The microbiological quality of milk is important not only for food safety, but it can also influence the quality of dairy products. In this study, our aim was to assess the microbiological status of the bulk milk of a milk-producing farm, and some natural and flavored (garlic, dill, onion) gomolya cheeses made from pasteurized milk produced by their own processing plant. We determined the number of coliform bacteria, Escherichia coli, Staphylococcus aureus, and molds of three milk and eight cheese samples. The tests were conducted between July and September, 2017. In bulk milk, the mean coliform count was 3.83±0.17 log10 CFU/ml; the mean E. coli count was 1.38±0.14 log10 CFU/ml; the mean mold count was 3.74±1.30 log10 CFU/ml; and the S. aureus count was <1.00 log10 CFU/ml, respectively. The mean coliform count in gomolya cheeses was 3.69±1.00 log10 CFU/g; the mean E. coli count was 2.63±0.58 log10 CFU/g; the mean S. aureus count was 3.69±1.35 log10 CFU/g and the mean mold count was 1.74±0.37 log10 CFU/g. The amount of coliforms detected in different flavored gomolya cheeses were significantly different (P<0.05). More than 10 CFU/g of E. coli was found only in the dill flavored cheeses, and S. aureus was found only in dill (3.66±1.86 log10 CFU/g) and onion (3.71±0.52 log10 CFU/g) flavored gomolya cheeses. Based on the obtained results, it was found that the amount of coliform bacteria and E. coli in bulk milk exceeded the limit set in regulation of the Hungarian Ministry of Health (MoH) 4/1998 (XI. 11.) and the amount of S. aureus was below the limit. For gomolya cheeses, the S. aureus count exceeded the limit. The amount of coliform bacteria remained above the limit in cheeses, except for the garlic flavored gomolya cheese. In cheeses, a larger E. coli count was detected than in the bulk milk, but there is no specific limit for cheeses in the regulation. The mold count exceeded the limit specified in the regulation in cheeses, but a lower value was detected relative to milk. The results show that, in the case of bulk milk and gomolya cheeses, certain detected quantities exceeded the limit values set forth in regulation of MoH 4/1998 (XI. 11.). The results indicate an inadequate microbiological state of the raw material and the finished products. The reasons for these are due to reduced technological hygiene or the inappropriate handling of raw material and finished products. In this study, we have summarized the results of our preliminary studies, which can provide a basis for further hygiene studies.

ABSTRACT: Transport of cooled raw milk in bulk has greatly improved the quality of the raw material collected by dairy plants as it reduces the proliferation of mesophilic microorganisms that cause milk acidity and hinder its processing. However, refrigeration has favored the growth of psychrotrophic microorganisms which are able to grow at low temperatures (below 10ºC) and that produce heat resistant enzymes which degrade some milk components, reducing milk shelf life and causing organoleptic changes. The aim of this paper was to evaluate the microbiological quality of raw milk in dairy farms and after its transport to the processing dairy plant, through plate counting of mesophilic and psychrotrophic microorganisms. Fourteen milk samples were collected from tanks of the dairy-farming properties, and one sample was collected from their milk transport tanker at the entrance of the processing plant. Our results showed that the mean number of mesophilic microorganisms was higher in samples collected straight from the dairy farm tanks than in the samples collected from the transportation tankers at the entrance of the plant. Of the 14 sampled tanks, 64.3% were non-compliant with legislation. The sample collected from the milk transportation tanker containing milk from all properties showed a higher mean number of psychrotrophic microorganisms than the dairy farm samples. We conclude that the milk from dairy properties showed a higher amount of mesophilic microorganisms, and after transportation, at the entrance of the processing plant, there is a higher amount of psychrotrophic microorganisms.

Two studies were undertaken to determine the microbiological status of sheep carcass meat and frozen, bulk-packed sheep meat produced in Australia. Samples were collected from 470 sheep carcasses and 415 cartons of frozen sheep trimmings over a period of approximately 12 months. Samples were collected from plants processing sheep carcasses for domestic or export markets. On carcasses, where bacterial counts were obtained, the mean of the log10 aerobic plate count (APC) was 3.92/cm2, the geometric mean of the most probable number (MPN) per square centimeter of Escherichia coli (biotype I) was 23, and the geometric mean of the coliform count was 38 MPN per cm2. A high percentage (75%) of samples was positive for E. coli (biotype I), 81% were positive for coliforms, 5.74% were positive for Salmonella spp., and 1.29% were positive for Campylobacter. Bacterial counts were higher on carcasses chilled over a weekend than on carcasses chilled for 24 h. The total number of bacteria on carcasses processed for domestic markets was similar to that on carcasses processed for export markets. E. coli O157 was not isolated from any of the 465 samples tested. Of the frozen export samples that tested positive, the mean of the log10 APC was 3.47/g, the geometric mean of the E. coli (biotype I) count was 9 MPN per g, and the geometric mean of the coliform count was 19 MPN per g. Of the frozen export samples tested, 48% were positive for E. coli (biotype I), 58% were positive for coliforms, and 6.5% were positive for Salmonella spp. E. coli O157 was recovered from 1 of 343 frozen sheep meat samples tested (0.29%). Bacterial counts were higher on samples of domestic product than on samples of export product. Results from both surveys are compared with data from similar studies conducted in other countries.

The aim of this study was to investigate the possible harmful effect of Sarcocystis parasites on bovine diaphragm meat quality. Meat samples were collected from 120 bulls aged 20–24 months. Meat quality was investigated using microbiological and physico-chemical (RP-HPLC, GC) methods 48 hours after slaughter. Sarcocystis infection was associated with increased fat content, lightness L* and drip loss, and decreased ash and protein percentages. Infection also had a significant effect on the amount of amino acids (AAs), which slowly decreased as the number of sarcocysts increased. The total amount of AAs correlated with glutamic acid content (R = 0.966, P &lt; 0.05). Heavily infected samples contained significantly lower amounts of putrescine, histamine, spermine and spermidine (P &lt; 0.05) and<br /> a noticeable increase in the total count of aerobic microorganisms, but no change in the numbers of E. coli and coliform bacteria in comparison with no infected samples. Sarcocysts in beef diaphragms did not cause serious changes in the technological quality of the meat, but the biological quality of infected meat was reduced.

ABSTRACT: Crambe is a rapeseed with high oil content and can be used as a winter cover or as a source of raw material for the production of biodiesel, however espite the growing interest in the culture, research on the subject is still incipient, especially concerning the seed production and analysis technology. The purpose of this study is to evaluate the physiological quality of crambe seeds, 'FMS Brilhante' cultivar, by testing the pH of exudate. Five seed lots were submitted to the determination of water content and the tests of germination and vigor (first count, emergence and tetrazolium). In the conduction of pH exudate test, temperatures (25 and 30oC), and periods of seed imbibition in water (15, 30 and 45 minutes) were tested. The experiment was conducted in a completely randomized manner, with four replicates, and the mean values were compared by the Tukey test at 5% probability; Pearson correlation between the pH of the exudate and initial tests was also made. Testing the pH of exudate is promising for separating lots of crambe seeds and the following combinations of 25°C/30 minutes or 30°C/45 minutes can be used.

Background and Aim: Pre-slaughter management and slaughter operations are considered critical factors for animal welfare and meat quality. Previous studies have found poor animal welfare management at municipal slaughterhouses in Ecuador, and little is known about how this affects the microbiological quality of the meat. Therefore, the aim of the study was to analyze the association of the microbiological quality of beef carcasses and animal welfare indicators in a municipal slaughterhouse in Ecuador. Materials and Methods: Data for 6 months were collected from a municipal slaughterhouse in Ecuador. Five trained researchers were strategically located along the slaughter process. A total of 351 animals were observed with regard to welfare indicators, and their carcasses were sampled to evaluate microbiological quality. Antemortem (slipping, falling, and vocalization) and postmortem animal welfare indicators (bleed interval, pH, temperature, and bruises) were measured. To determine the total aerobic bacteria (TAB) and Escherichia coli counts and the presence of Salmonella spp., we collected samples by swabbing four different points of each carcass. The association between microbiological quality and animal welfare indicators was studied using univariate and multivariate logistic regressions. Results: The mean TAB count was 5.3 log CFU/cm2, and the mean total count of E. coli was 2.4 log CFU/cm2. Salmonella spp. were isolated in 3.1% of the carcasses. An electric goad was used in all animals, 19.1% slipped at least once, and 19.9% vocalized. The mean pH of the carcasses was 7.2, and 79.2% of carcasses had bruises. Multivariate analysis showed that Salmonella spp. and the TAB count were associated with pH and the number of bruises (p = 0.01 in both cases). Conclusion: Although there was non-significant association between the majority of animal welfare indicators and microbiological quality, the poor management affecting animal welfare and carcass hygiene are worrisome.

Recently there has been considerable interest in Canada and the United States in the development of an effective, simple test for the estimation of Escherichia coli densities in shellfish and shellfish-growing waters as a better indication of "faecal" pollution than that provided by the coliform group. The fidelity of the E.C. confirmation test for the determination of E. coli densities was evaluated in 15 shellfish-growing areas which were classified as "polluted" or "unpolluted" according to generally accepted criteria. Coliform strains were isolated from 2765 E.C. gas positive confirmation test cultures and identified by IMViC tests. The mean fidelity of the test, as an indication of the presence of E. coli, was circa 90 per cent; contrary to expectations, however, the per cent recovery of E. coli was higher in unpolluted areas than in polluted areas, and varied considerably from region to region. Aerobacter aerogenes types I and II were the most common other E.C. gas positive coliform biotypes found. The usefulness of the confirmation test and the sanitary significance of these data are discussed.

The aim of the study was to synthesize the latest literature data on the content of toxic elements in tissues of hunted animals and to determine whether there is a health risk associated with consumption of these products in the light of current legal regulations. The data show the increased mean lead contents in kidneys of wild boar (1.1900 mg/kg) and roe deer (0.9060 mg/kg) and in the muscle tissue of wild boar, roe deer and deer; the contents are even up to 0.1240 mg/kg, 0.1760 mg/kg and 0.2200 mg/kg, respectively. In addition, the increased mean cadmium content indicates the possibility of the deposition of this element in muscle tissue of wild boars (0.0519 mg/kg) and deer (0.1000 mg/kg), as well as in the liver of wild boars (5.5730 mg/kg) and roe deer (6.4350 mg/kg) and in kidneys of game species, sometimes far above the applicable maximum levels. The highest mean cadmium contents in kidneys of wild boar, roe deer and deer are 49.5000 mg/kg, 80.2600 mg/kg and 4.9740 mg/kg, respectively. The mean values of the amount of arsenic do not usually exceed the legally established maximum levels of this element. Only the borderline value of arsenic in muscle tissue of roe deer is slightly exceeded and amounts to 0.2700 mg/kg. There is the possibility of the accumulation of larger amounts of mercury mainly in kidneys of deer and roe deer. The highest mean content of this element of 0.1001 mg/kg is demonstrated in the kidneys of deer. The research on the content of toxic elements in meat of hunted animals indicates the need to regularly monitor the quality of the obtained raw material as well as to exclude selected tissues of wild animals from consumption.

Background: The study aimed at assessing the knowledge of Bonendale populations related to safe drinking water; determine the microbiological quality and the household treatment practices of drinking water at the point of use (PoU). Methods: The study was descriptive and cross-sectional, conducted between November 2017 and November 2018 in Bonendale, Douala IV. A systematic random sampling method was used to select 237 households. Results: The main source of drinking water in the study population was dug wells (64.9%). The major activity around the boreholes was agriculture 50% (6/12) while agriculture and livestock rearing were practiced around 34.3% (12/35) and 42.8% (15/35) of dug wells respectively. Nearly half 110 (46.4%) of respondents had a good knowledge of physical properties of drinking water. Fifty-nine water samples out of a total of 60 tested (98.3%) were contaminated by coliform bacteria. Analysis of water samples indicated that 47.6% (10/21) of dug wells, 37.0% (10/27) of Camwater, and 41.2% (5/12) of boreholes had a total coliform count varying between 101-1000 CFU/100mL. And only a single Camwater water sample 1 (137%) had a coliform count of 0 CFU/100mL. Conclusion: These findings indicated the poor microbiological quality of point water used by Bonendale populations representing an important potential health risk of water-borne diseases. The results also reveal that a very high proportion of households treat their PoU before drinking.

The article presents the results of studies obtained in the study of the quality of milk processed with the use of high-frequency acoustic cavitation and dairy products produced with its use. The research was carried out with the use of General scientific and special research methods in the laboratory of the Department of technology of storage and processing of animal products of the K. A. Timiryazev and in the accredited laboratory of technochemical control of the all-Russian research Institute of dairy industry. It is shown that the treatment of cow's milk-raw materials high-frequency ultrasonic vibrations (above 45 kHz), the generated electric ultrasonic device immersion type pulse impact ouzo "Activator-150", the number of bacteria of group of intestinal sticks (coliforms) decreased by almost 40%, which allows to make a conclusion about the appropriateness of the selected method of exposure for the destruction of microorganisms E. coli and coliform bacteria. Processed, using high-frequency acoustic cavitation, cow's milk was used for the production of brine cheese-cheese. It is shown that cheese cheese had a high nutritional value, which is due to the preservation of essential substances in the raw material (in particular, calcium, which in the production of cheese cheese precipitates if pasteurized milk is used), elastic consistency, safety of consumer characteristics - microbiological and physico-chemical.

Introduction. The increasing bacterial contamination of water bodies requires an increase in water quality control’s reliability to ensure epidemic safety against waterborne infections. Therefore, researchers in both Russia and Europe came to the conclusion that it is necessary to search for indicator microorganisms that can more accurately suggest the presence of pathogens. microorganisms in water than traditional indicators. The aim of the study was to justify the introduction of indicator indices of fecal contamination “generalized coliform bacteria” and Escherichia coli to assess the safety of drinking water Material and methods. The article provides an analysis of domestic and international regulatory documents and literary materials regulating the quality of drinking water in terms of sanitary and microbiological indicators and assessment criteria. The results of many years of experimental and field research carried out by research organizations and practical organizations of the Federal Service for Supervision of Consumer Rights Protection and Human Welfare are presented. Results. On the territory of Russia, “general coliform bacteria,” is one of the indicator indices chosen according to the fermentation of lactose, determine the safety of drinking water. The water does not take into account pathogenic bacteria (Salmonella, Shigella) and a number of lactose-negative opportunistic bacteria, causative agents of intestinal infections. The study of microorganisms isolated from the feces of patients confirms the frequency of occurrence of lactose-negative microorganisms to varying from 20 to 100% of strains. With an annual trend towards a decrease in the percentage of non-standard drinking water samples in terms of microbiological indices, general intestinal infections (GII) of unknown etiology increase, i.e. risk of GII. If the quality of drinking water does not correspond to thermotolerant coliform bacteria (TCB), 95% of samples contain E. coli. Therefore, the determination of E. coli more reliably indicates the intake of fresh fecal contamination and provides efficiency in taking measures to eliminate an unfavorable situation than TCB. Conclusion. Reasons are given for the introduction of more reliable microbiological indicators of water safety control, such as - “generalized coliform bacteria” with the preservation of the abbreviation GCB, combining both lactose-positive and lactose-negative bacteria, determined by the sign of glucose fermentation, negative oxidase test and negative stain according to Gram and E. coli as an indicator of recent faecal contamination, which will allow the assessment of water quality for a wide range of bacteria of the order Enterobacterials, corresponding to the modern taxonomy of Enterobacteriaceae NCBI, will ensure harmonization with international requirements and the safety of drinking water for the population.
