The effect of combining nanosilver and new injection form of rexod on the periodontal tissues state on experimental periodontitis in rats

A representative and randomized study of the effect of a combination of nanosilver and a new injection form of Rexod® on the periodontal tissues state of animals (rats) in an experimental model of inflammatory pathology of periodontal tissues (experimental periodontitis) was carried out. It was shown that the improvement of the dental status of periodontal tissues against the background of the course application of a combination of nanosilver (topically) and a new injection form of Rexod® was accompanied by a decrease in the amount of gingival fluid of the sulcular gingival sulcus, an improvement in the clinical condition of the marginal periodontal mucosa (hyperemia was leveled, swelling decreased, the bleeding indexPBI Muhllemann-Cowell decreased, test Schiller-Pisarev results normalised). The decrease in the number of gum fluids is due to the decrease in antibacterial activity of pathogenic microflora and their products of their life-based hemodynamics of the peripheral microvascular network (exchange capabulars) of the periodontal, a decrease in perfusion pressure in them, reduced (narrowing) of their lumen, reduce their permeability and restoration of oxidation-reduction processes.

Abstract Background Granulocyte colony-stimulating factor (G-CSF) is an important immune factor that mediates bone metabolism by regulating the functions of osteoclasts and osteoblasts. Bone loss is a serious and progressive result of periodontitis. However, the mechanisms underlying the effects of G-CSF on periodontal inflammation have yet not been completely elucidated. Here, we examined whether an anti-G-CSF antibody could inhibit bone resorption in a model of experimental periodontitis and investigated the local expression of G-CSF in periodontal tissues. Methods Experimental periodontitis was induced in mice using ligatures. The levels of G-CSF in serum and bone marrow were measured; immunofluorescence was then performed to analyze the localization and expression of G-CSF in periodontal tissues. Mice with periodontitis were administered anti-G-CSF antibody by tail vein injection to assess the inhibition of bone resorption. Three-dimensional reconstruction was performed to measure bone destruction‐related parameters via micro-computed tomography analysis. Immunofluorescence staining was used to investigate the presence of osteocalcin-positive osteoblasts; tartrate-resistant acid phosphatase (TRAP) staining was used to observe osteoclast activity in alveolar bone. Results The level of G-CSF in serum was significantly elevated in mice with periodontitis. Immunofluorescence analyses showed that G-CSF was mostly expressed in the cell membrane of gingival epithelial cells; this expression was enhanced in the periodontitis group. Additionally, systemic administration of anti-G-CSF antibody significantly inhibited alveolar bone resorption, as evidenced by improvements in bone volume/total volume, bone surface area/bone volume, trabecular thickness, trabecular spacing, and trabecular pattern factor values. Immunofluorescence analysis revealed an enhanced number of osteocalcin-positive osteoblasts, while TRAP staining revealed reduction of osteoclast activity. Conclusions G-CSF expression levels were significantly up-regulated in the serum and gingival epithelial cells. Together, anti-G-CSF antibody administration could alleviates alveolar bone resorption, suggesting that G-CSF may be one of the essential immune factors that mediate the bone loss in periodontitis.

With diabetes, there is a violation of the microvasculature, a violation of the immune status, an increase in destructive processes in the bone tissue, which leads to the development and / or intensification of pathological processes. In the oral cavity, the most frequently diagnosed inflammatory periodontal diseases, chronic injuries of the oral mucosa (COP), recurrent aphthous stomatitis, COP candidiasis. The aim of our study was to study the dental status of patients with type 2 diabetes mellitus. Materials and Methods: A descriptive evaluative epidemiologically controlled, non-randomized study was conducted to determine the effect of type 2 diabetes on dental status. The main group (“case”) consisted of 68 patients with type 2 diabetes in the compensation phase (IA) and 56 patients in the subcompensation phase (IB). The comparison group included 60 patients without somatic pathology. All patients underwent a comprehensive dental examination, bacteriological and microscopic examination to identify fungi of the genus Candida. Results: in patients with subcompensated form of type 2 diabetes, there was an increase in the frequency of complaints of plaque on the tongue, burning sensation in the COP, dryness and changes in taste. Periodontal indices indicate that the patients of the main group have a high prevalence of inflammatory and destructive processes in the periodontal tissues (indices of PMA, KPI, Mullemann indices) with poor oral hygiene (OHI-S = 3.5 ± 0.4), which confirms the Friedman criterion (χ2 = 116.27; p = 0.0000) based on the calculation of Kendall's concordance coefficient (0.743). The diagnosis of oral mucosa candidiasis was confirmed in 74.6% of patients in the main group by bacteriological examination.

The article presents the results of a comparison of the microbial composition of the gingival fluid of dogs of different age groups. The practical significance of the work lies in the fact that the data obtained during the study allow us to predict the risk factors for damage to periodontal tissues, as well as to determine the choice of antiseptic and antibiotic, the duration of the course of their use.

Introduction. Activation of lipid peroxidation is one of the trigger mechanisms of periodontium injury, which is primary caused by cellular damage. Reactive oxygen and nitrogen species (RONS) are able to cause damage to a cell as well as final products of lipid peroxidation, including unsaturated aldehydes and other metabolites. Objective. The aim of the research was to determine the role of RONS and accumulation of lipid peroxidation derivatives in initial development and formation of chronical inflammatory process in periodontium. Methods. Experimental periodontitis was modeled in animals by injection of complex mixtures of microorganisms diluted in egg protein into periodontal tissues. The results of biochemical studies of free radical processes activity in blood serum were evaluated by content of diene, triene conjugates, TBA-active products and total quantity of metabolites of nitric oxide (NO2–+NO3–), which were determined on the 7th, 14th and 30th days of the experiment. Results. Generation of active forms of oxygen is more influential, providing longevity of inflammatory process. This pays attention to typical dynamics of changes in active processes of lipid peroxidation in the development and course of experimental periodontitis. The study of inflammatory process with a bacterial-immune component in the rats’ periodontal complex proved accumulation of lipid peroxidation and nitric oxide metabolites in blood serum.Conclusions. The preservation of increased lipid peroxidation and nitric oxide metabolites in blood serum of the experimental animals with acute periodontitis conduce enhance of alteration and delayed healing that result in its sequel into chronical periodontitis.

The peculiarities of metabolic and oxidation reduction processes as well as energoplastic metabolism during physiologic course of gestation in unfavourable cologie conditions are studied. It is established that in ecologically unfavourable region during physiologic pregnancy general degydrogenase activity of blood plasma increases, cell membranes are packed, general protein level increases. The decrease of general degydrogenase activity of blood plasma with the increase of pregnancy length, rarefaction of membranes, the reduction of relation coefficient of malate dehydrogenase and lactate degydrogenase activity, the increase of combined quantity relationship of reduced and oxidated equivalents can be considered to be biomarkers of physiologic pregnancy transfer to pathologic.
