The aim of the article is to study the interaction effect between NAA and BAP concentration on protocorm growth and to know the proper concentration for growth of Dendrobium  spectabile  orchid protocorm. This research method using an experimental method which is conducted in the Laboratory of Agriculture Faculty of Winaya Mukti University, Tanjungsari Subdistrict, Sumedang District. The experiment was conducted from June 2017 until September 2017. The experiment used was a Completely Randomized Design (CRD) with the factorial pattern, consisting of two factors and repeated twice. the first factor was the effect of NAA concentration which consisted of five levels, namely without NAA, 0.5 mg kg-1, 1.0 mg kg-1, 1.5 mg kg-1, and 2.0 mg kg-1.  The second factor is the BAP concentration which consists of five levels, namel without BAP, 1.0 mg kg-1, 2.0 mg kg-1, 3.0 mg kg-1, and 4.0 mg kg-1. Eksplant is protocorm from orchid D. spectabile which is grown on MS medium (Murashig and Skoog) half recipe as base medium accompanied by each treatment for 12 weeks. The experimental results show that there is an interaction between the effect of NAA and BAP concentration on the number of leaves only. Without NAA or 1.5 mg kg-1 NAA concentration with BAP 2.0 mg kg-1 gives more leaves.  Independent of NAA or BAP concentrations did not affect the number of buds, number of roots, root length, fresh and dry weight of plantlets, and also growth ability of plantlets.  BAP concentration only affect plant height, and the highest plantlet height is found without add of BAP in medium culture

<p><em>Indonesia has a high diversity of species of orchids. One of them is the Dendrobium orchid. Orchid seeds do not have endosperm should be grown in a medium that has enough nutrients. Orchid propagation in vitro is strongly influenced by the composition of the medium used. Research through experiments aimed at studying the effect of NAA, BAP, NAA and BAP combination of the subculture plantlets growth of  </em><em>hybrids</em><em> orchid Dendrobium biggibum X liniale in Vacin Went media. Research conducted at the Laboratory of Tissue Culture Center for Plant Conservation Bogor Botanical Gardens. </em><em>The experimental design used was completely randomized design (CRD) with two factors. The first factor was the concentration of Naphthalene Acetic Acid /NAA ( 0 ppm, 1 ppm, 3 ppm and 5 ppm). The second factor  was the type media of </em><em>Benzyl Amino Purine</em><em> /BAP ( 0 ppm, 1 ppm, 3 ppm and 5 ppm). </em><em>Each treatment was replicated eight times. Analysis of the data by F test level 5% and if there is a significant difference continued with Duncan Multiple (Duncan's Multiple Range Test) level of 5%. The result showed that </em><em>the addition  </em><em>of NAA 3 ppm </em><em>as much as 4,96 cm </em><em> and BAP 3 ppm </em><em>as much as 4,41 cm </em><em> give a significant effect on the increase of high plantlets</em><em> </em><em>and  so NAA 3 ppm </em><em>as much as 5,76 cm </em><em> effect  on  roots length , but did not significantly effect the number of leaves and roots.</em></p>

<em>A planting medium that rich in nutrients is needed for oil palm, especially in the nursery phase. This study aimed to determine the effect of various doses of EM4 and to get the best dose of EM4 on the growth of oil palm seedlings in the pre-nursery. The research method used was a non-factorial completely randomized design with 4 treatments, namely 0 ml EM4 (A1), 4 ml EM4 (A2), 8 ml EM4 (A3), and 12 ml EM4 (A4). Each treatment was repeated 6 times so that there were 24 experimental units. The data obtained were analyzed for variance at the 5% level, if the effect was real, it was followed by the least significant difference (LSD) test. Based on the results of the study, it could be concluded that application of various EM4 doses had a significant effect on plant height (4-12 MST), the number of leaves (4-8 MST), trunk girth (4-12 MST), and shoot wet weight but did not significantly affect the number of leaves (12 MST), the volume of roots, length of roots, root wet weight and shoot dry weight, dry weight root, the best dose was shown at a dose of 12 ml EM4.</em>

The propagation of Cavendish (Musa acuminata L.) seedling conventionally relatively took a longer time, not uniformed in quality and possibly has a disease that is derived from unhealthy mother stock. This research aimed to find the effect of IBA and BA on difference concentrations on the growth of plantlet, also to determine the optimum concentration of IBA and BA on the growth of Cavendish bud plantlets in vitro. This research had been carried out started from April to July 2018 in Tissue Culture Laboratory, Research Institute of Horticultural Seed Development of Pasir Banteng, Sumedang, West Java. Indonesia. The method used in this research was Completely Randomized Design with two factors. The first factor was IBA (i0 = 0 mg L-1, i1= 0,75 mg L-1, dan i2= 1,5 mg L1), and the second factor was BA (b1= 1 mg L-1 b2= 3 mg L-1 b3= 5 mg L-1 b4= 7 mg L-1), replicated three times. Duncan Multiple Range Test (DMRT) was used as a further test. The result showed an interaction between IBA and BA on the parameters of shoot growth and the number of leaves at 6 WAC. IBA showed an independent effect on the number of buds. The addition of 1,5 mg L-1 IBA and 3 mg L-1 BA was optimum for the growth of Cavendish bud explants.

The purpose of this research is determining the effect of some  treatment combinations and to determine the combination of best compost bunches empty palm oil and dolomite that could support growth seeds palm oil plantations in a main nursery. This research has been conducted in the experimental field, Agriculture Faculty, University of Riau from April to July 2015. The research used Completely Randomized Design Factorial that consisting of two factors. The first factor is giving compost bunches empty palm oil : (37,5, 75 and 112,5 g/polybag).  The  second  factor  is  giving  lime  dolomite  (CaMg  (CO3)2) : (9, 18 and 30 g/polybag).   Each combination made three replications and two plants. The data were statistically analyzed using analysis of variance and the means were tested with Duncan 5%. The research results show that combination of compost bunches empty palm oil and dolomite had show the real effect on the increase in diameter hump of seedlings , but the effect not real on the parameters height of seedlings,increase number of leaves, volume roots, ratio crown rootsand and dry weight of seedlings. Combination compost bunches empty palm oil 112,5 g/polybag and dolomite 18 g/polybag showed better results for the growth of palm oil seedlings.

<p>Porang (Amarphophallus. oncophyllus) is a type of plant tubers that have the potential and prospects for development in Indonesia. In addition to readily available, plants are also capable of producing carbohydrates and high harvest index. The last few years a very large porang needs. Porang plant propagation is vegetative and generative in which to make the seeds are ready to harvest takes between 4-6 months. This study aims to determine kinds of the appropriate medium for growing plants porang network and determine the concentration  of  each  medium,  so  we  get  a  superior  porang  plants.  Research  using factorial completely randomized design. The first factor is a growth regulator BAP concentration: 0 mg / l, 0.5 mg / l, 1.0 mg / l, 1.5 mg / l and 2.0 mg / l, while the second factor is the concentration of IBA 0 mg / l, 0.5 mg / l and 1.0 mg / l, in order to obtain 15 treatments and each treatment made 15 replications. Data were collected for the pengkalusan, contamination callus, callus mortality rate, number of buds, leaf buds height, number of leaves and root. The results showed that the addition of plant growth regulators (hormones) BAP in MS medium effect on the number of shoots, buds and the high number of young leaves. The addition of hormone IBA in MS medium pengkalusan affect the process and the number of roots. The addition of hormone BAP and IBA on MS medium did not significantly affect mortality contamination callus and callus. The interaction between BAP and IBA hormone given on MS medium did not significantly affect the pengkalusan, contamination callus, callus mortality rate, number of buds, leaf buds height, number of leaves and root</p> <p> </p>

ABSTRAK<br />Temu lawak (Curcuma xanthorrhiza) merupakan salah satu jenis<br />tanaman obat potensial untuk dikembangkan. Rimpangnya berguna untuk<br />mengobati penyakit hepatitis dan memperbaiki sistem kekebalan tubuh.<br />Untuk mendukung kegiatan plasma nutfah temulawak saat ini telah<br />dilakukan upaya konservasi secara in vitro di laboratorium Kultur Jaringan<br />Balai Penelitian Tanaman Obat dan Aromatik, Bogor mulai bulan Juni<br />2005 sampai April 2006. Bahan tanaman yang digunakan adalah mata<br />tunas temulawak yang telah tersedia secara in vitro. Media dasar yang<br />digunakan adalah Murashige dan Skoog (MS) yang diperkaya vitamin dari<br />group B. Sebagai sumber energi digunakan sukrosa sebanyak 30 g/l.<br />Perlakuan yang diuji adalah beberapa taraf konsentrasi paclobutrazol<br />yaitu: Paclobutrazol 1,0 mg/l; 3,0 mg/l dan 5,0 mg/l. Sebagai kontrol digu-<br />nakan media dasar MS tanpa paclobutrazol. Rancangan yang digunakan<br />adalah acak lengkap dengan tujuh ulangan. Parameter yang diamati adalah<br />jumlah tunas, panjang tunas, jumlah daun dan akar pada umur 4 dan 7<br />bulan serta penampilan kultur secara visual. Setelah dikonservasi selama<br />tujuh bulan, maka dilakukan uji regenerasi kultur setelah perlakuan<br />paclobutrazol ke dalam media MS + BA 0,1 mg/l. Hasil penelitian<br />menunjukkan bahwa penggunaan retardan paclobutrazol mampu menekan<br />pertumbuhan kultur dan dapat mengurangi periode sub kultur yang<br />biasanya setiap dua bulan menjadi tujuh bulan. Konsentrasi paclobutrazol<br />5,0 mg/l merupakan konsentrasi terbaik karena kultur masih mampu<br />beregenerasi normal setelah konservasi. Hasil aklimatisasi plantlet di<br />rumah kaca dapat tumbuh dengan baik. Plantlet tumbuh dan berkembang<br />tanpa menunjukkan adanya penyimpangan dalam penampilan visualnya.<br />Kata kunci: Temulawak, Curcuma xanthorrhiza, paclobutrazol, konser-<br />vasi regenerasi, in vitro, pertumbuhan, Jawa Barat<br />ABSTRACT<br />Effect of paclobutrazol on temulawak growth during in<br />vitro conservation<br />Temulawak (Curcuma xanthorrhiza) is one of medicinal plant<br />which is potential to be developed. The rhizome is used to heal hepatitis<br />and to improve the imune system of human body. To support the<br />medicinal germplasm conservation, in vitro conservation of temulawak<br />was conducted in Tissue Culture Laboratory of Indonesian Research<br />Institute for Medicinal and Aromatic, Bogor from June 2005 to April<br />2006. Sterile shoots in vitro were used as plant explants. The basic<br />medium used was Murashige and Skoog (MS) medium, supplemented<br />with vitamine from B group. Sucrose as carbon sources, was given 30 g/l<br />into the medium. The treatment tested were several concentrations of<br />paclobutrazol: (1) Paclobutrazol 0.0 mg/l; 1.0 mg/l; 3.0 mg/l; dan 5.0 mg/l.<br />The treatments were arranged in a completely randomized design with<br />seven replications. The parameters observed were number of shoots, shoot<br />length, number of leaves and roots during conservation. After seven<br />months conserved, shoots were regenerated into regeneration medium.<br />The result showed that paclobutrazol at 5.0 mg/l could reduce the plant<br />growth during seven months in conservatioan period and the culture could<br />regenerate normally after transferring into multiplication medium. This<br />technique can be applied to prolong the conservation culture. This study was aimed at assessing the effects of different rates of cattle urine and urea fertilizer and on the growth and production of three lemon basil accessions. A factorial completely randomized design with two factors was used. The first factor was the composition of cattle urine and urea fertilizer consisting of six levels, namely 100% cattle urine, 75% cattle urine + 25% urea, 50% cattle urine + 50% urea, 25% cattle urine + 75% urea, 100% urea, and 0% cattle urine + 0% urea. The second factor was lemon basil accession consisting of three levels, namely Bogor, Cianjur, and Sukabumi accessions. Results showed that Sukabumi acession had the highest number of leaves, leaf area, branch length, number of branches, branch diameter, fresh weight and dry weight. Urea and urine cattle compositions did not give significant effects on parameters measured. However, compared to no fertilizer tretament, other treatments gave significant differences in parameters measured. It was concluded that cattle urine could be used to substitute urea fertilizer.

This research was aim to identify the effect of Naphthylacetic Acid (NAA) and 6-Benzylaminopurin (BAP) on growth of Lilium longiflorum planlet. The research method was Completely Randomized Design Factorial 4 x 4 with 4 replication. First factor was concentration of NAA, consist of A0: 0 mg/l, A1: 0,75 mg/l, A2: 1,5 mg/l, and A3: 2,25 mg/l. Second factor was concentration of BAP, consist of : B0: 0 mg/l, B1: 0,3 mg/l, B2: 0,6 mg/l, and B3: 0,9 mg/l. The observed parameters were number of shoots, plant height, number of leaves, number of roots, and percentage of contamination. Data were process by Analysis of Variance and continued by Least Significance Different (LSD). The result showed that combination of NAA and BAP did not significantly affect the growth parameter of Lilium longiflorum. BAP treatment significantly affected the number of shoots and number of leaves. The number of roots only observed in NAA treatment; 0,3 mg/l BAP + 0,75 mg/l NAA; 0,3 mg/l BAP + 1,5 mg/l  NAA; and 0,6 mg/l BAP + 0,75 mg/l NAA. Percentage of fungal contamination was 7,5%. The conclusion of the result were the treatment 0f 0,3 mg/l BAP + 0,75 mg/l NAA was the best treatment for growth of Lilium longiflorum planlets with no contamination.

<p>ABSTRACT</p><p>In order to enhance the multiplication of  Protocorm Like Bodies (PLBs)  of Spathoglottis  plicataorchid  in vitro, several combinations of vitamin, sucrose concentration, and cytokinin were tested.   Thisexperiment was arranged in Completely Randomized Design (CRD) with two factors. The first factor was combination  of  vitamin  composition  and  sugar  concentrations  in  the medium  which  consisted  of  four different  combinations  (J1 = vitamin  MS + sucrose 30 g  L-1, J2 = vitamin B5 + sucrose 30 g  L-1, J3= vitamin  MS  + sucrose  40  g  L-1,  J4  =  vitamin  B5  +  sucrose  40  g  L-1).  The  second factor  was  seven combinations  of  cytokinin types and its  concentrations (S0 = without cytokinin (control), S1 =    20  μM BA, S2 = 40  μM BA, S3 = 20 μM kinetin,   S4 = 40  μM  kinetin, S5 = 75  ml L-1 coconut milk, and S6 = 150 ml L-1 coconut milk).  PLBs  were used as  explant  and were  grown on MS  solid medium containing various vitamin, sucrose, and cytokinin combinations  as mentioned above.  Growth and multiplication of PLBs were based on  the number of PLBs per explant, number of plantle ts per explant, number of roots formed, plantlet height and visual appearance and performa nce of the observations at 6  MST.  The best growth  and  multiplication  of  PLBs  orchid S.  plicata  was  produced  on  MS  medium  modified  with  B5 vitamins  and 30 g  L-1 sucrose, followed by those and  on MS medium enriched  with  75 ml  L-1 coconut milk and on MS medium supported by 20 μM BA.</p><p>Key words : B5, benzyl adenine, coconut water, in vitro , orchid, sugar concentration,  vitamin composition</p><p>ABSTRAK</p><p>Percobaan ini bertujuan untuk mendapatkan formulasi kom posisi vitamin dan konsentrasi gula medium,serta jenis dan  konsentrasi sitokinin terbaik dalam menginduksi pertumbuhan dan  multiplikasi  lini klon PLBsanggrek Spathoglottis plicata dalam jumlah yang besar  secara  in vitro. Percobaan ini menggunakan Rancangan Acak Lengkap (RAL) Faktorial.  Eksplan yang digunakan adalah PLBs  yang ditanam pada  medium Murashige dan  Skoog  (MS)  yang  telah   dilakukan  modifikasi  sesuai  dengan  perlakuan.  Faktor  pertama  adalah  formulasi komposisi vitamin dan konsentrasi gula yang terdiri dari empat macam formulasi yaitu J1 = vitamin  MS + gula 30 g L-1, J2 = vitamin B5 + gula 30 g  L-1, J3= vitamin MS + gula 40 g L-1, J4 = vitamin B5 + gula 40 g  L-1. Faktor kedua adalah penambahan sitokinin (3 jenis dengan 2 taraf konsentrasi) yang terdiri dari 7 kombinasi perlakuan yaitu S0 = tanpa sitokinin (kontrol), S1 = BA 20 μM, S2 = BA 40 μM, S3 = kinetin 20 μM,  S4 = kinetin 40 μM, S5 = air kelapa 75 ml L- 1, dan S6 = air kelapa 150 ml  L-1. Pertumbuhan dan multiplikasi PLBs anggrek S. plicataterbaik  dihasilkan   pada medium  MS  dengan  modifikasi  vitamin  B5  dan  konsentrasi  gula  30  g  L-1dan  pada medium MS dengan penambahan air kelapa 75 ml L- 1 serta pada medium MS dengan penambahan BA 20  μMdengan  kriteria  jumlah  PLBs akhir dan  jumlah  planlet  akhir  tertinggi,  jumlah  akar  dan  tinggi  tanaman serta penampilan visual hasil pengamatan pada 6 minggu setelah tanam.</p><p>Kata kunci : air kelapa, anggrek, B5, benzyl adenin, in vitro, komposisi vitamin, konsentrasi gula</p>

The purpose of this research is to determine the concentration of young coconut water that is appropriate for the growth of orchid plantlets (Dendrobium sp.) In vitro. This study used an experimental method, descriptive data analysis to compare several different concentrations of coconut water. The design of this study uses a completely randomized design (CRD). The treatments consist of 0% coconut water concentration (as a control), 15%, 30% and 60%. Each concentration was carried out 5 replications and each repetition consisted of 5 Dendrobium sp plantlets in each culture bottle conducted for 40 HST, for observing the root length carried out for 50 HST. The highest number of shoots and leaves were produced at the same concentration, namely 150 ml / L coconut water treatment (15% concentration) with an average of 2.8 shoots and the average number of leaves 10.8 leaves. The average number of roots and the longest root length was produced at a concentration of 600 ml / L coconut water (60% concentration) with an average of 6 roots, and the longest root length was 0.5 cm.Keywords: Young coconut water, (Cocos nucifera L.), Dendrobium sp., in vitro, growth.ABSTRAKTujuan penelitian ini ialah menentukan konsentrasi air kelapa muda yang tepat untuk pertumbuhan planlet anggrek (Dendrobium sp.) secara in vitro. Penelitian ini menggunakan metode eksperimen, analisis data secara deskriptif untuk membandingan beberapa konsentrasi air kelapa yang berbeda. Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Perlakukan terdiri dari konsentrasi air kelapa 0 % (sebagai kontrol), 15% , 30% dan 60%. Masing-masing konsentrasi dilakukan 5 kali ulangan dan setiap ulangan terdiri dari 5 planlet Dendrobium sp dalam setiap botol kultur yang dilakukan selama 40 HST, untuk pengamatan panjang akar dilakukan selama 50 HST. Jumlah tunas dan jumlah daun terbanyak dihasilkan pada konsentrasi yang sama, yaitu perlakuan air kelapa 150 ml/L (konsentrasi 15%)  dengan rata-rata jumlah tunas terbanyak 2,8 tunas dan rata-rata jumlah daun terbanyak 10,8 helai daun. Rata-rata jumlah akar terbanyak dan panjang akar terpanjang dihasilkan pada konsentrasi air kelapa 600 ml/L (Konsentrasi 60%) dengan rata-rata jumlah akar terbanyak sebanyak 6 akar, dan rata-rata panjang akar terpanjang 0,5 cm.Kata kunci : Air kelapa Muda (Cocos nucifera L.), Dendrobium sp., in vitro, pertumbuhan
