Downy mildew of peas is caused by the obligate parasite Peronospora pisi, which occurs sporadically throughout temperate pea-growing regions across the world. To screen pea lines against this biotrophic pathogen, a suitable and reproducible in vitro method using living plant material is required. Field screening can be influenced by environmental factors, thus giving variable results. The aim of this study was to develop a method that could reliably be used to screen pea cultivars against P. pisi in a laboratory setting. A range of bioassays were used to test various methods of inoculation, utilizing sporangia and naturally infested soil. Latent infection was achieved by planting seeds in soil collected from a site with a known history of P. pisi infection and directly inoculating young pea plants with sporangia. Out of the 108 plants which survived the experimental period, only two plants expressed visible signs of disease; however, through a two-step nested PCR process we detected latent infection in 24 plants. This research highlights the importance of considering the presence of latent infection when screening pea lines against downy mildew.

The Reformation of Prophecy presents and supports the case for viewing the prophet and biblical prophecy as a powerful lens by which to illuminate many aspects of the reforming work of the Protestant reformers in the sixteenth and seventeenth centuries. It provides a chronological and developmental analysis of the significance of the prophet and biblical prophecy across leading Protestant reformers in articulating a theology of the priesthood of all believers, a biblical model of the pastoral office, a biblical vision of the reform of worship, and biblical processes for discerning right interpretation of Scripture. Through the tool of the prophet and biblical prophecy, the reformers framed their work under, within, and in support of the authority of Scripture—for the true prophet speaks the Word of God alone and calls the people, their worship and their beliefs and practices, back to the Word of God. The book also demonstrates how interpretations and understandings of the prophet and biblical prophecy contributed to the formation and consolidation of distinctive confessional identities, especially around differences in their visions of sacred history, Christological exegesis of Old Testament prophecy, and interpretation of Old Testament metaphors. This book illuminates the significant shifts in the history of Protestant reformers’ engagement with the prophet and biblical prophecy—shifts from these serving as a tool to advance the priesthood of all believers to a tool to clarify and buttress clerical identity and authority to a site of polemical-confessional exchange concerning right interpretations of Scripture.

Wood-based mulches are used in avocado production and are being tested on Fraser fir for reduction of Phytophthora root rot, caused by Phytophthora cinnamomi. Research with avocado has suggested a role of microbial cellulase enzymes in pathogen suppression through effects on the cellulosic cell walls of Phytophthora. This work was conducted to determine whether cellulase activity could account for disease suppression in mulch systems. A standard curve was developed to correlate cellulase activity in mulches with concentrations of a cellulase product. Based on this curve, cellulase activity in mulch samples was equivalent to a cellulase enzyme concentration of 25 U ml–1 or greater of product. Sustained exposure of P. cinnamomi to cellulase at 10 to 50 U ml–1 significantly reduced sporangia production, but biomass was only reduced with concentrations over 100 U ml–1. In a lupine bioassay, cellulase was applied to infested soil at 100 or 1,000 U ml–1 with three timings. Cellulase activity diminished by 47% between 1 and 15 days after application. Cellulase applied at 100 U ml–1 2 weeks before planting yielded activity of 20.08 μmol glucose equivalents per gram of soil water (GE g–1 aq) at planting, a level equivalent to mulch samples. Cellulase activity at planting ranged from 3.35 to 48.67 μmol GE g–1 aq, but no treatment significantly affected disease progress. Based on in vitro assays, cellulase activity in mulch was sufficient to impair sporangia production of P. cinnamomi, but not always sufficient to impact vegetative biomass.

Inhibition of protein–DNA interactions represents an attractive strategy to modulate essential cellular functions. We reported the synthesis of unique oligoamide-based foldamers that adopt single helical conformations and mimic the negatively charged phosphate moieties of B-DNA. These mimics alter the activity of DNA interacting enzymes used as targets for cancer treatment, such as DNA topoisomerase I, and they are cytotoxic only in the presence of a transfection agent. The aim of our study was to improve internalization and selective delivery of these highly charged molecules to cancer cells. For this purpose, we synthesized an antibody-drug conjugate (ADC) using a DNA mimic as a payload to specifically target cancer cells overexpressing HER2. We report the bioconjugation of a 16-mer DNA mimic with trastuzumab and its functional validation in breast and ovarian cancer cells expressing various levels of HER2. Binding of the ADC to HER2 increased with the expression of the receptor. The ADC was internalized into cells and was more efficient than trastuzumab at inhibiting their growth in vitro. These results provide proof of concept that it is possible to site-specifically graft high molecular weight payloads such as DNA mimics onto monoclonal antibodies to improve their selective internalization and delivery in cancer cells.

Justice Amy Coney Barrett’s nomination and confirmation featured frequent references to her role as a mother. This article situates these references within the trajectory of American political development to demonstrate how motherhood operates as a mechanism for enforcing a white-centered racial order. Through a close analysis of both the history of politicized motherhood as well as Barrett’s nomination and confirmation hearings, I make a series of claims about motherhood and contemporary conservatism. First, conservatives stress the virtuousness of motherhood through a division between public and private spheres that valorizes the middle-class white mother. Second, conservatives emphasize certain mothering practices associated with the middle-class white family. Third, conservatives leverage an epistemological claim about the universality of mothering experiences to universalize white motherhood. Finally, this universalism obscures how motherhood operates as a site in which power distinguishes between good and bad mothers and allocates resources accordingly. By attending to what I call the “republican motherhood script” operating in contemporary conservatism, I argue that motherhood is an ideological apparatus for enforcing a racial order premised on white protectionism.

Though several powerful explorations of modern evangelical influence in American politics and culture have appeared in recent years (many of which illumine the seeming complications of evangelical influence in the Trump era), there is more work that needs to be done on the matter of evangelical understandings of and influence in American law enforcement. This article explores evangelical interest and influence in modern American policing. Drawing upon complementary interpretations of the “antistatist statist” nature of modern evangelicalism and the carceral state, this article offers a short history of modern evangelical understandings of law enforcement and an exploration of contemporary evangelical ministry to police officers. It argues that, in their entries into debates about law enforcement’s purpose in American life, evangelicals frame policing as both a divinely sanctioned activity and a site of sentimental engagement. Both frames expand the power and reach of policing, limiting evangelicals’ abilities to see and correct problems within the profession.

The gold standard for classification of neurodegenerative diseases is postmortem histopathology; however, the diagnostic odyssey of this case challenges such a clinicopathologic model. We evaluated a 60-year-old woman with a 7-year history of a progressive dystonia–ataxia syndrome with supranuclear gaze palsy, suspected to represent Niemann–Pick disease Type C. Postmortem evaluation unexpectedly demonstrated neurodegeneration with 4-repeat tau deposition in a distribution diagnostic of progressive supranuclear palsy (PSP). Whole-exome sequencing revealed a new heterozygous variant in TGM6, associated with spinocerebellar ataxia type 35 (SCA35). This novel TGM6 variant reduced transglutaminase activity in vitro, suggesting it was pathogenic. This case could be interpreted as expanding: (1) the PSP phenotype to include a spinocerebellar variant; (2) SCA35 as a tau proteinopathy; or (3) TGM6 as a novel genetic variant underlying a SCA35 phenotype with PSP pathology. None of these interpretations seem adequate. We instead hypothesize that impairment in the crosslinking of tau by the TGM6-encoded transglutaminase enzyme may compromise tau functionally and structurally, leading to its aggregation in a pattern currently classified as PSP. The lessons from this case study encourage a reassessment of our clinicopathology-based nosology.

Five commercial composts were evaluated to suppress the root-rot pathogens (Fusarium solani (Mart.) App. and Wr, Pythium ultimum Trow, Rhizoctonia solani Kuhn, and Sclerotium rolfsii Sacc.) of cucumber plants under in vitro and greenhouse conditions. In vitro tests showed that all tested unautoclaved and unfiltrated composts water extracts (CWEs) had inhibitor effect against pathogenic fungi, compared to autoclaved and filtrated ones. Also, the inhibitor effects of 40 bacteria and 15 fungi isolated from composts were tested against the mycelial growth of cucumber root-rot pathogens. Twenty two bacteria and twelve fungal isolates had antagonistic effect against root-rot pathogens. The antagonistic fungal isolates were identified as 6 isolates belong to the genus Aspergillus spp., 5 isolates belong to the genus Penicillium spp. and one isolate belong to the genus Chaetomium spp. Under greenhouse conditions, the obtained results in pot experiment using artificial infested soil with cucumber root-rot pathogens showed that the compost amended soil reduced the percentage of disease incidence, pathogenic fungi population, and improved the cucumber vegetative parameters as shoot length, root length, fresh weight, and dry weight. These results suggested that composts are consequently considered as control measure against cucumber root-rot pathogens.
