ABSTRACT: The objectives of the present project were to evaluate the sanity and germination of tobacco seed samples cultivated in Brazil and to identify potential pathogenic fungi to the culture. Thirty-four representatives samples of tobacco seeds were evaluated through germination and sanity test by the blotter-test. On the germination test, results were expressed in percentage of seedlings considered normal, abnormal and non-germinated seeds. The percentage of seeds germination varied between 54.5 and 90%. According to sanity test, it was verified the incidence of Alternaria alternata, Cladosporium spp., Fusarium spp., and Phoma spp. The incidence of A. alternata varied from 3 to 67%, and Fusarium spp. varied from 0 to 19%. There was significative positive correlation between the incidence of A. alternata and the percentage of seed germination. Species of Alternaria and Fusarium can be potential pathogens to tobacco culture and an alarm for the necessity of seed treatment of the seeds that are transporting these fungi.

Abstract: Priming is a commercial technique used to increase the speed and uniformity of seed germination. However, the physiological quality of primed seeds is usually negatively affected during storage. Tobacco seeds of the cultivar BAT 2101 were used to investigate whether primed seeds could be cryopreserved. The most suitable substances, type of drying, and reheating during priming and cryopreservation of tobacco seeds were studied. Seed priming was performed with water, spermidine, and potassium nitrate, and drying was carried out with silica gel and a saturated saline solution. Seeds were cryopreserved in liquid nitrogen for 24 h and reheated in a water bath for two and five minutes and at ambient temperature. Tobacco seeds primed with spermidine and water can be cryopreserved without loss of physiological quality when quickly dried on silica gel and reheated in a water bath for two minutes.

Abstract: Due to the non-uninform maturation of tobacco fruits and seeds, the priming technique has been used by the tobacco industry as a way of increasing the uniformity and germinations peed of seeds. Several factors can affect priming, including the time and environment in which the treatment is performed. In order to evaluate the effect of sodium hypochlorite as priming agent at different concentrations and the priming period on the physiological quality of the tobacco seeds, it was used a seed lot of commercial cultivar CSC 439 from the Virginia varietal group, widely used by the producing companies. The seeds were subjected to priming in aerated solution at different priming periods (180, 60, 30, 15, and 5 min) and concentrations of sodium hypochlorite (1%, 0.50%, 0.25%, and 0.10 %). The physiological quality was evaluated by the water contents, germination tests, first count, germination index, initial and final stands, emergence index and electrical conductivity. The priming with sodium hypochlorite did not affect germination, but allowed a higher percentage and speed of emergence when using the 1% concentration for 180, 30 and 15 min.

The use of gravity table can result in improved quality of seeds of several species, demonstrating the superiority of the quality attributes of seeds collected in the top positions in relation to the lower positions of the discharge zone of the gravity table. The availability of information on tobacco seeds, particularly on the action of gravity table, has not been addressed in the literature. The present study was to evaluate the influence of different regulations in the gravity table on the quality of tobacco seeds. The terminal edge of the machine of 50 cm width was divided into four parts plus the outlet for stones. The treatments were in the following fractions: original seed (obtained in the feed hopper), heavy seed (at the top), middle high, intermediate, and light seed (lower part), and the stones outlet of the gravity table. Each combination of regulation was in an independent adjustment with a total of seven adjusts. The gravity table, in the adjustments with high oscillation, was efficient in improving the physiological quality of seed lots of tobacco, by removing the fraction of light seeds discharged at the bottom of the terminal edge.

Desiccation tolerance in seeds depends on the species, development stage and drying conditions, especially the water removal rate. Coffea seeds are considered of intermediate performance, because they tolerate relative dehydration compared to orthodox seeds and are sensitive to low temperatures. The objective of this study was to verify the effect of different drying rates on the viability and storability of Coffea canephora seeds. A complete randomized experimental design was used, in a factorial 3 x 5 x 2 design, with three drying rates (fast, intermediate and slow), five final mean water contents after drying (51, 42, 33, 22 and 15 %) and two storage temperatures (10 and 20°C). The germination and seed vigor assessments, using radicle protrusion, cotyledon leaf opening, seedling emergence and emergence speed index, were performed shortly after drying and after two and four months storage. It was observed that with reduction in the water content there was reduction in the germination values and seed vigor, for all the drying rates. The greatest reductions in physiological quality occurred when the seeds were dried quickly and the best results were obtained at the intermediate drying rate. There was an effect of drying rate and storage temperature on the physiological quality of the seeds, and lower germination and vigor values were observed in seeds with lower water content stored at 20°C. C. canephora seeds were tolerant to desiccation down to 15 % water content and can be stored for four months at 10°C. A temperature of 20ºC can be used to store C. canephora seeds, as long as the water content is not reduced to values below 22 % water content.

The aim of this work was to correlate macronutrient content of soybean seeds to physiological quality of different cultivars. The work was developed in the Federal University of Pelotas, in the facilities of the Seed Science and Technology Graduate Program. The experimental design was randomized blocks in arranged in four replicates. The following soybean cultivars were used: BMX Apolo RR (12 lots), BMX Ativa RR (13 lots), BMX Energia RR (26 lots), BMX Força RR (24 lots), BMX Impacto RR (35 lots), BMX Magna RR (16 lots), BMX Turbo RR (44 lots), BMX Potência RR (82 lots) and NA 5909 RR (28 lots), with seeds produced in northwest Rio Grande do Sul. The determination of nutritional contents found in plant tissues of soybean seeds, were measured: Nitrogen (N), Phosphorus (P), Potassium (K), Magnesium (Mg), Calcium (Ca), Sulfur (S). The macronutrient contents of the seeds vary according to genetic characteristics of the cultivars, with higher oscillations of nitrogen, phosphorus, magnesium and sulfur contents present in soybean seeds. Potassium and calcium are defined as the most stable nutrients for the cultivars and seed lots analyzed. High concentrations of nitrogen, phosphorus and calcium are determinants for the physiological quality of soybean seeds.

The physiological quality is based on the genotype and may be accompanied since the first stages of selection through the study of genetic diversity. The purpose of this study was to evaluate the parental genetic contribution in the seed physiological quality in corn intervarietal crossbreeds. The intervarietal crossbreeds were performed using landraces genotypes, open pollinated varieties and commercial hybrids. The seeds obtained from the crossbreed, their equivalents and parentals were evaluated regarding their water percentage, germination, germination speed rate, accelerated aging, field emergence and electrical conductivity. The seeds obtained in the crossbreed are viable and vivid with heterosis when compared with the parentals. The accelerated aging test was more effective for selecting the strongest genotypes. The canonical variables were useful to group the genotypes that presented similar physiological characteristics. The genotypes MPA 01 x Pixurum 05, MPA 01 x AS1565, MPA 01 x SJC5886, SJC5886 x Pixurum 05 and Fundacep 35 x SCS 154 Fortuna were the best crossbreeds regarding the production of better physiological quality seeds.
