The present study aimed to characterize Pleurotus eryngii and P. pulmonarius ligninolytic enzymes and to determine their potential for polymer degradation in common agroforestry residues. The peak of laccase activity (36052.33 U L-1) was observed after P. pulmonarius cultivation on oak sawdust. The maximal Mn-dependent peroxidase activity was reached by P. eryngii (2511.36 U L-1), while the highest level of versatile peroxidase activity was noted in P. pulmonarius (3053.03 U L-1), after fermentation of corn stalks. The highest level of lignin loss (46.28%) was achieved after cultivation of P. pulmonarius on corn stalks, but the most selective degradation of lignocellulose polymers was observed after P. eryngii cultivation on wheat straw. The obtained results lead to the conclusion that the studied P. eryngii and P. pulmonarius strains are good producers of ligninolytic enzymes and effective and selective depolymerizers of agroforestry residues, and therefore their use would be beneficial in numerous environmentally friendly technologies.

To get a better insight into the ligninolytic system of Lenzites betulinus, the effect of wheat straw and oak sawdust, as carbon sources, on production of Mn-oxidizing peroxidases and laccase, under solid-state and submerged fermentation, was studied. Obtained results revealed considerable differences related to the both factors affecting enzyme activities. Wheat straw was more favorable carbon source for Mn-oxidizing peroxidases and oak sawdust for laccase activity. Solid-state fermentation of wheat straw was optimal for Mn-dependent peroxidase activity (72.1 Ul-1). In contrary to this, submerged fermentation of the same residue gave the highest level of versatile peroxidase activity (25.4 Ul-1). The peak of laccase activity was noted during solid-state fermentation of oak sawdust (32.3 Ul-1), while this enzyme was not detected under submerged fermentation of any plant residues.

AbstractEnzyme production by immobilized Phanerochaete chrysosporium was evaluated in airlift bioreactor and agitated cultures. Free mycelium and immobilized mycelium on alginate beads were tested in the decolourization of 50 and 500 mg/L of Remazol Brilliant Blue R. Dye concentration did not inhibit the fungi development in all tests. In addition, high decolourization percentage of dye was found with free mycelium (99%) in agitated flasks and with immobilized mycelium in airlift (98%). However, decolourization period by immobilized mycelium (120 h) was greater than that by the free mycelium (14 h). Important manganese peroxidase, lignine peroxidase and laccase activities were identified in decolourization process. Manganese peroxidase appeared to be promoted by high dye concentrations during the treatment with immobilized mycelium, but this enzyme was not detected with free mycelium in airlift. Bioreactor prompted also laccase and lignine peroxidase actions in both tests; free mycelium registered a maximum laccase action of 31.569 × 103 U/L in 70 h, whereas immobilized mycelium registered 1.680 × 103 U/L in 170 h, while lignine peroxidase secretion by free P. chrysosporium was higher (1.300 × 103 U/L) than immobilized mycelium (1.250 × 103 U/L). Maximum laccase activity coincided with the maximum percentage of decolourization, however, high peroxidase activity was identified from the start of dye treatment.

Abstract The effect of lignocellulose and lignin on growth of mycelium of mushroom Lentinula edodes and laccase activity in cultivation medium was studied. It was shown that cultivation of L. edodes mycelium in liquid nutrient medium with addition of 0.25-0.5% of kraft lignin increased mycelium biomass yield approximately two times compared with reference conditions without addition of lignin. Similar results were obtained in experiments in which 0.5% lignocellulose that remained after obtaining furfural, and 0.5% lignin that remained after obtaining furfural and glucose from wheat straw, were added to the nutrient medium. This effect was greater in the conditions of cultivation with good aeration, compared with static culture. Laccase activity in medium increased after addition of wheat straw lignocellulose or lignin only in the case of mycelium cultivation with aeration. In the case of mushroom cultivation on solid nutrient medium, addition of wheat straw lignocellulose and lignin promoted growth of mycelium only during the first 7 days of cultivation.

In order to meet the requirement of sustainable development, building materials are increasingly environmentally friendly. They can be partially or fully bio-based or recycled. This paper looks at the development of fully bio-based composites where agro-resources are valued as bio-based aggregates (hemp) and as binding materials (wheat). In a previous work, a feasibility study simultaneously investigated the processing and ratio of wheat straw required to ensure a gluing effect. In this paper, three kinds of hemp-straw composites are selected and compared with a hemp-polysaccharides composite. The gluing effect is analyzed chemically and via SEM. The developed composites were characterized multi-physically. They showed sufficiently high mechanical properties to be used as insulating materials. Furthermore, they showed good thermal performances with a low thermal conductivity (67.9–69.0 mW/(m · K) at 23 ° C, dry).

ABSTRACT A versatile peroxidase able to oxidize Mn2+ as well as phenolic and nonphenolic aromatic compounds is produced in peptone-containing liquid cultures of Pleurotus eryngiiencoded by the gene mnpl. The regulation of its transcript levels was investigated by Northern blotting of total RNA. High-peroxidase transcripts and activity were found in cultures grown in glucose-peptone medium, whereas only basal levels were detected in glucose-ammonium medium. The addition of more than 25 μM Mn2+ to the former medium did not result in detectable peroxidase transcripts or activity. Potential regulators were also added to isolated mycelium. In this way, it was shown that high transcript levels (in peroxidase-expressing mycelium) were maintained on peptone, whereas expression was not induced in short-term incubation experiments. Similar results were obtained with Mn2+ ions. Strong induction of mnpl expression was caused by exogenous H2O2 or by continuous H2O2 generation during redox cycling of menadione. By the use of the latter system in the presence of Fe3+, which catalyzes the reduction of H2O2 to hydroxyl radical, it was shown for the first time that the presence of this strong oxidant causes a rapid increase of the transcripts of a ligninolytic peroxidase. In conclusion, peptone and Mn2+ affect the levels of transcripts of this versatile peroxidase in culture, and reduced oxygen species induce short-term expression in isolated mycelium, probably via a stress response mechanism.

Five agro-substrates including date palm fibers (fibrillum), wheat straw, white sawdust and their combinations were investigated to grow Pleurotus eryngii. The longer mycelium complete time within bags was 20 days on sawdust (S4), in contrast, the shorter time for mycelium overgrew was completed after 15 days on date palm fiber (S5). In significant (p<0.05), S5 showed the higher growth intensity level (vigorous growth) than other substrates. Thus use of date palm wastes (S5 medium) may be useful for successfully cultivation king oyster mushroom in farm.International Journal of Environment Vol.5(3) 2016, pp.1-10
