.95, 0.98 aw), CO2 (400, 800, 1200 ppm) and temperature (30, 35 °C and 30, 33 °C for Fusarium verticillioides and F. graminearum, respectively) on fungal growth and mycotoxin production of acclimatised isolates of F. verticillioides and F. graminearum isolated from maize. To determine fungal growth, the colony diameters were measured on days 1, 3, 5, and 7. The mycotoxins produced were quantified using a quadrupole-time-of-flight mass spectrometer (QTOF-MS) combined with ultra-high-performance liquid chromatography (UHPLC) system. For F. verticillioides, the optimum conditions for growth of fumonisin B1 (FB1), and fumonisin B2 (FB2) were 30 °C + 0.98 aw + 400 ppm CO2. These conditions were also optimum for F. graminearum growth, and zearalenone (ZEA) and deoxynivalenol (DON) production. Since 30 °C and 400 ppm CO2 were the baseline treatments, it was hence concluded that the elevated temperature and CO2 levels tested did not seem to significantly impact fungal growth and mycotoxin production of acclimatised Fusarium isolates. To the best of our knowledge thus far, the present work described for the first time the effects of simulated climate change conditions on fungal growth and mycotoxin production of acclimatised isolates of F. verticillioides and F. graminearum.

Investigating the in vitro fumonisin biosynthesis and the genetic structure of Fusarium verticillioides populations can provide important insights into the relationships between strains originating from various world regions. In this study, 90 F. verticillioides strains isolated from maize in five Mediterranean countries (Italy, Spain, Tunisia, Egypt and Iran) were analyzed to investigate their ability to in vitro biosynthesize fumonisin B1, fumonisin B2 and fumonisin B3 and to characterize their genetic profile. In general, 80% of the analyzed strains were able to biosynthesize fumonisins (range 0.03–69.84 μg/g). Populations from Italy, Spain, Tunisia and Iran showed a similar percentage of fumonisin producing strains (>90%); conversely, the Egyptian population showed a lower level of producing strains (46%). Significant differences in fumonisin biosynthesis were detected among strains isolated in the same country and among strains isolated from different countries. A portion of the divergent FUM1 gene and of intergenic regions FUM6-FUM7 and FUM7-FUM8 were sequenced to evaluate strain diversity among populations. A high level of genetic uniformity inside the populations analyzed was detected. Apparently, neither geographical origin nor fumonisin production ability were correlated to the genetic diversity of the strain set. However, four strains from Egypt differed from the remaining strains.
