Dystocia causes great financial losses: due to dystocia milk production is decreased and the probability of calf loss is increased. There are many factors that may cause dystocia. One of the factors –often investigated in beef cattle- is pelvic measurements. There have not been inner pelvic measurement comparisons in dairy breeds in Hungary. After comparing the imported, primiparous cows, Jersey turned out to have the smallest absolute inner pelvic measures. According to their age and weight, Brown Swiss cows had the largest pelvic dimensions. Ayrshires, Norwegian and Swedish Red, the three dairy breeds which share similar genetic background did not differ in most measures. Holsteins were closest to the seemingly ideal 1:1 horizontal and vertical diameter ratio; however this breed suffers the most from dystocia. Jerseys, despite having the smallest pelvic area are famous of their calving ease, perhaps not by coincidence. Though this dairy breed is the lightest, when pelvic area was compared in ratio of body weight Jerseys were not smaller than the 130 and 114 kg heavier Norwegian and Swedish Red cows.

Body weight and seven body dimensions measuring the length, height, width and depth were taken at birth and every 180 days up to 720 days of age on 49 pure Canadian, 49 first cross of 1/2 Brown Swiss 1/2 Canadian, and 30 backcross of 1/4 Brown Swiss 3/4 Canadian heifers. The animals were raised up to 180 days of age at the Deschambault Station, then half the pure Canadian and the first cross were transferred to the La Pocatière Station. At birth, the pure Canadian calves weighed 29.2 kg and were lighter than the first and backcrosses by 6.9 kg (24%) and 4.8 kg (16%), respectively. The two crosses exceeded the pure Canadian in all the dimensions, the differences being significant in height at shoulder and width at rumps. At 360 days of age the two crosses were about 6% heavier and 4% higher at shoulder than the pure Canadian. With the exception of body length after 1 yr of age in which the backcross was slightly inferior to the pure Canadian, the two crosses were generally larger than the pure Canadian, with the backcross being intermediate between the first cross and the pure Canadian. The effects of season of birth and age of dam were found to be non-significant on most of the traits studied in this experiment.

The aim of this study was to analyze the mineral composition of milk, the amino acid composition of milk protein, and the productivity per 305 days of lactation in crossbred cows compared to pure-breed local Ukrainian analogues. The research was conducted at cows of the Ukrainian Black-Spotted (UBS) dairy breed and crossbred cows of the first-generation obtained as a result of crossing with Brown Swiss breed and at cows of the Ukrainian Red-Spotted (URS) dairy breed and crossbred cows of the first-generation obtained by crossing of the URS dairy breed with Montbeliarde breed. In both farms there were two groups of pure-breed and crossbred cows-analogues. The mineral composition of milk and its technological properties in crossbred cows were higher compared to the pure-breed analogues. The fat content of milk crossbred cows exceeded by 0.08-0.16%, protein exceeded by 0.15-0.22%, and the main trace elements include Ca and P – by 2.66-8.83 and 2.12-7.81 mg/100 g of milk, respectively. Milk protein of crossbred cows was marked by a more balanced composition of essential amino acids (EAAs) and their ratio suitable for processing. The amount of EAAs in 100 g of milk protein of crossbred cows was 3.11-6.25% higher compared to local analogues. Crossbred cows dominated pure-breed analogues by the mineral composition of milk. The authors recommend the use of Montbeliarde or Brown Swiss breeds in commercial herds to improve the technological properties of milk.

The aim of this work was to study bioenergetic and ethological indicators in crossbred first-calf heifers compared to purebred first-calf heifers. The research was conducted on the first-calf heifers of Ukrainian Black-Pied dairy breed and first-generation crossbreeds obtained as a result of crossing with Brown Swiss breed, as well as, on the first-calf heifers of Ukrainian Red-Pied dairy breed and first-generation crossbreeds obtained as a result of crossing Ukrainian Red-Pied dairy with Montbéliarde breed. The purebred Black-and Red-Pied first-calf heifers prevailed over crossbreed first-calf heifers on the indicator of milk yield for 305 days of lactation by 106.45 and 218.36 kg, respectively. Accordingly, the indicators of average daily milk yield were also higher by 0.35 and 0.72 kg and the indicators of average daily milk yield adjusted by 4 % fat content per 0.15 and 0.16 kg. At the same time, the indicator of average fat content in milk prevailed in crossbreeds of Black-Pied and Brown Swiss breed by 0.08 % compared to purebred Black-Pied breeds, and in Red-Pied crossbreeds with Montbéliarde breed by 0.16 % compared to purebred Red-Pied breeds. The purebred Black-Pied first-calf heifers had an advantage by 2.04 MJ compared to crossbreeds, while the Red-Pied first-calf heifers had a slight advantage by 0.06 MJ compared to crossbreed first-calf heifers on the indicator of daily cost of exchange energy (EE). The purebred Black-and Red-Pied first-calf heifers had higher cost of exchange energy allocated with milk compared to crossbreed first-calf heifers by 0.42 and 0.82 MJ. They also had higher cost of exchange energy rates for heat products by 0.79 and 0.25 MJ. According to the energy index indicator, purebred Black - and Red-Pied first-calf heifers prevailed over crossbreeds first-calf heifers by 0.16 and 0.07 %, respectively. The same trend was observed in relation to the productive index indicator. The purebred Black-and Red-Pied first-calf heifers had a slight advantage by 0.001 and 0.002 kg adjusted by 4 % of milk fat content per 1 MJ. According to the indicators of daily behavioral reactions, no special differences between a purebred and crossbreed were found.

Background Studying cardiac electrophysiology in isolated perfused beating murine hearts is a well-established method. The range of normal values for left atrial action potential durations (LA-APD), activation times (LA-AT) and effective refractory periods (atrial ERP) in murine wildtype (WT) is not well known. Purpose This study aimed to establish reference values for LA-APD, LA-AT and atrial ERP and to identify factors that influence these electrophysiological parameters in wildtype (WT) mice. Method We combined results from isolated beating heart Langendorff experiments carried out in WT between 2005 and 2019 using an octopolar catheter inserted into the right atrium and a monophasic action potential electrode recording from the LA epicardium. Electrophysiological parameters (LA-APD at 50%, 70%, 90% repolarization (APD50, APD70, APD90), LA-AT and atrial ERP) at different pacing cycle lengths (PCL) were summarized. We analyzed effects of PCL, genetic background, age, gender, heart weight to body weight ratio (HW/BW), LA weight to body weight ratio (LAW/BW) as well as coronary flow and temperature as experimental conditions. Results Electrophysiological parameters from 222 isolated hearts (114 female, mean age 6.6 ± 0.25 months, range 2.47-17.7 months) of different backgrounds (77 C57BL/6, 23 FVB/N, 33 MF1, 69 129/Sv and 20 Swiss agouti) were combined. Coronary flow rate, flow temperature and start of isolation to cannulation time were constant experimental conditions over the timespan of experiments. LA-APD was longer while LA-AT decreased with longer PCL throughout all genetic backgrounds (Figure 1A). Genetic background showed strong effects on all electrophysiological parameters. LA activation was delayed in 129/Sv compared to other backgrounds (Figure 1D). LA-APD70 and atrial ERP were significantly shorter in Swiss agouti background compared to others. LA-APD70 was also significantly prolonged in 129/Sv background compared to MF1 (Figure 1C). Atrial ERP was longer in FVB/N compared to other backgrounds. Age effects were compared in groups. Atrial ERP was significantly longer in mice ≤ 3 months compared to all older mice. Atrial ERP was also significantly prolonged (+ 3.4ms, + 13.5%) in female mice compared to males (Figure 1B). Conclusion This dataset summarizes left atrial electrophysiological parameters in the beating mouse heart and can serve as a reference for design and interpretation of electrophysiological experiments in murine models of commonly used genetic backgrounds. We confirm that cycle length, genetic background, age and gender affect atrial electrophysiological parameters. Awareness of these will support successful experimental design. Abstract Figure 1

In contrast to oocytes matured in vitro, porcine embryos that result from in vivo maturation and fertilization have a high developmental competence and readily make the transition from oocyte to blastocyst. This observation led us to investigate the transcript profile differences between in vivo- and in vitro-matured porcine oocytes. For the in vivo-matured group, oviducts of 3 gilts of similar genetic background were flushed 2 days after detection of standing oestrus. MII oocytes were collected in pools of 10 and snap frozen in liquid nitrogen for RNA isolation. The in vitro-matured oocytes were obtained by euthanizing 3 gilts, again with a similar genetic background and recovering the ovaries. Follicles (2 to 8 mm in size) were aspirated and oocytes with multiple layers of cumulus cells and uniform cytoplasm were placed in M-199 supplemented with LH, FSH and epidermal growth factor for 42 h. Upon maturation, cumulus cells were stripped and the healthy MII oocytes were collected in pools of 10 and snap frozen. Total RNA was extracted from 3 pools of 10 oocytes for both treatments using an All prep DNA/RNA micro isolation kit (Qiagen, Valencia, CA, USA). Complementary DNA was synthesized using oligo (dT′) primed reverse transcriptase with superscript III (Invitrogen, Carlsbad, CA, USA). Second-strand cDNA was synthesized using DNA polymerase I and sequenced using Illumina Genome Analyzer II. All reads were aligned to a custom-built porcine transcriptome. There were over 18 million reads in the 2 maturation groups that tiled to the 34 433-member transcriptome: 1317 transcripts were detected with a P ≤ 0.1 (Students t-test), a minimum of 7 reads in at least 1 of the treatments and ≥2-fold difference. Real-time PCR was used on selected transcripts. Comparative CT Method was used on an IQ real-time PCR system with the Bio–Rad SYBR green mix. Statistical differences were determined using the Proc general linear model procedure of SAS (SAS Institute Inc., Cary, NC) and means separated with a l.s.d. (P ≤ 0.05). The misrepresented transcripts from the sequencing data were also characterized using the functional annotation tool DAVID. Twelve pathways were overrepresented in the in vitro-matured oocytes (the top 4 are pathways to cancer, spliceosome, cell cycle and ubiquitin-mediated proteolysis). Eight pathways were underrepresented in the in vitro-matured oocytes (the top 4 are cytoskeleton regulation, T-cell receptor signaling pathway, ubiquitin-mediated proteolysis and cell cycle). Eight transcripts were selected for real-time PCR. ZP2 was higher in the in vitro-matured oocytes as determined by both sequencing and real time. ATG4, HSP90, UBAP2 and SOX4 were not different, regardless of assay. SLC7A3, MRPS36 and PDHX2 were not different based on sequencing, but based on real-time MRPS36 and PDHX2, were higher in the in vivo group and SLC7A3 was higher in the in vitro group. In conclusion, there is an abundance of misregulated transcripts and altered pathways in in vitro-matured oocytes. This dataset is a tool that may provide clues to improve the in vitro maturation process so that in vitro-matured oocytes will be more like their in vivo-matured counterparts, thus improving developmental competence. The objective of this review is to evaluate examination tools and how they can be used to assess the breeding potential of beef heifers prior to service. Reproductive tract examination techniques, pelvic measurement, and antral follicle counts (AFC) are methods that are used and could be of value to farmers where they can either identify pubescent heifers and/or those with the best reproductive potential. Reproductive tract scoring (RTS) to assess the pubertal state and therefore reproductive status of replacement heifers is explored alongside further adaptions of this method. The use of pelvic measurement is considered: first, where a small pelvic area or an abnormal shaped pelvic inlet is identified, which may result in elimination from the breeding cohort to reduce the potential risk of dystocia and poor performance; and second, since it may be advocated for use in conjunction with other methods. AFC are included since they too may help identify the reproductive potential of heifers. Evaluation of these examination tools demonstrates that the use of modified RTS methods, to include ultrasound examination to detect the absence of a corpus luteum (CL), absence of a follicle ≥ 13 mm and the uterine horn diameter as well as pelvic measurements, could add value to heifer breeding soundness examinations (HBSE) when carried out.
